Abstract In this paper,the sequences of second exon of DQA1 and DQB1 were amplified byPCR primers,and hybridized with 17 or 23 sequence specific oligonucleotide probes labeled byisotope respectively.The distribution of 10 DQA1 alleles and 17 DQB1 alleles was observed inHunan Hans population.The results showed that the most of DQAI and DQB1 alleles were ex-pressed in Hans,however,their frequencies are different from Caucasoids.
2.Expression of some oncogenes and point mutation of c-Ha-ras1 during hepatocarcinogenesis in rats

Objective To study the expressions of oncogenes c-Ha-ras, c-ki-ras, pan-ras and c-myc and point mutation of c-Ha-ras1 during hepatocarcinogenesis in rats. Methods Immunohistochemistry, in situ hybridization and microdissection of tissue (MDT)-PCR-SSCP were used to detect the oncogene expressions and point mutation of c-Ha-ras1 in both Solt-Farber model and DEN-induced liver cancer model. Results The overexpression of c-Ha-ras was closely associated with the formation and proliferation of the precancerous basophilic hepatocyte foci, while that of c-myc with the growth of the oval cell foci. The abnormalities of IGF-Ⅱ played an important role in the evolution of precancerous foci/nodules towards hepatocellular carcinoma (HCC). The overexpression of fms was only associated with HCC of some rats. Conclusion Hepatocarcinogenesis in rats was related with the overexpression of c-Ha-ras, c-myc, IGF-Ⅱand fms and the point mutation of c-Ha-ras1, and overexpression of these oncogenes was associated with morphological evolution.
3.Quantitative study on morphologic features and proliferative activity during DEN-induced hepatocarcinogenesis in rats

Objective To explore the relationship between morphologic evolution and proliferative activity during hepatocarcinogenesis in rats. Methods Imaging analysis technique was used to detect the morphologic parameters of cells in hepatic lesions in both Solt-Farber model and diethylnitrosamine (DEN)-induced liver cancer model. Immunohistochemistry was employed to detect proliferating cell nuclear antigen (PCNA) and bromodeoxyuridine (BrdU). Results The oval cells were identified as irregular small proliferating cells in size of one-eighth of and with a nucleus/cytoplasm ratio of 6 times of the normal hepatocyte by image analysis. The morphometric parameters of basophil hepatocyte in precancerous foci and nodule were similar to those of the liver cancer cell. PCNA and BrdU positive cells were mainly localized within the proliferative foci and hepatocellular carcinoma (HCC) tissues. There was a better consistency between the development of hepatic lesions and cellular proliferative activity. Conclusion The morphologic evolution is closely related to proliferative activity during hepatocarcinogenesis in rats.
4.Implantation of arc-track private lock pedicle orthopedic fixation system Ⅱ for treatment of spinal diseases in 86 cases

OBJECTIVE: To investigate the effects of arc-track private lock pedicle orthopedic fixation system Ⅱ (ALPF Ⅱ) on treatment of spinal diseases. METHODS: A total of 86 patients with spinal diseases were treated using self-made ALPF in the Wendeng Orthopaedic Hospital and were included in this study. Of these patients, 14 suffered from cervical spinal stenosis complicated by cervical vertebral destabilization, 29 from thoracolumbar fractures and dislocations, 15 from lumbar spinal stenosis complicated by lumbar vertebral destabilization, 2 from lumbar spondylolisthesis, 8 from spinal tuberculosis, 6 from ankylosing spondylitis, 9 from idiopathic scoliosis, and 3 from congenital scoliosis. According to conditions, different therapeutic regimens were selected. Postoperatively, regular follow-up was performed to observe vertebral healing, intervertebral height, spinal column sequence, and neurological function recovery. RESULTS: All patients were followed up for 9-30 months (average 12 months). The improvement rate of neurological function, spinal mobility, back pain, and melosalgia was 94.1%, 65.9%, 92.1%, and 87.4%, respectively. The postoperative anterior and posterior vertebral heights were apparently recovered, and kyphotic angle was well corrected. No screw, rod loosening or breakage was found. CONCLUSION: Self-made ALPF Ⅱ is an internal fixation method for treatment of spinal diseases. It provides good reduction, reliable curative effects, less complications, and no biocompatibilities.
5.Effects of nordihydroguaiaretic acid on genomic methylation pattern of malignant glioma cells and its significance

Objective To investigate the genomic methylation pattern of a malignant glioma cell line in the process of differentiation induced by nordihydroguaiaretic acid (NDGA). Methods Methylation-sensitive arbitrarily primed PCR was used to study the genomic methylation changes. Results Fragments of genomic DNA of PCR products in control groups digested with MspⅠ were smaller than those with HpaⅡ. No large fragment could be identified and at least three fragments of different sizes were demonstrated in the control group. In NDGA treatment group, comparing MspⅠ digestion with HpaⅡ digestion, the amount of PCR products was smaller with more DNA bands. The amount of PCR products in NDGA treatment group was increased with more DNA bands compared with that in the control groups. Conclusion The genomic methylation level in SHG-44 cells was increased by NDGA in the differentiation process of SHG-44 cells. It suggests that genomic methylation pattern may be one of the targets for glioma cell differentiation induced by NDGA.
6.Effects of NDGA on the growth of a human malignant glioma cell line CHG-5

Objective To investigate the effects of nordihydroguaiaretic acid (NDGA) on the growth of a human malignant glioma cell line CHG 5 in vitro and in vivo . Methods Colorimetric MTT assay, flow cytometry, and light microscopy were used to investigate the proliferation in vitro , cell cycles, apoptosis of CHG 5 cells, and the growth of xenografted tumor in nude mice. Results NDGA significantly inhibited the proliferation of CHG 5 cells in vitro . Cells in G 0/G 1 phase increased, but cells in S, G 2/M phases decreased, and apoptotic cells increased significantly. After treatment of NDGA (50 mg/kg, intraperitoneally) at 5 d after the inoculation of tumor cells, the xenografted tumor volume reduced remarkably without causing significant toxic and side effects. Conclusion The inhibitory effect of NDGA on the growth of CHG 5 cells may be correlated with the regulation of cell cycles and induction of apoptosis.
7.Activation of SDF-1/CXCR4 axis induces proliferation,migration,and tubulogenesis of human endothelial progenitor cells

Objective To explore the effect of chemokine SDF-1 on the proliferation,migration and in vitro of human endothelial progenitor cells(EPCs).Methods The expression of CXCR4 and SDF-1 on EPCs was detected with immunocytochemistry.Proliferation,migration and in vitro tubulogenesis of EPCs was detected by MTT,Millicell chemotaxis and three-diamensional in vitro Matrigel assays,respectively.Results SDF-1 and CXCR4 were expressed on EPCs.SDF-1 induced proliferation,migration and tubulogenesis of EPCs.However,AMD3100 abolished the effects induced by SDF-1.Conclusion The SDF-1/CXCR4 axis possibly plays a critical role in regulating vasculogenesis of EPCs.

9.Construction of tissue microarray with astrocytic tumor microvessels of human brain

Objective To construct tissue microarray with astrocytic tumor microvessels(AstMV) of human brain and investigate astrocytic tumor angiogenesis.Methods Thirty-six specimens containing different microvessels from 200 specimens were selected as donor blocks,which was followed by punching,sampling and seeding samples with tissue microarray apparatus.The tissue microarrays were used to detect the expression of CD34,FⅧ-RAg and CD105.Results Tissue microarray of AstMV was successfully constructed and an instrument for location was made.CD34,FⅧ-RAg and CD105 were expressed on endothelial cell membrane or(and) plasma.There were difference in microvessel number between astrocytic tumors and between endothelial markers.In the gradeⅡ,Ⅲ and Ⅳ astrocytomas,microvessels labeled by CD34 and FⅧ-RAg antibodies were more than those detected by CD105 antibody.There were number difference in microvessels labeled by CD34 and FⅧ-RAg antibodies between grade Ⅰ and gradeⅡ,Ⅲ and Ⅳ tumors.Conclusion Construction of tissue microarray containing different tumor microvessels might be of significance in evaluating tumor angiogenesis.This study may be the basis for constructing digital tumor microvessel models and exploring the mathematic relation between astrocytic tumor microvessels and vascular growth factors.
10.Clinical efficacy of Traditional Chinese and Western medicine in treatment of 60 cases with angina pectoris

Objective To explore the clinical effect of traditional Chinese medicine and western medicine in treatment of patients with angina pectoris. Methods 120 cases with angina pectoris admitted in the Fourth Hospital of Baotou City from May 2012 to August 2013 were randomly divided into treatment group (n=60)and control group (n=60). All patients were given secondary prevention of coronary heart disease,and besides this, treatment group were added nitrates medications and traditional Chinese medicine treatment,while control group were only given nitrates medications.The treatment course was 14 days. The efficacy of angina pectoris,nitroglycerin loss rate and electrocardiogram efficacy were compared and the safety were evaluated between two groups. Results The efficacy of angina pectoris in treatment group was 90.00%,while control group was 71.67%,the difference was statistically significant(P<0.05 ). ECG efficiency in treatment group was 68.33%,while control group was 48.33%,the difference was statistically significant(P<0.05 ). The reduction of nitroglycerin was 86.67%,while control group was 61.67%,the difference was statistically significant(P<0.05). In addition,there were no adverse reaction in two groups. Conclusion Compared with nitrates medications,combined Chinese and Western medicine in treatment of patients with angina pectoris on basis of secondary prevention of coronary heart disease has better therapeutically effective rate.
