1.Improvement of insertion of gastric lavage canal after tracheal intubation in patients with severe organophosphorns pesticide intoxication

Objective To improve the success rate of insertion of gastric lavage canal in patientswith tracheal intubation. Methods 70 patients with organophosphoms pesticide intoxication who neededgastric lavage after tracheal intubation were randomized into the test group and the control group, 35 easesin each. In test group, lavage canal was inserted through bite block orally and the bedside was raised till 70to 80 in angle. While in the control group, lavage canal was inserted directly through mouth with the posi-tion of hypsokinesis. The success rate of intubation was compared between the two groups. Results One-time success rate was 94.2% in the test group,which was statistically different from that of the control group(37.1%), P < 0.01. The complication of intubation in the test group was also lower than that of the controlgroup,which had statistical difference. Conclusions Through improvement of position to raise the bedsidetill 70 to 80 in angle, the insertion of gastric lavage canal through bite block orally was much better than thenormal one and it is worth applying.
2.Epidemiological analysis of measles in China between 2005 and 2013

Objective To analyze the epidemiologic characteristics and epidemic intensity of measles in China between 2005 and 2013 , and to provide evidence for reasonable allocation of health resources and measles elimination in China . Methods Descriptive epidemiological methods were applied to measles surveillance data and demographic information during this study.Results A total of 597 594 cases, including 366 deaths, were reported from all over China , among whom 355 409 were males (59.47%) and 242 185 were females (40.53%).The average annual incidence was 5.07/100 000 and the average annual mortality was 0.0031/100 000.The majority of cases were children under 10 years old, accouning for 64.64%.Conclusion The incidence of measles in China is decreasing .Measles mainly occurrs in spring and there is significant difference between age and gender groups , and obvious regional differences are also found .
3.Construction and expression analysis of the zebrafish heart-specific transgenetic vector based on Tol2 transposable element.

In an effort to generate a desired expression construct for making heart-specific expression transgenic zebrafish, a Tol2 plasmid, which can drive EGFP reporter gene specifically expressed in the heart, was modified using subcloning technology. An IRES fragment bearing multiple cloning site (MCS) was amplified directly from pIRES2-EGFP plasmid and was inserted between the CMLC2 promoter and EGFP fragment of the pDestTol2CG vector. This recombinant expression plasmid pTol2-CMLC2-IRES-EGFP can drive any interested gene specifically expressed in the zebrafish heart along with EGFP reporter gene. To test the effectiveness of this new expression plasmid, we constructed pTol2-CMLC2-RED-IRES-EGFP plasmid by inserting another reporter gene DsRed-Monome into MCS downstream of the CMLC2 promoter and injected this transgenic recombinant plasmid into one-cell stage embryos of zebrafish. Under fluorescence microscope, both the red fluorescence and the green fluorescence produced by pTol2-CMLC2-RED-IRES-EGFP were detected specifically in the heart tissue in the same expression pattern. This novel expression construct pTol2-CMLC2-IRES-EGFP will become an important tool for our research on identifying heart development candidate genes' function using zebrafish as a model.

4.Optimization of a fluorescent qPCR detection for RNA of SARS-CoV-2.

We optimized a fluorescent quantitative polymerase chain reaction (qPCR) assay system for rapid and real time detection of SARS-CoV-2 RNA. The results show that the lowest dilution of RNA samples used for the detection of SARS-CoV-2 RNA could reach 1/10 000 (the initial value is set as 10 ng/μL). Moreover, the cycle threshold (Ct) for samples of clinically diagnosed COVID-19 was lower than 35 or 40. The sensitivity of this method was satisfactory. The results were consistent with those of the COVID-19 detection kit on the market under the same conditions, but the number of cycles required was shortened by about 2. Therefore, the optimized assay developed in this study can be used in screening and early clinical diagnosis. Our work provides a tool to facilitate rapid clinical diagnosis of COVID-19.
