1.Influence of Mycoplasma pneumoniae infection on the expression of interleukin-18 in BALB/c mice

Objective To investigate the expression of interluekin(IL)-18 in BALB/c mice serum and bronchoalveolar fuid infec-ted by mycoplasma pneumonia(MP) and to clarify the relationship between IL-18 and MP infection .Methods A total of 72 BABL/c mice were randomly divided into control group ,MP infection group and treatment group ,each group with 24 mice .MP infection models were established with ripping cultured MP into nasal cavity of mice .Clarithromycin was used at the fifth day in the treat-ment group .The blood of eyeball and bronchoalveolar lavage fluid were collected at the first ,fifth ,ninth day in the three groups ,and detected IL-18 .At the third ,seventh ,forth ,twenty-first ,30th day ,one mice was selected to isolate lung tissue ,and make pathologi-cal section to observe lesion degree .Results Pathological section indicated that lung tissue of mice in the control group had no in-flammatory response ,but lung tissue in the M P infection group had serious inflammatory response ,lung tissue in the treatment group at seventh day had mild inflammatory response .Compared with that of the control group ,the IL-18 in the serum and bron-choalveolar lavage fluid significantly increased at the first ,fifth ,ninth day ,the differences were statistically significant (tfirst day =10 .08 ,tfifth day =9 .14 ,tninth day = 10 .59 ,P< 0 .05) .The IL-18 levels in the serum and bronchoalveolar lavage fluid at fifth day and ninth day in the treatment group were significantly lower than those of the MP infection group(tfifth day =12 .96 ,tninth day =17 .95 ,P<0 .05) .Conclusion MP infection causes IL-18 level increasing ,it is effective to treat with clarithromycin ,and IL-18 might correlate with pneumonia severity .
2.Expression of serum anti-IMP1 autoantibody IgG in patients with hepatocellular carcinoma and itsrelationship with clinicopathological parameters

Objective To investigate serum anti-insulin-like growth factor-Ⅱ mRNA binding protein1(IMP1) autoantibody IgG expression in hepatocellular carcinoma(HCC) patient and its relationship with clinicopathological parameters.Methods ELISA method was used to detect serum anti-IMP1 autoantibody IgG in 120 cases of HCC,then the detection results were compared those in 115 healthy individuals(control group).The relationship between serum anti-IMP1 autoantibody IgG level with clinicopathological parameters in HCC patients was analyzed.Results The serum anti-IMP1 autoantibody IgG levels in the HCC group and control group were(1.512±0.685),(1.080±0.301)μg/mL respectively,the HCC group was significantly higher than the control group with statistical difference(P<0.001);serum anti-IMP1 autoantibody IgG in HCC patients was related with the tumor size,tumor differentiation degree,TNM grade and intrahepatic metastasis(P<0.001),and had no obvious relation with the age,sex,drinking and HBV infection(P>0.05).Conclusion Serum anti-IMP1 autoantibody IgG expression increased is increased in HCC patient.The detection of serum anti-IgG autoantibody IMP1 level in HCC is conducive to judge the malignancy degree of HCC.

OBJECTIVE To analyze 61 cases occupational exposure risk factors,the preventive measures and reduce occupational exposure risk.METHODS The record of 61 cases of occupational exposure such as job seniority,occupation,exposed location,place,method,damage degree,processing,preventive medicine and follow-up results register will be statisticully analysed according to the registered items from Jan 2007 to Dec 2008.RESULTS The factors of occupational exposuve were needle treatment,violating operution standard and deficient of protective measures.Both ward and hand were the main place of occupational exposure.CONCLUSIONS Strengthening of effective preventive measues is key to minimize the risk of occupational exposure and reduce the incidence of infection.

5.Expression and Significance of Anti EZH2 Autoantibodies IgG in Hepatocellular Carcinoma Patient's Serum Samples

Objective To detect the anti EZH2 autoantibodies IgG expression in hepatocellular carcinoma patient's serum samples and investigate the relationship of anti EZH2 autoantibodies IgG expression with clinicopathological parameters.Methods ELISA method was used to detect the anti EZH2 autoantibodies in serum of 120 cases of HCC patients and compared with 115 healthy controls.Results Serum anti EZH2 antibodies in patients with hepatocellular carcinoma group the content of IgG was 2.015±0.568 μg/ml group,anti EZH2 antibodies IgG content control health was 1.185±0.289 μg/ml.There was significant difference between two groups (t=12.48,P<0.001).Anti EZH2 antibodies of IgG in serum of pa ticnts with hepatocellular carcinoma was up-regulated and the degree of tumor size,tumor the degree of differentiation,TNM grading and intrahepatic metastasis (t=8.485 ~ 57.93,P<0.001).And age,sex,drinking,whether HBV infection had no significant difference (t=1.22~5.20,P>0.05).Conclusion The expression of anti EZH2 autoantibodies in serum was related to the occurrence and development of hepatocellular carcinoma,and the detection of serum anti EZH2 autoantibodies in serum of liver cancer may be helpful to judge the malignant behavior and prognosis of hepatocellular carcinoma (HCC).which may be used as a refcrcnce for the evaluation of the prognosis of HCC patients with IgG antibody against IgG.

OBJECTIVEThe aim of this study is to analyze the three-dimensional crystal structure of SMU.2055 protein, a putative acetyltransferase from the major caries pathogen Streptococcus mutans (S. mutans). The design and selection of the structure-based small molecule inhibitors are also studied.
METHODSThe three-dimensional crystal structure of SMU.2055 protein was obtained by structural genomics research methods of gene cloning and expression, protein purification with Ni²⁺-chelating affinity chromatography, crystal screening, and X-ray diffraction data collection. An inhibitor virtual model matching with its target protein structure was set up using computer-aided drug design methods, virtual screening and fine docking, and Libdock and Autodock procedures.
RESULTSThe crystal of SMU.2055 protein was obtained, and its three-dimensional crystal structure was analyzed. This crystal was diffracted to a resolution of 0.23 nm. It belongs to orthorhombic space group C222(1), with unit cell parameters of a = 9.20 nm, b = 9.46 nm, and c = 19.39 nm. The asymmetric unit contained four molecules, with a solvent content of 56.7%. Moreover, five small molecule compounds, whose structure matched with that of the target protein in high degree, were designed and selected.
CONCLUSIONProtein crystallography research of S. mutans SMU.2055 helps to understand the structures and functions of proteins from S. mutans at the atomic level. These five compounds may be considered as effective inhibitors to SMU.2055. The virtual model of small molecule inhibitors we built will lay a foundation to the anticaries research based on the crystal structure of proteins.

8.Analysis on the risk factors of radiation pneumonitis in lung cancer patients

Objective To explore the relative factors of radiation pneumonia(RP) in patients with lung cancer after radiotherapy,and find predictors of reasonable.Methods 98 cases received radical radiotherapy of non-small cell lung cancer patients were included,prescription dose of 60-68 Gy,clinical symptoms,chest X-ray,HRCT were used to detect RP.The clinical factors and treatment parameters were collected.Analysis of univariate and multivariate were used for chnical data and treatment planning indicators,assessment of RP related factors.Results 26 cases (26.53 %) were RP in 2 above (≥2).The univariate anaiysis revealed that many parameteers (chronic obstructive pulmonary disease,whether chemotherapy,tumor volume,V20,V25,V30) were significantly associated with RP (x2 =12.253,x2 =8.802,t =3.758,t =3.432,t =3.210,t =3.190,P < 0.05).Multivariate analysis revealed that COPD and V20>25 %,V30>18 % were associated with RP (x2 =4.17,24.18,3.56,P =0.045,0.041,0.000).Conclusion Lung V20,V30,patients with chronic lung disease are the independence of the influencing factors of RP.In patients with chronic obstructive pulmonary disease the dose of normal tissue should be strictly controled,V20 ≤ 25 %,V30 ≤ 18 %.
9.Simple effect analysis on the influence of relaxation training time on relaxation effect of hospitalized patients with coronary heart disease

Objective To explore the influence of different relaxation training time on relaxation effect of patients with coronary heart disease. Methods 108 patients with coronary heart disease were randomly divided into the morning group, the night group and the morning-night group, each group adopted morning, night and morning-night relaxation training. The relaxation effect was compared between these groups. Results Significant difference existed in sleep quality, anxiety condition and hypnotic dose of the patients in these groups. The time factor played a significant role in the effect of relaxation. The morning group was different in sleep quality improvement and anxiety condition alleviation from the night group and the morning-night group. Conclusions Relaxation training at night or morning-night could effec-tively help improve the sleep quality of patients, decreasing the level of anxiety and tendency of hypnotic in pa-tients with coronary heart disease.
10.Expression of ANGPTL3 in children with primary nephrotic syndrome

Objective To examine the expression of angiopoietin-like protein(ANGPTL)3 in kidneys from children with primary nephrotic syndrome. Methods Immunohistochemistry for ANGPTL3 was performed in kidney biopsies from patients with nephrotic syndrome or hematuria, including MCD (n=31), MN(n=6), FSGS (n=6), TBMN (n=10), IgA nephropathy (IgAN) with mesangial proliferation (n=16). Normal renal tissue of 2 cases with nephrectomy for tumor were used as control. According to the episode, four groups were divided ("12 months"). The expression was quantitatively examined with IMS color image analysis system, using positive index (PI) as sediment degree of ANGPTL3 in glomeruli or tubules.
