1.Relationships between the polymorphism of methylenetetrahydrofolate reductase gene C677T and hypertension,cardiac structure and function

Objective Elevated plasma homocysteine(Hcy) level has been associated with the increased risk for cardiovascular disease.The level of plasma Hcy is related to methylene tetrahydrofolate reductase(MTHFR) gene C677T polymorphism.The aim of this study is to evaluate the relationships between MTHFR gene C677T polymorphism and hypertension,cardiac structure and function.Methods Genotype of MTHFR gene C677T polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) analysis and decided by electrophoresis and sequencing in 695 patients with essential hypertension and 509 age-matched normal controls.Echocardiography was performed in patients with essential hypertension.Results In hypertension group,the frequencies of TT genotype and T allele were significantly higher than those in control group(26.5% vs 20.6%,48.7% vs 42.4%;P=0.015,0.002).The essential hypertension odds ratio(OR) associated with MTHFR 677T allele was 1.300(95%CI:1.166 to 1.450).Logistic regression analysis indicated that MTHFR gene C677T polymorphism was an independent variable of hypertension(P=0.027).End diastolic dimension(EDD),interventricular septal thickness(IVST),left ventricular posterior wall thickness(LVPWT),left ventricular mass(LVM) and left ventricular mass index(LVMI) were higher in hypertensive patients with T allele than that in those with CC genotype,but all had no statistical significance(P=0.428,0.118,0.064,0.077,0.054).Conclusion The 677T allele in MTHFR gene may be a risk factor for hypertension.Hypertensive patients with MTHFR 677T had a trend of hypertrophy,but the relationship has no statistical significance.The relationship between MTHFR gene C677T polymorphism and left ventricular hypertrophy need to be further studied.

5.Clinical Study of Pingyi Mixture on Resisting Tumour Transfer after Surgery

Objective To observe the effect of Pingyi(PY) Mixture combined with chemotherapy to Ⅱ~Ⅲ patients after tumour surgery which can resist transference and recurrence of tumour.Method According to the random principle,120 patients who had suffered the surgery of commonly seen tumour were divided into observed group and matched group.Observed group adopted PY Mixture combined with chemotherapy and matched group adopted simple chemotherapy.After 3 periodic treatments,the effect of the patients after tumour surgery was observed including the ratio of recurrence and transference of observed group within 1 or 2 years,survival ratio,quality of life,body weight,immunological indexes,the toxic and sub-effect,the blood rheology behavior,and compared with simple chemotherapy.Results The ratio of recurrence and transference of observed group within 1 or 2 years was respectively 25.74% and 31.43%,which was obviously lower than that of control group,which was 32.00%,52.00%(P
7.Effect of lipopolysaccharide on mRNA expression of Toll-like receptor 4 and CD14 in lens epithelial cells

AIM: To investigate the effect of lipopolysaccharide (LPS) on the mRNA expression of Toll-like receptor 4 (TLR4) and CD14 in lens epithelial cells (LECs).METHODS: The cultured LECs were incubated with defferent concentrations of LPS for defferent times.The mRNA expression of TLR4 and CD14 in LECs were detected by reverse transcription polymerase chain reaction (RT-PCR).RESULTS: The mRNA expression of TLR4 in the LECs treated with LPS at concentrations of 50 μg/L, 100 μg/L, 200 μg/L, 500 μg/L or 1 000 μg/L was higher than that in the LECs without LPS treatment (P<0.01), respectively.The mRNA expression of TLR4 in the LECs incubated with 100 μg/L LPS for 24 h, 48 h or 72 h was higher than that in LECs without LPS treatment at same time points (P<0.01).The mRNA expression of CD14 in the LECs incubated with LPS at concentration of 100 μg/L for 24 h was also higher than that in the LECs without LPS treatment (P<0.01).CONCLUSION: LPS enhances the mRNA expression of TLR4 and CD14 in LECs, which may be related to intraocular response, as well as to the formation and development of after-cataract.

AIM: To investigate the effect of natural medicinal monomer elemene (Ele) on apoptosis of lens epithelial cells (LEC) and its the cellular and molecular mechanism in vitro. METHODS: The bovine LEC were cultured with Ele and used to observe the ultrastructure changes under transmission electron microscope and to detect DNA content and mitochondrial transmenbrane potential (??m) changes by flow cytometry. RESULTS: The typical morphological changes of LEC apoptosis in Ele group were observed under transmission electron microscope, such as chromatin condensation and aggregation at the nuclear periphery, and nuclear fragmentation as well. 9.Advantage Analysis of Qilian Fuzheng Capsule in Maintenance Treatment of Advanced Non-small Cell Lung Cancer

Objective To observe the effect and analyze the advantage of Qilian Fuzheng capsule in maintenance treatment of advanced non-small cell lung cancer. Methods The advanced non-small cell lung cancer patients were randomly divided into observation group and control group, 32 cases in each group. The control group was given the best supportive care, and the observation group was given Qilian Fuzheng capsule 4 capsules three times daily additionally. The tumor control rate, quality of life, PFS and OS of the two groups were compared by follow-up. Results Qilian Fuzheng capsule maintenance treatment of advanced non-small cell lung cancer improved the quality of life. PFS of observation group and control group was 5.4 months and 4.1 months, respectively (P<0.05), and OS of the two groups was 12.3 months and 10.6 months, respectively (P>0.05). The tumor control rate of observation group was 77.42% (24/31), control group was 60.00% (18/30), with no significant difference (P>0.05). Conclusion Qilian Fuzheng capsule is a feasible application in maintenance treatment of advanced non-small cell lung cancer.
10.Analysis on the risk factors of radiation pneumonitis in lung cancer patients

Objective To explore the relative factors of radiation pneumonia(RP) in patients with lung cancer after radiotherapy,and find predictors of reasonable.Methods 98 cases received radical radiotherapy of non-small cell lung cancer patients were included,prescription dose of 60-68 Gy,clinical symptoms,chest X-ray,HRCT were used to detect RP.The clinical factors and treatment parameters were collected.Analysis of univariate and multivariate were used for chnical data and treatment planning indicators,assessment of RP related factors.Results 26 cases (26.53 %) were RP in 2 above (≥2).The univariate anaiysis revealed that many parameteers (chronic obstructive pulmonary disease,whether chemotherapy,tumor volume,V20,V25,V30) were significantly associated with RP (x2 =12.253,x2 =8.802,t =3.758,t =3.432,t =3.210,t =3.190,P < 0.05).Multivariate analysis revealed that COPD and V20>25 %,V30>18 % were associated with RP (x2 =4.17,24.18,3.56,P =0.045,0.041,0.000).Conclusion Lung V20,V30,patients with chronic lung disease are the independence of the influencing factors of RP.In patients with chronic obstructive pulmonary disease the dose of normal tissue should be strictly controled,V20 ≤ 25 %,V30 ≤ 18 %.
