1.Study on adsorptive properties of spherical activated carbon on several kinds of active ingredient from Chinese materia medica

ObjectTo study the adsorptive properties of spherical activated carbon on some effective ingredients from Chinese materia medica (CMM) and to approach the application feasibility to CMM purification. Methods The spherical activated carbon with high strength has been successfully prepared by the carbonation and activation of polymeric resin. The apparent and theoretical adsorption capacities under the condition of static operation were investigated by Langmuir monolayer adsorption model. Elution by 75% alcohol was studied. Results The static adsorption capacities of the berberine hydrochloride and dioscin were 35.46 mg/g and 47.12 mg/g, and theoretical adsorption capacities were 96.16 mg/g and 102.04 mg/g, respectively. The apparent adsorption amount of rutin was 40.88% mg/g. The elution ratio were 83.71%, 91.45%, and 87.69%, respectively. Conclusion The spherical activated carbon adsorbent shows better comprehensive adsorption properties and can be used to purification.
2.The comparistion of application of three adhesive membrane in cesarean section

Objective To investigate the effect of adhesive membrane that has anti-side leakage and can be drained during the operation of cegarean section.Methods 300 cageg of lying-in women with cessrean section were randomly divided into three groups,the test 1 group,test 2 group and the control group.The test 1 group used adhesive membrane specially adopted in cerebral surgery,the test 2 group used the"U"shape adhesive membrane that had anti-side leakage and could be drained during the operation,the control group used the routine adhesive membrane,and then the contamination by amniotic fluid and blood of the thine groups was observed.Resulls Compared with test 1 group and the control group,the pollution by amniofic fluid and blood was significanfly decreased in the test 2 group,P<0.01.Conclusiom This modified designed"U"shape adhesive membrane can drain amniotic fluid and blood during cesarean section in time and simply.
3.Effects of Tiopronin Combined with Lamivudine on the Treatment of Pulmonary Tuberculosis Combined with Chronic Hepatitis B

OBJECTIVE:To investigate clinical efficacy and safety of tiopronin combined with lamivudine in the treatment of pulmonary tuberculosis complicated with chronic hepatitis B. METHODS:150 cases diagnosed as pulmonary tuberculosis with chronic hepatitis B were randomly divided into group A(drug combination group),group B(lamivudine group)and the group C (control group),with 50 cases in each group. 3 groups were given isoniazid+rifapentine+ethambutol+levofloxacin(2HTELfx/4HT) anti-TB treatment and liver protection treatment,etc. Group B was additionally given Lamivudine tablet orally,0.1 g,qd;group A was additionally given Tiopronin tablet 0.3 g,tid,on the basis of group B. The treatment course of 3 groups lasted for 6 months. Liver damage,serum fibrosis indexes of 3 groups were observed in 3 groups before and after treatment as well as hepatitis B virolo-gy indexes,clinical efficacy and the occurrence of ADR after treatment. RESULTS:After treatment,serum levels of ALT,AST and TBIL weresignificantly increased in group C,significantly decreased in group A,with statistical significance(P<0.05).There was no statistical significance in above indexes of group B before and after treatment(P>0.05). 4.Quality Standard Study for Tiaojing Pills

Objective To establish the quality standard for Tiaojing Pills. Methods Radix Angelicae Sinensis and Rhizoma Chuanxiong in Tiaojing Pills were identified by TLC and the content of paeoniflorin was determined by HPLC. Results Radix Angelicae Sinensis and Rhizoma Chuanxiong could be identified by TLC. Paeoniflorin showed a good linearity in the range of 0.086 88~0.868 8 ?g,r=0.999 6.The average recovery was 101.28 %,and RSD was 1.31 %. Conclusion The established methods are simple,convenient and reproducible,and can be used for the quality control of Tiaojing Pills.
5.Promotion of calcium oxalate crystallization after human renal tubular epithelial cell injury

Objective To investigate the injury caused by hydrogen peroxide (H2O2) on human renal tubular epithelial cell (HKC) and its effect on calcium oxalate (CaOxa) crystal crystallization time before and after the injury. Methods The injury degree of HKC by H2O2 was measured by detecting the cell survival rate and the concentration change of malonaldehyde (MDA). CaOxa crystallization was investigated by scanning electron microscopy (SEM). Results Control cells induced only a small amount of calcium oxalate dihydrate (COD) crystals, while the injured cells not only induced calcium oxalate monohydrate (COM) crystals, but also increased the number and aggregation of CaOxa crystals. After incubating with CaOxa supersaturated solution, the control group HKC cells could be injured as well. Conclusions H2O2 can cause oxidative damage on HKC. The injured HKC promotes the nucleation and aggregation of COM crystals. In the body environment, the long-term presence of crystals in urinary tract is a risk factor for stone formation.
6.Research in effect of centralized training of basic operational skills for nursing students before nursing practice

Objective To discuss effect of centralized training of basic operational skills for nursing students before nursing practice. Methods 176 student nurses in 2007 were set as the control group,320 nursing students in 2008 as the experimental group. The control group began practice after routine pre -practice education, based upon this, the experimental group carried out centralized training of basic operational skills for one week, only those who passed the examination could began practice, those who failed the examination should continued the training till they passed the examination. Time cost to master the operational skills, operational assessment results and evaluation of nursing practice by teachers were compared between the two groups. Results Compared to the control group, time cost to master the operational skills shortened, operational assessment results and evaluation of nursing practice by teachers improved in the experimental group. Conclusions Through centralized training of basic operational skills, time cost to master the operating skills of basic nursing by nursing students shortened, operational capability of nursing students can be improved, and the clinical nursing teaching quality can be promoted.
7.Effect of recombinant adenovirus vector expressing human endostatin on endothelial cell proliferation

BACKGROUND: Scar hypertrophy is always followed by the wound healing in burn and trauma. Endothelial cells play a key role in scar hypertrophy, so inhibitory growth of endothelial cells can relieve scar hypertrophy to a certain degree. OBJECTIVE: To construct a recombinant adenovirus vector expressing human endostatin (Ad/hEnd), and to investigate the cooperative effect of Ad/hEnd and keratinocyte on endothelial cell proliferation. DESIGN, TIME AND SETTING: Observational study, which was performed in the State Key Laboratory of Trauma, Burn and Combined Injury, Institute of Burn Research, Southwest Hospital of the Third Military Medical University of Chinese PLA between September 2006 and May 2007. MATERIALS: pAdTrack-CMV and pAdEasy-1 were obtained from Stratagene Company, USA; 293 cell and Ecoli.DH5α were stored in our laboratory. METHODS: The endostatin gene sequence was obtained by reverse transcription-polymerase chain reaction (RT-PCR) and polymerase chain reaction (PCR) based on mRNA of human fetal hepatic tissue and inserted into the adenovims shuttle plasmid pAdTrack-CMV to obtain recombinant plasmid pAdTrack-ES. After identification, positive recon was transformed into pAdeasy 1 recipient virus to screen positive clones. The adenovirus Ad/hEnd was generated from 293 cells and identified by PCR and fluorescence microscope. Then the keratinocytes were infected with Ad/hEnd, and co-cultured with endothelial cells by nest dish culture method. The content of endostatin was detected, and the non-transfection keratinocytes were used as the controls. MAIN OUTCOME MEASURES: Homologous recombination and identification of pAd/hEnd; generation and identification of Ad/hEnd; endostatin expression after 293 cell transfection; purification and titer measurement of Ad/hEnd; content of endostatin in culture solution; apoptotic percentage of endothelial cells; inhibitory ratio of endothelial cells. RESULTS: Ad/hEnd was constructed and the virus titer was generally up to 1.65×1012 PFU/L. Ad/hEnd-infected keratinocytes could effectively express and secrete endostatin of which the content reached 226 μg/L after 3 days of co-culture. The apoptotic percentage and inhibitory ratio of the endothelial cells co-cultured with Ad/hEnd-infected keratinocytes were significantly higher than those in control group (P<0.05). CONCLUSION: Ad/hEnd-infected keratinocytes co-cultured with endothelial cells can promote apoptosis and inhibit proliferation of endothelial cells through excretion of endostatin.
8.Clinical Observation of Acupuncture at Cranial Sutures plus Rehabilitation Training for Cerebral Infarction

ObjectiveTo observe the clinical efficacy of acupuncture at cranial sutures (cranial acupuncture) plus rehabilitation training in treating cerebral infarction.MethodSixty patients with cerebral infarction were randomized into an acupuncture-rehabilitation group (cranial acupuncture plus rehabilitation training) and a rehabilitation group (dryrehabilitation training), 30 cases in each group, to respectively receive cranial acupuncture plus rehabilitation training and dry rehabilitation training. The Fugl-Meyer Assessment (FMA) and Barthel Index (BI) were adopted to respectively evaluate the motor function of limbs and activities of daily living (ADL); the content of hypersensitive C-reactive protein (hs-CRP) was also observed before and after treatment for the evaluation of therapeutic efficacy.ResultThe acupuncture-rehabilitation group wassuperior to the rehabilitation group in improving the motor function and ADL, and down-regulating the hs-CRP content (P<0.05).ConclusionCranial acupuncture plus rehabilitation training is the optimal protocol in treating cerebral infarction, and it canproduce a more significant efficacy than dry rehabilitation training.
9.Study on quality standards of decoction pieces of salt Alpinia.

OBJECTIVETo establish the quality criteria for decoction pieces of salt Alpinia.
METHODDecoction pieces of salt Alpinia were measured with moisture, total ash, acid-insoluble ash, water-extract and volatile oils according to the procedures recorded in the Chinese Pharmacopoeia 2010. The content of Nootkatone was determined by HPLC, and NaCl, by chloridion electrode method.
RESULTWe obtained results of total ash, acid-insoluble ash, water-extract and volatile oils of 10 batches of decoction pieces of salt Alpinia moisture; Meanwhile we set the HPLC and chloridion electrode method.
CONCLUSIONThis research established a fine quality standard for decoction pieces of salt Alpinia.

10.The three-dimensional culture of adult mesenchymal stem cells for intervertebral disc tissue engineering.

Intervertebral disc (IVD) degeneration is one of the major causes of low back pain. As current clinical treatments are aimed at restoring biomechanical function and providing symptomatic relief, the methods focused on biological repair have aroused interest and several tissue engineering approaches using different cell types have been proposed. Owing to the unsuitable nature of degenerate cells for tissue engineering, attention has been given to the use of mesenchymal stem cells (MSCs). In this connection, we have made a study on the characteristics of MSCs derived from adult bone marrow and on the feasibility of constructing IVD tissue-engineering cell under a Three-Dimensional Pellet Culture System. The human bone marrow MSCs were isolated and purified with density gradient solution and attachment-independent culture system. MSCs isolated using this method are a homogeneous population as indicated by morphology and other criteria. They have the capacity for self-renewal and proliferation, and the multilineage potential to differentiate.
