1.Effects of electric stimulation on motor function and the expression of Rho kinase following cerebral infarction

Objective To investigate the effects of electric stimulation on motor function and expression of Rho kinase following cerebral infarction in rats. Methods Acute cerebral infarction was modeled in adult male Sprague-Dawley ( SD) rats using the permanent middle cerebral artery occlusion ( PMCAO) technique. The rats were randomly divided into sham operation, control, unilateral electric stimulation and bilateral electric stimulation groups ( each group had 36 rats). Electric stimulation was applied to the paralyzed ( unilateral or bilateral) limbs in the last two groups. Motor function recovery and the expression of Rho kinase were examined using a beam walking test ( BWT) and immunohistochemistry respectively at the 3rd, 7th, 14th and 21st day after stimulation. In addition, the cerebral infarction volume was also determined by 2, 3, 5-triphenyl tetrazolium chloride (TTC) staining at different time points. Results Motor function improved significantly in the electric stimulation groups compared with the control group, and the effect in the bilateral electric stimulation group was better than that in the unilateral electric stimulation group. The expression of Rho kinase decreased remarkably in the electric stimulation groups, and it was significantly lower in the bilateral group than in the unilateral electric stimulation group. No difference in cerebral infarction volume was found at the 3rd day. At the 21st day, the cerebral infarction volume had decreased significantly in both stimulation groups compared with the control group, but no difference was seen between the unilateral and bilateral electric stimulation groups. Conclusions Early electric stimulation, especially bilateral electric stimulation, can improve motor function after cerebral infarction and reduce cerebral infarction volume, which may be associated with down-regulation of the expression of Rho kinase in the border zone of the infarction.
2.Influence of mindfulness on anxiety and depression of gynecological oncology patients

Objective To investigate the effect of mindfulness on anxiety and depression of gynecological oncology patients.Methods Using objective methods,80 patients with gynecological oncology were divided to the usual care group and mindfulness training group according to random number table,40 cases in each group.The self -rated anxiety scale and self -rated depression scale were used to compare the effects at three times.Results There were 3 patients lost and 77 patients were analyzed finally.The ANOVA showed that there were significant differences on anxiety and depression scores of SAS (F =4.614,P =0.035) and SDS (F =4.070,P =0.044) between the two groups,and the scores of SAS (F =15.115,P <0.001) and SDS (F =7.034,P =0.002) had interaction effects. Conclusion Mindfulness can adjust the psychology state of gynecological oncology patients and decreased the level of anxiety and depression.
3.Clinical prognosis of acute myocardial infarction with normal coronary arteries

Objective To evaluate the clinical outcome of patients who suffered from acute myocardial infarction with angiographically normal coronary arteries (MINC) Methods Forty patients (mean age: 52 6?12 2 years; range: 30-75 years) admitted with acute myocardial infarction had normal coronary arteries after AMI, defined by normal or insignificant 50% diameter stenosis) The risk factors, clinical data, ventricular functions and the long term outcomes were compared between Group A and Group B Results Hypertension and previous angina were more frequent in Group B Patients in Group A had a lower number of risk factors associated with them than patients in Group B LVEF during hospital was higher in Group A than in Group B The mean follow up period was 67?40 months in Group A and 68?34 months in Group B Incidencs of rehospitalization, repeat intervention therapies, heart failures, and total heart events were fewer in Group A than in Group B Conclusion These data suggest a favorable long term prognosis and fewer risk factors in patients who suffered from acute myocardial infarction with normal or nearly normal coronary arteriogram
4.Expression and significance of Hsa-miR-181a in endometrial carcinoma

Objective To explore the expression and significance of hsa-miR-181a (miR-181a) in can-ceration progression of endometrial carcinoma. Methods A total of 75 formalin-fixed paraffin-embedded tissue specimens were studied in this study , of which , 13 were normal endometrium , 18 were endometrial hyperplasia , 44 were endometrial carcinoma. After total RNA had been extracted , real-time PCR was applied to detect the ex-pression level of miR-181a in endometrial tissue in each group. Results miR-181a expression in formalin-fixed paraffin-embedded tissue specimens can be detected. Expression of miR-181a in endometrial carcinoma was high-er than that in endometrial hyperplasia , its expression in endometrial hyperplasia was also higher than that in normal endometrium, and the difference was statistically significant (P < 0.05). The expression of miR-181a in endometrial carcinoma was associated with FIGO stages (P < 0.05). Conclusion The up-regulation of miR-181a expression in women with endometrial carcinoma may play the role of oncogenes. Abnormal expression of miR-181a is probably associated with the occurrence and development of endometrial carcinoma.
5.Clinical study of levonorgestrel-releasing intrauterine system for adenomyosis

Objective To investigate the clinical efficacy of levonorgestrel- releasing intrauterine system (LNG-IUS) in the treatment of adenomyosis and the influence of ovarian function. Methods Eighty patients with adenomyosis were followed up for 0, 1, 3, 6, 12 months after treating with LNG-IUS. The menstrual blood volume, dysmenorrheal, size of uteri, CA125 and EMAb level, hepatic function, serum glucose and serum lipids were observed and evaluated. Serum- levels of FSH and LH were tested before insertion and followed up for 6, 12 months after insertion, respectively. Results After inserting LNG-IUS,dysmenorrhea was obviously alleviated, and it was significantly decreased after 6 months. The menstrual blood volume was (200. 0 ±60)ml, (40. 0 ± 15)ml and (28 ±7)ml in 0, 6, 12 months after the insertion.Serum CA125 and positive rate of EMAb also significantly reduced [CA125: (50. 69 ± 10. 00) IU/L vs (18. 60 ±3.55)IU/L;EMAb:80. 0% vs 3. 8%, P <0. 05]. The volume of uterus reduced without significant changed (P >0. 05). There were little changes in the levels of hepatic function, serum glycosum, serum lipids, FSH and LH at 6 and 12 months after LNG-IUS(P > 0. 05). Conclusions LUG-IUS is an effective and safe therapy for adenomyosis with dysmenorrheal and menorrhagia, and it has no significant effects on ovarian function.
6.Gene expression profiling reveals signal transduction and abnormal expression of cytokine gene in patients with primary Sjogren syndrome

Objective To investigate signal transduction and cytokine gene expression pattern in peripheral blood mononuclear cell (PBMC) between primary Sjogren syndrome patients and healthy controls. Methods The PBMC of 3 patients with Sjogren syndrome and 3 healthy volunteers with consistent age were collected. The total RNA extracted from the PBMC was carried the reverse transcription and in vitro transcription (IVT), then labeled with Cy5/Cy3 and hybridized on the gene chips. After scanning and data extraction with LuxScan 3.0, differentially expressed genes were analyzed with SAM method. The online tools of molecule analysis system (MAS) was used for biological knowledge mining. The difference of signal transduction and cytokine gene expression of each patient and control were compared. Results Statistical difference was calculated between the patient and control group in the following four pathway: the chemokine-cytokine receptor interaction pathway[(FASLG(Fas ligand), CCL5, CCR3, IL-4R, CXCL10 (CXC chemokine ligand 10), TNFRSF19L (tumor necrosis factor supeffamily 19 ligand), CX3CR1 (CX3C chemokine receptor 1)], adhension molecule pathway [interleukin adhesion molecules-1 (ICAM-1)], Jak-STAT signal pathway [STAT4 (signal transducer and activation of transcription 4), CISH (chromogenic in situ hybridization), IL-4R], Toll-like receptor signal pathway(CCL5, CXCL10). Among these, 4 genes were up-regulated and 6 genes were down-regulated. Conclusion Understanding of differently expressed genes should help us disclose the potential molecular mechanism underlying the development process of pathogenesis of primary Sjogren syndrome.
7.Comparative advantages of different methods of collecting nasopharyngeal secretions for rapid detection of influenza virus

Objective To compare the effectiveness of different methods of collecting nasopharyngeal secre-tions by nasopharynx swab and nasopharyngeal underpressure suction catheter for rapid detection of influenza virus. Methods Nasopharyngeal secretions as the experimental samples of 1042 patients with acute respiratory tract disea-ses were collected by nasopharynx swab and nasopharyngeal suction catheter, and gold immunochromatographic assay (GICA) kit was applied for the detection of influenza viruses. Results The use of the above two methods collecting nasopharyngeal secretions as samples for rapid detection of influenza virus would get the same results. The difference between the two methods had no statistical significance( P > 0.05 ). Conclnsions Nasopharynx swab is a reliable method for rapid detection of influenza virus, which is fast and convenient, compared with nasopharyngeal suction catheter.
9.Effects of starfish saponins on insulinsignaling pathway in muscle of NAFLD rats

Aim To investigate the effects of starfish saponins(Sfs) on insulin signaling pathway in orotic acid-induced NAFLD rats.Methods 1% orotic acid was used to establish NAFLD model in male Wistar rats for six weeks.The NAFLD rats were randomly divided into two groups(eight rats in each group) and then fed with the corresponding diets: Model group(1% orotic acid)and Sfs group(1% orotic acid containing 0.04% starfish saponins).After starfish saponins feeding for 8 weeks, hepatic lipids content, liver function indices and relevant protein expression in muscle insulin signaling pathway were measured.Results Compared with model group, starfish saponins reduced hepatic lipids content and improved liver functions.In addition, it effectively ameliorated insulin resistance by improving insulin signaling pathway and improved glucose uptake in muscle.Conclusion The amelioration effect of starfish saponins on impaired insulin signaling pathway in muscle is observed in orotic acid-induced NAFLD rats.
10.Expression of regulatory T cells and helper T cells in human IgA nephropathy and its significance

Objective To investigate the effects of CD4+CD25high regulatory T cells(Treg)and the imbalance of helper T lymphocyte subsets(Th1/Th2)on the immunological mechanism of IgA nephropathy(IgAN)patients. Methods The percentage of Treg and helper T cells subpopulation (Th1/Th2)in the peripheral blood of IgAN patients and healthy controls was examined by flow cytometry.The FOXP3 expression was detected through intracellular staining.The correlation of Treg or Th1/Th2 with clinical parameters of IgAN was analyzed by Spearman or Pearson rank correlation test. Results The percentages of Treg and Th2 cells were significantly higher in peripheral blood of IgAN patients compared to that of healthy controls[Treg (2.14±0.82)%vs[1.59±0.53)%,Th2(2.57±0.72)%vs(1.81±1.10)%,all P<0.05].Th1/Th2 ratio was significantly reduced in IgAN patients(5.75±1.89 vs 12.73±9.79,P<0.05).The percentage of circulating Treg cells was positively correlated with serunl IgA concentration(r=0.397,P<0.05),and was negatively correlated with eGFR(r=-0.376,P<0.05).The percentage of circulating Th2 cells was positively correlated with serum IgA(r=0.468,P<0.05). Conclusions There is a disorder of T lymphocyte population in the peripheral blood of IgAN patients.The increased Treg and Th2 cells may play an important role in the pathogenesis of IgAN.
