Human interleukin-15 (IL-15) is a proinflammatory cytokine to suppress neutrophil apoptosis, which is a potential therapeutic agent. The modulatory effect of TNFα was investigated in IL-15-induced suppression of human neutrophil apoptosis. TNFα was shown to reverse the ability of IL-15 to delay neutrophil apoptosis within certain time course. Moreover, this reverse effect by TNFα might be associated with a reduction of the expression of the anti-apoptotic Bcl-Xl protein detected by Western blotting. It is concluded that TNFα can be used to modulate IL-15-induced suppression of neutrophil apoptosis within certain time course.

Human interleukin-15 (hIL-15) is an important cytokine to activate endothelial cells and can be regulated by many other cytokines. The aim of this study is to examine the ability of interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha) to induce the production of human interleukin-15 (hIL-15) and IL-15 receptor (IL-15Ralpha) by human umbilical vein endothelial cells (HUVECs). The data are summarized as follows: 1. Northern blot revealed that IL-15 mRNA was up-regulated by IFN-gamma and TNF-alpha. 2. Intracellular IL-15 protein was visualized by fluorescence microscopy, whereas the expression of IL-15 on the surface of HUVECs was detected by fluorescence activated cell sorting (FACS), and no detectable IL-15 in the medium was verified by ELISA. 3. IL-15Ralpha was detected on the surface of HUVECs by FACS after IFN-gamma and TNF-alpha stimulation, whereas Western blotting revealed that the elevated expression on surface IL-15Ralpha was not due to the increased protein expression. The conclusion demonstrated from our results is that IFN-gamma and TNF-alpha play an important role in regulating the expression of IL-15 and IL-15Ralpha on the surface of HUVECs.

Objective To investigate the correlation between developmental function and pathological behavior in children with autism. Methods A total of 154 children with autism were tested with Revised Chinese Version Psycho-Educational Profile (C-PEP) during 2011 to 2014. Results The scores of fine movement, hand-eye coordination, and oral language of developmental function were lower in the girls than in the boys (t>2.120, P<0.05). Among the children with autism aged 2-7 years, the developmental function was better in the older group than in the younger group (P<0.05);however, it was not better in the children aged 8-10 years and>10 years than in those aged 6-7 years. There was no difference in the scores of pathological behavior between the girls and the boys (P>0.05). Some pathological behaviors were milder in the older group than in the younger group (P<0.05). The scores ofP(pass) in developmental function were positively correlated with the scores ofA(absence) in pathological behavior (P<0.01), and negatively correlated with the scores ofS(severe) in pathological behavior (P<0.01). The scores ofF(fail) in developmental function were negatively correlated with the scores ofA(absence) in patho-logical behavior (P<0.01), and positively correlated with the scores ofS(severe) in pathological behavior (P<0.01). Conclusion Develop-mental function of children with autism is highly correlated with pathological behavior, and the rehabilitation and education of children with autism aged above 7 years should be concerned.

Objective: To evaluate cardiopulmonary exercise testing (CPET) on sildenaifl effect for treating the patients with pulmonary arterial hypertension (PAH). Methods: A total of 25 PAH patients received sildenaifl treatment in our hospital from 2012-01 to 2014-01 were enrolled as PAH group, in addition, there were a Control group including 24 healthy subjects. The CPET, echocardiography, NYHA function class, 6-mimute walking distance (6MWD) and plasma levels of NT-proBNP at the baseline, (6-12) months and (13-18) months after sildenaifl treatment were assessed and compared between 2 groups. Results: Compared with Control group, PAH group showed decreased aerobic capacity (peakVO?2, Peak O2pulse) and ventilation efifciency (PETCO2@AT, VE?/VC?O2@AT), allP<0.05. At (8±2) months after sildenaifl treatment, aerobic capacity and ventilation efifciency was improved, meanwhile, NYHA function class, 6MWD and plasma levels of NT-proBNP were improved, allP<0.05. At (16±2) months after sildenaifl treatment, 6MWD was similar,P=0.26, while peak VO?2 and peak O2 pulse were decreased than they were at (8±2) months after sildenaifl treatment,P=0.04 and 0.06; the ventilation efifciency was elevated (as presented by increased VE?/VC?O2@AT and decreased PETCO2@AT,P=0.04 and P=0.04); plasma level of NT-proBNP was increased,P=0.05. Conclusion: CPET can effectively evaluate sildenaifl effect for treating PAH patients and therefore and guide the drugs therapy.

Objective:To investigate the effect of oxygen-driven nebulization inhalation combined with invasive mechanical ventilation on sputum viscosity and inflammatory indicators in patients with severe pneumonia complicated by respiratory failure. Methods:A total of 300 patients with severe pneumonia complicated by respiratory failure who received treatment at Lanxi Traditional Chinese Medicine Hospital from June 2021 to June 2023 were included in this prospective study. Using the random number table method, the patients were divided into a control group (conventional invasive mechanical ventilation, n = 150) and a treatment group (conventional invasive mechanical ventilation + oxygen-driven aerosol inhalation, n = 150). The sputum viscosity, inflammatory indicators, and blood gas analysis indicators were compared between the two groups before and after treatment. The occurrence of ventilator-associated pneumonia, mechanical ventilation time, intensive care unit stay duration, and the length of hospital stay were recorded. Results:After treatment, the sputum viscosity grade of the treatment group was significantly different from that of the control group ( Z = 7.09, P < 0.05). The serum levels of interleukin-6 and interleukin-10 in the treatment group were significantly lower, with values of (8.19 ± 0.91) μg/L and (19.26 ± 2.17) μg/L, respectively, compared with (12.01 ± 1.34) μg/L and (32.57 ± 3.85) μg/L in the control group ( t = 28.88, 36.88, both P < 0.05). The arterial partial pressure of oxygen in the treatment group post-treatment was (95.75 ± 3.51) mmHg (1 mmHg = 0.133 kPa), which was significantly higher than (90.14 ± 3.64) mmHg in the control group ( t = 13.58, P < 0.001). The arterial partial pressure of carbon dioxide in the treatment group was (40.65 ± 4.03) mmHg, which was significantly lower than (44.81 ± 4.12) mmHg in the control group ( t = 8.84, P < 0.001). The incidence of ventilator- associated pneumonia in the treatment group was significantly lower than that in the control group ( χ2 = 14.00, P < 0.001). The mechanical ventilation time, intensive care unit stay duration, and the length of hospital stay in the treatment group were significantly shorter compared with the control group ( t = 25.82, 18.23, 15.28, all P < 0.001). Conclusion:Oxygen-driven aerosol inhalation combined with invasive mechanical ventilation can effectively dilute sputum, reduce the patient's inflammatory response, improve blood gas analysis indicators, lower the risk of ventilator- associated pneumonia, and thereby facilitate recovery from severe pneumonia complicated by respiratory failure.
