Objective To investigate the effect of artesunate on the expression of heat shock protein 47(Hsp47)in pulmonary fi-brosis in rats.Methods Thirty SD male rats were randomly divided into control group,model group and artesunate intervention group (intervention group).Rats in control group were intrachacheally instilled with saline (0.4 mL),while rats in other two groups 5 mg/kg bleomycin.Rats in both control group and model group were intraperitoneal injected with 1 mL saline per day sub-sequently.And the rats in intervented group were intraperitoneal injected with 10 mg/100 g artesunate per day.The body weight of day 0,day 14 and day 28 were recorded.All the rats were sacrificed on day 28.The pulmonary coefficient and the hydroxyproline content were observed.The expression of Hsp47mRNA and CollagenⅠmRNA were evaluated by RT-PCR.The Hsp47 expression was also detected by Western Blot.The pathological changes were analyzed by hamatoxylin-eosin (HE)stain and Masson stain.Re-sults (1)The body weight of control group grew much faster than other groups (P <0.05).(2)Pulmonary coefficient:model group (12.31±1.89)mg/g>intervention group (8.54±0.67)mg/g>control group(4.81±0.38)mg/g (P <0.05).(3)Ashcroft Scores:model group(5.70±1.09 )>control group (3.08±0.56),model group(5.70± 1.09)>intervention group (4.01 ± 1.25).There was no significant difference between control group and intervention group(P >0.05).(4)Hydroxyproline content:model group (620.33±66.16)μg/g>control group(379.00±35.51)μg/g,model group (620.33±66.16)μg/g >intervention group(429.00± 36.51)μg/g (P <0.05),there was no significant difference between control group and intervention group.(5)Hsp47mRNA expres-sion:model group and intervention group > control group (P <0.05),there was no difference between the first two groups(P >0.05).CollagenⅠmRNA:model group > intervention group>control group (P <0.05).(6)Western blot showed that Hsp47 ex-pression of model group was the highest in all three groups.Conclusion These results indicate that artesunate might inhibit pulmo-nary fibrosis and depress CollagenⅠproduction,and it′s probably a result of Hsp47 down regulation.

Objective To discuss the effect of psychological and behavioral intervention on sleep quality and self efficacy in patients with type 2 diabetes mellitus complicated with sleep disorder.Methods 80 patients with type 2 diabetes mellitus complicated with sleep disorder in our hospital were divided into control group (with 40 cases) and intervention group (with 40 cases)according to the random digital table method.The control group were given routine nursing,while the intervention group were given psychological and behavioral intervention based on the control group.The changes of sleep quality and self efficacy,scores of anxiety and depression and fasting blood glucose before and after intervention were compared between the two groups.Results The differences in the scores of PSQI and GESE were not statistically significant between the control group and the intervention group before intervention (P >0.05),which were significantly improved after intervention[(8.26 ±1.32)points,(29.65 ±3.01)points;(5.62 ± 1.03)points,(34.24 ±4.15)points;t =15.386,10.502,28.087,14.751,all P <0.05],and the scores in the inter-vention group were better than those in the control group (t =9.972,5.663,both P <0.05).There were not obvious differences in the scores of SAS and SDS between the control group and the intervention group before intervention(all P >0.05),which were significantly decreased after intervention[(41.24 ±3.21)points,(42.52 ±3.01 )points;(32.36 ±2.12)points,(34.24 ±4.15)points;t =19.674,12.550,27.589,19.546,all P <0.05)],and the scores in the intervention group were significantly lower than those in the control group (t =14.599,10.215,both P <0.05).There was no obvious difference in fasting blood glucose between the two groups before intervention (P >0.05),which were significantly decreased after intervention[(8.76 ±1.06)mmol/L,(6.25 ±1.32)mmol/L,t =3.646,8.955,all P <0.05],and the level in the intervention group was significantly lower than that in the control group (t =9.377,P <0.05).Conclusion The psychological and behavioral intervention can effectively improve the
sleep quality of patients with type 2 diabetes mellitus complicated with sleep disorder,improve the self efficacy,and relieve anxiety and other negative emotions,then decrease the fasting blood glucose level,which has promotion value in the clinical nursing.

OBJECTIVE: To investigate the situation and future trend of use of drugs in this hospital after medical re- form.METHODS:Using the principle of sequencing analysis of DUD system of drug,we analysed the data of drug consumption in our hospital during the period 1999-2000.RESULTS: The policy of medical reform has affected the component ratio of drugs used, the consumption of peroral drugs being increased.The incidence of cardiovascular diseases was high in this re- gion.CONCLUSION:Most of the drugs used after medical reform were those assigned in medical insurance,which will improve the rationality and effectiveness of medication.

OBJECTlVE To explore the mechanisms of tau hyperphosphorylation induced by calyculin A ( CA) in neuroblastoma cells and the effect of melatonin. METHODS N2a cells were treated with CA 5 nmol·L-1 , or CA with melatonin 50 μmol·L-1 , or CA with vitamin E ( Vit E ) 50 μmol·L-1 for 12 h. The level of tau phosphorylation at Ser422 ( recognized by R145d antibody) site and the level of phosphorylated c-Jun N-terminal kinases ( p-JNK ) and phosphorylated mitogen-activated protein kinase kinase 4 ( p-MKK4 ) were detected with immunoblotting, the level of malondialdehyde ( MDA ) was assayed with fluorimetry, and the activity of p38-mitogen activated protein kinase ( p38MAPK ) was assayed by radioimmunobloting. RESULTS CA treatment increased the level of phosphorylated tau at Ser422 site (1.70±0.19, 1.0, P<0.01), and melatonin attenuated the effect of CA (0.98±0.12, 1.70± 0.19, P<0.01). ln addition, CA treatment increased the level of MDA (μmol·g-1 protein:0.241±0.006, 0.141±0.006, P<0.01) and melatonin antagonized the increase of MDA induced by CA (μmol·g-1 protein:0.172±0.004, 0.193±0.005, 0.241±0.006, P<0.01) . CA treatment increased the level of p-JNK (1.91±0.27, 1, P<0.01) and p-MKK4 (1.81±0.09, 1, P<0.01) and melatonin antagonized the effect of CA induced increase of p-JNK (1.11±0.15, 1.91±0.27, P<0.01) and p-MKK4 (1.14±0.06, 1.81±0.09, P<0.01) without changing the level or activity of p38MAPK. Both JNK inhibitor ( SP600125 ) and MKK4/JNK transduction pathway inhibitor antagonized CA induced tau phosphorylation at Ser422 site and JNK phosphorylation. CONCLUSlON lnhibiton of JNK phosphorylation is possibly involved in the protection of melatonin on CA-induced tau hyperphosphorylation at Ser422 site.

Systematic pharmacological evaluation of material basis for the efficacy of Chinese herbal formula is one of the key issues in Chinese materia medica(CMM),which is in urgent need of solution.However,its scientific connotation is still controversial in academic circles.The material basis for the efficacy of Chinese herbal formula is the total non-endogenous bioactive compound(derived either directly or indirectly from this formula) exerting the therapeutic effects,which is generated by the interaction of the formula with human body and reaches certain concentration in the target organs or tissues.Systematic evaluation of material basis for the efficacy of Chinese herbal formula is the cornerstone of modernization and internationalization of CMM,while the quality control of CMM is its prerequisite.

OBJECTIVE To discuss the importance of communication in infection control.METHODS To eliminate the obstacles in infection control,we used different communication skills and manners to gain the aid from the leadership.RESULTS Both the leadership and everyone else in the hospital supported our work in infection prevention and control,for they had known the importance of infection control,especially in hospital.We communicated with them about the knowledge in this aspect in different means.CONCLUSIONS The professional technician should take different ways in communication to solve the problems in every department in the daily work,which can help us to achieve success in infection control.

Objective To establish a RP- HPLC method for the determinatio n of protocatechualdehyde in Danqi tablet. Methods LiChrospher RP- 18 column w as used, the mobile phase was MeOH- 1.5 % Hac(1∶ 9) with a flow rate of 1.0 mL/ min, and the detection wavelength was at 280nm. Results There was a goo d linearity within the range of 0.030～ 0.150 ? g of protocatechualdehyde. The average recovery was 103.08 % and RSD was 0.796 % (n=5). Conclusion This method can be used for the quality control of Danqi tablet .

Objective To explore the significance of established Tp0259-PCR in diagnosing primary syphilis.To provide basis for establishing new PCR diagnosis method of Treponema pallidum.Methods Specific primers were designed by the gene sequences of Tp0259 which was obtained from Genbank.Tp0259-PCR was established under the optimization reaction condition.Using the new construction of amplification method to detect the common pathogenic microorganism in urinary and reproductive tract infec-tion.42 clinical secretion specimens suspected primary syphilis were tested by Tp0259-PCR.Serum specimens from the same pa-tients were tested by TPPA.Results Only specific amplicons could be found in amplifying the Treponema pallidum by Tp0259-PCR.The detection rate was 73.8% using Tp0259-PCR which was apparently higher than 57.1% using TPPA(P <0.05 ).The specificity of the established method was 100% comparing with TPPA.Conclusion The established method of Tp0259-PCR has high sensibility and specificity.This method might be better than serological method in diagnosis of primary syphilis.but its clinical diagnosis value need to be repeatedly verified by more clinical specimens.

The proliferation of vascular smooth muscle cells is closely related to the pathogenesis of a variety of diseases, such as atherosclerosis, hypertension, diabetes, vascular restenosis, and etc. A proper model for smooth muscle cell proliferation can be served as an important tool for exploring the molecular mechanisms of smooth muscle cell proliferation and the screening of potential inhibitory agents. Various factors can induce the proliferation of smooth muscle cells with different mechanisms and pathological significance. The ox-LDL, AGEs, angiotensin II (AngⅡ), high glucose, tumor necrosis factor (TNF) and etc. have been successfully applied in the establishment of smooth muscle cell proliferation model. This article summarized the most commonly used models for smooth muscle cell proliferation. The inhibitory effects of Chinese herbal medicine, especially the isolated pure compounds were discussed. It was aimed to provide references for the screening of smooth muscle cells based medications as well as studies on related research and development of Chinese herbal medicine.
