〔Abstract〕 The paper introduces the informatization management system in specialist clinic blood-taking room, which compares with the process before informatization from the aspects of error rates of blood samples, patients satisfaction, hospital labor costs and so on to highlight the advantages of informatization management.The progressiveness, existing problems and corresponding countermeasures are discussed, informatization management could improve work efficiency and patients satisfaction.

Objective:To investigate the relationship between pemphigus vulgaris(PV) and HLA-DR,DQ haplotypes in Han nations of northeast China.Methods:Polymerase chain reaction-sequence specific primers(PCR-SSP) method was used to detect the HLA-DRB1 and DQB1 alleles of 27 PV patients of Han nation of northeast China, analysed haplotyes and compared with 99 healthy controls.Results:Compared with control group, the frequencies of the haplotypes of HLA-DRB1*140x-DQB1*0503,DRB1*140x-DQB1*0201,DRB1*120x-DQB1*0503 and DRB1*140x-DQB1*0302 increased significantly in PV group. After statistical test, the difference between the two groups was significant.Conclusion:The special haplotypes may contribute to genetic susceptibility to PV in northeast Chinese.

Objective To study the anti-proliferation and apoptosis induction effect of cisplatin combined with Astragalus on human laryngeal cancer Hep-2 cells.Methods The proliferation inhibition of cisplatin and Astragalus alone or in combination on Laryngeal cancer Hep-2cells was measured by MTT assay.Effects of cisplatin and Astragalus alone or in combination on apoptosis of Hep-2 cells were detected by flow cytometry (FCM).Western blot was used to analyze the expressions of Bcl-2 and Bax proteins.Results The inhibition ratio of proliferation of Hep-2 cells was (53.83 ± 17.12) ％ in the astragalus group,(69.12 ± 27.12)％ in the cisplatin group,and (84.55 ± 27.84)％ in the cisplatin combined with Astragalus group,and was significantly greater than the control group (0％) (t =16.87,16.67,40.90,P ＜0.01),respectively.The apoptotic ratio of Hep-2 cells was (38.2 ± 13.6)％ in the astragalus group,(67.2 ± 17.8)％ in the cisplatin group,and (86.4 ± 25.1)％] in the cisplatin combined with Astragalus group,and was significantly greater than control group (17.1 ± 1.3) ％ (t =8.11,12.77,24.92,P ＜0.05),respectively.The effect in the combination group is better than the other group (t =11.33,9.37,P ＜ 0.01).The expressions of Bcl-2 and Bax proteins were changed.Conclusions The tumor-killing effect of cisplatin on laryngeal cancer Hep-2 cells could be enhanced significantly by the combination application of astragalus by the way of regulating the expression of Bcl-2/Bax.

Objeetive To study the differences of the biological characteristics and immune regulation function of adipose mesenchymal stem cells (AMSCs)from psoriasis patients and healthy people.Methods AMSCs were isolated and cultured from human psoriatic and healthy adipose tissue,the phenotypes and cell cycle of AMSCs taken from three generation were detected by flow eytometry.Alkaline phosphate enzyme staining and oil red o staining were used respectively to identify their adipogenic and osteogenic capacity.Next,the levels of inflammation antimicrobial proinflammatory factor were detected by PCR and ELISA.Then gene expression profile of AMSCs were screen by gene expression profile chip,as so to bolting the the gene array related with immunology gene.Results There was no significant change in cell morphology,and cell surface markers were expressed high for CD29,CD44,CD73,while lower for CD31,CD45 and HLA-DR.AMSCs of psoriasis patients and healthy people both had the ability of adipogenic and osteogenic differentiation.But the cell cycle showed the third generation AMSCs proliferation rates were slower than that of normal control,as compared with healthy controls,adipogenic differentiation ability was stronger.What'more,the level of inflammatory cytokines in psoriasis group was lower than that in controls such asIL-10,IDO,TGF-β,on the contrary the levels of proinflammatory factor in psoriasis group were higher than that in controls,such as TNF-α,IFN-γ.In addition,gene chip results suggested that psoriasis group AMSCs had obvious expression differences on JAK-STAT pathway with healthy controls.Conclusions Compared with the control,there are significant differences in patients AMSCs proliferation and adipogenic differentiation ability,immune inflammation suppression control ability is weaken,this phenomenon may be associated with JAK-STAT immune pathways related to downgrade.

Objective To establish the mouse models of focal cerebral ischemic/reperfusion (I/R) injury combined with diabetes mellitus (DM),and investigate the possible mechanism of brain injury.Methods Eighty healthy C57BL/6J mouse were randomly divided into normal group and DM group;normal saline or streptozotocin was given to the mice in the two groups via intraperitoneal injection,and the concentration of glucose in peripheral blood was measured at different time points (2,4,6 and 8 weeks after injection);and then,the mice in the two groups were subdivided into normal sham-operated group,normal I/R group,DM sham-operated group and DM+I/R group (n=20).The focal I/R models were induced by thread occlusion method;the neuroethology assessment was determined by Zea Longa method in the normal I/R group and DM+I/R group;the morphology of the brain tissues in the four groups was observed by HE staining;changes of mRNA expressions of interleukin (IL)-1β,IL-6 and tumor necrosis factor (TNF)-α were analyzed by quantitative real-time PCR.Results (1)The glucose concentration in DM group was stable at a high level within the 8 weeks;as compared with normal group,DM group had significantly higher glucose concentration at each time point (P＜0.05).(2) The assessment scores of DM+I/R group (2.800±0.092) were significantly higher than those of I/R group (1.750±0.123,P＜0.05).(3) Typical pathological changes were presented by HE staining,DM+I/R group presented worse damage to the brain as compared with normal I/R group.(4) Quantitative real-time PCR showed that the IL-1β and TNF-α mRNA expressions in the normal I/R group was significantly higher than those in the normal sham-operated group (P＜0.05);the mRNA expressions of IL-1β,IL-6 and TNF-α in DM+I/R group were significantly increased as compared with those in the DM sham-operated and normal I/R group (P＜0.05).Conclusion The simple and reliable focal cerebral I/R combined with DM models are successfully established;inflammation reaction mediated by inflammatory factors might play an important role in this aggravating damage.

Objective To explore the effect of postoperative rehabilitation nursing for hypertensive intracerebral hemorrhage treated by minimally invasive hematoma evacuation.Methods A total of 100 cases with hypertensive intracerebral hemorrhage treated by minimally invasive hematoma evacuation were randomly divided into two groups.Patients in the control group were given routine nursing,while the experimental group applied rehabilitation care.Hematoma removal,deterioration rate and mortality,related score of disease and rehabilitation before and after intervention,the incidence of related clinical adverse events and family nursing satisfaction were compared.Results The 3 d hematoma clearance rate and overall hematoma clearance rate of the experimental group were significantly higher than that of the control group (P ＜ 0.05).The scores of GSS and AS were significantly higher,and the scores of SSS,FM and ADL were significantly lower than that of the control group(P ＜ 0.05).The incidence of overall adverse events was significantly lower,and nursing satisfaction were significantly higher than that in the control group (P ＜ 0.01).Conclusion Effect of postoperative rehabilitation nursing for patients with hypertensive intracerebral hemorrhage treated by minimally invasive hematoma is significant,and it can improve satisfaction of family members.

Objective To investigate whether cerebral ischemia reperfusion (I/R) injury combined with diabetes mellitus (DM) could elevate concentrations of serum high mobility group box 1 (HMGB 1) and its related inflammatory factors,and to explore the underlying mechanism of cerebral I/R injury combined with DM by blocking HMGB 1.Methods One hundred and forty healthy male C57BL/6 mice were randomly divided into four groups:normoglycemia (NG) group (n=25),NG+I/R group (n=25),hyperglycemia (HG) group (n=25),and HG+I/R group (n=65);40 mice in the HG+I/R group were chosen and divided into anti-HMGB1 group and IgG control group (n=20).High-fat feeding and i.p.injection of streptozotocin were used to establish HG mouse models,and then,middle cerebral artery occlusion was performed to establish the HG+I/R mouse models.Mice were treated with tail intravenous injection of 30 μg/g anti-HMGB 1 polyclonal antibody or control IgG 1 h before ischemia as previously described.After accomplishment of animal models,serum HMGB1 concentrations were evaluated by ELISA,the permeability of blood brain barrier (BBB) was observed by Evans-blue fluorescence quantitative method,quantitative real-time PCR was introduced to detect the mRNA expressions of interleukin (IL)-1β,IL-6,and inducible nitric oxide synthase (iNOS).Morphology changes of damaged brains were observed by HE staining.Results The serum HMGB 1 level in the HG+I/R group was significantly higher than that in the NG+I/R group (P＜0.05).HG+I/R group had significantly higher BBB permeability than NG+I/R group (P＜0.05),while anti-HMGB1 group had significantly lower BBB permeability than HG+I/R group (P＜0.05).As compared with the NG group,the HG+I/R group had unclear cellular structures in the brain tissues with necrosis neurocytes and interstitial edema,while the cellular structures were obviously improved in the anti-HMGB1 group.Expressions ofIL-1β,IL-6 and iNOS in the HG+I/R group were significantly elevated as compared with those in the NG+I/R group (P＜0.05),while those in the anti-HMGBl group had significantly decreased levels ofIL-1β and iNOS as compared with those in the HG+I/R group (P＜0.05),and the IL-6 level showed no significant difference between the two groups (P＜0.05).Conclusion The pathogenesis of DM could increase concentration of serum HMGB 1,and anti-HMGB 1 mAb could alleviate brain injury by blocking HMGB 1,and render a new promising therapeutic way for DM patients suffered with ischemic stroke.

Objective: To analyze the association between different sleep behaviors and overweight and obesity of junior high school students in Yangzhou City, and to provide a basis for policies and interventions related to adolescent health management. Methods: A total of 1 589 students in grades 7-9 from two middle schools in Yangzhou City were selected using the cluster sampling method and were administered with sleep time, bedtime, social jetlag difference, and sleep habits. Results: Totally 64.38% were sleep deprived during the school days, 86.78% went to bed too late, 46.51% had a social jetlag of ≥1 h, and 37.44% took a nap every day(Incluldes holidays and school days). Social jetlag length was statistically different between grades( F =6.97, P < 0.01 ). Girls[(0.95±0.65)h] shown significantly higher social jetlag than the boys[(0.76±0.59)h]( t=6.19, P <0.01). Later bedtime on weekends, later wake up time on weekends and poor sleep behavior were risk factors for overweight and obesity in junior high school students( OR=1.20, 1.14, 1.04, P <0.05). Conclusion Junior high school students had less sleep and later bedtimes with the increase of grade, and weekend bedtimes,wake up times and poor sleep behavior were independently associated with the risk of overweight and obesity in junior high school students. Parents and schools should be instructed to pay attention to their sleep health and carry out adolescent sleep health guidance.
