1.Clinical observation on the effect of modified Yiweitang on anorexia of the Spleen and Stomach Yin deficient type

Objective To observe the curative effect of modified Yiweitang on anorexia of the Spleen and Stomach Yin deficient type. Methods90 patients with anorexia belonging to spleen yin deficiency type were randomized in to 2 groups, by 1: 1 ratio. According to the age of children, appropriate dose of modified Yiwei decoction (treatment group) and Erbao Granule (control group), 3 times/d, for 4 weeks were given to the children. Clinical efficacy of the treatment group was observed before and after the treatment and compared with the control group. Results(①) symptom scores: Vomiting (0.061±0.024), sweating (0.140±0.004), stool (0.024±0.011), urine (0.026±0.010) in treatment group after treatment, showing statistical significance (P<0.05) with the control group. ② disease treatment: The total effective rate was 93.3% and 91.1% in the treatment group and the control group, the difference was not statistically significant (X2=0.886, P>0.05). ③symptoms treatment: The total effective rate was 95.6% and 66.7% in the treatment group and the control group respectively, the difference was statistically significant (x2=0.625, P<0.05). Conclusion The treatment of modified Yiweitang was better than Erbao Granule in treating children with anorexia belonging to stomach yin deficiency.
2.Determination of Protocatechualdehyde in Danqi Tablet by HPLC

Objective To establish a RP- HPLC method for the determinatio n of protocatechualdehyde in Danqi tablet. Methods LiChrospher RP- 18 column w as used, the mobile phase was MeOH- 1.5 % Hac(1∶ 9) with a flow rate of 1.0 mL/ min, and the detection wavelength was at 280nm. Results There was a goo d linearity within the range of 0.030~ 0.150 ? g of protocatechualdehyde. The average recovery was 103.08 % and RSD was 0.796 % (n=5). Conclusion This method can be used for the quality control of Danqi tablet .
3.Determination of Icarin in Naoling Granule by HPLC

Objective: To establish a method for determinating icarin content in Naoling Granule. Methods: Water-soluble ethyl-acetate-extracting polyamide column purification was adopted. The content of icarin was determined by HPLC. C18 column was used, mobile phase was methanol:0.1mol/mL sodium phosphate dibasic (6:5) and detection wavelength was at 270nm. Results: Icarin has a good linearity in the range of 0.24~1.2?g ( r=0.9998). The average recovery was 98.23 %and RSD was 1.2 %(N=5). Conclusion: The method was simple with good reproducibility and can be used for the content determination of icarin in Naoling Granule.
4.Association between serum cystatin C and carotid intima -media thickness in patients with type 2 diabetes mellitus

Objective To evaluate the association between serum cystatin C and carotid atherosclerosis in patients with type 2 diabetes mellitus.Methods A total of 318 patients with type 2 diabetes mellitus were enrolled in this study.According to the results of carotid color Doppler ultrasound in the patients were randomly divided into two groups:CIMT abnormal group (CIMT was equal to or larger than 0.9 mm,with or without associated with carotid artery plaque formation)CIMT and normal group (CIMT <0.9 mm,and it was not associated with carotid artery atherosclerotic plaque formation.Differences in serum cystatin C,serum lipid and lipoprotein,fasting plasma glucose and other biochemical markers between the two groups were analyzed by using Student's t test.The correlated factors of the carotid thickness and plaque appearance were analyzed by Logistic regression analysis.Results The serum cystatin C levels of the CIMT abnormal group were higher than the normal CIMT group[(1.24 ±0.22)mg/L vs.(1.05 ±0.41)mg/L, t =4.949,P <0.001).Multivariate logistic regression analysis showed adjusted for gender,age and other factors,ser-um cystatin C was independently associated factor of CIMT (OR =2.563,95% CI:1.026 ~6.403,P =0.044). Conclusion Our data suggest that serum cystatin C is significantly associated with carotid atherosclerosis.
5.A new sesquiterpenoid from fungus Colletotrichum sp. and its cytotoxicity.

A novel sesquiterpenoid (1) and three known compounds identified as isoaltenuene (2), altenuene (3), and alternariol 4, 10-O-dimethyl ether (4), were isolated in our investigation of the cytotoxic constituents from solid cultures of the endophytic fungus Colletotrichum sp. The structures of these compounds were elucidated through spectroscopic data analysis. All compounds exhibited cytotoxic activity against lung cancer cell line A549, breast cancer cell line MDA-MB-231 and pancreatic cancer cell line PANC-1. Compound 4 could induce the PANC-1 cells inflation or death, but couldn't induce apoptosis at the IC50 of 60.2 microg x mL(-1).
6.Differences of Ventilation Function in Patients Between Chronic Right Heart Failure Secondary to Pulmonary Arterial Hypertension and Chronic Whole Heart Failure Secondary to Left Heart Diseases

Objective:To study the differences of ventilation function in patients between chronic right heart failure secondary to pulmonary arterial hypertension and chronic whole heart failure secondary to left heart diseases.
Methods:A total of 102 patients with clinical stable chronic heart failure (CHF) were studied. The patients were divided into 2 groups:Whole heart failure (WHF) group, n=41 and Right heart failure (RHF) group, n=61. The ventilation function test at rest and cardiopulmonary exercise test (CPX) were conducted and compared between 2 groups.
Results: The rest ventilation function was similar between 2 groups. For CPX examination, compared with WHF group, RHF group had the lower peak minute ventilation (VE), higher anaerobic threshold and peak minute ventilation/CO2 production (VE/VCO2), higher peak dead space volume/tidal volume (VD/VT), all P<0.05.
Conclusion:At exercise condition, RHF patients had decreased ventilation function and ventilation index due to severe abnormality of ventilation/perfusion (V/Q).
7.Isocryptotanshinone Induced Apoptosis and Activated MAPK Signaling in Human Breast Cancer MCF-7 Cells.

PURPOSE: Isocryptotanshinone (ICTS) is a natural bioactive product that is isolated from the roots of the widely used medical herb Salvia miltiorrhiza. However, few reports exist on the mechanisms underlying the therapeutic effects of ICTS. Here, we report that ICTS has anticancer activity and describe the mechanism underlying this effect. METHODS: The antiproliferative effect of ICTS was determined using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) and clonogenic assays. The effect of ICTS on the cell cycle was measured using flow cytometry. Apoptosis was determined by Hoechst 33342 staining, DNA fragmentation assays, and Western blotting for apoptotic proteins. Finally, the effect of ICTS on mitogen-activated protein kinases (MAPKs) was determined by Western blotting. RESULTS: ICTS significantly inhibited proliferation of MCF-7 and MDA-MB-231 human breast cancer cells, HepG2 human liver cancer cells, and A549 human lung cancer cells in vitro. Among the tested cell lines, MCF-7 cells showed the highest sensitivity to ICTS. ICTS significantly inhibited colony formation by MCF-7 cells. Furthermore, exposure of MCF-7 cells to ICTS induced cell cycle arrest at the G1 phase and decreased mitochondrial membrane potential. Hoechst 33342 staining and Western blot analysis for apoptotic proteins suggested that ICTS induced apoptosis in MCF-7 cells. In addition, ICTS activated MAPK signaling in MCF-7 cells by inducing time- and concentration-dependent phosphorylation of JNK, ERK, and p38 MAPK. CONCLUSION: Our results suggest that ICTS inhibited MCF-7 cell proliferation by inducing apoptosis and activating MAPK signaling pathways.

8.Evaluation of Cardiopulmonary Exercise Testing on Sildenafil Effect for Treating the Patients With Pulmonary Arterial Hypertension

Objective: To evaluate cardiopulmonary exercise testing (CPET) on sildenaifl effect for treating the patients with pulmonary arterial hypertension (PAH). Methods: A total of 25 PAH patients received sildenaifl treatment in our hospital from 2012-01 to 2014-01 were enrolled as PAH group, in addition, there were a Control group including 24 healthy subjects. The CPET, echocardiography, NYHA function class, 6-mimute walking distance (6MWD) and plasma levels of NT-proBNP at the baseline, (6-12) months and (13-18) months after sildenaifl treatment were assessed and compared between 2 groups. Results: Compared with Control group, PAH group showed decreased aerobic capacity (peakVO?2, Peak O2pulse) and ventilation efifciency (PETCO2@AT, VE?/VC?O2@AT), allP<0.05. At (8±2) months after sildenaifl treatment, aerobic capacity and ventilation efifciency was improved, meanwhile, NYHA function class, 6MWD and plasma levels of NT-proBNP were improved, allP<0.05. At (16±2) months after sildenaifl treatment, 6MWD was similar,P=0.26, while peak VO?2 and peak O2 pulse were decreased than they were at (8±2) months after sildenaifl treatment,P=0.04 and 0.06; the ventilation efifciency was elevated (as presented by increased VE?/VC?O2@AT and decreased PETCO2@AT,P=0.04 and P=0.04); plasma level of NT-proBNP was increased,P=0.05. Conclusion: CPET can effectively evaluate sildenaifl effect for treating PAH patients and therefore and guide the drugs therapy.
9.Comparison of Applicability Test of Sterile Examination Method for Artemether Injection

Objective:To develop a more suitable sterile examination method for artemether injection in order to control its quality better. Methods:The direct inoculation and the membrane filtration was respectively used for the sterile examination of artemether in-jection. Results:In the direct inoculation method, Staphylococcus aureus, Bacillus subtilis and Clostridium perfringens grew well in the test tubes containing artemether injection in 24 h, and Escherichia coli, Aspergillus niger and Candida albicans grew well in 48 h. In the membrane filtration method, Staphylococcus aureus grew well in 24 h in the test group containing artemether injection, and the other 5 strains grew well in 48 h. Conclusion:The positive strains can grow well in the applicability test of both methods. However, due to the simpler operation and higher accuracy of membrane filtration method, it is recommended to be used for the sterile test of artemether injection.
