Objective:To evaluate the in vitro and in vivo immunostimulatory activity and the safety of ethanol extract of wild Artemisia rupestris L. (EEWAR). Methods:Bone marrow dendritic cells (BMDCs) from C57BL/6 mice were treated with different concentrations of EEWAR in vitro and the expression of CD40 and CD80 on BMDCs was detected by flow cytometry. ICR mice were subcutaneously immunized with different concentrations of EEWAR in combination with ovalbumin (OVA) or OVA alone. Aluminum adjuvant was used as the positive control. OVA-specific IgG antibodies in mouse serum samples were measured by ELISA following immunization. T cell proliferation in spleen tissues was detected by MTT method. Acute toxicity test was conducted in ICR mice to analyze the safety of EEWAR. Results:In vitro experiment showed that EEWAR at the concentrations of 10-20 μg/ml increased the expression of CD40 and CD80 on BMDCs ( P<0.05), and had no significant effect on the morphology of BMDCs; EEWAR at the concentrations of 100-200 μg/ml significantly promoted the expression of CD40 and CD80 on BMDCs ( P<0.01), but had a certain influence on the morphology of BMDCs. In vivo experiment showed that EEWAR enhanced the production of IgG, IgG 1 and IgG 2a antibodies against OVA and the proliferation of splenocytes ( P<0.05). In the acute toxicity test, EEWAR at the concentrations of 50-5 000 μg/ml had no side effects on mouse body weight and was relatively safe. Conclusions:EEWAR could promote the maturation of DCs and enhance the humoral and cellular immune responses when used as an adjuvant to OVA. It was safe in a certain dose range. This study provided reference for further research on EEWAR as a new-generation adjuvant.

AIM:To observe the effects of liraglutide on the level of microRNA-33 (miR-33) and the expres-sion of AMP-activated protein kinase ( AMPK ) and apoptosis-related proteins in mice with type 2 diabetes mellitus (T2DM), and to explore its possible mechanism .METHODS:High-fat diet and intraperitoneal injection of streptozocin were used to establish the type 2 diabetic model in C57BL/6 mice.The mice were randomly divided into 4 groups ( n=15 ):in control group , the normal mice were subcutaneously injected with equivalent volume of saline ;in model group , the T2DM mice were subcutaneously injected with equivalent volume of saline ; in low-and high-dose liraglutide treatment groups, the T2DM mice were subcutaneously injected with 100 and 200 μg? kg -1? d-1, respectively.After 4 weeks of administration, the levels of FBG, TG, TC, HDL-C, LDL-C, ALT and AST were determined.HE staining was used to ob-serve the pathological changes of the liver tissues .The protein level of cleaved caspase-3 in the liver tissue was detected by the technique of immunofluorescence .The protein levels of p-AMPK/AMPK and apoptosis-related proteins were detected by Western blot .The expression of miR-33 in the liver tissues was detected by real-time PCR.RESULTS: Compared with model group, the contents of FBG, TG, TC, LDL-C, ALT and AST were decreased significantly , while the content of HDL-C was increased significantly in low-dose liraglutide group and high-dose liraglutide group ( P<0.05 ) .The protein levels of phosphorylated AMPK and Bcl-2 were up-regulated significantly , and the expression of cleaved caspase-3 was down-regulated significantly (P<0.05).The level of miR-33 was decreased significantly (P<0.01).CONCLUSION:Liraglutide alleviates liver injury in type 2 diabetic mice , and the mechanism may be associated with reducing the level of miR-33 and increasing the phosphorylation of AMPK in the liver tissues , thereby inhibiting hepatocyte apoptosis .

Objective To determine the pharmacokinetics and pharmacodynamics of chloroprocaine with or without epinephrine for epidural blockade. Methods Twenty ASA physical status Ⅰ or Ⅱ patients of both sexes aged 37-55 yrs weighing 45-80 kg undergoing elective lower abdominal surgery were randomly divided into 2 groups (n = 10 each): group C chloroprocaine without epinephrine and group CE chloroprocaine with epinephrine. The epidural catheter was inserted into epidural space at T12-L4 and advanced toward head for about 4 cm. After correct epidural placement was confirmed,3% chloroprocaine 6 mg?kg-1 with or without 1:200 000 epinephrine was injected into epidural space over 2 min. Onset time of analgesia , motor blockade and degree of motor blockade at 20 min after epidural injection (Bromage scale 0 = no motor block, 3 = unable to flex hip, knee and ankle) were recorded. Blood samples were taken from radial artery before and at 3, 6, 9, 11, 13, 15, 17, 20, 30, 45, 60, 90 min after epidural injection for determination of plasma chloroprocaine concentration by HPLC. Results The two groups were comparable with respect to demographic data. There were no significant differences in the pharmokinetics and pharmacodynamics of chloroprocaine between the two groups. The Cmax was (0.491?0.47) mg?L-1 and (0.32?0.22) mg?L-1; The Tmax was (8?3) min and (9?4) min; the AUC was (10?6) ?g?min?ml-1 and (7?4) ?g?min?ml-1;the K was 0.32?0.21 min-1 and 0.36?0.32 min-1 in group C and CE respectively. Conclusion Epinephrine 1:200 000 does not affect the pharmacokinetics and pharmacodynamics of 3 % chloroprocaine for epidural block.

Purpose To observe the effect of paeonol on inflammatory factors after focal cerebral ischemiareperfusion injury in rats and its possible mechanism of paeonol.Methods Focal cerebral ischemic model was established by MCAO in rats.After 2 hours of cerebral ischemia and 22 hours of reperfusion,the expression of TNF-α and IL-1β in brain was determined by immunohistochemistry.The content of TNF-α and IL-1β in serum was measured by ELISA.Results Compared with model group,the levels of TNF-α and IL-1β in brain and serum in rats in the paeonol groups were obviously decreased.Conclusion Paeonol could relieve brain damage induced by focal cerebral ischemia-reperfusion,which may be related to inhibiting the production of inflammatory factors.

Objective To investigate the correlations between obsessive-compulsive symptoms and autism-spectrum quotient and explore if systemizing is the mediator between them. Methods With Padua Inventory (PZ) ,AQ and Systemizing Quotient (SQ), 1901 college students were assessed with stratified random sampling survey. Results Compared with the femal students, score of systemizing in male students had a significant increase ( (26. 773 ± 9.225 )vs (22.017 ± 7 ± 934), P ＜0.01 ). AQ total score was significantly positive correlated with PI total score( r=0. 336, P＜0.01 ),and correlation was also found among SQ,AQ and PI in different degrees. the mediating effect of systemizing was significant. The regression equation was AQ = 18. 942 +0. 064PI -0. 022SQ (P ＜ 0.01 ). Conclusion There are positive correlation between obsessive-compulsive symptoms and AQ in college students, and systemizing is the mediator between above symptoms.

Objective To evaluate the pharmacokinetic parameters and bioavailability of Cyclobuxine D in transdermal patch in Newzealand rabbits by determining concentration-time curve and by comparing with the pharmacokinetics of Cyclobuxine Dinjection and suspension.Methods Precolumn derivatization RP-HPLC was used to detect the concentration of Clovirobuxine D in rabbits plasma at different time,and software 3p87 was used to analyze the pharmacokinetics parameter.Results In contrast to oral delivery,relatively steadily sustained blood concentration with minimal fluctuation and prolonged peak time were presented in the rabbits over a long period after transdermal administration.The absolute bioavailability of Cyclobuxine D was 30.472 %.Conclusion Cyclobuxine D Patch exhibits good controlled-release properties and maintains appropriate blood concentration for a prolonged time.

Objective: To explore the role of clinical pharmacists in the treatment of artial fibrillation complicated with digoxin poisoning and the entry points of pharmaceutical care.Methods: Clinical pharmacists participated in the therapy for a patient with atrial fibrillation and digoxin poisoning, monitored the blood concentration of digoxin combined with the characteristics of the patient, analyzed the causes of digoxin poisoning in terms of the underlying diseases, renal function and combined medication, and performed beneficial pharmaceutical care.Results: The suggestions of clinical pharmacists were adopted by doctors, digoxin was withdrawn timely and the drug poisoning was corrected.

Objective To investigate the current situation of radiation protection capability and personal protection in the clinical practice of interventional radiology in grade-Ⅲ hospital.Methods A total of 108 medical staffs including physicians,technicians and nurses,who worked in interventional room,CT/ MR room,interventional catheterization room,radiotherapy department or radiology department in a grade-Ⅲ hospital of Guangdong province during the period from June 2014 to November 2015,were enrolled in this questionnaire investigation.The contents of self-mnade questionnaire included general demographic data,personal radiation exposure and protection.By using self-made questionnaire about the radiation protection capability of interventional work (both I-CVI and S-CVI being 0.9) the interventional radiation protection capability of the hospital was evaluated.Results In a certain grade-Ⅲ hospital of Guangdong province,the protection capability in shielding facilities,operating time and distance protection was quite strong,but the health-care leave system was lack,the occupational hazard detection was insufficient,and the protection and training system was poorly executed.In aspect of personal protection,the usage rate of lead apron in interventional procedures was only 72.2％,moreover,the rate of not wearing a radiation detector was up to 4.6％,and 9.3％ of medical staff didn't know the correct wearing position of a radiation detector.Conclusion The medical institution is lack of enough attention to the personal radiation protection as well as to the occupational health of interventional medical staff.In part of the medical staff,the consciousness of radiation protection is weak and the protection knowledge is insufficient,they are lack of adequate attention to occupational protection.All these issues need to be further improved.

Objective To detect and analyze the HBV?related indexes in the urine of HBV transgenic mice and further understand the biological characteristics of transgenic mice, and to clarify the tissue sources of HBV?related indexes. Methods HBV?related indexes in the urine of transgenic mice were tested using enzyme?linked immune sorbent assay ( ELISA ) and fluorescence quantitative PCR ( real?time RCR ) . The tissue sources were confirmed by several experiments, i. e. hydrodynamic transfection of mice, RNA interference to inhibit HBV?expression in the transgenic mice, and to infect normal mice with HBV?positive serum from patients. Results Expression of HBsAg, HBeAg and HBV?DNA was present in the urine of transgenic mice, of which the HBsAg expression level was high (6674 ± 619?8 IU/mL), but lower than that in the serum (16470 ± 2704 IU/mL). The level of HBsAg expression in the urine of male mice was higher than that in female mice. The level of HBeAg expression in the urine was lower and the HBeAg positive rate of urine was higher than that of blood, and the levels of HBeAg expression showed significant inter?individual and inter?sexual differences. HBV?DNA level reached 103 -105 copy/mL in the urine, but no related antibody expression was detected. The experiments such as hydrodynamic infection test indicated that the HBV?related indexes in the urine are derived from replication in the kidneys rather than secreted from the liver, entered into the blood circulation, and discharged from the urine. The kidneys are an independent expression site of HBV. Conclusions The expression of HBV?related indexes is present in the urine of transgenic mice and it is a long?term expression along with the age in months, of which the expression levels of HBsAg and HBV?DNA are rather high and stable. HBsAg titer in the urine of the male mice is higher than that of female mice. HBeAg expression level in the male mice is more stable compared with that in female mice. No expressions of various kinds of antibodies have been found in the urine. The kidneys are an independent expression site of HBV.
