1.Clinical efficacy on neurogenic bladder after spinal cord injury treated with rehabilitation training and acupuncture-moxibustion.

OBJECTIVETo compare the differences in the clinical efficacy on neurogenic bladder after spinal cord injury (SCD between the matrix needling technique combined with rehabilitation training and rehabilitation training in the patients.
METHODSSixty patients of SCI neurogenic bladder were randomized into an observation group (29 cases) and a control group (31 cases). In the control group, the conventional rehabilitation therapy, the intermittent catheterization and bladder function training were adopted, once every day, for 4 weeks totally. In the observation group, on the basic treatment of the rehabilitation as the control group, the matrix needling technique and acupuncture at sanyin points were applied. The treatment of 8 days made one session, at the interval of 2 days, continuously for 3 sessions, meaning 4 weeks totally. The bladder function score, residual urine volume and bladder capacity were compared before and after treatment in the two groups.
RESULTSThe bladder function score was reduced after treatment as compared with that before treatment in either group (both P<0. 05). The score in the observation group was lower than that in the control group after treatment (P<0. 05). No matter which spinal segment was injured, the residual urine volume after treatment was reduced as compared with that before treatment and the bladder capacity was increased (P<0. 05). The residual urine volume in the observation group was less than that in the control group after treatment and the bladder capacity was more increased (P< 0. 05).
CONCLUSIONThe conventional rehabilitation program in combination with the matrix needling technique and acupuncture at sanyin points achieve the significant efficacy on SCI neurogenic bladder.

2.Correlation between Aurora-A and breast carcinoma treatment efficacy of paclitaxel

Objective To explore the expression of Aurora-A in breast carcinoma and the relationship with breast carcinoma treatment efficacy of paclitaxel. Methods The expression of Aurora-A were tested by immunohistochemical SP method in 67 cases of breast carcinoma. The relationship of Aurora-A with treatment efficacy of paclitaxel was analysed. Results The expression rate of Aurora-A was 73.13%(49/67)in breast carcinoma. The treatment effective rate of breast carcinoma was 77.78%(14/18) and the rate of nonresponder was 11.11%(2/18) in negative expression of Aurora-A;but the treatment effective rate was 34.69%(17/49) and the rate of nonresponder was 34.69%(17/49) in positive expression of Aurora-A. There were obvious correlations between expression of Aurora-A and treatment efficacy of paclitaxel. Statistical significance could be found between them (P = 0.013 ). Conclusion There is manifest correlation between the high expression of Aurora-A and resistance of paclitaxel in breast carcinoma.
3.Single-step purification of recombinant human interleukin-6 employing DEAE-Seph-arose weak anionic-exchange chromatography

Abstract Objective: To prepare high-purification and high-specific activity of recombinant human interleukin-6 (rhIL-6). Methods: The rhIL-6 was obstained from inclusion body expressed by IPTG-induced pT7.7hIL-6 expressed vector using extracting,denature and refolding techniques. The rhTL-6 was further purified by anionic-exchange chromatography. Activity of rhIL-6 was measured by 3H-TdR method. Results: After a single-step purification,the product purity reach 95% and it's specific activity was 3.0 x 10~8 U/mg. Conclusion:This scheme of puri-fication was an easy way requiring rhTL-6.
4."Observations on the Efficacy of ""Three Yin Points"" Acupuncture plus Medicinal Spreading Moxibustion in Treating Erectile Dysfunction"

Objective To investigate the clinical efficacy of three yin points acupuncture plus medicinal spreading moxibustion in treating erectile dysfunction.Methods Sixty patients with erectile dysfunction meeting the inclusion criteria were randomly allocated to two groups, 30 cases each. The control group took sildenafil at 1 hr before sexual intercourse. The treatment group received three yin points acupuncture plus medicinal spreading moxibustion, 8 days as one course of treatment, for three consecutive courses with 2 days of rest between the courses. The erectile function was scored in the two groups before and after treatment. The clinical therapeutic effects were compared.Results The total efficacy rate was 93.3% in the treatment group and 80.0% in the control group; there was no statistically significant difference between the two groups (P>0.05). The marked efficacy rate was 83.3% in the treatmentn group and 60.0% in the control group; there was a statistically significant difference between the two groups (P<0.05). in the treatment group, there was a statistically significant pre-/post-treatment difference in the erectile function score after 20 days of treatment (P<0.05) and statistically a very significant pre-/post-treatment difference in the erectile function score after 30 days of treatment (P<0.01). In the control group, the overall therapeutic effect was stable after treatment and there was a statistically significant pre-/post-treatment difference in the erectile function score (P<0.05). There was no statistically significant post-treatment difference in the erectile function score between the treatment and control groups (P>0.05).Conclusions Both acupuncture-moxibustion and sildenafil have a therapeutic effect on erectiledysfunction. Sildenafil produces a rapid and stable effect. Acupuncture and moxibustion produce a slower effect but have an advantage in the marked efficacy rate. Acupuncture and moxibustion are safe, of fewer contraindications and suitable for most of the population.
5.Effect of LSD1 knock-down by small hairpin RNA on the apoptosis and cell cycle of human acute myelogenous leukemia cells

Objective To investigate the effect of down-regulation of lysine specific demethylase 1 (LSD1) by shRNA on the apoptosis and cell cycle of human acute myelogenous leukemia cells.Methods The lentiviral vector-mediated LSD1-shRNA was transfected into human acute promyelocytic leukemia HL-60 cells and acute monocytic leukemia SHI-1 cells.The expressions of LSD1 mRNA and protein were examined by real time quantitative PCR and Western blot,respectively.The flow cytometry was applied to detect the apoptosis and cell cycle distribution after AnnexinV-PE/7-AAD and PI dying,respectively.Results The expressions of LSD1 mRNA and protein in HL-60 and SHI-1 LSD1-shRNA group were significantly decreased compared with the blank control group and the negative shRNA group (P < 0.01,respectively).The apoptosis levels of HL-60 and SHI-1 cells were significantly increased after knockdown of LSD1 (P < 0.01).Moreover,the cell cycle distribution in the G0/G1 phases was also significantly increased(P < 0.01).Conclusion LSDI-shRNA promotes cell apoptosis and increases the percentage of cells in the G0/G1 phases of human acute myelogenous leukemia cells.
6.Hemorrheological Changes in Lacunar Cerebral Infarction Patients With Arteriopathy of Small Artery

【Objective】To observe the hemorrheological changes in lacunar cerebral infarction(LCI) patients with arteriopathy of middle and small arteries.【Methods】The hemorrheological changes of cerebral arteries in 85 LCI patients without arteriopathy of large artery confirmed by cervical vessels color ultrasonography and magnetic resonance image(MRI)were observed by transcranial Doppler ultrasonography(TCD).The correlation of the hemorrheological changes with age and blood pressure(BP) of arteries was investigated.The above indexes were compared with those in 25 healthy volunteers confirmed by encephalography in the same age group.【Results】The mean blood flow rates of cerebral middle,anterior and posterior artery in LCI patients were(62.09?16.90)cm?s~(-1),(50.42?13.11)(cm?s~(-1),)and(32.33?7.55)cm?s~(-1) respectively,lower than(69.65?19.20)cm?s~(-1),(57.75?16.75)cm?s~(-1),and(38.75?8.81)cm?s~(1) respectively in the healthy volunteers(P0.05).【Conclusion】The arteriopathy of small artery in LCI patients can cause the mild decrease of blood flow rate and the increase of pulsating index.There exists a correlation of pulsating index with age and arterial blood pressure.

Objective To construct a recombinant expression vector of human interleukin-24(hIL-24) gene and express it in E.coli M15,and to evaluate the bioactivity of IL-24 fusion protein.Methods The human IL-24 cDNA fragment was amplified from plasmid by polymerase chain reaction(PCR),and sequenced.PQE/hIL-24 was constructed by gene rearrangement,then it was transformed into E.coli M15.The expression of the target protein was induced with IPTG and purified by Ni2+-NTA agarose column.The expressed recombinant hIL-24(rhIL-24) was identified by SDS-PAGE and Western blotting.Normal peripheral blood mononuclear cells(PBMCs) were cultivated with the expression protein for 48 and 72 h,the levels of IL-6,IFN-? and TNF-? of PBMCs stimulated with rhIL-24 were detected by ELISA.Results The recombinant prokaryotic expression vector PQE/IL-24 was constructed successfully and expressed stably in E.coli M15.At about 18 400 of molecular weight,there was an induced protein band.The levels of IFN-?,IL-6 and TNF-? in the group of cultivated with the expression protein were obviously higher than those in the groups without the expresson protein(P

10.Evaluation on effect of rosuvastatin in treatment of lower extremity atherosclerotic disease in elderly

Objective To observe the clinical effect of rosuvastatin in the treatment of lower extremity atherosclerotic disease in the elderly,and to provide clinical basis of the application of statins in lower extremity atherosclerotic disease in the elderly.Methods 90 patients diagnosed as lower extremity atherosclerotic disease (aged 60 years or over)were randomly divided into simvastatin group and rosuvastatin group (n=45),and they were treated with simvastatin and rosuvastatin,respectively,based on the regular treatment.The total cholesterol (TC),low density lipoprotein-cholesterol (LDL-C), and high sensitivity C-reactive protein (hs-CRP)of the patients in two groups were detected at the begining of treatment and the ending of 32 weeks;at the same time,the lower limb artery intima thickness, strength of hardened, plaque, and artery stenosis degree scores were compared.Results 32 weeks after treatment,the levels of TC,LDL-C and hs-CRP of the patients in two groups were decreased statistically compared with before treatment (P <0.05);the levels of TC,LDC-C,and hs-CRP in rosuvastatin group were lower than those in simvastatin group (P < 0.05 ). 32 weeks after treatment in two groups,compared with before treatment,the lining thickness and plaque,stenosis scores were decreased (P <0.05);the lining thickness,plaque and stenosis scores in rosuvastatin group were lower than those in simvastatin group (P <0.05);the gardening strength grades had no statistical difference between before and after treatment. Conclusion Statins is lipid-lowering and resistant to lower limb arteriosclerosis,and rosuvastatin is better than simvastatin.
