1.Effect of Combination of Epigallocatechin-3-gallate and Cisplatin on Proliferation and Apoptosis of SW620

Objective To investigate the effect of different composed combination of EGCG and cisplatin on human colon adenocarcinoma cell line SW620 and the related mechanisms. Methods SW620 cells at exponential phase were treated with cisplatin(5 mmol.L-1)and EGCG(0, 25, 50 and 100 mmol.L-1).Seventy hours after the cultivation, cell proliferation was detected with MTT;the percentage of apoptotic cells was determined by flow cytometry;expression levels of JAK2, p-JAK2, p-STAT3, STAT3, Bax, Bcl-2, cleavage-PARP, and cleavage-caspase3 were detected by Western blotting. Results The SW620 cell proliferation rate was(95±5)%,(67±5)%,(42±5)%(22±4)% and the apoptotic rat was 5%, 15%, 35%, 57%in the 0, 25,50, 100 mmol.L-1 EGCG groups, respectively.Moreover, EGCG up-regulated the expression of Bax, cleavage-PARP and cleavage-caspase3(P<0.05)and down-regulated the expression of p-JAK2, p-STAT3 and Bcl-2(P<0.05)in a dose-dependent manner, but had no influence on the expression of JAK2 and STAT3(P>0.05). Conclusion EGCG can promote cisplatin-induced apoptosis and proliferation inhibition of human colon adenocarcinoma cell lines of SW620 by suppressing JAK2/STAT3 gignaling pathway.

Epithelial-mesenchymal transition (EMT) is the basis of biological process of embryonic development,and is also closely related with tumor cell in situ invasion and distant metastasis.Recent studies find that EMT and cancer stem cells (CSCs) have a close relationship.CSCs have a strong ability to selfrenewal,tumorigenicity and cell differentiation potential.Identifing the markers,in-depth study of CSCs resistance,may provide a new path for cancer treatment.
3.Determination of Saponins Content in Compound Danshen Prescription by HPLC

Objective To develop a method for the determination of four saponins of panax notoginseng in Compound Dan-shen Prescription (CDP) by HPLC. Methods The conditions for the experiment were: determination of notoginsenoside R1, ginsenoside Rg1 and ginsenoside Re with acetonitrile - 0. 05 % phosphoric acid (19 : 81) as mobile phase and the detection wave at 203nm, determination of ginsenoside Rb1 with acetonitrile - water (33 : 67) as mobile phase and the detection wave at 203nm. Results A good linearity for notoginsenoside R1 was in the range of 0. 24 ~ 3. 6?g ( r = 0. 9997), 1. 2 ~ 18?g (r = 0. 9994) for ginsenoside Rg1, 0. 08 ~ 1. 2?g(r = 0. 9997) for ginsenoside Re and 0. 25-3. 72?g (r = 0. 9998) for ginsenoside Rb1. The stability of the four saponins is well kept at 4℃ for four months. Conclusion This method is simple, rapid, accurate and with good reproducibility and can be used for the quality control of CDP.
4.Experimental Study on Effect of Huangqi Danshen Drop Pill on Pituitary - Induced Acute Myocardium Ischemia

Objective To observe the effect of Huangqi Danshen Drop Pill(HDDP) on pituitary (Pit) induced acute myocardium ischemia. Methods Acute myocardium ischemia rat model was established by sublingual venous injection of Pit(0. 35U/kg). The changes of the T wave ( II channel ECG) and serum contents of lactate dehydrogenase (LDH), CK, superoxide dismutase (SOD) and malondialdehyde (MDA) were observed in various times after Pit injection. Results HDDP could significantly lower the height of T waves, increase SOD activity and decrease MDA content. Conclusion HDDP can improve acute myocardium ischemia induced by Pit.
5.Pharmacokinetics and pharmacodynamks of chloroprocaine with or without epinephrine for epidural blockade

Objective To determine the pharmacokinetics and pharmacodynamics of chloroprocaine with or without epinephrine for epidural blockade. Methods Twenty ASA physical status Ⅰ or Ⅱ patients of both sexes aged 37-55 yrs weighing 45-80 kg undergoing elective lower abdominal surgery were randomly divided into 2 groups (n = 10 each): group C chloroprocaine without epinephrine and group CE chloroprocaine with epinephrine. The epidural catheter was inserted into epidural space at T12-L4 and advanced toward head for about 4 cm. After correct epidural placement was confirmed,3% chloroprocaine 6 mg?kg-1 with or without 1:200 000 epinephrine was injected into epidural space over 2 min. Onset time of analgesia , motor blockade and degree of motor blockade at 20 min after epidural injection (Bromage scale 0 = no motor block, 3 = unable to flex hip, knee and ankle) were recorded. Blood samples were taken from radial artery before and at 3, 6, 9, 11, 13, 15, 17, 20, 30, 45, 60, 90 min after epidural injection for determination of plasma chloroprocaine concentration by HPLC. Results The two groups were comparable with respect to demographic data. There were no significant differences in the pharmokinetics and pharmacodynamics of chloroprocaine between the two groups. The Cmax was (0.491?0.47) mg?L-1 and (0.32?0.22) mg?L-1; The Tmax was (8?3) min and (9?4) min; the AUC was (10?6) ?g?min?ml-1 and (7?4) ?g?min?ml-1;the K was 0.32?0.21 min-1 and 0.36?0.32 min-1 in group C and CE respectively. Conclusion Epinephrine 1:200 000 does not affect the pharmacokinetics and pharmacodynamics of 3 % chloroprocaine for epidural block.
6.Liraglutide ameliorates liver injury of type 2 diabetic mice via micro-RNA-33-AMPK pathway

AIM:To observe the effects of liraglutide on the level of microRNA-33 (miR-33) and the expres-sion of AMP-activated protein kinase ( AMPK ) and apoptosis-related proteins in mice with type 2 diabetes mellitus (T2DM), and to explore its possible mechanism .METHODS:High-fat diet and intraperitoneal injection of streptozocin were used to establish the type 2 diabetic model in C57BL/6 mice.The mice were randomly divided into 4 groups ( n=15 ):in control group , the normal mice were subcutaneously injected with equivalent volume of saline ;in model group , the T2DM mice were subcutaneously injected with equivalent volume of saline ; in low-and high-dose liraglutide treatment groups, the T2DM mice were subcutaneously injected with 100 and 200 μg? kg -1? d-1, respectively.After 4 weeks of administration, the levels of FBG, TG, TC, HDL-C, LDL-C, ALT and AST were determined.HE staining was used to ob-serve the pathological changes of the liver tissues .The protein level of cleaved caspase-3 in the liver tissue was detected by the technique of immunofluorescence .The protein levels of p-AMPK/AMPK and apoptosis-related proteins were detected by Western blot .The expression of miR-33 in the liver tissues was detected by real-time PCR.RESULTS: Compared with model group, the contents of FBG, TG, TC, LDL-C, ALT and AST were decreased significantly , while the content of HDL-C was increased significantly in low-dose liraglutide group and high-dose liraglutide group ( P<0.05 ) .The protein levels of phosphorylated AMPK and Bcl-2 were up-regulated significantly , and the expression of cleaved caspase-3 was down-regulated significantly (P<0.05).The level of miR-33 was decreased significantly (P<0.01).CONCLUSION:Liraglutide alleviates liver injury in type 2 diabetic mice , and the mechanism may be associated with reducing the level of miR-33 and increasing the phosphorylation of AMPK in the liver tissues , thereby inhibiting hepatocyte apoptosis .

9.Effects of Different Processing Methods on the Antiosteoporotic Effective Compositions of Psoralea corylifolia L.

This study aimed at evaluating the effect of different processing methods on the contents of antiosteoporotic compositions in P.corylifolia.HPLC method adopted to analyze the contents of antiosteoporotic compositions,including coumarin (psoralen and isopsoralen),flavonoids (neobavaisoflavone,corylifolin,corylifolinin and corylifol A) and meroterpenes (bakuchiol) in P.corylifolia.The contents were analyzed before and after the processing methods.Results:The results showed that the content of coumarin,flavonoids and meroterpenes obviously changed in different processed P.Corylifolia in comparison with the crude drugs.In conclusion,it was demonstrated that the therapeutic efficacy of osteoporosis was enhanced by CP method which increased all of the three compositions.The testimony tallied with the Chinese medical theory:stir-frying with salt-water could enhance the effect of the tonification of spleen and kidney.
10.Experimental Study on Liver Regeneration Following Portal Branch Ligation in Rats

Objective To study liver regeneration of the non-ligated liver lobes following portal branch ligation (PBL). Methods Sixty male Wistar rats were randomly divided into PBL group and sham operation (SO) group. Under ether anesthesia, the rats were subjected to PBL and sham operation, respectively. The animals were sacrificed on the 1st, 2nd, 3rd, 7th and 14th day respectively. The blood sample was collected from heart and the livers were harvested to determine serum alanine aminotransferase (ALT) levels and total liver weight, respectively. The hepatic histopathology was studied through light microscopy. The number of liver cell nuclear mitosis index was counted. The number of proliferative cell nuclear antigen (PCNA) index was counted by immunohistochemistry. The hepatic ultrastructural changes were studied under electron microscope. Results Elevated serum ALT level was observed in the first postoperative day in PBL group compared with SO group (P
