AIM: To construct the liver-specific transgene vectors encoding wild-type as well as most common disease mutant STBXATP7B cDNA under the control of mouse albumin promoter and to explore their expression. METHODS: Two Kbpala/Alb-STBXATP7B mutants containing the Arg778Leu and His1069Gln mutations were constructed using site-directed mutagenesis system plus site-subcloning technique. The vectors expressed wild-type and mutants of human STBXATP7B in mouse liver cells were obtained and transiently transfected into BRL and BHK cell lines. Western blotting analysis was utilized to detect the expression of human STBXATP7B. RESULTS: Enzyme analysis and sequencing analysis confirmed that the target genes were STBXATP7B and in right position. The results of Western blotting showed that the gene products were expressed specifically in liver cells. CONCLUSION: The Kbpala/Alb-STBXATP7B vectors were constructed successfully and the liver-specific expression of human STBXATP7B proteins were conformed.

Objective 　To screen for gene mutation of exon 18 in Chinese patients with Wilson disease. Methods　PCR-SSCP was used to screen exon 18 in 45 Wilson disease patients among 39 Chinese families and 10 normal controls. Those with abnormality were further analyzed by necleotide sequence analysis. Results　There were 16 mobility shift with two different styles in exon 18. All abnormal mobility shifts were sequence analysed. No gene mutation was found. Conclusions　Our result suggest that, contrary to findings in Caucasians, exon 18 is not a frequent mutation point in Chinese patients with Wilson disease.

Objective To explore the correlation between blood glucose and postoperative recovery of pancreatic cancer patients with diabetes.Methods According to the level of glycemic control after operation,68 cases of pancreatic cancer patients with diabetes were divided into two groups,including 34 cases of intensive control group (blood glucose 4.4 ～ 6.1 mol/L),34 cases of control group (blood glucose 6.1 ～11.1 mol/L).Fasting blood glucose (FBG),fasting insulin (FINS) and C reaction protein (CRP) in all patients were detected at 1st,3rd and 7th day after operation,and the recovery condition,postoperative complications and other clinical data were compared and analyzed.Results FBG,FINS and CRP in intensive control group were (6.94 ± 0.94) mmol/L,(17.38 ± 7.37) mmol/L,(108.33 ± 37.25) mg/L,and in control group were (7.81 ± 1.36) mmol/L,(23.73 ± 8.25) mmol/L,(131.51 ± 42.34) mg/L at 3rd day after operation.There were significant difference in serum FBG,FINS and CRP level between the two groups at 3rd day after operation (P ＜ 0.05).Compared with control group,postoperative duration of fever [(1.4 ± 0.8) vs (2.5 ± 1.1) d],duration of antibiotics [(3.1 ± 0.7) vs (4.6 ± 0.8) d] and time of anal exhaust [(2.5 ±0.5) vs 93.7 ± 0.8)d] in intensive group were significantly reduced (P ＜ 0.05).The wound infection rate in intensive control group was significantly decreased (5.9％ vs 23.5％,P ＜0.05),the rate of mortality and other postoperative complications between the two groups was not statistically significant (P＞0.05).Conclusions Intensive glycemic control can contribute to improve insulin resistance,reduce inflammation reaction and improve the prognosis of postoperative pancreatic cancer patients with diabetes,and it does not increase the postoperative complication rate.

Purpose:To investigate the expression of PCNA、p53、C erbB 2 in colorectal carcinoma and its relationship with host immune reactions. Methods:The expression of PCNA、p53、C erbB 2 was detected by Envision immunohistochemical method in 42 colorectal carcinoma, 35 pericarcinomatous and 10 normal coloerctal mucosa respectively. Host immune reactions (pericarcinomatous fibrosis, mononuclear infiltration and lymph nodes reaction) were also observed. Results:(1) The positive expression of PCNA、p53、C erbB 2 in colorectal carcinoma was significantly higher than in corresponding pericarcinomatous mucosa and normal colorectal mucosa respectively( P 0.05). Objective To investigate the change and role of nitric oxide (NO), nitric oxide synthase (NOS) in serum of children with Epilepsy.Methods The serum NO and NOS levels of 58 epileptic children and 23 healthy children were measured with enzyme linked immunosorbent assay (ELISA). Objective To study the gene mutation and clinical characteristic of hereditary spinocerebellar ataxia type 7 (SCA7).Methods The SCA7 (CAG) trinucleotide repeat mutations were detected by polymerase chain reaction(PCR) and polyacrylamide gel electrophoresis technique in 24 patients with autosomal dominant SCA from 15 families, 20 sporadic SCA patients and 41 normal persons from the same family and 30 healthy persons from different family,the abnormal allele fragments were sequenced by ABI 373 DNA sequencing machine.Results 24 patients with SCA had CAG repeat numbers of SCA 7 allele from 9~18.Normal alleles of SCA 7 had CAG repeat number from 9 to 19. One sporadic SCA patient had one abnormal SCA 7 allele with the CAG repeat expanded to 63 repeats, being confirmed by DNA sequencing.Conclusion CAG expansions were pathogenic cause of SCA 7. The technique of gene mutation detection could provide an effective way for the prediction of asymptomatic and genetic counseling,which was a basis for gene typing.

[Objective] To discuss the cure effect of acupoints application combined with TCM decoctions and auricular point buried-bean on pernicious vomiting. [Method] Choose 65 such cases from April 2011 to March 2012 as control group and another 65 from Nov. 2011 to Sep. 2013 as treatment group. The control one take oral y TCM decoction combined with auricular point buried-bean, the treatment one added with TCM acupoints applica-tion;observe the result after 1 course. [Result] In treatment group, 46 cases were cured, 18 better, 1 was not cured, the total effective rate 98.5%;in con-trol one, they were respectively 40,11,14 and 78.4%; the comparison of them had difference of statistical meaning.[Conclusion] The acupoint application combined with TCM decoction and auricular point buried-bean has obvious cure effect on pernicious vomiting.

Objective To explore the effects of acute hypoxia on the expression of neuropeptide Y (NPY) mRNA in the rat hypothalamus. Methods A total of 78 Wistar rats were randomly divided into 13 groups ( n =6 in each group): control group and 12 experimental groups. Rats in the experimental groups were exposed to hypoxia in the hypobaric chamber simulating the altitudes of 3 000, 4 000, 5 000, and 6 000 m for 1, 2, and 3 d, respectively. The expression of NPY mRNA in the rat hypothalamus under the condition of acute hypoxia was detected by semi quantitative reverse transcription polymerase chain reaction (RT PCR). Results ① Expression of NPY mRNA was detected in the rat hypothalamus in the experimental groups and the control group. ② Under the condition of acute hypoxia for the same time, the expression of NPY mRNA in the rat hypothalamus showed the tendency of decrease at increasing altitudes; ③ At the same altitude, expression of NPY mRNA in the rat hypothalamus decreased gradually with the increasing time for acute hypoxia. Conclusion Acute hypoxia can decrease the expression of NPY mRNA in the rat hypothalamus.

Objective To investigate the clinic diagnosis value of CT scan and the determination of serum NSE, Cyfra21-1 for SCLC. Methods ① The CT findings and pathologic basis of 220 cases with SCLC were analyzed retrospectively.② Between February 2000 and December 2001, 120 consecutive patients with pulmonary lesions by CT scan, which suspiciously suffered lung cancer, were studied prospectively. Serum samples were obtained by venous puncture from all patients 1 week prior to surgery or puncture biopsy. The serum concentration of NSE, Cyfra21-1 were determinated using ELISA. According to the pathologic result, the diagnostic value of CT scan, NSE and Cyfra21-1 were evaluated.Results ① Most CT findings of SCLC were central and spindly or eggplant-shape mass, hilar and/or mediastinal lymph nodes enlargement.The obstructive pulmonary lesions were found rarely.② Using CT scan, NSE and Cyfra21-1 alone, the sensitivity were 75%,72.5% and 32.5%,the specificity were 73.75%, 83.75% and 48.75%,the accuracy were 74.16%, 81.67% and 43.33% respectively. With combination of CT scan and NSE, the diagnostic accuracy was 78.3%, and the sensitivity was 87.5%. However, CT scan combined with Cyfra21-1 can not be improve the diagnostic value. Conclusion SCLC has specific CT findings. The sensitivity of CT scan was 75%, combinated with NSE, its accuracy was higher(78.3%), and the sensitivity was 87.5%.
