Guidelines of Prevention and treatment of Children's Influenza A (H1N1) with TCM includes four parts of prevention, diagnosis, clinic treatment and nursing. The drug prophylaxis stresses on administration by identified body without diseases, which includes normal constitution, qi-deficiency constitution, inner-heat constitution and phlegm-damp constitution, children of different constitution are given different appropriate drugs. The treatment bases on syndrome differentiation, such as mild syndrome, severe syndrome, crises and convalescence. The syndrome-classification includes wind-heat invading the exterior, dampness obstructing defensive qi, the heat-toxin blocking the lung, exuberant toxin in both qi and ying, deficiency of heart-yang, the evil sinking into jueyin, residual evil remaining in the body and the deficiency of both qi and yin. The nursing emphasizes the isolation, keeping body warm, bland diet and nursing during convalescence.

Objective To investigate the effects of HINT1 on the growth of and apoptosis in malignant melanoma in nude mouse models established by subcutaneous xenotransplantation of human melanoma A375 cells. Methods Three groups of nude mice were subcutaneously inoculated with A375 cells transfected with pcDNA 3.1/myc-His (-) A-HINT1 (HINT1-A375), A375 cells transfected with pcDNA 3.1/myc-His (-) A empty vector (neo-A375), and untransfected A375 cells, respectively. Then, the growth of transplanted tumors was observed and tumor formation rate was calculated. Thirty-three days after the inoculation, mice were killed and tumor tissue was obtained followed by the examination of tumor weight and volume. Histopathology was performed to observe the morphological features of tumor cells and in situ end labeling technique (TUNEL)was carried out to assess the apoptosis in transplanted tumor cells. Results The tumor formation rate was consistently 100% in the three groups. The transplanted tumor in HINT1 group grew more slowly than that in the other two groups, and significant difference was observed as early as day 18 (P < 0.01 ). Lower tumor weight and volume were noted in the HINT1 group compared with the neo group and A375 cell group (0.04 ±0.00 g vs. 0.23 ± 0.00 g and 0.29 ± 0.03 g, 0.06 ± 0.04 cm3 vs. 0.34 ± 0.15 cm3 and 0.43 ± 0.19 cm3 respectively, all P < 0.01 ). Histopathology revealed smaller tumor nests, slight atypia of tumor cells with no obvious pathologic mitoses or necrosis in HINT1 group in comparison with the other two groups. Immunohistochemistry and TUNEL revealed that the percentage of apoptotic cells in HINT1 group was statistically higher than that in the neo group and A375 cell group (12.87% ± 1.18% vs. 3.22% ± 0.49% and 3.00% ± 0.53%, both P <0.01 ). Conclusion High expression of HINTl could inhibit the growth of and promote the apoptosis in malignant melanoma in nude mice subcutaneous xenotransplantation models, suggesting that HINT1 gene might be responsible for tumor suppression in human malignant melanoma.

Objective To investigate the effects of chemokine CCL18 on the proliferation and invasion of a human melanoma cell line A375. Methods Human peripheral blood monocytes were isolated from healthy volunteers, cultured in vitro and divided into two groups to be induced by IL-4 for 48 hours or remain untreated. A375 cells were classified into 3 groups to be cultured with IL-4-induced monocytes, untreated monocytes or CCL18 of 200 g/L for various durations. A375 cells receiving no treatment served as the control.MTT assay was performed to detect the proliferation of A375 cells, chemotaxis test and Matrigel-transwell assay to evaluate the chemotaxis and invasion ability of A375 cells, and chicken chorioalllantoic memebrane (CAM)was used to detect the effect of CCL18 on tumor angiogenesis. Results The proliferation of A375 cells was statistically accelerated by IL-4-induced monocytes and untreated monocytes, but unaffected by CCL18.Matrigel-transwell assay revealed that IL-4-induced monocytes and CCL18 promoted the chemotaxis and invasion ability of A375 cells (all P<0.05). Tumor angiogenesis was also increased by IL-4-induced monocytes and CCL18 (both P < 0.05). Conclusion The chemokine CCL18 can promote the invasion ability of A375 cells and tumor angiogenesis.

Objective To explore the prognostic value of N-terminal pro-brain natriuretic peptide ( NT-pro-BNP) levels in critically ill infants. Methods Eighty-one critically ill infants were enrolled from January 2013 to January 2014 in pediatric intensive care unit. The minimum of pediatric critical illness score ( PCIS) and the number of dysfunction organs were calculated within 24 hour after admission. According to PCIS,the critically ill infants were divided into extremely critical group(PCIS≤70,n=25),critical group (PCIS 71-80,n=30)and non-critical group(PCIS>80,n=26). According to the prognosis, the critically ill infants were divided into survival group (n=68)and death group(n=13). The serum NT-pro-BNP levels were determined on the first day, third day and convalescent phase. The relationships of serum NT-pro-BNP levels with PCIS and the number of dysfunction organs and prognosis were observed. Results The study showed statistical significances of serum NT-pro-BNP levels among the extremely critical group, critical group and non-critical group, whether on the first day,or on the third day and convalescent phase(P<0. 01). There were statistical significances of serum NT-pro-BNP levels among different stages of the disease in each group(P<0. 01). Compared with survival group,PCIS was significantly lower and the serum NT-pro-BNP levels and the number of dysfunction organs were significantly higher in death group. The serum NT-pro-BNP level on the third day was higher than that on the first day in death group ( P<0. 01 ) , while no significant difference was found in survival group. The serum NT-pro-BNP levels on the first day and the third day and PCIS were negatively correlated(r= -0. 59,P<0. 01;r= -0. 66,P<0. 01). The serum NT-pro-BNP levels on the first day and the third day and the number of dysfunction organs were positively correlated(r=0. 40,P<0. 05;r=0. 57,P<0. 01). Conclusion The serum NT-pro-BNP levels of the critically ill infants are correlated with disease severity, and can be useful for assessing the severity of critical illness.
