[Objective]To explore the change and its significance of the expression of vascular endothelial growth factor(VEGF) messenger RNA and bone morphogenic protein-2(BMP-2) of the local femoral head in the nontraumatic osteonecrosis of femoral head(NONFH).[Method]Sixteen samples of femoral heads of NONFH were collected as the experimental group and fresh 10 samples of femoral heads of femoral neck fracture as control group, overall examples were collected from total articular replacement arthroplasty. The samples were splitted in coronal plane and get one bone block from necrosis area and another one from healthy area, make them into microtome section after the process of immobility and decalcification.Their pathological change was observed by using optical microscope and electron microscope and detect the expression of BMP-2 and VEGFmRNA in femoral head through making use of immunohistochemistry and in-situ hybridization technique.[Result]The organization structure of experimental group was disorganized, cracked and the bone trabecula was rarefactive and non-intact and there was a great number of empty lacuna in bone trabecula. While there was the reverse situation in the control group. The intensity and area of positive expression of BMP-2 and VEGFmRNA in femoral head of the experimental group were lower than that of control group obviously. The result showed statistical significance(P

Serum metalloproteinase matrix (M MP )-9, tissue inhibitor of metalloproteinase (TIMP)-1 and type Ⅳ collagen level s were measured in patients with diabetic nephropathy (DN). The severity of DN w as determined by urinary albumin excretion rate. The results show that signific ant increases of serum TIMP-1 and type Ⅳ collagen depend on severity of DN, wh ile MMP-9 tends to decline. TIMP-1 and MMP-9 seem to participate in the patha genesis of DN.

Chinese Journal of Cancer Biotherapy 1994;0(01):-
Abnormal elevation and abnormal isoenzyme of non-specific esterase (?-NAE) are intimately associated with the genesis and development of cancer.Anti-GCBAP is a new kind of biological response modifier(BRM) which is separated from gastric cancer cells immunized animal spleen. As determined by clonogenic assay. Anti-GCBAP displayed highly-potent, specific cytotoxicity regarding to target cancer cells. In the present study, we have further investigated the effect of Anti-GCBAP on the ?-NAE isoenzymes in gastric cancer-bearing nude mice. The results demonstrated that in vivo adminstration of Anti-GCBAP can not only inhibit tumor growth in tumor-bearing mice, compared with control(p

Hoechst 33342 is a kind of fluorescent dye used for evaluating the cell cycle and apoptosis in a variety of benign and malignant cell lines, such as BC3H-1 monocytes,HI-60 cells,HT-144 melanoma cells and hepatoma cells. The inhibitor of topoisomerase I,the alternation of TATA box binding protein.the intraeellular accumulation of E2F-l protein and the mitochondrial dysfunction may play an important roles in the apoptosis pathway indueed bv Hoechst 33342.

Objective To detect the expression of vascular endothelial growth factor(VEGF)-C and VEGF-D in normal gastric tissue and gastric carcinoma tissue, and further investigate the relationship between VEGF-C,VEGF-D and lymph vessel density (LVD),lymph node metastasis. Methods The expression of VEGF-C and VEGF-D in gastric carcinoma tissues were examined by using SP immunohistochemical method in 30 normal gastric tissue and 75 gastric carcinoma tissue respectively. The lymph vessel were marked by LYVE-1 and calculated LVD.Results The rate of positive expression of VEGF-C and VEGF-D in gastric carcinoma tissue was significantly higher than that in normal gastric tissue, there was significant difference [73.3 % (55/75) vs.33.3 % (10/30),66.7 % (50/75) vs.30.0 % (9/30),P <0.01].The level of LVD in gastric carcinoma tissue with lymph node metastasis (40 cases) was significantly higher than that in without lymph node metastasis (35 cases)[(8.85 ± 1.24)/HP vs.(4.75 ± 1.04)/HP](P =0.000).The level of LVD increased when the expression level of VEGF-C and VEGF-D gradually went up.Conclusions The level of LVD increases in gastric carcinoma tissue.VEGF-C and VEGF-D are two significant factors for LVD and lymph node metastasis.

Objective To analyze the expression profile of specific miRNA of gastric cancer tissues. Methods miRNA were isolated from five paired gastric cancer tissues and corresponding paracancerous normal tissues. To identify the profile of gastric cancer,miRNA hybridization and cluster analysis were processed by significance analysis of microarrays (SAM,version 2.1) and cluster software. The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b were validated by fluorescence Real-Time PCR using taqman probe.Further hsa-miR-30c taget gene was predictived by online software. Results Revealed by miRNA microarray detection, there were 16 miRNA with differential expression in gastric cancer tissues and para-cancerous normal tissues, among which HS_100,hsa-miR-27a,hsa-miR-214*,solexa-555-1991 and hsa-miR-196 were significantly up-regulated,and hsa-miR-502-3p,hsa-miR-29c*,hsa-miR-64,hsa-miR-193b,hsa-miR-551b,hsa-miR-30c,hsa-miR-582-5p,hsa-miR-625*,hsa-miR-660,hsa-miR-363 and hsa-miR-486-5p were down-regulation.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b was in high concordance with microarray. Conclusion There is specific miRNA expression profile in gastric cancer tissues, among which differentially-expressed miRNAs may be involved in the regulation of gastric cancer progress and serve as new potential biomarkers.

Objective Using ?-cyanoacrylate to establish the orthotopic transplantation tumor model from the subcutaneous model of human hepatocellular carcinomas(HCC)in nude mice. Methods The subcutaneous animal tumor model was established by transplanting BEL-7402 cell into the subcutaneous of nude mice. Then the orthotopic transplantation tumor model was established by injecting the subcutaneous transplanted tumor into liver(indirect orthotopic model). Results Using ?-cyanoacrylate to establish the indirect orthotopic model successfully and the survival rates of the transplantation tumor were 95.8 %. Conclusion It was compared with the traditional methods that the indirect orthotopic transplantation tumor model by using ?-cyanoacrylate to establish is an easy method with high successful rate and easy to spread.
