Objective: To study the inhibition and mechanism of tea polyphenols (TP) on apoptosis induced by oxidized low density lipoprotein (ox-LDL) in human umbilical vein endothelial cells (HUVEC). Method: HUVEC cells were administered with TP (25 ?g/ml), ox-LDL (200 ?g/ml)+TP (25 ?g/ml), ox-LDL (200 ?g/ml) respectively, cells with equal volume of solvent as control. Cell viability was determined by MTT assay, apoptosis by acridine orange fluorescence dyeing, and expressions of Bcl-2、Bax and caspase-3 were analyzed by Western blotting. Results: Ox-LDL could inhibit HUVEC cell proliferation. Objective To investigate the influence of acupuncture therapy on neural stem cell (NCS) proliferation and differentiation in the hippocampus of the senescence-accelerated prone-8 (SAP8) mouse. Methods Twenty-four SAP8 mice were randomly and equally divided into a model group and an acupuncture group. Twelve senescence-accelerated resistant ( SAR1 ) mice served as the control group. Acupuncture was administered at the Baihui (DU20) point to mice in the acupuncture group once daily for 21 consecutive days. Bromodeoxyuridine (BrdU) was used to detect the proliferation and differentiation of NCSs in the hippocampus through double-labeled immunofluoresSAR1 group, the positive cells in the SAP8 group were significantly fewer Compared to the SAP8 group, positive cells markers of neuron or glia cell positive cells appeared. Compared to the SAR1 group, the expression of BrdU/GFAP increased in the SAP8 group, and decreased after acupuncture but not significantly. BrdU/S-100 β cells decreased significantly in the SAP8 group, and increased after acupuncture, but again not significantly. Compared to the SAR1 group BrdU/GalC positive cells increased significantly in the SAP8 group and decreased significantly after acupuncture.Conclusions After acupuncture treatment for 21 days, the differentiation of hippocampal NSCs into oligodendrocytes was inhibited, but there was little effect on their differentiating into neurons and immature astrocytes.
3.Effect of systemic lupus erythematosus on right heart function evaluated by parameters recommended by the American Society of Echocardiography

Objective To evaluate the right heart dysfunction in SLE patients with/ without pulmonary arterial hypertension (PAH) using the parameters recommended by the American Society of Echocardiography (ASE),and to examine whether the right heart dysfunction is directly related to elevated pulmonary arterial systolic pressure(PASP) and pulmonary vascular resistance(PVR).Methods Study population composited of 43 patients with SLE.The patients were divided into two groups according to the PASP measured by echocardiography:Group A was 24 patients with PASP ≤35 mm Hg,Group B was 19patients with PASP>35 mm Hg.Twenty-two healthy subjects with age and gender matched were set as control group.Routine transthoracic echocardiography study was performed on all patients and 26 parameters were measured in order to compare the differences of the parameters among the three groups.Results There were significant differences in 17 parameters among the three groups (P <0.05).Tricuspid annular plane systolic excurtion(TAPSE) and E/A ratio of tricuspid flow velocities in SLE patients without PAH significantly decreased compared to control group (P =0.04 and 0.03).There were significant differences in 11 parameters between SLE with and without PAH group (P <0.05).Multivariate logistic regression analysis indicated that after adjustment for age and gender,the patients with elevated PVR associated with a 6.18-fold increase in right ventricular dysfunction compared to the patients with normal PVR (P =0.02).Conclusions The impairment of right ventricular function in SLE patients was directly related to PVR and PASP.Elevation of PVR was an important predictor for right heart dysfunction.
4.Influence of acupuncture and moxibustion on proliferation and differentiation of neural stem cells in olfactory bulb of senescence-accelerated mouse P8.

This study was carried out to defect the effects of acupuncture and moxibustion therapy on the proliferation and differentiation of neural stem cells in the olfactory bulb of senescence-accelerated mouse P8. Immunofluorescence double staining methods(BrdU/Nestin, BrdU/MAP-2 and BrdU/beta-tubulin III) were used. The results show that both acupuncture and moxibustion could improve proliferation of neural stem cells, but only moxibustion could induce differentiation of neural stem cells into neuron.

5.Prediction model of platelet transfusion refractoriness in patients with hematological disorders

【Objective】 To explore the risk factors for platelet transfusion refractoriness(PTR)in patients with hematological disorders, construct a prediction model and validate the model efficacy. 【Methods】 Patients with hematological disorders who received platelet transfusion therapy in the Chengdu Second People′s Hospital from December 2021 to December 2022 were retrospectively included to judge the effectiveness of platelet transfusion and screened for risk factors by univariate and multivariate logistic regression. A prediction model for PTR was constructed using receiver operating characteristic(ROC) curve, calibration curve and decision curve(DCA) to assess the differentiation, calibration and clinical value of the model, respectively. 【Results】 A total of 334 hematological patients were included, including 168 males and 176 females, with a PTR incidence of 40.4%. Univariate and multivariate logistic regression analysis showed that platelet transfusion volume, erythrocyte transfusion volume, and neutrophil ratio were risk factors for PTR(P<0.05). A prediction model for PTR in hematological patients was established based on these risk factors. The area under the model′s curve was 0.8377(95% CI: 0.723-0.772), the sensitivity was 58.52%, and the specificity was 89.95%. The calibration curve showed that the S∶P was 0.964, the maximum absolute difference Emax was 0.032, and the average absolute difference Eavg was 0.009. The DCA analysis showed that the model had clinical application value when the risk threshold ranged from 0.2 to 0.9. 【Conclusion】 The PTR prediction model based on platelet transfusion volume, erythrocyte transfusion volume and neutrophil ratio can provide a basis for effective platelet transfusion in hematological patients.
