1.INHIBITORY EFFECT OF BOANMYCIN ON THE GROWTH OF COLON CARCINOMA 26 AND HEPATIC METASTASIS IN MICE

AIM To investigate the therapeutic effect of boanmycin (BAM, bleomycin A6) on colon carcinoma 26 and its hepatic metastasis in mice. METHODS A series of models including subcutaneous transplantation, orthotopic transplantation in cecum subserosa, intra-hepatic transplantation of tumor, and intra-splenic transplantation of tumor accompanied with hepatic metastases were employed. In addition, LEICA Q 500IW image analysis system was used to determine the area of metastatic lesions in the liver in histopathological sections. RESULTS BAM at 5 mg*kg-1 and 2.5 mg*kg-1 inhibited the growth of subcutaneous tumors by 78.7% and 61.9%, and the orthotopic tumors by 99.4% and 90.0%; and the intra-hepatic tumors by 86.9% and 75.7%, respectively. Determined by the numbers of metastatic nodules, BAM at 10 mg*kg-1 and 5 mg*kg-1 inhibited hepatic metastases from intra-splenic transplantation by 97.6% and 56.8%; and evaluated by image analysis of metastatic lesions, by 100% and 63.3%, respectively. CONCLUSION Boanmycin is highly effective in a panel of models including the subcutaneous, orthotopic, intra-hepatic transplanted tumors, and hepatic metastases of murine colon carcinoma 26.
2.Correlation between coronary artery disease and levels of glycosylated hemoglobin and endothelin

Objective To analysis the correlation between coronary atherosclerosis and levels of glycated hemoglobin (HbA1c) and endothelin(ET ) ,furthermore to explore the clinical significance of the two indexes in the department of cardiology .Methods 572 inpatients were divided into four groups according to the blood glucose level and coronary angiography results :negative control group with 40 patients ,simplex DM group with 50 patients ,simplex CHD group with 262 patients ,CHD+DM group with 220 pa-tients .The HbA1c and ET levels in the four groups were monitored ,and the correlations between the two indexes and coronary ath-erosclerosis were analysed by Logistic regression analyses .Results HbA1c level :control group and CHD group < DM group
3.APPLICATION OF LASER HOLOGRAPHIC STEREOVISION TEST CHART IN PATIENTS AFTER PHACOEMULSIFICATIONS

The purpose of this study was to investigate the function of stereo visual acuity in patients after phacoemulsifications and intraocular lens implantation. Mensuration of stereopsis with laser holographic stereovision test chart was done in 80 cases who had undergone phacoemulsifications and intraocular lens implantation. In this group, 43 patients were male, 37 were female .Ages of the patients were 52 to 81 years. Single eye operation was done in 18 cases, while both eye operation in 62 cases. The corrected vision was all over 0 4 in both eyes.In the 80 cases 67 (83 7%) had near stereopsis and 13 cases (16 3%)did not have; 71 cases (88 8%) had distance stereopsis and 9 cases (11 2%)did not have. Function of stereopsis weakens gradually following aging and is closely related with vision. The function of sterovision would recover to varying degrees after phacoemulsifications and intraocular lens implantation. Recovery of distance stereopsis was better than that of near stereopsis.The results of this study show that the status of stereopsis is an important index to evaluate the curative effect in patients after phacoemulsifications and intraocular lens implantation.
4.Analysis of Bone Mineral Density of Lumbar Spine in Middle-old-aged Female with Quantitative Computed Tomography

Objective To explore the clinical value and the difference of bone mineral density(BMD) in every vertebra of lumbar spine in middle old-aged Chinese female with quantitative computed tomography (QCT).Methods The BMD in every vertebra of lumbar spine was investigated by QCT in 725 healthy females aged from 35 to 98 years. They were divided into groups every five years.The BMD in vertebra of lumbar spine (L 1~L 4) were measured by QCT and the groups tested for significant differences,using SPSS10.0 software for the analysis.Results Lumbar spine (L 1~L 4) BMD declined with aging. 5.Cultured homologous dermal fibroblasts of rabbit in aPVR animal model

Objectives:To establish animal model of anterior proliferative vitreoretinopathy (aPVR) with cultured homologous dermal fibroblasts of rabbit, and to provide evidence why hypotony was caused by aPVR. Methods:Animal models of aPVR were established with cultured homologous dermal fibroblasts on pigmented rabbits. Rabbits were sacrificed on the 14th, 28th and 56th day after the operation to prepare naked eyes and to receive histological examinations. Results:Naked eye examination showed that the peripheral retina was detached by dragging in the experimental group 28 and 56 days postoperatively. Microscopic examination showed atrophy or absence of the non-pigmented ciliary epithelium on the 28th and 56th postoperative day in the experimental group. Conclusions:The epiciliary membrane in aPVR dragged the ciliary body, made atrophy of non-pigmented epithelium, which perhaps was the main cause of hypotony.
6.Effect evaluation of admission test combined with artificial rupture of membrane to predict delivery and fetal outcome

Objective To evaluate the value of admission test of fetal heart rate and artificial rup-ture of membrane in predicting fetal distress in labor. Methods 658 pregnant women who had been pregnant for 33+5 to 43 weeks and delivered spontaneously in 24 hours were divided into low risk group(598 cases) and hign risk group(60 cases) according to their admission test. The admission test, result of deliv-ery, amniotic fluid contamination and Apgar scores of newborns were analyzed. Results 515 cases were reactive type(78.27%), 72 cases were equivocal type(10.94%) and 71 cases were ominous type(10.79%). Total incidence of neonatal asphyxia were 6.53%,and it was 1.94% and 33.08% in reactive type subjects and ominous type subjects, respectively. The rates of anmiotic fluid contamination were 20. 97% and 67.61% in reactive type subjects and ominous type subjects (P<0.01). The incidence of reactive type and equivocal type of amniotic fluid contamination and neonatal asphyxia were significantly lower in the low risk group than those in the high-risk group (P<0.05). Conclusions Reactive type of admission test can ensure safety of fetus during the subsequent four to five hours. Admission test of fetal heart rate and artifi-cial rupture of membrane can be used to predict fetal outcome in labor.
7.Expression and clinical significance of MST1 in cervical cancer

Objective To investigate the expression and clinical significance of mammalian sterile 20-like kinase 1 (MST1) in cervical cancer. Methods Immunohistochemical method was applied to detect the expression level of MST1 protein in specimens of cervical cancer tissues (n=139) and pericarcinomatous tissues (n=20, with≥4 cm distance from the primary tumor's edge). Western blot assay and qPCR were used to detect the protein and mRNA transcription expression levels of MST1 in 20 pairs of cervical cancer tissues and pericarcinomatous tissues, respectively. The correlation between MST1 expression, clinic pathological features and the prognosis were analyzed. Results MST1 was mainly expressed in cytoplasm. The positive expression rate of MST1 was significantly lower in cervical cancer tissues (27%, 38/139) than that in pericarcinomatous tissues (80%, 16/20,χ2=21.62, P<0.01). The expressions levels of MST1 protein and mRNA were both lower in the cervical cancer tissues (P<0.01). In cervical cancer, the positive expression rate of MST1 inⅠb+Ⅱa stage was higher than that ofⅡb+Ⅳstage (P<0.05), the positive expression rate of MST1 in lymph node metastasis was lower than that of without lymph node metastasis (P < 0.05). Values of age, tumor size, histological type and differentiation degree showed no significant difference to positive expression rate of MST1. Moreover, the negative expression of MST1 displayed a significantly poorer overall survival time than that of positive expression of MST1 (Log-rank χ2=28.35, P < 0.01). Conclusion MST1 shows a lower expression in cervical cancer, which may be a new target for clinical treatment and prognosis of cervical cancer.
8.The diagnostic criteria of latent autoimmune diabetes of adults

Objective To evaluate the clinical diagnostic criteria of latent autoimmune diabetes of adults(LADA).Methods A total of 268 subjects consisted of 168 patients with type 2 diabetes(T2DM) and of 100 normal controls were involved in the study.The body mass index(BMI) was calculated by formula(kg/m~2) and the levels of fasting blood glucose(FBG),postprandial blood glucose(PBG),fasting C-peptide(FC-P),postprandial Cpeptide(PC-P),GAD antibody(GADA),ICA,IAA and IA-2 antibody were detected. The patients with positive antibody in T2DM group were considered as LADA and the others in T2DM group still as T2DM.Results The GADA,ICA,IA-2 antibody and IAA antibody were detected in 21.18%,14.29%,1.19% and 0.60% of patients with T2DM,respectively.The indexes such as the age of onset ≤ 40 years old,overt symptom,ketosis,without chronic complication and hypertension,BMI90.00%).Conclusion The GADA is the most specific antibody in the diagnosis of patients with autoimmune diabetes.The indexes such as the age of onset≤40 years,overt symptom,ketosis,without chronic complication and hypertension,BMI
9.Pars repair by internal fixation and bone graft through the Wiltse approach via Quadrant channel

Objective To investigate the clinical effects of pars repair by the pedicle screws,laminal hooks and auto iliac bone graft through the Wiltse approach via Quadrant channel for the treatment of the adolescent spodylolysis.Methods From November 2007 to April 2011,pars defects of 11 patients (10 males and 1 female) were repaired with pedicle screws,laminal hooks and auto iliac bone graft via Quadrant channel through the Wiltse approach.The mean age of 11 patients was 17.3 years (range 15-19).Mean course was 9.9 months (range 6-14 months).All patients have bilateral pars defect of L5.There was no poroma across the pars that have been proved by X-ray film and 3-D CT before the operation.There were 4 patients with Ⅰ grade spondylolisthesis.The degeneration of the intervertebral disc assessed by Pfirrmann grade of all patients was no more than Ⅰ grade.Visual analogue scale (VAS) and Oswestry disability index (ODI) were compared before and after operation.The fusion of pars defect was observed by X-ray film and 3-D CT.Results The mean operation time was 85.5±12.3 min (range 70-110 min); intraoperatve blood loss was 80-150 ml (mean 105 ± 19.4 ml).Two patients were lost in follow-up after operation and the others had been followed up for 12 to 24 months (mean 16 months).All patients had significant pain relief and function recovery after operation.Mean postoperative VAS score was 1.5± 1.0 compared with 4.5± 1.0 of preoperation (t=4.603,P=0.002).ODI was decreased from 49.3%± 14.2% of preoperation to 12.0%±4.9% of the last follow-up (t=7.344,P=0.000).With the Macnab grades,7 patients are excellent,2 patients are good.3-D CT showed that all pars defect were fused in the last follow-up.Conclusion Pars repair by the pedicle screws,laminal hooks and auto iliac bone graft through the Wiltse approach via Quadrant channel can protect the paravertebral muscle,relief patients from pain,and improve function.
10.An Analysis of Loneliness and Its Related Influencing Factors in Individuals with Speech Disability

Objective To study the prevalence and associated factors of loneliness in individuals with speech disability.Methods Using multi-stage stratified random cluster sampling method,170 community-residing ver-bally disabled persons were selected and administered with a general information questionnaire,one single -item loneliness self-rating question and social support scale.A total of 204 study questionnaires were distributed to the subjects;170 subjects(mean age:43.1±13.7 years)completed the survey.Results As high as 46.47% (79/170)of these verbally disabled individuals reported to feel lonely often.Females (OR=2.45),unemployment (OR=2.95), first and second degrees of disability (OR=4.35),co-existence of chronic illnesses (OR=6.50)and low utiliza-tion of social support (OR=2.58)were significantly associated with the increased risk of loneliness in persons with speech disability (P =0.002~0.046).Conclusion Loneliness is highly prevalent in individuals with speech disabili-ty.Verbally disabled persons,who are female,unemployed,severely disabled,and chronically ill and have a low use of social support,are the target population of mental health services.
