1.Effects of exposure to simulated sunlight on the morphology of rabbits' retina

Objective To investigate the pathological features of rabbit' retina at the early stage of sunlight-induced injury.Methods Twenty New Zealand rabbits without any eye diseases were randomly allocated to 4 groups(5 each): animals in group A were fed under normal illumination environment,the animals in group B were fed under normal illumination environment and exposed to constant simulated sunlight,the animals in group C were fed under low illumination environment,and the animals in group D were fed under low illumination environment and exposed to constant simulated sunlight.The pathological changes in rabbits' retina were observed with light and transmission electron microscope.Results The structure of retina in group A and group C were histologically well-defined,uniform in staining,and with regular cell shape.In the animals in group B,vesiculation occurred in the ganglion cell layer,inner nuclear layer and outer nuclear layer,and some of the mitochondria showed swelling.The intramembranous structure of external segments became disorganized.In the animals in group D,vesiculation and swelling occurred in the ganglion cell layer and inner nuclear layer,with disruption of the cell membrane.Vesiculation and swelling were also found in the outer nuclear layer,with reduction in cell quantity.The structure was disorganized,with disorganized cell membrane,obvious swelling of mitochondria,disorganized intramembranous structure of external segments,and loss and vesiculation of the laminal structure.Conclusion The rabbit' retina will be injured when it is exposed to 30 000 lux constant simulated sunlight for 30 minutes.Rabbits fed in a low illumination environment and exposed to constant simulated sunlight may suffer severer injury than those fed in a normal environment and exposed to constant simulated sunlight.

A number of biosimilars will be soon marketing due to the expiration of patent protection of their originators.Unlike classical chemical drugs,biotherapeutics are proteins with large and complex molecules produced by biological high-tech with sophisticated manufactural procedures.This leads to the difficulties of the copy of biosimilars from their originators,and the almost inevitable distinctions between the two products.Thus biosimilars are only similar but not identical to their reference drugs in terms of stracture,action,and safety.It is important for physicians to understand these differences before using biosimilars.Optimal management by a national regulatory authority with rigorous standards is very important in all aspects of authorization of the development,production,marketing,and postmarketing surveillances for biosimilars.

4.Effects of Diosgenin on VEGF and AP-1 Expression in Synovial Tissues of CIA Rats

This paper was aimed to study effects of diosgenin on expressions of activator protein-1 (AP-1) and vascular endothelial growth factor (VEGF) in synovial tissues of collagen-induced arthritis (CIA) rats induced by bovine type II collagen, in order to investigate the possible mechanism of herbal medicine diosgenin in the treatment of rheumatoid arthritis (RA). After the CIA rats models were successfully established, rats were randomly divided into the blank control group, CIA model group, diosgenin group, and positive medicine control (tripterygium) group. The in situ hybridization was used to detect the expressions of AP-1 (c-fos and c-jun) in synovial tissues of the knee joint. The real-time PCR was used to detect the VEGF mRNA expression in synovial tissues of rats’ knee joints. The results showed that c-jun and c-fos, VEGF mRNA expressions in synovial tissues of rats’ knee joints were obviously higher than that of the blank control group (P<0.01). After treatment of diosgenin and tripterygium, the expressions of c-jun and c-fos, VEGF mRNA were significantly reduced (P<0.01). It was concluded that diosgenin may regulate the expression of VEGF in synovial tissues through c-jun and c-fos of AP-1 in order to inhibit synovial angiogenesis for the treatment of RA.
5.Effect of Xiaozheng Yiliu Decoction on the cytotoxic activity of cisplatin against human ovarian cancerOVCAR/DDP cells and its action mechanism

Objective To study the cytotoxic activity of Chinese compound prescription (Xiaozheng Yiliu Decoction) combined with cisplatin on human ovarian cancer OVCAR/DDP cells and its molecular mechanism. Methods The xenograft model was established by injection of OVCAR/DDP cells. Forty nude mice were randomly allocated into four groupsincluding control group, cisplatin treatment group, low-dose Xiaozheng Yiliu plus cisplatin treatment group,and high-dose Xiaozheng Yiliu plus cisplatin treatment group.The apoptosis was determined by TUNEL analysis.The survivin expression was detected by RT-PCR analysis.Results The inhibit rates in cisplatin treatment group, low-dose Xiaozheng Yiliu plus cisplatin treatment group,and high-dose Xiaozheng Yiliu plus cisplatin treatment group,were(5.56±1.14) %,(18.42±7.73) %,and (31.59±12.58) %,respectively,and there were significant differences among the groups(F=8.64,P<0.01 ).The TUNEL analysis showed that the percentages of cell apoptosis were(2.35±1.04) %,(4.56±1.61) %,(12.42±3.68) %,(21.59±5.17) % in four different groups,respectively.After treated by Xiaozheng Yiliu Decoction,the mRNA expression of survivin was found to be down-regulated by RT-PCR analysis. Conclusion Xiaozheng Yiliu Decoction could iucrease the cytotoxic activity of cisplatin on ovarian cancer OVCAR/DDP cells, which may be related to the down-regulation of survivin gene expression.
6.Effect of ionizing radiation on the expressions of STAT3 target genes in A549 cells

Objective To investigate the effect of ionizing radiation on the expression of transcription factor STAT3 target genes in the pulmonary adenocarcinoma cell line A549.Methods The expressions of VEGF,Bcl-2 and Survivin in A549 cells with or without STAT3 inhibition were detected by RT-PCR and/or Western blot 24 h after 2 or 4 Gy of γ-ray irradiation.Results After γ-ray irradiation with 2 or 4 Gy,the expression of VEGF and Survivin increased significantly.However,the expression of Bcl-2 was not affected by γ-ray irradiation.Furthermore,the increased expression of VEGF and Survivin induced by radiation was found to be inhibited after radiation-induced activation of STAT3 was blocked by AG490 inhibiter.Conclusions γ-ray irradiation could up-regulate the expression of Survivin and VEGF via activating STAT3.The increase of Survivin might partly inhibit the radiation-induced apoptosis of A549 cells,and the up-regulation of VEGF might contribute to the tumor angiogenesis.
7.Laparoscopic cholecystectomy combined with intraoperative cholangiography and endoscopic sphincterotomy for the treatment of choledocholithiasis

ObjectiveTo evaluate the effect of laparoscopic cholecystectomy (LC) combined with intraoperative cholangiography (IOC) and intraoperative endoscopic sphincterotomy (IOEST) for the diagnosis and treatment of choledocolithiasis. Methods Statistical analysis was carried out for 106 patients with cholecystocholedocolithiasis diagnosed and treated by LC-IOC-IOEST. Results Sixty-four (60.4%, 64/106 ) patients with preoperative diagnosis of simple gall stone were found with complicated choledocholithisis; LC-IOC -IOEST was successfully performed in 99(93.4%, 99/106) cases. Six cases (6.1%) suffered from mild postoperative acute pancreatitis. Duodenal perforation, bile leakage, peumothorax (0.9%) developed in one each. Duodenal adenocarcinoma was overlooked in one case, and 2 cases (1.8%) suffered from gastric paralysis. Conclusion LC combined with IOC and IOEST was a safe, effective approach to the diagnosis and minimally invasive treatment of choledocholithiasis.
8.Bifidobacteria relieve CPFX-induced testosterone reduction in mice

Objective To explore if bacillus bifidus relieve CPFX-induced testosterone reduction in mouse testes.Methods Twenty-four male mices were divided into 4 groups, then administered saline for 6 days (Sal6 group), CPFX for 6 days (A6 group), CPFX for 6 days followed by bifidobacteria treatment for the next 6 days (A6+P6 group), CPFX for 6 days and then saline for the next 6 days (A6+Sal6 group).We detected serum levels of testosterone by RIA, as well as levels of steroidogenic enzymes mRNA [cholesterol side-chain cleavage enzyme (P450scc) and steroidogenic acute regulatory protein (StAR)] and NF-E2-related factor2 (Nrf2) mRNA in testes by real-time PCR, Nrf2, heme oxygenase-1 (HO-1), and 4-hydroxy-2-nonenal (4-HNE) by Western blot and4-HNE by Immunohistochemistry.Results The A6 group had significantly lower serum testosterone levels compared with the Sal6 group (P<0.001), the A6+P6 group had significantly higher compared with the A6 (P<0.001) and A6+Sal6 groups (P<0.01).The A6 group had significantly lower StAR mRNA compared with the Sal6 group (P<0.001), the A6+P6 group had significantly higher level compared with the A6 (P<0.01) and A6+Sal6 groups (P<0.01).The A6 group had significantly lower P450scc mRNA as compared with the Sal6 group (P<0.001), the A6+P6 group had significantly higher compared with the A6 (P<0.001) and A6+Sal6 groups (P<0.05).The A6 group had significantly lower Nrf2 compared with the Sal6 group (P<0.001), the A6+P6 group had significantly higher compared with the A6(P<0.01) and A6+Sal6 groups (P<0.05).The A6 group higher 4-HNE expression compared with the Sal6 group, the A6+P6 group had significantly lower compared with the A6 (P<0.01) and A6+Sal6 groups (P<0.05).Conclusions Bifidobacteria the reduction of CPFX-induced testosterone reduction, and these effects may potentially explained by Nrf2 inflammatory signaling pathway.
9.Isolation, identification and antimicrobial resistance of Campylobacter jejuni isolates from poultry in Jiangsu Province

The aim of the research is to study the prevalence and antimicrobial resistance of Campylabocter jejuni isolated from poultry in Jiangsu Province.A total of 753 samples from poultry meat and cloacal swabs were investigated,after the pure culture and the polymerase chain reaction of the mapA gene,207 isolates were examined for antimicrobial resistance by using K-B method according to World Health Organization.Results showed that all isolates performed different degree of antimicrobial resistance except meropenem,gentamycin,kanamycin,florfenicol and fosfomycin,the resistance rates of 194 strains to trimethoprim,norfloxacin,ceftriaxone and tetracycline were 100%,84.02%,80.9% and 79.4% respectively,1 strain isolated from Xuzhou was resistant to 92.6 % antibiotics.The multi-drug resistance appeared and the advantage of drug-resistant spectrum was LIN/CTX/CRO/NOR/CIP/T/TE,the resistant type focused on 40%-60%.The research provided evidence for surveillance of the antimicrobial resistance of C.jejuni and highlighted the need to employ more prudent use of critically important antimicrobial.
10.Effects of Total Saponins from Rhizoma Dioscreae Nipponicae on VEGF and AP-1 in Rat Synovial Cell Strain

Objective To study the effects of medicated serum with total saponins from Rhizoma Dioscreae Nipponicae (RDN) on VEGF mRNA expression and AP-1 activity in rat synovial cell strain RSC-364 induced by IL-17 and TNF-α. To investigate the mechanism about total saponin from RDN inhibition of angiogenesis. Methods Medicated serum of total saponins from RDN and tripterygium (positive control) were prepared. Rat synovial cells RSC-364 were divided into four groups: the blank control,IL-17+TNF-α model,tripterygium medicated serum,and total saponins medicated serum groups. After one hour of incubation,all groups except for the blank control were incubated with both IL-17(10 μg·L-1 ) and TNF-α(10 μg·L-1 ) for 24 hours. VEGF mRNA expression in RSC-364 was detected by PrimeScriptTM real-time quantitative PCR (RT-PCR) detection kit,and the AP-1 DNA-binding activity was detected by electrophoretic mobility shift assay (EMSA). Results Compared with the control blank group,both of the VEGF mRNA expression and AP-1 activity in rat synovial cell strain RSC-364 induced by IL-17 and TNF-α increased remarkably (P<0. 05,P<0.01). The VEGF mRNA expression and AP-1 activity in tripterygium medicated serum group and total saponins medicated serum group were remarkably lower than those of the model control group (P<0.05). There was no significant difference between the two medicated serum groups. Conclusion Serum medicated with total saponins from RDN can remarkably decrease VEGF mRNA expression and AP-1 activity,indicating that the total saponins from RDN could influence VEGF secretion by inhibiting the AP-1 signal transduction pathway,VEGF is the key factor of angiogenesis,thereby to restrain angiogenesis.
