1.Effect of Auricularia auriculajudae polysaccharide on cell immunity of tumor-bearing mice

AIM: To study the influence of Auricularia auriculajudae polysaccharide (APP) on celular immunity of mice bearing tumor. METHODS: Influence of AAP on lymphocyte proliferaton and IL-2 production of H_(22) mice and was observed by cell cultivation. And the influence of AAP on free Ca~(2+) concentration in lymphocytes of H_(22) mice through laser sacanning confocal microscope (LSCM). RESULTS: AAP can lengthen the average survival time of H_(22) tumor-bearing mice. The research of immune function indicated that AAP can improve the tumor-bearing mice's celular immunity the increase in lymphocytes, and the product of IL-2 and Ca~(2+) concentration inside lymphocytes. CONCLUSION:AAP has a function of anti-tumor. The mechanism has a relation to raising the celular immunity.
2.Effect of galanthamine on motor function of patients with cerebral infarction

ObjectiveTo explore the effect of galanthamine on motor function of patients with cerebral infarction.Methods78 patients were randomly divided into the galanthamine treatment group (40 cases) and the control group (38 cases). Patients in the control group were given clinical routine treatment and rehabilitation training, while the galanthamine treatment group took galanthamine orally in addition. Evaluations were done in pre-treatment and post 6 weeks. Motor function was assessed with Fugl-Meyer motor scale (FMMS).ResultsThe increase of FMMS scores in each group had a significant difference ( P<0.05-0.01), while the process of scores in the galanthamine treatment group was superior to that in the control group (P<0.01).Conclusions Galanthamine hydrobromide is an effect drug on improving activities of motor function and ameliorating the therapeutic efficacy of rehabilitation to patients with cerebral infarction.
3.The relation research on resilience, personality and family environment of male juvenile delinquents with high traumatic experiences

Objective To explore the resilience traits of male juvenile delinquents with high traumatic experiences and the relationship on resilience and personality,family environment.Methods 147 male juvenile delinquents selected by self-made traumatic experience questionnaire were surveyed with the Resilience Scale for Chinese Adolescent,Eysenck Personality Questionnaire and Family Environment Scale.The data were statistically analyzed by Pearson correlation analysis and stepwise Regression Analysis.Results The total scores of resilience in male juvenile delinquents with high trauma experiences were (75.98 ± 14.03).Besides the scores of goal planning (3.47±4.23),the scores of affect control(1.94±2.85),positive thinking(2.31±3.47),family support(1.89±3.84)and help seeking (2.92±3.38)were all below 3.Differences of goal planning,positive thinking,family support had been found in male juvenile delinquents with high trauma experiences on the factors including educational level,rural or urban family,whether only-child family,whether single-parent family.The delinquents' resilience was correlated with their personality(r=-0.357-0.175) and family environment (r=-0.185-0.272) in several dimensions (P <0.05).Neuroticism,family conflict and psychoticism predicted resilience negatively (β=-2.354,-0.135,-0.194,P<0.05).Family cohesion predicted resilience positively(β=1.389,P<0.001).Conclusion The resilience of male juvenile delinquents with high traumatic experiences is poor and personality,and family environment has significant influences on their resiliences.

Objective To investigate the role of MHC class Ⅱ transactivator(CⅡTA) in upregulating the expression of HLA class Ⅱ antigen in HepG2 cells.Methods Eukaryotic expression vector EBS-NPL-CⅡTA containing CⅡTA cDNA or the empty vector EBS-NPL was transfected into HepG2 cells respectively.CⅡTA mRNA and HLA class Ⅱ antigen(HLA-DR,DP,DQ) were respectively detecued by RT-PCR,indirect cell immunofluorescence technique and flow cytometry in original HepG2 cells,HepG2 cells transfected with EBS-NPL-CⅡTA or the empty vector EBS-NPL.Results The expression of CⅡTA mRNA and HLA class Ⅱ antigen(HLA-DR,DP,DQ) were not observed in original HepG2 cells and HepG2 cells transfected with empty vector,but in the HepG2 cells transfected with EBS-NPL-CⅡTA.Conclusion CⅡTA is a switching factor of mastering the expression of HLA class Ⅱ antigen in HepG2 cells.The lack of CⅡTA expression in HepG2 cells contributes to no expression of HLAⅡ antigen.
6.Functional study of the different haplotypes cDNA in the coding region of CⅡTA gene

Objective To study the function of 4 different haplotypes cDNA which are constructed by two non-homonymy single nueleotide polymorphism (SNP) sites C19170G (Leu45Val) and C30799G (Ala500Gly) in the coding region of human CⅡTA gene. Methods HeLa cells were transfeeted with eu-karyotic expression vectors containing four different haplotypes cDNA. C Ⅱ TA mRNA and HLA classⅡanti-gen (HLA-DR, DP, DQ) were respectively detected by RT-PCR and indirect cell immunofluoreseence tech-nique in the untransfected and transfeeted with four eukaryotic expression vectors and empty vectors HeLa cells. The quantity of HLA classⅡ antigen were analyzed by flow eytometry. Results No expression of CⅡTA mRNA and HLA class Ⅱ antigen were observed on original HeLa cells and empty vector transfected cells. CⅡTA mRNA expression was emerged, and the expression of HLA class Ⅱ antigen were observed in the HeLa cells transfected with eukaryotic expression vectors containing four different haplotypes cDNA. And there were not significantly different with the levels of HLA class Ⅱ antigen expression among HeLa cells transfected with eukaryotic expression vectors containing four different haplotypes cDNA ( P > 0.05 ). Con-dusion The SNP of Chinese at the sites C19170G(Leu45Val) and C30799G(Ala500Gly) in the coding site of C Ⅱ TA gene did not influence capability of CⅡTA trans-aetivating HLA class Ⅱgene expression.

Abstract Objective:To study the expression and its biologic specification of the Ku80 gene molecular cloning.Methods: Constructingrecombinant sense or antisense Ku80 expression vectors (Ku80/pCAGGS) and transferring into LCA .Those Ku80 mRNA and protein were de-teited in the Ku80s-LCA, Ku80as-LCA and by Northern-blotting and or Western-blotting.Results: Those Ku80 mRNA were detected in the Ku80s-LCA and Ku80as-LCA by Northern-blotting. Only the sense Ku80 protein was defected in the Ku80s-LCA , but wasn' t detected in the Ku80as-LCA by Western-blotting.Conclusion:The sense or antisense Ku80 were not only transferred in to LCA ,but also expressed in the mR-NA . Only the sense Ku80 protein was expressed in the Ku80s-LCA , but wasn' t expressed in the Ku80as-LCA . The Ku80 is a candidate tar-get for cancer gene therapy.
9.Analysis and treatment of abnormal results of Sysmex CA-1500 automated coagulation analyzer

Objective To analyze the parametric errors during the use of Sysmex CA-1500 automated coagulation analyzer and their treatment measures .Methods Blood samples with test results showing “*”and without figure results were collected .Sysmex CA-1500 automated coagulation analyzer was employed to retest prothrombin time (PT ) ,activated partial thromboplastin time (APTT) and fibrinogen(FIB) .Results FIB ,PT and APTT were found abnormal in 226 cases(95 .77% ) ,5 cases(2 .12% ) and 5 cases(2 .12% ) ,respectively ,with FIB the mainly .ERR008 ,ERR004 ,ERR016 ,ERR002 ,ERR032 and ERR100 were found abnormal in 227 cases(76 .95% ) ,64 cases(21 .69% ) ,one case(0 .34% ) ,one case(0 .34% ) ,one case(0 .34% ) and one case(0 .34% ) ,respec-tively ,with ERR008 the mainly .Difference of no-error results between the initial and repeat test of PT and APTT showed no statis-tical significance(P>0 .05) ,while those of FIB demonstrated statistical significance (P<0 .01) .Conclusion Samples with paramet-ric errors during the use of Sysmex CA-1500 automated coagulation analyzer should be retested to promote the quality of blood co-agulation detection .
10.Comparison of therapeutic effects of different tiotropium bromide regimens in adult patients with asthma

Objective To analyze the clinical effect and the related performance of different tiotropium bromide in the treatment of adult asthma patients.Methods58 patients in the Second hospital of Shandong University from August 2014 to May 2016 admitted to the adult bronchial asthma, were randomly divided into observation group and control group, each of the 29 cases of patients, two patients were taken on the basis of routine treatment of tiotropium bromide in the treatment, the observation group in each given tiotropium 5 g treatment.The control group took a day sooner or later given tiotropium 2.5 g treatment, analysis of clinical effect of two groups were compared and the related performance.ResultsThis group of patients with different delivery methods after treatment, the observation group and the control group of arterial blood gas analysis results have significant difference compared to the clinical effect compared with statistical difference (P<0.05);treatment group the effective rate of treatment group and control the efficiency compared with significant differences (P<0.05);the two groups of patients after treatment, clinical symptoms were observed in groups of patients with asthma remission time were (6.24±1.92) d, while the control group was (8.33±2.47) d, the two groups were compared with statistical difference (P<0.05).ConclusionDifferent tiotropium dosing regimen for adult patients with asthma curative effect has significant clinical effect, compared with 5 per night given tiotropium Gtherapy and its clinical effect is more obvious.
