Objective To investigate the relationship between the level of tumor necrosis factor (TNF)? and the clinical features of multiple sclerosis(MS). Methods The TNF? level of 58 MS patients (MS group) was determined by means of sandwich enzyme-linked immunosorbent assay (ELISA). The correlative analysis between level of TNF? and the Expanded Disability Status Scale (EDSS), the clinical data (including the duration of disease,the number of relapses and the age of onset) were take in MS patients. Objective To study the changes of the T-lymphocyte subsets in patients with Guillain-Barré syndrome(GBS)before and after intravenous immunoglobulin treatment(IVIG),and to explore the possible mechanism of the IVIG curing GBS further.Methods Chose 31 cases of clinically confirmed GBS were enrolled and compared before and after the treatment.According to the effect of the therapy,31 cases of the total were sub-divided into effective and ineffective groups.Relative counting of peripheral blood T-lymphocyte subsets was preformed with flow cytometry.Results ①The percentage of CD8+ T and CD4+CD29+ T cell was significantly lower(CD8+T:28.77%±11.02% vs 31.84%±12.35%,CD4+CD29+T:56.71%±12.44% vs 62.40%±12.72%,t=2.995,3.919,P<0.05)after therapy,while the rate of CD4+/CD8+T and the percentage of CD4+CD45RA+T cell increased notably(t=2.368,3.860,P<0.05);but there was no notable difference in the percentage of CD3+T and CD4+T cell.②The percentage of CD8+T and CD4+CD29+ T cell was significantly lower(t=2.144,3.343,P<0.05)after the treatment,while the rate of CD4+/CD8+T and the percentage of CD4+CD45RA+T cell increased notably(t=2.159,3.277,P<0.05)in the good curative effect group,but there was no change in the bad curative effect group.③61.29%(19/31)of the patients significantly improved by IVIG,and there was no death case.Conclusions T-lymphocyte subsets change in a varing degree after IVIG treatment in acute GBS patients,which lays an immunological foundation for the further study of pathogenesis and mechanism of IVIG curing GBS;effective on GBS,IVIG can actively suppress pathogenetic condition and promote the recovery of nrevous function.
3.The Role of NF-kappa B Signaling Pathway in TNF-α-induced Gastric Cancer Cell Apoptosis

Objective To investigate the role of nuclear factor kappa-B(NF-κB)signaling pathway in tumor necrosis factor α(TNF-α)-induced apoptosis in gastric cancer cells.Methods Human gastric cancer cell line SGC7901 was treated with TNF-α for 24 hours.MTT assay,flowcytometry and Western blot was used to detect the cell viability and apoptosis.Transient transfection was performed by using Lipofectamine 2000 reagent.The cells of experimental group and control group were respectively transfected with mutant IκB cDNA and vectors.Results Under the treatment of 100,1 000 or 10 000 U/ml TNF-α,the cell viability of SGC7901 cells was(99.2±0.6)%,(92.0±2.7)% and(97.9±2.2)%,respectively.Further study showed that TNF-α engagement led to rapid activation of NF-κB signaling pathway.Blockage of TNF-α-induced NF-κB activation by transient transfection with a mutant IκB enhanced the sensitivity of cells towards TNF-α-induced apoptosis.Conclusion In human gastric cancer cells,activation of NF-κB signaling pathway by TNF-α might be responsible for the resistance to TNF-α-induced apoptosis.Blockage of NF-κB significantly enhanced the apoptosis induced by TNF-α.
4.Oxaliplatin Enhances TRAIL-induced Apoptosis in Gastric Cancer Cells by Inhibiting PI3K/Akt Pathway

Objective To investigate the role of phosphoinositide 3-kinases (PI3K)/Akt signaling pathway in TRAIL-induced cell apoptosis, and the effect of oxaliplatin on TRAIL-induced apoptosis in gastric cancer BGC823 cells. Methods Cell proliferation was roeasured using MTT assay. Cell apoptosis was determined by flow eytoroetry. The expression of Akt and phospbor-Akt were determined by Western blotting. Results 100 ng/mL TRAIL caused little cell apoptosis in BGC823 cells. TRAIL activated P13K/Akt pathway. Pretreated with PI3K in- hibitor LY294002 (25 μmol/L)for 1 h followed by exposure to TRAIL for 16 h,the cell apoptosis was obviously higher (12.7%±3,1% vs 3.5%±1.1% ,P 〈 0.05) than that without the treatment of LY294002. Treatment with 38 μg/mL oxaliplatin blocked the activation of P13K/ Akt signaling, and enhanced the sensitivity of cells to TRAIl,, the rate of cell apoptosis increased to 35.5%±4.5% (P 〈 0.05 ). Conclusion Oxaliplatin enhanced the sensitivity of gastric cancer BGC823 cells to TRAIL by inhibiting TRAIL-induced the activation of PI3K/Akt pathway.
5.Neuroimaging and its correlation with clinical aspects of cerebral palsy

Objective To investigate neuroimaging and its correlation with clinical aspects of cerebral palsy (CP).Methods A retrospective study of 295 children with CP was conducted.Magnetic resonance imaging (MRI),the gross motor function classification system (GMFCS) and intelligence testing were administered,and any correlations among these measures was analysed.Results Among the 295 cases,257 presented abnormal MRIs (87.1%) due to brain maldevelopment (n =11),periventricular leukomalacia (PVL) (n =173),cortical/subcortical lesions (n =17),basal ganglia lesions (n =26),cerebellar maldevelopment (n =11) or others lesions (n =19).Thirty-nine presented with normal MRIs.About 26% were rated at GMFCS level 1,18% at level 2,17% at level 3,19% at level 4 and 20% at level 5.Almost 82% of the children presented with brain maldevelopment and 84.4% with PVL-induced spastic bilateral paralysis.In 41% of the children with cortical or subcortical lesions,induced spastic hemiplegia was observed,whereas 47% had induced spastic bilateral paralysis.In 77% of the children with basal ganglia lesions induced involuntary movement was observed,and all of those with maldevelopment of the cerebellum were ataxic.Most of those with spastic hemiplegic,bilateral paralysis,involuntary movement and ataxia were on GMFCS levels 1 or 2,with only 3.7%,33.5%,64.1% and 46.2% respectively on GMFCS level 4 or 5.Among those in whom the MRI revealed brain maldevelopment,9.1% were on GMFCS level 1 or 2.The corresponding percentage for PVL was 43.9%,for cortical or subcortical lesions 58.8%,for basal ganglia lesions 19.2% and for cerebellar maldevelopment 27.3%.The balance in each category were on GMFCS level 4 or 5.Epilepsy was most common in the children with brain maldevelopment (36.4%) or cortical or subcortical lesions (41.2%).Mental retardation was most common in cases of brain maldevelopment (45.5%),cortical or subcortical lesions (41.2%) or cerebellum maldevelopment (36.4%).The incidence of epilepsy and mental retardation was higher among the children on levels 4 and 5 than on levels 1 and 2.Conclusions Neuroimaging correlates significantly with the type of CP and GMFCS level.Epilepsy and mental retardation are most common in children with brain maldevelopment or lesions.The incidence of epilepsy and mental retardation is higher among children rated at GMFCS level 4 or 5 than among those on levels 1and 2.
6.Study on Acute Toxicity and Effect of Aconite Roots Based on Disease/Syndrome

This study was aimed to observe influence of disease/syndrome on the toxicity and effect of aconite roots, in order to reveal relationship among disease/syndrome, toxicity and effect. The mice model of cold syn-drome was establish by wind-cold stimulation. The pain model was established by intraperitioneal injection of glacial acetic acid. Then, LD50 and ED50 of analgesic effect were compared, as well as the therapeutic index (TI) of crude aconite roots. The rat model of cold syndrome was also established by wind-cold stimulation. And the rheumatoid arthritis (RA) model was established by intracutaneous injection of CII and CFA. TD50 of cardiac toxic-ity and arthroncus degree of prepared aconite roots were compared among the normal rats, RA rats, RA with wind-cold stimulation rats according to the recording of lead II ECG. The results showed that after wind-cold stimulation, mice and rats appeared with symptoms which were similar to Chinese medicine cold syndrome. Com-pared with normal mice, LD50 and TI increased, but ED50 decreased in the group of wind-cold stimulation after using powders of crude aconite roots. Compared with normal rats, TD50 of cardiac toxicity and arthroncus degree in-creased in groups of RA and RA with wind-cold stimulation after using prepared aconite roots. It was concluded that in the case of disease/syndrome state, the toxicity of aconite roots decreased, but its effect increased. It sug-gested that there is a significant correlation among disease/syndrome, toxicity and effect.
7.The expression and clinical significance of FGFR2, and c-Cbl in gastric carcinoma

Background and purpose: FGFR2 is a receptor tyrosine kinase and c-Cbl is a new RING finger type of ubiquitin ligase in the ubiquitin-proteasomes path. The purpose of this study was to evaluate the expression and significance of FGFR2 and c-Cbl in gastric carcinoma. Methods: The expression of FGFR2 and c-Cbl were detected by immunohistochemical method of SP. Results: The positive expression rates of FGFR2 and c-Cbl were 77.4%,71.0% in gastric carcinoma, respectively, both were higher than those normal tissue (P<0.05);The expression of FGFR2 and c-Cbl were positively correlated with depth of invasion and TNM staging, and there was a positive relationship between the expressions of FGFR2 and c-Cbl. Conclusion. The expressions of FGFR2 and c-Cbi were associated with some clinicopathologic features in gastric carcinoma, indicating that their expression may be the prognostic factors for gastric carcinoma.
8.Comparative Study on the Application of Sofren Injection in Respiratory Department before and after Intervention

Objective: To compare the application of Sofren injection in respiratory department before and after the intervention to provide reference for clinical application and pharmacy management.Methods: A retrospective study was conducted on the medical records of inpatients treated with Sofren injection, and then intervene the found problems.Comparative study of Sofren injection in respiratory department before and after the intervention was performed.Results: Among the 584 cases in the retrospective study, those with off-indications were 438 ones (75%), those with inappropriate solvents were 555 ones (95.03%), and those with improper course were 196 ones (33.56%).After the intervention in the clinical practice, the improvement rate of improper solvents was 98.70%, that of off-indications was 29.85%, and that of unsuitable course was 77.71%.Conclusion: Through comparative study on the main problems before the intervention and the effective performance of specific solutions, the application of Sofren injection in respiratory department is greatly improved, which provides reference for the special management of other drugs.
9.Relationship between the Sensitivity of Gastric Cancer Cells to Arsenic Trioxide and the Increase of Reactive Oxygen Species Level

Objective To investigate the relationship between the sensitivity of gastric cancer cells to arsenic trioxide(As2O3)and the level of reactive oxygen species(ROS)production.Methods The viability of gastric cancer cell lines MGC803,BGC823 and SGC7901 treated with As2O3 was determined by MTT assay.ROS levels of the gastric cancer cells before and after the treatment of As2O3 were detected by flow cytometry.Results Cell growth was significantly inhibited by As2O3 in time-and dose-dependent manner in three gastric cancer cell lines.The IC50(72 h)of As2O3 for MGC803,BGC823 and SGC7901 was about 2.8,3.1 and 10.2 μmol/L,respectively.IC50(72 h)of MGC803 and BGC823 was lower than that of SGC7901(P 0.01),gastric cancer cell line SGC7901 was less sensitive than the others.The inherent ROS level of MGC803,BGC823 and SGC7901 was 20.3±2.0,64.2±3.3 and 57.7±2.0.After treatment with As2O3 5 μmol/L for 24 h,the peak level of ROS in MGC803 and BGC823 cells increased to 100.8±3.8 and 103.5±2.3,compared with inherent ROS level,the difference had statistical significance(P 0.001,P 0.01),but the inherent ROS level in SGC7901 cells was 56.5±2.4(P 0.05).Conclusion The sensitivity of gastric cancer cells to arsenic trioxide is associated with the increase of reactive oxygen species level.
10.Role of Akt and SRC Pathways in Exosome?mediated Proliferation of Homologous Lung Adenocarninoma Cells

Objective To explore the effect of Exosomes isolated from the A549 lung cancer cells on the proliferation of these cells and their ho?mologous tumor cells,HCC827,and the role of the PI3K/Akt and SRC signaling pathways in this process. Methods Exosomes were isolated from the supernatant after density gradient centrifugation of A549 cells. The Exosomes morphology was observed by transmission electron microscopy. The expression of the Exosome?specific proteins was analyzed using Western blotting. Cell proliferation was investigated using the MTT assay. Re?sults The A549?derived Exosomes were 30?100 nm in diameter and had a bilayer membrane.Western blotting showed that CD9 was detected in these Exosomes. The isolated Exosomes promoted the proliferation of the A549 and the HCC827 cells in a dose?and time?dependent manner,ac?companied by the activation of Akt and SRC. Conclusion Exosomes isolated from A549 cells promote the proliferation of the secreting cells and the homologous tumor cells in a dose?and time?dependent manner. The mechanism may be related to the activation of Akt and SRC.
