1.The research of acetated Ringer solution and succinylated gelatin in the fluid resuscitation of sepsis patients

Objective To research the clinical significance of acetated Ringer solution and succinylated gelatin in the fluid resuscitation of sepsis patients.Method Fifty-nine sepsis patients were divided into two groups:lactic acid Ringer solution and hetastarch (group A,27 cases)and acetated Ringer solution and suecinylated gelatin (group B,32 cases),compared the fluid resuscitation and other parameter.Results There were no significant difference in the central venous pressure,mean arterial pressure and the dose of noradrenaline (P > 0.05).The activated partial thrombeplastin time in group A was longer than that in group B[(58±10),(74±13)s vs(48±7),(54 ±11) s in 6,24 hours],fibrinogen concentration was lower than that in group B [(3.3 ±0.8),(1.6±0.3)g/L vs (4.2±1.1),(2.1±0.2)g/L in 6,24 hours] (P<0.05).Serum lactic acid,creatinine,calcium concentration in group A [(6.9±0.8)retool/L,(289 ~ 27)μ mol/L,(2.1 ±0.3)mmol/L]were higher than those in group B [(3.2 ±1.1)mmol/L, (193 ±42)μmol/L, (1.7±0.2)mmol/L](P<0.05),there was no difference in APACHE Ⅲ scores on the third day between two groups (P>0.05).Conclusions Acetated Ringer solution and succinylated gelatin in the fluid resuscitation of sepsis patients has the same effect,but fewer impacts on serum coagulation,serum lactic acid and renal function than lactic acid Ringer solution and hetastareh.They may be more suitable for fluid resuscitation of sepsis patients,but hypecalcemia is easy to happen,and there is no significant evidences proving the prognosis.
2.Research of reformative prone position ventilation in patients with acute respiratory distress syndrome

Objective To investigate the effect of reformative ventilation method (head-high leg-low 30°-declivate prone position ) in the ventilation of acute respiratory distress syndrome ( ARDS ) patients, and compared with normal prone position ventilation. Methods Forty-four patients of ARDS underwent ventilation were divided into prone position group (22 cases) and reformative prone position group (22cases) by random digits talle. Continuous 30° semireclining position was kept in normal time. The patients of the two groups underwent different prone position ventilation 4 hours every day,and indexes of hemodynamics and respiration before,during and after prone position ventilation were recorded and analyzed. Results After prone position ventilation, oxygenation index of two groups improved similarly (P > 0.05 ). In prone position group, lung compliance decreased [2 h, ( 25.6 ± 5.8 ) ml/cm H2O ( 1 cm H2O = 0.098 kPa) vs. ( 37.2 ± 20.5 )ml/cm H2O] ,average airway pressure increased [2 h, ( 18.5 ± 3.9) cm H2O vs. ( 15.6 ± 5.3 )cm H2O] ,heart rate increased [2 h, ( 112.0 ± 16.2 ) beats/min vs. ( 102.0 ± 11.3 ) beats/min], mean arterial blood pressure decreased [2 h, (86.0 ± 6.7) mm Hg ( 1 mm Hg = 0.133 kPa) vs. (93.5 ± 7.5 ) mm Hg] ,central venous pressure increased[2 h, ( 15.5 ± 3.3 ) cm H2O vs. ( 12.6 ± 4.3 ) cm H2O]. These changes persistently existed during prone position ventilation (P < 0.05 ),and they disappeared in 1 hour when recovered to 30 °semireclining position. Conclusion Reformative prone position ventilation not only has same effect on improving oxygenation index, but also obviates adverse effect on lung compliance and hemodynamics.
3.The influence and mechanism of cisplatin on autophagy and proliferation of endometrial cancer

Objective To explore the effects of cisplatin on the proliferation and autophagy of endometrial carcinoma Ishikawa cells.Methods Ishikawa cell proliferation was detected by MTS assay after the cells were treated with CDDP.To assess the level of autophagy,transmission electron microscope and Western blot were used to detect LC3 and Beclin1 expression;fluorescence microscopy was used to observe the fluorescence aggregation of green fluorescent protein and microtubule associated protein 1 light chain 3 fusionprotein (GFP-LC3).Results Cisplatin of 10 g/mL inhibited the proliferation of Ishikawa cells,with an increase of time and concentration,the inhibition of cell proliferation was significantly elevated (P < 0.01).Transmission electron microscopy showed that under a condition of cisplatin on Ishikawa endometrial cancer cell,autophagy occurred.With an increase of concentration and dosage,Western blot showed that autophagy related protein LC3 expression was up-regulated,but becline-1 had no obvious change.LC3 expression level was higher in 12h-treatment with 20 μg/mL cisplatin group than in the control group,and was higher in 24h-treatment group than in 12h-treatment group.Conclusions Cisplatin inhibits proliferation of Ishikawa cells and induces autophagy of the cells in a time-and dose-dependent manner.Autophagy related MAP-LC3 is involved in the molecular mechanisms of autophagy induced by cisplatin in endometrial carciHom.
4.Expression and assaying of thymic stromal lymphopoietin in nasal mucosa of murine model of allergic rhinitis

OBJECTIVE To study the content and distribution characteristics of thymic stromal lymphopoietin (TSLP) in nasal mucosa of murine model of allergic rhinitis.METHODS 6-week-old BALB/c mice were divided into control group and experimental group.Mice of experimental group were sensitized by means of dropping OVA in nasal cavity and intraperitoneal injection of OVA. Meanwhile,mice of control group were dealt with physiological saline instead of OVA.The content of TSLP in nasal mucosa of mice was tested by means of real-time PCR and its distribution was detected by means of immunohistochemistry staining.RESULTS The content of TSLP was obviously higher in nasal mucosa of experimental group than that of control group.It was distributed mainly in lamina propria and on the surface of epithelium.CONCLUSION TSLP may play an important role in allergic rhinitis of the mice.
5.Design, synthesis of novel N, N'-bis-(halogenophenyl)-4- methoxybenzene-1, 3-disulfonamides and evaluation of their anti-platelet aggregation activity.

Combining the structural features of picotamide and linotroban, a series of N,N'-bis-(halogenophenyl)-4-methoxybenzene-1, 3-disulfonamides were designed and synthesized on the basic principles of drug design. The structures of target compounds were confirmed by IR, 1H NMR and HR-MS, and the in vitro antiplatelet aggregation activity was evaluated by Born turbidimetric method with adenosine diphosphate (ADP) as the platelet aggregation inducers. The assay results showed that twelve compounds (4b, 4f, 4l, 5b, 5d-5g, 5j, 5k, 5m and 5n) were found to have superior anti-platelet aggregation activities than the positive drug picotamide. The preliminary structure-activity relationship (SAR) has been explored.
6.Establishment and characteristics of multidrug-resistant Lewis lung cancer cell line.

BACKGROUNDTo establish an ADM resistant Lewis lung cancer cell line and to investigate its biological characteristics.
METHODSThe multidrug-resistant cell line was gradually induced by ADM from Lewis lung cancer cell line (L3-8), its growth characteristics and cancinogenicity were observed. The fluorescent density of ADM and Rh-B in the cells were analyzed by IPP image analysis system. The ADM concentration in cells was assayed with fluorescent meter. The IC50 was evaluated by MTT assay and the drug resistant index was counted.
RESULTSThe drug resistant Lewis lung cancer cell line, L3-8/ADM, which can grow in the medium containing 0.4mg/l ADM was acquired after 14 months selective culture. Its tumor generatic rate was 10/10. When the L3-8/ADM was cultured in 0.15mg/l ADM and general 1640 medium, the doubling time was 22.0h and 21.8h separately. After culturing in 4mg/l ADM and Rh-B medium, the fluorescent density ratio of L3-8/ADM and L3-8 were 2.82:1 and 2.65:1 separately. The ADM concentration of L3-8/ADM was only 64.7% of its parental, but there was no significant difference in their ADM release rate. Except for ADM, L3-8/ADM was resistant to DNR, VCR, MIT and CDDP in different degrees.
CONCLUSIONSL3-8/ADM cell is a typical MDR cell line. The cell membrane obstruction of drug infiltration might be the cause of drug resistance. This study will provide a basis for the establishment of lung cancer MDR animal model.
7.Effect of doxazosin and metoprolol on vascuIar remodeling in rats with hypertension induced by abdominal aorta coarctation

OBJECTIVE To investigate the effect of doxazosin(DOX) and metoprolol( MET) on vascular remodeling in rats with abdominal aorta coarctation (AAC). METHODS An animal model was established by AAC. Two weeks later, the rats were treated with DOX (10 mg.kg-1 per day) or MET (20 mg.kg-1 per day) for six weeks. Blood pressure was measured using carotid artery intubation with a MP150 polygraph. The media thickness, wall cross-sectional area and thickness / internal diameter ratio were calculated by morphometry. Vascular fibrosis was evaluated by Masson′s trichrome staining. The collagen and fibronectin expression in vascules was measured by Western blotting. RESULTS Compared with the sham group 〔(17.6±0.5)kPa〕, the mean arterial blood pressure in the model group〔(23.3±0.7)kPa〕 was significantly increased(P<0.05), but was lowered by DOX 〔(20.5±0.7)kPa〕 and MET 〔(19.0±0.4) kPa〕 (P<0.05). Moreover, HE staining showed that tunica media thickness, artery vessel area and thickness / inner diameter in the model group were increased by 39.5%, 46.4% and 27.0%(P<0.05), respectively. The tunica media thickness was decreased by 16.0% and 26.1%( P<0.05), respectively, the artery vessel area by 22.8% and 26.1%(P<0.05), respectively, and the thick-ness / inner diameter by 17.0% and 26.0%( P<0.05) when the rats were treated with DOX and MET. Masson staining showed that the collagen accumulation in vascules increased, suggesting that AAC induced fibrosis. Meanwhile, vascular fibrosis induced by AAC was also reduced by MET or DOX. Western blotting also proved that the increase of collagen and fibronectin induced by AAC could be attenuated by DOX and MET(P<0.05). CONCLUSION DOX and MET are effective in suppressing the role of norepi-nephrine in vassels, which can attenuate AAC-induced vassels remodeling by preventing the binding between norepinephrine and adrenoceptors.
8.The capacity and biological characteristic of the p75 neurotrophin receptor-positive ectomesenchymal stem cell in vitro

Objective To investigate the biological characteristic of ectomesenchymal stem cells (EMSC) and the differentiation of p75+EMSC and influencing factors.Methods The immunohistochemical staining method was used to observe the migration of EMSC from 12.5 d SD rat embryonic facial process.Then EMSC was labeled by p75 neurotrophin receptor,and the cell cycle and stem cell surface antigens of the p75 + EMSC was examined.Results Immunohistochemical staining showed that stem cells from the cranial neural crest migrated to embryonic rat facial process at 12.5 days.The sorting rate of the p75+EMSC was 6.1%.The proportion of the p75 +EMSCs' S/G2/M phase was stable during subculture.The special substances CD29,CD146 and Stro-1 were marked for p75 +EMSC,and the expressions of the markers were all higher(>90%) in p75+EMSC.Conclusions The embryonic tooth did not start to grow on the conception 12.5 days of SD rats.The p75 + EMSC after sorting had stable proliferation ability and had stem cell characteristics during subculture and didn't start differentiation.
9.Construction and clinical application of nursing assessment information model to prevent deep vein thrombosis

Objective To introduce the construction and clinical application of nursing assessment information model to prevent deep vein thrombosis ( DVT) . Methods The electronic nursing assessment sheet was designed and perfected, the correct rate and nursing assessment length of DVT were compared. Results After the application of DVT nursing assessment information model, the correct rate of DVT assessment increased from 80%to 98% (χ2 =49. 64,P<0. 01);the assessment length decreased from (5. 36 ± 3. 02)min to (1. 24 ± 2. 89)min (t=7. 63,P<0. 05). Conclusions The application of DVT nursing assessment information model can improve nursing staff members'work efficacy and protect patients'safety.
10.Influences of reading therapy on the improvement of empathy and communication skills of nurses in cancer hospitals

Objective To discuss the influences of reading therapy on empathy and communication skills of nurses in cancer hospital. Methods From January 2016 to January 2017, a total of 60 nurses were recruited voluntarily in 3 phases to participate in reading therapy training, and there were 16 times of training in each course. Before and after training,Jefferson Scale of Empathy-Health Professionals and Nurses' Clinical Communication Competency Scale were used to evaluate the nurses' empathy and communication skills and compare the effects of reading therapy. Results After the training, the total score of nurses' empathy increased from (110.43±13.53) to (118.68±11.93), the score of nurses' clinical communication competency increased from (3.81±0.37) to (4.09±0.38). The differences were statistically significant (t=-3.54,-3.60;P< 0.05). Conclusions Reading therapy can improve the level of empathy and communication skills of nurses in cancer hospitals and promote the harmony of nurses and patients.
