1.Tanreqing Injection in the Treatment of Children's Viral Pneumonia:Observation of Clinical Efficacy

OBJECTIVE:To observe the clinical efficacy of Tanreqing injection in the treatment of children's viral pneumonia.METHODS:A total of sixty children with viral pneumonia were randomly divided into 2 groups:the treatment group(n=30)were treated with Tanreqing injection(20mL)plus Glucose(250mL)qd for 7d while the control group(n=30)with Cibavirin(10~15mg?kg-1)plus Glucose(250mL)bid for 7d.The primary outcome measures were clinical efficacy and adverse drug effects(ADRs).RESULTS:The effective rate was 100% in the treatment group as compared with 80% in the control group,showing significant differences between the two groups(P

AIM:To investigate whether hypoxic preconditioning(HPC)protects cardiomyoblast H9c2 cells against oxidative injury,and to discuss whether calreticulin(CRT)contribute to this protection through p38 MAPK signaling pathway.METHODS:Cardiomyoblast H9c2 cells were randomly divided into eight groups as follows:hydrogen peroxide stress(H2O2);brief hypoxic exposure of 20 min to simulate hypoxic preconditioning(HPC);20 min of hypoxic exposure followed by 24 h of normoxic reoxygenation before hydrogen peroxide stress(HPC+H2O2),SB203580(the specific inhibitors of p38 MAPK)+HPC+H2O2,antisense oligonucleotides transfection of calreticulin(AS),AS+H2O2,AS+HPC+H2O2 and control.Morphological studies,estimation of lactate dehydrogenase(LDH)leakage and flow cytometry were employed to assess the cell apoptosis and necrosis.RT-PCR and Western blotting analysis was used to detect calreticulin expression and phosphorylation of p38 MAPK.RESULTS:The results obtained are as follows:(1)HPC relieved cell injury caused by H2O2.Compared with those in H2O2 group,apoptosis rate and LDH leakage in culture medium in HPC + H2O2 group decreased 13.4% and 44.0%,respectively(P

objective To assess relationship between microvascular changes of hemodynamics of the fingertip and evolution of hypertension in patients with essential hypertension(EH)by enhanced power Doppler imaging(E-Flow).Methods The right ring fingertip'S blood flow peak systolic velocity(PSV),end-diastolic velocity(EDV),resistance index(RI)of finger ventral arteriole and nail bed arteriole in thirty-nine patients with EH were separately observed and were made contrast with 35 age-matched healthy subjects by E-Flow imaging.Results Compared with traditional colour Doppler flow imaging and power Doppler imaging,E-Flow imaging was clearer to demonstrate the distribution and movement of the fingertip'S microvascular.It was shown that the PSV and EDV in the ring finger ventral arteriole and nail bed arteriole of EH were decreased(P<0.05),but the RI increased(P<0.05),along with the evolution of EH.Conclusions Fingertip'S microvascular changes of hemodynamics could be a criterion to detect obstruction alternations of essential hypertension.E-Flow imaging iS superior to traditional color Doppler flow imaging and power Doppler imaging in differentiating microvascular and could provide a new method in assessing microvacular changes of patients with EH for clinic.
4.A brief review on the progress of mouse models of liver cancer

Liver cancer remains one of the leading cause of cancer death in the world.Animal models, especially mouse models, are important tools for studying the biological characteristics, pathogenesis, new drug screening and therapy of liver cancer.Up to now, although the development of various animal models accelerates the research of liver cancer, all the existing models have their own disadvantages.Lacking of economical and applicable animal models that can mimic the human liver cancer seriously restrict the further study of liver cancer.With the development of genetically modified technologies, it provides a fast, easy and reliable method to establish liver cancer models.In this review, we describe the different types of mouse models used in liver cancer research, with emphasis on genetically engineered mice used in this field, which may open an avenue for functional cancer genomics and generation of liver cancer models by using gene editing technologies.
5.High level secretory expression of recombinant human tissue factor in Pichia pastoris

Objective To construct a eukaryotic expression vector in Pichia pastoris containing human tissue factor( hTF) gene,in order to achieve high level secretory expression in extracellular.Methods Expression plasmid, pGAPZaA-hTF, was constructed by inserting the synthesized sequence encoding human extracellular tissue factor into yeast expression vector pGAPZaA and transformed into Pichia pastoris SMD1168H with electroporation.Having been selected by Zeocin, transformants containing hTF cDNA were expressed in YPD, extracellular proteins were detected by SDS-PAGE and confirmed by Western bolt.Results Successfully constructed the recombinant pGAPZaA-hTF expression system in Pichia pastoris.SDS-PAGE showed that the molecular weight of the expression product was about 37 -40 kDa.Western-blot indicated that it was human extracellular tissue factor.The crude yield of total protein in medium was up to 1 g/L, more than 80% of which was hTF.Conclusion Truncated rTF gene is expressed in Pichia pastoris and the active products are secreted into the medium which have the same activation as the native TF.
6.Survey on needs of main caregivers of children with cerebral palsy

Objective To investigate the needs of the main caregivers of children with cerebral palsy and probe into the nursing strategies. Method One hundred and fifteen main caregivers of children with cerebral palsy participated in the survey by a general state questionnaire and the family needs questionnaire. Results The score by the family needs questionnaire was (96.13 ± 19.47), with an average score of (3.00 ± 0.61). And the top needs of the main caregivers were information and econony need. Conclusions The needs of the main caregivers of children with cerebral palsy are high. More attention should be paid to community and family-based rehabilitation and supports to those care-givers.
7.Establishment of hepatocellular carcinoma model in mouse by interventional ultrasound

ObjectiveTo study the feasibility and stability of hepatocellular carcinoma model established in mouse with the guidance of ultrasound, and to explore the value of ultrasonography in hepatocellular carcinoma model.MethodsThe type of H22 cells, whose concentration was 2×107/ml, were injected into the left liver of 30 mice with the guidance of ultrasound. After injection, the mice were examined by ultrasound on the tenth and twentieth day, and implemented laparotomy on the twentieth day. The tumors were examined pathologically. The formation rate and growth of tumors were evaluated, and the ultrasonic features of tumors were observed.ResultsThe formation rate of implanted tumors was 96.7% (29/30), and the peritumoral blood flow could be detected. The diameters measured by ultrasound and explorative laparotomy were (8.4±0.3) mm and (8.6±0.2) mm, respectively (P>0.05). The pathological examination showed that the tumors were hepatocellular carcinoma.ConclusionEstablishing mouse model of hepatocellular carcinoma by ultrasound-guided is simple and minimally invasive, and it provides an ideal model of high formation rate for the study of hepatocellular carcinoma.
8.Value of the time-intensity and gamma curves of the contrast-enhanced ultrasound in diagnosis of hepatocellular carcinoma

Objective To investigate the value and the enhanced mode of time-intensity curve(TIC)combined with gamma curve of contrast-enhanced ultrasound in diagnosis of hepatocellular carcinoma.Methods The ultrasonic contrast images of 34 patients with hepatocellular carcinoma by the gamma curve were obtained by the time before,increased branch slope parameter, curve sharpness, gamma modulus, peak time,peak intensity, the average pre-peak intensity, increased branch slope and pre-peak area.Results Compared with the peripheral hepatic parenchyma, the curve of the tumor tissue ascended as well as declined more rapidly, peaked earlier with a more intensive peak value.There were significant differences between the two groups in all the data mentioned-above (P<0.05 ).Conclusions TIC and gamma fit quantitative analysis can reflect the different perfusion characteristics between hepatocellular carcinoma and normal hepatic parenchyma objectively and accurately,so as to provide the quantitative criterion and basis for the diagnosis and hemodynamic study of the hepatocellular carcinoma.
9.Analysis of the effect of clinical application of vaccine for esophageal cancer

Objective:To analyze the effects of the clinical applications of vaccine for esophageal cancer to provide treatment ba-sis. Methods:Tumor-soluble antigen was extracted from clinical samples of esophageal cancer tissue. Vaccine for esophageal cancer was prepared using antigen and superantigen staphylococcal enterotoxin C (SEC). One group of patients with esophageal cancer was treated with vaccine after surgery (treatment group). Another group of patients with esophageal cancer was treated using routine meth-ods (control group). After five years, the patients were followed up to analyze survival. Results:After five years of follow up, the sur-vival rates of treatment and control groups were 173/328 (49.71%) and 67/326 (20.55%), respectively. Survival rates significantly dif-fered between the two groups (P<0.05). Furthermore, the survival rates of the female patients were higher than those of the male pa-tients (P<0.05). In the treatment group, the survival rates of patients with low-differentiation cancer were higher than those of patients with high-differentiation cancer (P<0.05). In the control group, the survival rates of patients with low-differentiation cancer were lower than those of patients with high-differentiation cancer (P<0.05). Conclusion:Tumor vaccine prepared with esophageal cancer antigen and superantigen SEC could increase the survival rate of patients with esophageal cancer, particularly female patients and those with low-differentiation cancer.
10.Simultaneous Determination of Toxic Alkaloids and Rat Poisons in Toxic Samples by Dispersive Liquid-liquid Micro-extraction Combined with Gas Chromatography-Mass Spectrometry

A novel method for simultaneous determination of 3 rat poisons ( tetramine, bromadiolone, brodifacoum) and 5 toxic alkaloids ( hyoscyamine, scopolamine, gelsemine, strychnine, brucine ) in toxic samples by dispersive liquid-liquid micro-extraction ( DLLME ) coupled with gas chromatography-mass spectrometry was established. A mixture extractant containing 100 μL trichloromethane and 600 μL methanol was injected into the prepared sample to form an emulsion and the extraction process was accomplished. After centrifuged at 8000 r/min for 5 min, the settled drop of trichloromethane solvent was transferred to a conical insert within a GC autosampler vessel, and analyzed by GC-MS. Factors affecting extraction efficiency such as the type and volume of extractant, dispersive agent, extraction time, pH value and salt concentration of extraction system were studied. The limits of detection(LODs) were from 0. 003 to 1 μg/L in water sample, urine sample and rice wine sample. LODs were from 0. 002 to 0. 2 μg/kg in rice sample. The recoveries of toxic samples were in the range of 81. 0%-110%. The relative standard deviations( RSDs) were lower than 7%. The proposed method was sensitive, effective, and suitable for the simultaneous determination of toxic alkaloids and rat poisons in food poisoning sample.
