Objective To detect the effects of siRNA targeting CDX2 gene expression on of BCR-ABL, caspase and Bax expressions, and the mechanisms thereof. Methods According to the earlier experiments, siRNA specifically targeting CDX2 gene (CDX2-siRNA) and the negative control sequence (CDX2-siRNA-NC) were selected, and then were transfected into K562 cells by Roche X-tremeGENE HP DNA Transfection Reagent. The flow cytometry analysis was used to detect the effects of siRNA on cell apoptosis. The expressions of BCR-ABL, caspase-9, Bax mRNA and protein were tested by RT-PCR and Western blot assay. Results MTT and flow cytometry analysis showed that after the silence of CDX2 gene expression, the proliferation of K562 cells was prohibited and the apoptotic rate of K562 cells was distinctly increased compared with that of normal cell group, but the negative control group had no significant change. According to the RT-PCR and Western blot assay, in comparison with the normal cell group and the negative control group, the expression levels of BCR-ABL mRNA and protein were obviously decreased, and the difference was statistic significance. On the other hand, the expressions of caspase-9 and Bax mRNA and protein were significantly higher than those of other two groups (P<0.05). Conclusion CDX2-siRNA can promote apoptosis of K562 cells obviously, and the mechanism is related with the down-regulation of BCR-ABL and the up-regulation of caspase-9 and Bax.

AIM: To study the distribution of nitric oxide synthase (NOS) in normal lung of rats, and the changes in distribution and activity of NOS in lung of rats suffering from limb ischemia reperfusion (LIR). METHODS: Using tourniquet, the model rats undergoing ischemia reperfusion were made. Using ?-ANDPH-d histochemical method, computer analysis of image and spectrophotography, the distribution of NOS in normal lungs of rats, and the changes in distributon and activity of NOS in lung of rats suffering from limb ischemia reperfusion (LIR) were observed. RESULTS: Histochemically, in normal rats, the NOS positive staining was found in epithelial cells of respiratory duct including bronchi, alveolar duct as well as in endothelium of pulmonary vessels, and the negative staining was found in pneumonocytes. In LIR rats, the above NOS staining were significantly stronger than that in normal one, and positive staining was also found in vascular smooth muscle and pneumonocytes. Biochemically, the results showed that the values of lung NOS, NO in LIR group increased significantly compared with the normal control. CONCLUSION: NO may play an important role not only in physiology of lung but also in pathophysiology of acute lung injury following LIR. [

9.PACK-CXL:corneal collagen cross-linking for infectious keratitis
Chinese Journal of Experimental Ophthalmology 2018;36(7):570-575
Infectious keratitis is one of the leading causes of irreversible vision loss. With the emergence of multidrug-resistant pathogen, despite appropriate diagnosis and antimicrobial treatment, infectious keratitis can still result in corneal opacity and even melting and perforation. Therefore,it is imperative to develop novel treatment for infectious keratitis.More recently, several articles have demonstrated the effectiveness of collagen cross-linking (CXL) in treating infectious keratitis via direct antimicrobial activity and inhibition of corneal enzymatic degradation. In order to highlight the use of CXL tor infectious keratitis treatment,a new term was proposed at the 9th CXL congress to rename its use in infections as photoactivated chromophore for infectious keratitis-CXL (PACK-CXL).Based on the current literature,PACK-CXL appears to be effective for bacterial keratitis, especially on inhibiting corneal perforation. The effect was limited for fungal and amoebic keratitis, and maybe aggravative on viral keratitis. In this article,we will review the basic profile, technical operation, prevention of complication of CXL,the principle mechanism of PACK-CXL and its clinical application for bacterial, fungal keratitis, amoebic and viral keratitis.
10.Uncovering the etiology of developmental dyslexia and sights for localized diagnosis and intervention system
Chinese Journal of School Health 2019;40(10):1445-1448
Abstract
Developmental dyslexia (DD) is a common learning disability that can cause long-term psychological and physical damage to children. However, up to now, the underlying mechanism of DD is still unclear, and there is no gold standard diagnosis and treatment measures. Compared with western countries, research on Chinese DD is lagging behind, which greatly affects the prognosis of Chinese DD children. Extensive research on DD is needed for developing a diagnostic system suitable for children with developmental disorders in China.
