1.THE ANTITUMOR ACTION OF ADRIAMYCIN-POLY I:C COMPLEX IN VIVO AND IN VITRO

The binding of adriamycin to poly I:C, a double-stranded thetic homoribopolymor,and antitumor activity of adriamycin-Poly I:C complex were studied. Results showed that the affinity for adriamycin to poly I:C is no linear relationship. Adriamycin binding rate( % ) = 20.6 + 0.2268?Poly I:C(W)/adriamycin(W)? The increased therapeu-tic effect of the complex was comparable to that of the free and the Poly I:C alone on bearing Sl80 mice. In vitro cytotoxic effect of the complex is lower than free adriamycin following lh exposure. The complex is recommended for clinical therapeutic studies.
2.Clinical significance of measuring angel of lesser pancreas in diagnosis of neoplasm in the head of pancreas

Objective To evaluate the clinical significance of measuring angel of lesser pancreas(ALP) by ultrasound in diagnosis of neoplasm in the head of pancreas.Methods ALP was measured by ultrasound in 41 cases of pancreatic neoplasm,and in 52 normal cases as contrast.All of the cases were divided into three groups: group A,32 cases which were proven having mass in the head of pancreas by ultrasound; group B,9 cases which were discovered mass in the head of pancreas by CT or MRI,but not diagnosed by ultrasound; group C,52 cases as contract without presence of any mass in the head of pancreas.Results The mean value of ALP in group A,group B and group C was 83??9?,78??9?,and 43??5?, respectively.The angel in group A and group B was significantly wider than that in group C (P60?.Conclusions The widing of ALP by ultrasound is a sensitive sign for mass in the head of pancreas.

Among the different x-ray features of breast carcinoma, mass and calcification are the most significant reference parameters in the screening and diagnosis of early carcinoma. Mammographic follow-up after conservation therapy is the other important value of mammography for breast carcinoma. Calcification is the most important sign to show residue or recurrence of conservation therapy.
4.Determination of Norcantharidin using high performance liquid chromatography

ABSTRACT AIM To determine Norcantharidin in its tables. METHODS A PHLC method was adopted, using Polaris C18 column(5?,4.6?250 mm) and UV detector at 211 nm wavelength. The mobile phase is consisted of water: methanol(85 : 15 V/V), adjust the pH to 3.1 with phosphoric acid. RESULT Excellent liner relationship was ob- tained from the range of 25 to 1 000 mg ? L-1. The minimal limit of Norcantharidin was 0.2 mg ? L-1. Recovery was 100.84% and RDS was 1.335%. COUCLUSION Practice suggested that this method was rapid, accuracy and suitable for Norcantharidin determining.
5.THE EXPRESSION OF LYSOZYME IN CARCINOMA OF AMPULLA CORRELATED WITH INVASION,METASTASIS AND PROGNOSIS

Objective:To investigate the expression of lysozyme in ampullary carcinoma, and its relation with invasion, metastasis and prognosis. Methods: 36 cases were analyzed with immunohistochemistry of lysozyme. Results: The lysozyme-positive rate in patients with lymph node metastasis (30%) was significantly lower than that in patients without metastasis (71%), while that in patients with advanced cancer invading the pancreas (35%), was significantly lower than that in patients with cancer without invasion of the pancreas. The Survival rate of lysozyme-positive patients (66.7%) was significantly higher than that of lysozyme-negative patients (14.3%). Conclusion: The expression of lysozyme in carcinoma of ampulla is associated with invasion, metastasis and prognosis. Lysozyme is important in typing of cancer of ampulla.
6.In vitro effects of sonodynamic management on inhibition of ovarian cancer cells

Objective To study the in vitro effects of sonodynamic management on human ovarian cancer cell line HRA. Methods Hematoporphyrin was selected as a sonosensitizer and ultrasound with certain intensity was used to activate hematoporphyrin.Then MTT assay and clony-forming efficiency assay were applied to determine the growth inhibitory effects of ovarian cancer cells. Electron microscope was applied to detect the morphological changes of the cells. Cell apoptosis was analyzed using flow cytometry. Results Hematoporphyrin had no significant inhibitory effects on the cells. However, when hematoporphyrin was used before ultrasound,it could enhance the killing effects of ultrasound(P
7.Application research on Disease Severity Assessment Scale in dividing nursing level of patients with different diseases

Objective To explore clinical practice value of Disease Severity Evaluation Scale (DSAS), and divide patient's nursing level according to the results of DSAS. Methods The assessment group was set up for evaluation of the scale, DSAS was used to determine the nursing level of 3 713 hospitalized patients (age > 18 years old) from 17 clinical departments, from April to May 2014. Results Nursing level determined by DSAS was compared with the standard nursing level, kappa coefficient was 0.72, P < 0.05, suggesting that the consistency was better. According to the kappa coefficient, 17 clinical departments could be divided into three categories, namely the applicability of DSAS were different in different departments. Conclusions DSAS can be used as a objective basis for division of nursing level and has a certain clinical significance. But in order to make it guide clinical work better, scoring system with department characteristic should be developed on the basis of DSAS.
8.Screening of the pORF3 binding protein of HEV in human liver cells by using yeast double hybridization system

HEV is a positive strand RNA virus containing 3 open reading frames(ORFs), while the function of its binding protein pORF3 is poorly understood. In our study, the interactive protein of the pORF3 from human liver cDNA library was screened by the SOS recruitment system, and the construction of the pSOS bait-ORF3 plasmid was identified. The expression of the pORF3 was analyzed by Western blotting, and the detection result of the self activation of bait protein was obtained as expected. Totally, 8 putative interacting proteins had been successively screened. Some of these cellular binding proteins of pORF3, especially the important immunological pathway kinase p110δ, may advance our understanding of the role in ORF3 protein of HEV, and the possible implications of these candidate proteins for the further research are discussed.
9.Simple and effective method for monitoring ambulatory electrocardio-gram in toads

[ ABSTRACT] AIM:To screen out a suitable lead for monitoring the ambulatory electrocardiogram ( ECG) in un-restrained toad, and to investigate its practicability.METHODS:After subcutaneously implanting the electrodes in toads under anaesthesia, the ambulatory ECG of 5 leads were monitored with BL-420S data acquisition and analysis system, and the leads which could well express the waveform in ECG were screened out.The recovery process of the toads from the arti-ficial hibernation within 6 h, the day-to-day stability of the heart rate ( HR) and the heart rate variability ( HRV) in 5 suc-cessive days of hibernation, and the HR and HRV after freeze-thawing process were monitored to determine its practicabili-ty.RESULTS:Two out of 5 leads showed better ECG waveforms.Compared with 6 h post hibernation, lowered HR at 0 h and 1 h was observed, and the standard deviation of normal R-R intervals ( SDNN) was significantly increased ( P<0.05 or P<0.01), but the HR and SDNN from 2 to 5 h showed no significant difference, suggesting that the cardiac function reached the steady state after 2 h recovery.The HR at 2 h and 4 h on day 4 and day 5 decreased significantly compared with that on day 1 (P<0.05 or P<0.01), followed with a significant increase in SDNN (P<0.05 or P<0.01), sugges-ting that the ECG remained stable within 3 d.The HR increased, while SDNN decreased significantly at 1 h and 12 h post-thawing compared with that at pre-freeze (P<0.05), indicating the damaged cardiac function after freeze-thawing process. CONCLUSION:The method of subcutaneously implanting electrodes is suitable for effectively monitoring the ambulatory ECG in toads.
10.Cloning, expression and functional analysis of the dhbC gene from the siderophore producing bacterium Bacillus subtilis CAS15.

We amplified dhbC gene from the siderophore producing bacterium CAS15 by PCR. After ligated the PCR product to pMD18-T vector and then sequenced, we obtained a 1197 bp fragment. The blast result showed that the nucleotide acids of dhbC gene (Accession No. FJ194456) of CAS15 shared 99.7% identity with that of dhbC gene of Bacillus subtilis (GenBank Accession No. Z99120), and was predicted to encode a 43.8 kD polypeptide with 398 amino acid residues. We cloned the dhbC gene into expression vector pET-30a(+) and then transformed into Escherichia coli BL21(DE3) via calcium chloride transformation method, and obtained the recombinant E. coli BL21(DE3)/pET-30a-dhbC. Induced by 1 mmol/L IPTG the fusion protein 6His-DhbC, a 48.8 kD polypeptide was successfully expressed mainly in soluble form in E. coli BL21(DE3), and the amount reached highest at 30 degrees C for 4 h. According to the N-terminal fusion 6 His-tag, we purified the recombinant polypeptide by Ni2+ metal affinity chromatography and finally identified it by Western blotting. The result indicated that the recombinant DhbC had the antigenicity to rabbit anti-his-tag polyclonal antibody, which provides the basis for the study of practical utilization in production and the biocontrol mechanism of B. subtilis. Finally, we deleted dhbC gene by gene knockout and then retransformed it into the dhbC gene-delected mutant, which confirmed that dhbC gene play an important role in siderophore biosynthesis.
