1.Development of an Enhanced Colloidal Gold Immunochromatographic Assay for Detection of Imidaclothiz

An enhanced gold immunochromatographic assay ( GICA ) with simplicity, rapidity and high sensitivity was developed to detect imidaclothiz by using the high affinity between biotin and streptavidin. The 13-nm AuNPs were double-labeled with anti-imidaclothiz antibody and biotinylated DNA, and the 41-nm AuNPs were labeled with streptavidin to prepare an enhanced gold immunochromatographic test strip for imidaclothiz. The working conditions of the strip were systematically optimized, and the sensitivity, specificity, precision and accuracy were assessed by testing the cross-reactivity ( CR) , spiked recovery and validation with high performance liquid chromatography ( HPLC) . Under the optimal conditions, the detection could be completed in 10 min with visual result, and the limit of detection ( LOD ) was 25 ng/mL. The analysis showed no cross-reactivity with analogues of imidaclothiz except for imidacloprid. The detection results of GICA agreed with the spiked concentrations of imidaclothiz at spiked levels of 0 . 05 , 0 . 5 and 5 μg/g in river water, rice, cucumber, tomato, pear, cabbage and apple samples. The detection results of GICA for imidaclothiz in unknown concentration river water and pear samples were consistent with that of HPLC.

4.Production and identification of monoclonal antibodies against pesticide imidacloprid.

To produce high-affinity monoclonal antibodies against pesticide imidacloprid, we synthesized the haptens 1-[(6-Carboxylethylthio-3-pyridinyl) methyl] -N-nitro-imidazolidinimine (named as H1) and 1-[(6-Chloro-3-pyridinyl) methyl]-3-carboxylpropyl-N-nitro-2-imidazolidinimine (termed as H2). And then the haptens were coupled to bovine serum albumin (BSA) and ovalbumin (OVA) for immunogen (H1-BSA) and coating antigen (H2-OVA) respectively by NHS ester method. BALB/c mice were immunized with H1-BSA conjugate. We obtained two hybridoma cell lines 2F11/A9 and 2G6/G12 secreting antibody specific for imidacloprid from the conventional hybridoma technology. The result showed that the subtypes of obtained monoclonal antibodies were IgG3 and IgG1, respectively, and the titers of ascites were up to 1:128 000. The indirect competitive ELISA indicated the IC50 values of 5.3 and 28.3 ng/mL with detection limits of 1.1 ng/mL and 7.7 ng/mL, respectively. Two monoclonal antibodies had no apparent cross reactivity with six analogous compounds. Thus, two prepared monoclonal antibodies had a very high affinity and specificity, and it could be used to develop ELISA for rapid determination of imidacloprid residue and laid a solid foundation for research and development of products for immunoassay.

5.Psychological hindrance and its treatment in chronic prostatitis patients.

OBJECTIVETo improve the knowledge of psychological hindrance in chronic prostatitis (CP) patients and study the treatment strategy of it.
METHODSTwo hundred and fifty-eight CP patients with psychological hindrance, aged 16 to 65 years (average 33 years), were divided into the psychotherapy group (148 cases) and the control (110 cases). The control were only treated with antibiotics and Chinese traditional medicines, while the psychotherapy group were treated with cognitive therapy besides the above medicines.
RESULTSThe symptoms in the psychotherapy group were improved obviously compared with those in the control. Both the cure rate (P < 0.05) and the effective rate (P < 0.001) of the psychotherapy group were higher than those of the control.
CONCLUSIONPsychotherapy is an effective way to treat CP patient with psychological hindrance.

6.Comparative Study on the Topological Attributes of Deficiency and Excess Pattern of Cerebral Small Vessel Disease Mild Cognitive Impairment Based on fMRI Graph Theory Network

Objective This study aimed to explore the differences in global and local brain network topological properties between deficient pattern(DP)and excess pattern(EP)of mild vascular cognitive impairment caused by subcortical small vessel disease based on graph theory network.Methods Patients were recruited prospectively and were classified with DP and EP subtype.The global small-world topological attributes and local nodes were calculated for the comparison of DP,EP,and healthy controls(CN)using the GRETNA platform.Results The three groups all had small-world attributes,but only the patients in EP had a significantly lower small world attribute δ in the range of 0.05-0.26 than the control group(P<0.05).The node efficiency and node strength indicators of multiple brain region were able to significantly distinguish the DP group from the EP group.However,there was no positive brain region in the node efficiency of the DP patients(P>0.05),and only a few brain regions showed increased node strength efficiency(P<0.05).Conclusion The results indicate that the syndrome of DP and EP have significantly different neuroimaging phenotypes,providing a basis for further research of biological classification based on Chinese Medicine syndromes.
7.PDGF-B immunogen preparation and the suppressive effect of anti-PDGF-B ascite antibody on the proliferation of hepG2 cells.

To observe the immunogenicity of hPDGF-B immunogens that were synthesized with the fusional expression vector pET28-Trx and to test the suppressive effect of these specific antibodies induced by both of immunogens on proliferation of human HepG2 hepatoma cells. First, we chose 2 antigenic epitopes hPDGF-BΔ103-118aa and hPDGF-BΔ152-167aa from human PDGF-B and inserted these 2 coding regions into the empty vector plasmid pET28-Trx, separately. Second, mice were immunized with purified recombinant proteins to generate polyclonal antibody. Then we intraperitoneally injected mice bearing hepatoma 22 (H22) tumor cells to prepare antibody ascites. ELISA and Western blot were used to detect the titer and the utility of the antibody, respectively. Finally, HepG2 cells were exposed to PDGF-BB protein or anti-PDGF-B ascite antibody in different dilution concentrations groups and the proliferation of HepG2 cells was quantified by CCK8 assay. As the results, we identified mice that could produce high drop of neutralizing antibodies against hPDGF-B induced by both two recombinant proteins. Two anti-PDGF-B ascite antibodies could markedly inhibit the proliferation of HepG2 cells by blocking the stimulating effect of PDGF-BB protein. Our results suggest that Trx-PDGF-B recombinant protein as immunogen provides a new method for the preparation of PDGF-B vaccine, and also a new idea for the treatment of hepatocellular carcinoma in clinical practice.
