1.Effect of BRL-37344 on ion channel in guinea pig cardiac myocytes

Objective To study the biological effect of ?3 adrenoceptor-activated electrophysiological activities in cardiac myocytes. Methods Changes of transient outward potassium ( Ito) and L-type calcium current ( ICa-L) in primary cultured guinea pig cardiacmyocytes were detected with the standard whole-cell patch clamp technique about 5 -10 min before and after administration of ?3 adrenoceptor agonist,4--2-hydroxy-( 3-chlorophenyl) ethyl-aminopropylphenoxyacetate ( BRL-37344; BRL). Results BRL could significantly increased the Ito in guinea pig cardiac myocytes with the I-V curve up-moved. When the membrane potential reached to + 80 mV,the density current increased from 6. 11 ? 1. 03 pA/pF to 8. 46 ? 2. 07 pA/pF ( n = 6,P =0. 013 064). BRL ( 10 -6mol/L) could significantly increase the ICa-Lin guinea pig cardiac myocytes,which was ( 2. 30 ?0. 75) -fold higher than in control group( n =5,P =0. 0063). When the membrane potential increased to + 10 mV ,the current,increased from 89. 25 ? 17. 83 pA to 205. 00 ? 72. 24 pA ( n = 5,P = 0. 006 3). Conclusion BRL-37344 can increase the transient Itoand ICa-L,thus further regulating the cardiac activities.

4.Exploration on the efficacies of Yoga on neurosis patients in hospital

Objective To explore the efficacies of Yoga on neurosis patients. Methods A total of 102 neurosis inpatients were divided into a study group and a control group. The patients in the control group were in treatment only using drug. The patients in the study group were guided to practice the Yoga exercise for 1 month in addition to the treatment of drugs. The efficacies of Yoga were surveyed by NOSIE (Nurses' Observation Scale for Inpatient Evaluation) and HAMD (Hamilton Depression Rating Scale) before and after the exercise. Results After 1 month of exercise in the study group, the NOSIE results showed that, scores of social ability (37.75±2.23), social interesting (17.54±5.14), personal health factors (28.37±3.13) were higher than those before Yoga exercises, especially the score of social interesting was higher than that before Yoga exercises (P<0.01). Scores of irritability, delay and depression factors were lower than those before Yoga exercises (P<0.01). Compared with the control group, there were significant differences in the decrease of the scores of HA MD and NOSIE (P<0.01). Conclusions Yoga is beneficial to body function, psychological function and social function of neurosis patients.

6.Impurity removal technology of Tongan injection in liquid preparation process.

In order to effectively remove the invalid impurities in Tongan injection, optimize the optimal parameters of the impurity removal technology of liquid mixing process, in this paper, taking Tongan injection as the research object, with the contents of celandine alkali, and sinomenine, solids reduction efficiency, and related substances inspection as the evaluation indexes, the removal of impurities and related substances by the combined process of refrigeration, coction and activated carbon adsorption were investigated, the feasibility of the impurity removal method was definited and the process parameters were optimized. The optimized process parameters were as follows: refrigerated for 36 h, boiled for 15 min, activated carbon dosage of 0.3%, temperature 100 degrees C, adsorption time 10 min. It can effectively remove the tannin, and other impurities, thus ensure the quality and safety of products.

Objective To investigate effects of atorvastatin on the development from macrophages (HMDM) to foam cells.Methods Monocytes were isolated from human peripheral blood by Ficoll-Hypaque density gradient centrifugation and plastic adsorptive process.The isolated cells were stimulated by phorbol 12-myristate 13-acetate (PMA) (50 nmol/L) for 48 h and transformed to macrophages.Macrophages were co-incubated with 80 mg/L ox- idized low density lipoprotein (ox-LDL) and atorvastatin (0-100 ?mol/L),respectively for 0,6,12 and 24 h. Total cholesterol (TC),free cholesterol (FC) and protein (Pro) in cultured cells were quantitatively analyzed by high performance chromatography (HPLC) analysis and modified Lowry protein assay.Results When macropha- ges were incubated with 80 mg/L ox-LDL,the ratio of TC/Pro was greater than 20,and large amount of lipid drop- lets were displayed indicating the formation of foam cells.Atorvastatin decreased TC/Pro ratio in foam cells in a concentration and time dependent manner (0-100 ?mol/L)(P

Aim To investigate the effect of hypercholesterolemia on L-type Ca2+(ICa-L) current and intracellular calcium concentration ([Ca2+]i) in single ventricular myocytes of hypercholesterolemic rats.Methods 12 male wistar rats were randomly divided into two dietary groups:a group fed a normal diet(n=6)and a group fed high-cholesterol diet(n=6) for 4 weeks,respectively. The level of serum lipid was examined.Zymolytic method was used to isolate single ventricular myocytes from hypercholesterolemic and normal rats,which were loaded with Ca2+-sensitive fluorescent indicator Fluo-3/AM.[Ca2+]i represented by fluorescent intensity(FI)was measured by laser scanning confocal microscope.Whole cell patch clamp technique was used to record ICa-L.Results There was no significant influence exhibited on TG level.However, the serum total cholesterol(TC)level of hypercholesterolemic rats was much higher than that of model control group; at the test potential of 0 mV, ICa-L decreased from(-8.56?1.29)pA/pF(Control)to(-5.24?0.90) pA/pF(HC)(n=6 in each group,P
10.Animal study for airway inflammation triggered by gastroesophageal reflux.

BACKGROUNDGastroesophageal reflux disease with extra-esophageal symptoms, especially those with respiratory distress was attracting more and more attention. The related mechanisms were still in controversy. The purpose of the work was to explore airway inflammation triggered by gastroesophageal reflux.
METHODSSixteen Sprague-Dawley rats were used as study group and 9 as control. In the study group, a plastic extender with a trumpet-shaped distal end was inserted into the lower esophagus to dilate the cardia, the pylorus was ligated. One ml of 0.1 mol/L hydrochloric acid was injected into the stomach. While a simple laparotomy was performed for control animals. All animals from two groups were sacrificed 24 hours after operation. Then tracheotomy was carried and the bronchoalveolar lavage fluid was collected in all animals. Cells in the fluid were counted and levels of interleukin (IL)-5, -6, -8 in it were measured.
RESULTSCompared with control group, the study group presented a neutrophil pattern of airway inflammation and an elevated concentration of IL-5, -6, -8 with no significant difference regarding eosinophil count.
CONCLUSIONThe gastroesophageal reflux-triggered airway inflammation is characterized by a neutrophilic airway inflammation which differed from that caused by asthma, and enhanced levels of IL-5, -6 and -8, which are similar to that caused by asthma.
