2.Effects of shenmai injection on lipid peroxidation in the lung following with ischemia/ reperfusion (I/R) injury of limb.

AIMTo explore the effects of shenmai (Chinese transitional medicine) injection on lipid peroxidation in the lung following with ischemia/reperfusion (I/R) injury of limb.
METHODSThe models of I/R injury of limb were constructed in rabbits. Superoxide dismutase (SOD) and malondialdehyde (MDA) in into and out-flowing pulmonary blood (IPB, OPB) and lung tissue were measured, as well as the effects of shenmai injection were observed.
RESULTSCompared with sham group, the activity of SOD in IPB, OPB and lung tissue were decreased, and the content of MDA was increased after 4 h ischemia followed by 4 h reperfusion. SOD increased and MDA decreased significantly by icy shenmai injection 30 min before reperfusion. The correlation analysis indicated that MDA was negatively correlated with SOD .
CONCLUSIONOxygen free radicals metabolic confusion of lung occurred in the course of I/R, shenmai injection can alleviate lipid peroxidation, get rid of free radicals and inhibit the damage of lung.

3.Cis-CA1P inhibits tumor cell proliferation and prevents blood vessel formation.

OBJECTIVETo investigate the effects of cis-combretastatin-A1 phosphate (cis-CA1P) on tumor cell proliferation, and its effects on the blood vessel formations.
METHODSMTT and IC50 values were used to assess the inhibitory effects of cis-CA1P on tumor cell proliferation. Chicken embryo chorioallantoic membrane and thoracic aorta annulations isolated from rats were used to investigate the effects of cis-CAIP on the blood vessel formation.
RESULTSCis-CA1P concentration-dependently inhibited the proliferations of several cancer cell lines, including human gastric carcinoma cell line MGC-803, human leukemic monocyte lymphoma cell line U937, human melanoma cell line A375, human colon cancer cell line HCT116, human breast carcinoma cell line MDA-MB-231, and human leukemia cell line K562. Cis-CAIP significantly decreased the formation of blood vessels in chicken embryo chorioallantoic membrane and in thoracic aorta annulations.
CONCLUSIONCis-CA1P inhibits cancer cell proliferation and prevents blood vessel formation.

4.Study on the effect and mechanism of agmatine on blood brain barrier with cerebral ischemic reperfusion injury rats

Objective To observe the effect of agmatine on brain ede-ma and examine the relation between blood brain barrier ( BBB) per-meability and aquaporin 4 ( AQP4 ) and aquaporin 9 ( AQP9 ) in rats with cerebral ischemic reperfusion.Methods Health male Sprague-Dawley rats were randomly divided into sham group , model group and test ( agmatine ) group.And 2 h after middle cerebral artery occlusion, giving agmatine ( 50 mg · kg -1 ) in test group or 0.9%NaCl in sham and model groups by intraperitoneal injection, respec-tively.Brain edema and the size of the brain infarct were assayed by brain water content and BBB permeability and 2,3,5 -triphenyltet-razoliumchloridstaining( TTC).The morphological changes of neurons were observed by hematoxylin and eosin staining and transmission electron microscopy.And the expression of AQP4 and AQP9 were assessed by Western -blotting.Results The BBB permeability de-creased significantly and the morphological changes of neurons were reduced significantly and the expression of AQP4 and AQP9 decreased significantly in rats with agmatine.Conclusion Agmatine reduce the injury of the BBB by decreasing the expression of AQP4 and AQP9 , and relieve the neuron injury induced by brain edema.

6.HPA distribution characteristics of platelet donor population in Mudanjiang area of China and establishment of its database.

This study was aimed to explore the distribution characteristics of the human platelet antigen (HPA) gene of human platelet donors and its polymorphism in Mudanjiang area of Heilongjiang Province in China, to determine platelet antigen system with clinical significance by judging the rate of incompatibility of HPA, as well as to establish a database of donors' HPA. The genotyping of 154 unrelated platelet donors was performed by means of PCR-SSP. The frequencies of gene and genotype were calculated and compared with that in other areas. The results showed that the genes 1a-17a of HPA-a were all expressed in the 154 healthy and unrelated platelet donors. Only genes 1b, 2b, 3b, 5b, 6b and 15b of HPA-b were expressed while genes 4b, 7b-14b, 16b were not expressed. Among the genotypes, aa homozygosity was predominant and HPA15 had the greatest heterozygosity, while HPA3 had lower heterozygosity. There were 23 combined types of HPA, 5 of them had a rate higher than 10%, and the frequencies of the other 18 were lower than 8%. HPA genotype frequencies showed a good consistency to Hardy-Weinberg equilibrium. It is concluded that the distribution of the allele polymorphism of HPA1-HPA17 in Mudanjiang area has its own characteristics, compared with other areas and some countries, the local HPA genotype database of platelet donors is established in Mudanjiang area, which can provide the matching donors for clinical use with immunological significance.

7.Detection of fetal short tandem repeat genotype in maternal plasma by multiplex fluorescent polymerase chain reaction.

OBJECTIVETo study the feasibility of using free fetal DNA in maternal plasma for non-invasive prenatal diagnosis.
METHODSMaternal DNA extracted from plasma samples of 10 pregnant women at early pregnancy, medium pregnancy and late pregnancy and their husband's DNA isolated from whole blood samples were used to detect genotype by multiplex fluorescent PCR at nine different polymorphic short tandem repeat(STR) loci.
RESULTSFetus-specific alleles were found in maternal plasma samples studied. By the application of these polymorphic short repeat sequences, every pregnant women/husband pair was informative in at least three of nine loci. Paternally inherited fetal alleles were detected in 23 of 30 plasma samples. They are 6/10 cases in early pregnancy, 8/10 cases in middle pregnancy and 9/10 cases in late pregnancy respectively.
CONCLUSIONFluorescent multiplex PCR can be used for amplification of male and female fetal STRs in maternal plasma to obtain genetic information, which may have implication for non-invasive prenatal diagnosis of certain hereditary diseases independent of the fetal sex.

8.The affection and significance of NO on the expression of P-selectin in renal injury following hind limb ischemia/reperfusion in rats.

AIMTo probe into the affection and significance of NO on the expression of P-selectin in renal injury following hind limb ischemia/reperfusion in rats.
METHODSIn accordance with the conventional approaches of our department, the model rats were prepared after they were made to undergo 4 hours or ischemia followed by 4 hours of reperfusion of hind limbs. The Wistar rats were divided into four groups randomly: Control group, LI/R group, L-Arg group and L-NAME group. And then in those four groups of Wistar rats, a series of values of measurement were determined such as: Plasma concentrations of nitric oxide (NO), blood urea nitrogen (BUN) and creatinine (Cr). Furthermore, biochemically there came to the assessment of the values including myeloperoxidase (MPO), NO, total nitric oxide synthase (tNOS), inducible NOS (iNOS) and constitutive NOS (cNOS) of renal tissue in different groups. By the methods of electrophoresis and biochemistry, the urine protein was mensurated. The immunohistochemical method was used to detect the expression of P-selectin protein. The morphologic changes were observed with a microscope.
RESULTSAfter hind limbs had suffered from ischemia/reperfusion for 4 hours, there was the occurrence of a series of results such as in the following which were based on the comparison between plasm of LI/R group and control group. The values of NO, BUN and Cr increased significantly, and the trend of indexes such as NO in renal tissue was similar to that in plasma. The values of MPO, tNOS and iNOS in renal tissue all increased significantly after reperfusion, while cNOS decreased distinctly. The urine protein appeared, especially large molecular weight protein. Renal pathology revealed that after LI/R there were edema and infiltration of polymorphonuclear neutrophil (PMN). Immunohistochemically, the expression of P-selectin was upregulated significantly. Compared with LI/R rats, all injury changes were alleviated in L-Arg group. Morphologic changes were mild. Both the content of urine protein and the percentage of apoptosis cell decreased. The expression of P-selectin was downregulated. In L-NAME group, all injury changes got worse. Immunohistochemical results showed strong positive staining of P-selectin.
CONCLUSIONThe renal injury after LI/R may relate to the strong expression of P-selectin. NO may have protective affection by decreasing the expression of P-selectin and alleviating the adhesion, aggregation and infiltration of neutrophils.

9.Healthcare-associated infection and drug resistance of Acinetobacter baumannii in neonatal intensive care units

Objective To investigate the clinical characteristics and antimicrobial resistance of Acinetobacter baumannii(A.baumannii) in neonatal intensive care units (NICUs).Methods The clinical isolation and antimicrobial resistance of A.baumannii causing healthcare-associated infection(HAD in 4 NICUs of a hospital from October 2012 to October 2014 were analyzed statistically.Results A total of 11 640 neonates were admitted in 4 NICUs,500(4.3 %) developed HAI,51 (10.2 %) developed 52 cases of A.baumannii infection.Distribution of A.baumannii infection was as follows:NICU of extremely premature infants,premature infants,full-term infants,and surgical NICU were 42,1,4,and 5 cases respectively.Incidences of A.baumannii HAI in 4 seasons were compared,difference was statistically significant(x2 =16.05,P<0.05),infection mainly occurred in the spring and summer.A.baumannii had high resistance rates to β-1actam antibiotics (such as piperacillin/sulbactam,cefepime,imipenem)and gentamycin(>90 %),resistance rate to amikacin was the lowest (51.9 %).Among 52 strains of A.baumannii,46 were multidrug-resistant strains,and 3 were extensively drug-resistant strains.Conclusion A.baumannii HAI is most serious in NICU of extremely premature infants,resistance rates to commonly used antimicrobial agents are high.
10.Epidemiological characteristics and related influencing factors on Japanese encephalitis in Hainan province

Objective To understand the epidemiological characteristics and the influencing factors of Japanese encephalitis (Abbreviated JE) at different periods in Hainan province, and to provide evidence for JE control strategies. Methods Data on JE had been collected since 1952 and related incidence, mortality, fatality rates of JE and the rate of JE vaccination were calculated for four periods: the natural epidemic period (1952-1978), period for inactivated JE vaccine promotion (1979-1986), national immunization program period (1987-1990) and the promotion of live-attenuated JE vaccine period (1991-2007). History on the immunity of JE and data from the monitoring programs on cases and insect vectors were also analyzed. Results The average annual incidence in natural epidemic period was 7.12/100 000, which was also the highest among the 4 periods with the mortality rate as 0.89/100 000. The average annual incidence was 0.11/100 000 which was the lowest while the mortality rate was 0.01/100 000 in the period when live-attenuated JE vaccination was under promotion. Since the establishment of JE surveillance system was set up in Hainan province in 2002, the specimen collection rate of JE reported suspected cases had reached 92.8%, while the incidence of confirmed JE cases fluctuated between 0.05/100 000 and 0.2/100 000. Of the confirmed cases, 86% did not have clear history of JE vaccination. The annual average protective rate reached 95.69% in 2004-2007. The density of insect vectors was high in Hainan province. The infection rate of JE in host animal swine, kept rising from April to October, and in some surveillance points, the rate reached 99.3%. The density of insect vector plotted in trans-curve with the infection of host animals. Conclusion The application of live-attenuated JE vaccine was found to have had significant protective effect on JE in susceptible children. It helped reducing the incidence and controlling the epidemics of JE. It appears feasible to include live-attenuated JE vaccine into the expanded immunization program.
