Journal of Shanghai Jiaotong University(Medical Science) 2006;0(09):-
Objective To explore the effects of thiamine and riboflavin on H2O2-induced DNA oxidative damage in human umbilical vein endothelial cell line ECV304.Methods ECV304 cells were incubated with 10,100,500,1000 mg/L of thiamine or 20,100,300,500 nmol/L of riboflavin for 24 h,and then oxidative damage of cells were induced by 25 mol/L H2O2 for 30 min.DNA damage was detected with single cell gel electrophoresis(SCGE)assay.ECV304 cells incubated without H2O2,thiamine and riboflavin were served as negative controls, and those incubated with H2O2 and without thiamine and riboflavin were served as positive controls. Results H2O2 induced DNA damage, and the indices of percent of DNA damage cells, percent of tail DNA,tail length and Olive tail moment were increased. The indices of cells pretreated with 10,100,500 mg/L of thiamine or 20,100,300 nmol/L riboflavin were significantly decreased(P0.05).Conclusion Proper supplementation of thiamine and riboflavin may decrease H2O2-induced DNA oxidative damage, while excess thiamine and riboflavin supplementation may be harmful to DNA and enhance the susceptibility to H2O2 potentially.

Objective To detect levels of serum platelet activating factor(PAF),thrombomodulin(TM) and white blood cell(WBC),platelet count(PLT) in neonates with meconium aspiration syndrome(MAS),and observe changes of mediators of inflammation and function of endotheliocute. Methods All cases were taken vein blood in 24 h and 72-96 h after birth.Surm PAF and TM were detected by EILSA technique,at the same time, blood cell counts were determined. Results PAF and WBC in neonates with MAS increased, which were relevant to the patients′ condition.TM of neonates with MAS increased significantly, especially in 72-96 h after birth and(aggrava)-ted with the patients′ condition. Conclusion Neonates with MAS have inflammatory reaction and injured endotheliocyte, which are(inte)-raction.

Chinese Journal of Experimental Ophthalmology 2011;29(2):151-154
Background Epidemiological investigation in human has been conclusive. In postmenopausal women, the incidence of cataract is higher than men at the same age. In addition, hormone replacement therapy may protect against the development of cataract. However, this role of androgen is not clear. Objective This study was to explore the effects of estrogen and androgen on anti-oxidative ability of lens after ovariectomy. Methods Fifty-six three-month-old clean female Wistar rats were randomly divided into normal control group, sham operation group, castration group, estrogen eyedrops group;estrogen injection group;androgen eyedrops group;androgen injection group and 8 rats for each. Ovariectomy was performed in the rats of castration group and gonadal hormone application group, and estradiol benzoate solution or testosterone propionate solution were utilized topically or systemly in 5 months after ovariectomy for 6 weeks respectively. Only abdominal cut was curried out in sham operation group. The lenses of rats were examined weekly under the slit lamp. The serum estrogen and androgen levels of rats were detected before, after operation and 6 weeks following the administration of gonadal hormone. The contents of superoxide dismutase( SOD) , glutathione( GSH) ,malondialdehyde( MDA) and water-soluble protein ( WSP) in rat lens homogenate were detected at the end of the experiment. Utilization of animals complied with the Regulation for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results No opacity of lenses was found during the experiment duration in various groups. The serum estradiol levels of rats in sham group were insignificantly different from normal groups in various time points( P>0. 05). The evident decline of serum estradiol was detected in the rats of castration group and gonadal hormone application groups compared with sham group in 5 months after operation( all P<0. 01). However,at the sixth weeks after the system use of estradiol or testosterone, the serum estradiol levels were significantly higher than the castration group and topical application groups of gonadal hormone(P<0. 01). The contents of SOD,GSH and WSP in lenses were considerably increased, but the MDA level in lenses was decreased after system use of estrogen ( P<0. 01). The activity of SOD and GSH were lower after system use of testosterone in comparison with castration rats ( P < 0. 05 ). Conclusion Estrogen can protect lens against oxidation damage. However, androgen, to a certain extent, may contribute to the development of oxidative damage in OVX female rats.

Objective To investigate the inducing effects of selenium dioxide(SeO2 ) on the apoptosis in human cervical carcino-ma cell line Hela and its influence on the expression of apoptosis-related proteins caspase-3 and P53 .Methods Hela cells were trea-ted with different concentrations of SeO2 for 24 h in vitro ;the morphological changes of Hela cells were observed by the optical mi-croscope;the influence of SeO2 on the cell proliferation and vitality was examined by the MTT assay ;the flow cytometry was em-ployed to detect the cell apoptosis rate ;the expressions of caspase-3 and P53 proteins in Hela cells were determined by the Western blot analysis .Results Under the optical microscopy ,SeO2 generated the obvious influence on the cell growth morphology ,a large number of cells became rounded and shrunken ,and lost the normal form ,while the adherence cell number was evidently decreased and the proliferation was slowed down ;the MTT results showed that SeO2 markedly inhibited the cell proliferation and viability in a dose-dependent manner ,in which ,the cell apoptosis rates induced by the 0 ,1 .875 ,3 .750 ,7 .500 ,15 .000 and 30 .000 μmol/L con-centrations of SeO2 were 3 .12% ,30 .56% ,33 .42% ,37 .50% ,45 .43% and 69 .38% respectively ,which revealing the obviously in-creasing trend;the Western blot assay revealed that SeO2 could up-regulate the caspase-3 and P53 levels ,and reached the peak value at the concentration of 7 .500μmol/L .Conclusion SeO2 could induce the cervical cancer cell apoptosis possibly by up-regulating the expressions of caspase-3 and p53 in Hela cells .

OBJECTIVETo investigate the effects of Hovenia dulcis extract on mRNA expression of MMP-13 and TIMP-1 mRNA in hepatic tissue in experimental rats.
METHOD48 male Sprague-Dawley rats were randomly divided into 2 groups: normal group (16) and model group (32), hepatic fibrosis was induced by CCl4 for 6 weeks in rats, 8 rats were sacrificed at the end of the 6th week from every group respectively, HE staining of hepatic tissue was performed; In the model group, rats randomly subdivided into 3 groups: spontaneous recovery group, control group and medication administration group, 8 rats were sacrificed at the end of the 12th week from every group respectively, the mRNA levels of MMP-13 and TIMP-1 in hepatic tissue were assayed by semi-quantitative RT-PCR.
RESULTThe mRNA expression of MMP-13 among the 4 groups were not statistically significant, but the mRNA expression of TIMP-1 among the 4 groups were statiscally significant. The levels of TIMP-1 mRNA were significantly increased in control group and medication administration group compared, with those in the model group (P < 0.05), and reverse effects of medication administration groups were significantly high than those of control group (P < 0.05).
CONCLUSIONInhibition of the mRNA expression of TIMP-1 may be the mechanism of reversing hepatic fibrosis H. dulcis, for thus collogen degradation system was recoveried gradually.
