Via activating G-protein receptor transduction pathways,nitric oxide (NO),an important signal molecule,has a complex and diverse role which is closely related with activity and gene expression of nitric oxide synthetase (NOS).NO is involved in tumor related events such as cellular proliferation,migration,especially the angiogenic process,and it is closely related with the occurrence and progression of prostate cancer.NO donors and NOS inhibitors have anti-cancer and radio-chemotherapy enhancement roles.
2.Progress of in vitro amplification of hematopoietic stem cells

In vitro proliferation of hematopoietic stem cells(HSCs) is performed on the basis ofthe simulation of in vitro blood system by adding growth factors and promoting components. Several protocols for expansion of HSCs in vitro are currently in development. In recent years, with the understanding of the molecular and cellular mechanisms regulating HSCs maintenance and expansion, tremendous progress has been made in this field. In this paper, research progress in the study of the factors determining the in vitro HSCs proliferation is reviewed.
3.Clinical therapeutic effect of rehabilitative therapy on dysphagia after cerebral stroke

Objective:To explore the clinical therapeutic effect of comprehensive rehabilitative treatment on dyspha-gia after acute cerebral stroke .Methods :A total of 78 patients with dysphagia after acute cerebral stroke ,who visi-ted to our hospital from Jun 2011 to Jun 2012 ,were selected ,randomly and equally divided into Vitalstim electrical stimulus plus swallowing training group (rehabilitation treatment group ) and traditional swallowing training group (routine training group) .Patients received Kubota's water drinking test before and after treatment .Therapeutic effects were observed ,compared and statistically analyzed .Results:Total effective rate of rehabilitation treatment group was significantly higher than that of routine training group (92.3% vs .74.3% ,χ2 = 4.191 , P= 0.041 );mean declining amplitude of swallowing function score in rehabilitation group was significantly higher than that of routine training group [ (2.0 ± 0.5) scores vs .(1.1 ± 0.6) scores , t=5.284 , P=0.032] .Conclusion:Comprehen-sive rehabilitation training can significantly improve dysphagia caused by cerebral stroke .
4. Inhibitory effect of combined transfection of antisense oligonucleotides of VEGF-C and COX-2 on the expression of VEGF-C in breast carcinoma cell line

Objective: To investigate the effect of combined transfection of antisense oligonucleotides (ASODNs) of vascular endothelial growth factor-C (VEGF-C) and cyclooxygenase-2 (COX-2) by cationic liposome (LIP) on the expression of VEGF-C in breast carcinoma MDA-MB-231 cells. Methods: Cytotoxicity of ASODNs on MDA-MB-231 cells was measured by MTT assay and the optimal concentration for transfection was determined. The transcription of VEGF-C in breast carcinoma cells was detected by RT-PCR. The protein expression of VEGF-C was detected by immunohistochemical staining and confocal laser scanning microscopy in MDA-MB-231 cells after co-transfection of VEGF-C and COX-2 ASODN. Results: No cytotoxic effect was found when ASODNs/LIP was transfected at 0.2 mmol/L + 2.5 mL/L. The inhibitory ratio on VEGF-C mRNA transcription was 70.37% in the combined transfection group, which was significantly higher than VEGF-C transfection group and COX-2 transfection group (48.15% and 42.21%, P < 0.01). The protein expression of VEGF-C was significantly lower in the combined transfection group than that in only VEGF-C or COX-2 transfection group (P < 0.01). Conclusion: Combined transfection of VEGF-C and COX-2 ASODN markedly decreases the expressions of VEGF-C at mRNA and protein levels in MDA-MB-231 cells. COX-2 maybe involved in the induction of VEGF-C expression in breast cancer.
5. Expression of CDX-2, PTEN, E-cadherin and NM23 in gastric cancer and its clinical significance

Objective: To investigate the expression of CDX-2, PTEN, E-cadherin and NM23 in gastric cancer and its clinical significance. Methods: Immunohistochemical S-P method was used to detect expression of CDX-2, PTEN, E-cadherin and NM23 in 77 gastric cancer specimens and 30 normal stomach mucosal tissues, and then the relationship of the expression with clinical pathology parameters was analyzed. Results: The positive rates of CDX-2, PTEN, E-cadherin and NM23 were significantly higher in gastric cancer tissues than those in the normal gastric tissues (P<0.05). The expression of CDX-2, PTEN, E-cadherin and NM23 was correlated with the differentiation, histological type, infiltration, lymph node metastasis and TNM stage of the tumors (P<0.05), but not with patients' gender, age or tumor size(P>0.05). The l-,3- and 5-year survival rates of CDX-2, PTEN, E-cadherin and NM23 positive patients were higher than those of negative patients(P<0.05, P<0.01). Conclusion: The expression of CDX-2, PTEN, E-cadherin, NM23 in gastric cancer is related to the differentiation, infiltration, lymph node metastasis and TNM stage of gastric cancer, and detecting their expression in gastric cancer may help to judge the prognosis of gastric cancer.
6.Application of large aperture spiral CT in radiotherapy simulation positioning

Objective:To discuss application of large aperture 16 row spiral CT in radiotherapy simulation positioning. Methods: To apply CT simulation positioning with large aperture 16 row spiral CT for the breast tangential field by early breast cancer after breast conserving surgery and radical radiotherapy. Compare CT-simulation and X-ray conventional positioning technique and Compare large aperture 16 row spiral CT and single row spiral CT image. Results: The application of large aperture 16 row spiral CT avoided the error because of body limited. Large aperture 16 row spiral CT simulation position validation error is superior to X-ray positioning. Its image quality is better than that of single row spiral CT. Conclusion:The application of large aperture 16 row spiral CT make simulation positioning more accurate, planned and treatment more accurate, it can provide guarantee of accurate simulation positioning for accurate plan and treatment.
7.Influence of tumor volume change on intensity modulated radiotherapy for nasopharyngeal carcinoma analyzed by repeated CT simulation

Objective To analyze the influence of tumor volume change on intensity modulated radiotherapy(IMRT)for nasopharyngeal carcinoma by analyzing the repeated CT simulation.Methods Twenty nasopharyngeal carcinoma patients undergoing IMRT from July 2011 to November 2012 were selected in the study.The first CT simulation was conducted prior to radiotherapy,and the repeated CT simulation was finished after radiotherapy of 30 Gy.The first and the repeated CT images were fused in treatment planning system and GTV volume shrink rate was calculated.The original plan was used to recalculate the dose distribution on repeated CT.The dose volume histogram was used to calculate the dose difference of organs at risk including the brain stem and spinal cord.Results Compared with the first CT,GTV volume shrink rate of the repeated CT simulation was 28.7%,the maximum dose,1 cm3 volume and the average dose percentage of the brain stem and spinal cord were increased(t=0.83-3.17,P<0.05).Conclusions GTV volume shrinked significantly after radiotherapy of 30 Gy in IMRT for nasopharyngeal carcinoma.The dose of the organs at risk increased accordingly.
8.Errors of two CT simulation positioning methods in intensity-modulated radiotherapy: a comparative study

Objective To compare the errors of final isocenter marking method and reference point marking method for CT simulation positioning in intensity-modulated radiotherapy (IMRT).Methods From 2009 to 2012,327 patients with head and neck cancer for IMRT underwent CT simulation positioning using the Philips Brilliance CT Big Bore scanner and Philips Tumor LOC workstation and were divided into final isocenter marking group (n =208) and reference point marking group (n =119) according to positioning methods.Target volume delineation and treatment plan design were performed on the Varian Eclipse treatment planning system (TPS).Before treatment,kilovoltage cone-beam CT scans and registration were performed with the Varian EX on-board imager system to obtain beam position errors in the right-left (RL),superior-inferior (SI),and anterior-posterior (AP) directions,and then comparisons of errors between the two groups were made by independent-samples t test.Finally,the TPS was used to measure the changes in the doses to the organs at risk after moving isocenters in the RL,SI,and AP directions among 5 patients with nasopharyngeal carcinoma.Results The mean beam position errors in the three directions were less in the final isocenter marking group than in the reference point marking group (P =0.02,0.01,0.03).After moving isocenters in the three directions,the target dose was reduced and the dose to the normal tissue around the target tumor was increased significantly.The error in the AP direction had the maximum influence on the spinal cord and brainstem.Conclusions Final isocenter marking method leads to less beam position error than reference point marking method in CT simulation positioning.Small isocenter motion can cause large changes in the doses to the organs at risk.
9.The effect of recombinant human CTLA-4Ig fusion protein on T lymphocytes in active rheumatoid arthritis patients

Objective To discuss the therapeutic effect of recombinant human CTLA-4Ig fusion protein (rhCTLA-4Ig) on rheumatoid arthritis (RA) patients and the change of T helper cells 17 (Th1 7 ) and regulatory T (Treg) cells.Methods Forty-eight active RA patients were randomly divided into the treatment group and the control group.Treatment group received 12 weeks of infusions of rhCTLA-4Ig ( 10 mg/kg),while the control group received 12 weeks of infusions of placebo.The changes of expression levels of Th17cell were detected by flow cytometry.The changes of expression level of FoxP3 in monocytes of the peripheral blood were examined by RT-PCR.T test and x2 test were used for statistical analysis.Results ① Eighteen of 24 cases in treatment group showed good ACR20 changes after 12 weeks,the effective rate (75%) was significantly higher than that of the control group ( 1/24,4% ).After treatment,DAS28 score of the treatment group (3.0±0.7) was significantly lower than that of the control group(6.9±0.7) (t=-12.39,P<0.01 ).② Compared with the Th17 monocytes of the peripheral blood from active RA patients of treatment group after therapy (0.22±0.20)%,the Th17 monocytes of peripheral blood from active RA patients of the control group (1.63±0.47)% were higher (t=5.61,P<0.05 ).③ Compared with the FoxP3 mRNA of the control group (0.24±0.05),that of the treatment group (0.88±0.18) was significantly higher after therapy (t=7.56,P<0.01 ).Conclusion The clinical parameters of RA patients have improved significantly after the treatment,and the Th17 and Treg cells balance is also restored after rhCTLA-4Ig therapy.
10.Analysis and treatment of explicit and occult Descemet's membrane detachment after phacoemulsification

AIM: The reasons of explicit and occult Descemet's membrane detachment ( DMD ) after phacoemulsification were analyzed and the therapeutic measures were further provided.
METHODS:A total of 31 cases diagnosed as explicit and occult DMD between Jan. 2013 and Apr. 2016 received the injection of sterile air into anterior chamber by 1-3 times, and were followed up for 3mo to observe the recoveries of cornea.
RESULTS: A total of 30 cases recovered with no visual impairment and other related complications. One case which refused being treated resulted in bullous keratitis.CONCLUSION:Both the internal and external factors can influence the operation effect of explicit and occult DMD after phacoemulsification. Early detection and timely treatment play important roles in improving patients'visual acuity. Early examination of anterior segment OCT or UBM for intractable corneal edema is helpful to find the occult DMD with different degree and work out the targeted therapies.
