1.Progress of in vitro amplification of hematopoietic stem cells

In vitro proliferation of hematopoietic stem cells(HSCs) is performed on the basis ofthe simulation of in vitro blood system by adding growth factors and promoting components. Several protocols for expansion of HSCs in vitro are currently in development. In recent years, with the understanding of the molecular and cellular mechanisms regulating HSCs maintenance and expansion, tremendous progress has been made in this field. In this paper, research progress in the study of the factors determining the in vitro HSCs proliferation is reviewed.
2.Clinical therapeutic effect of rehabilitative therapy on dysphagia after cerebral stroke

Objective:To explore the clinical therapeutic effect of comprehensive rehabilitative treatment on dyspha-gia after acute cerebral stroke .Methods :A total of 78 patients with dysphagia after acute cerebral stroke ,who visi-ted to our hospital from Jun 2011 to Jun 2012 ,were selected ,randomly and equally divided into Vitalstim electrical stimulus plus swallowing training group (rehabilitation treatment group ) and traditional swallowing training group (routine training group) .Patients received Kubota's water drinking test before and after treatment .Therapeutic effects were observed ,compared and statistically analyzed .Results:Total effective rate of rehabilitation treatment group was significantly higher than that of routine training group (92.3% vs .74.3% ,χ2 = 4.191 , P= 0.041 );mean declining amplitude of swallowing function score in rehabilitation group was significantly higher than that of routine training group [ (2.0 ± 0.5) scores vs .(1.1 ± 0.6) scores , t=5.284 , P=0.032] .Conclusion:Comprehen-sive rehabilitation training can significantly improve dysphagia caused by cerebral stroke .
3.Recent progress of targeted small molecular CDK9 degraders based on PROTAC technology

CDKs proteins are a kind of cell cycle protein-dependent kinases, which serve as important roles in controlling cell division and transcriptional stages. Among them, CDK9, as a key regulator responsible for the transcriptional elongation of cells, drives the development of various malignant cells and is considered as an important target in the field of anti-tumor drug development. However, the CDK family proteins feature high conservativeness and similarity in structure, leading to the poor selectivity and severe side effects for traditional small-molecular CDK9 inhibitors, which has limited their clinical applications. In view of this, there is an urgent need to investigate CDK9 targets through a novel strategy. The PROTAC is an emerging drug discovery strategy that the degrader could specifically recognize the target protein through indirect linkage with ubiquitin ligases and ultimately eliminate the target protein through the ubiquitination degradation system. This paper provides a brief overview of the structure and function of CDK9 protein, its relationship with the poor prognosis of clinical diseases, as well as the currently reported small molecular inhibitors. The latest research progress on the targeted degradation of CDK9 protein based on PROTAC technology is highlighted. Finally, the development prospects of this target protein in this novel technology field are summarized and prospected, aiming to provide a reference for the development of antitumor drugs in this direction.

5.Quality assessment of Periplocae Cortex from different habitats by UPLC fingerprint and quantitative analysis.

Ultra performance liquid chromatography (UPLC) was employed for simultaneous determination of three components and fingerprint analysis of Periplocae Cortex with gradient elution of mehtanol and water containing 0.1% phosphoric acid as mobile phase. Three components including chlorogenic acid, 4-methoxysalicylaldehyde and periplocoside were well separated under the analytical condition. Seventeen peaks were selected as the common peaks of 30 batches of Periplocae Cortex. The results showed that there is a significant difference in contents of periplocoside between the samples collected from Henan and Shanxi province. Based on the results of three components quantification and fingerprint analysis, hierarchical clustering analysis ( HCA) and principle component analysis (PCA) were used to further prove the differences between two group samples, and the results indicated that quality of Periplocae Cortex from Shanxi was more stable than that from Henan. The established UPLC fingerprint and quantitative analysis methods could be used efficiently in the quality control of Periplocae Cortex, and this study might contribute to the reasonable clinical application.

6.Correction of manual monitor unit calculation for asymmetric fields using Varian enhanced dynamic wedge

Objective To investigate the correction of manual monitor unit calculation for asymmetric fields using the Varian enhanced dynamic wedge.Methods Monitor unit (MU) was calculated when the field sizes ranged from 6 cm × 6 cm to 20 cm × 20 cm at a depth of 5 cm using Varian Eclipse and both 6 MV and 10 MV X-rays data from Varian Clinac 23EX for all seven available EDW angles,including 10°15°,20°,25°,30°,45°and 60° The field size was kept fixed,and the distance between geometry center of field and isocenter was increased in increments of 1 cm,ranging from -9 cm to 4 cm.When the field size was the same,the correction factor was defined as the ratio of MU calculated for asymmetric field to monitor unit calculated for symmetric field.To ensure the correction factors obtained above could be used in routine manual calculation for EDW fields,measurements were made at a depth of 5 cm for 30°and 45°EDW with field size of 10 cm × 10 cm using 6 MV X-rays.Results The correction factor was independent of field dimensions,so the average value was adopted to make practical calculation.Without correction,the maximum error was 18% for 30°,and 30% for 45.After the results of monitor unit calculation were corrected,the largest error was - 1.8% and - 1.7% for 30° and 45°EDW,respectively.The magnitude of errors was within the clinical tolerance limits.Conclusions For asymmetric EDW fields,there is very large difference between the prescribed dose by manual calculation using EDW factors measured for symmetric fields and that delivered during treatment in order to obtain correct dose to reference point.The errors are decreased to be acceptable after correction.The method of correction is simple and independent of machine specific beam parameters.
7.Application of transcutaneous electric nerve stimulation in operation of thyroid tumors.

OBJECTIVETo study the effect of transcutaneous electric nerve stimulation for decreasing stress response and strengthening the anesthetic effect in the operation of thyroid tumors.
METHODSSixty patients with thyroid tumors were randomly divided into an observation group and a control group, 30 cases in each group. All of them were given modified one-point cervical plexus nerve obstruction. The stimulation at bilateral Hegu (LI 4) and Quchi (LI 11) with HANS was added to the observation group from the beginning to end of operation.
RESULTSThe blood pressure, heart rate and jerking reflection were decreased significantly as compared with the control group. The oxygen saturation and respiratory frequency did not change.
CONCLUSIONCardiovascular stress response can be decreased with HANS in the operation of thyroid tumors and anesthetic effect can be strengthened.

9.Let us understand clinical outcome in surveillance of resistance of bacteria

It is an important task to lab of clinical microbiology to surveille multi-drag or pan-drug resistant strains,such as penicillin-or macrolide-resistant Streptococcus pneumoniae,methicillin-resistant Staphylococcus aureus(MRSA),3rd generation cephalosporin-or quinolone-resistant Enterobacteriaceae, carbapenem-resistant Pseudomonas aeruginosa or Acinetobacter baumannii,and so on.We must understand characteristics of these resistant strains to guide doctors' empirical therapy of infective diseases.
10.Clinical analysis and surgery of traumatic implantation cysts of the iris

Objective To explore the clinical characteristics, surgical treatment and related factors of traumatic implantation cyst of the iris. Design Retrospective cases series. Participants Thirty-six cases of traumatic implantation cyst of the iris. Methods Thir- ty-six cases of traumatic implantation cyst of the iris were reviewed. Main Outcome Measures Age, surgical history, history of disease, surgical method of patients. Results All cases of traumatic implantation cyst of the iris were secondary to perforating injury. 10 cases had been undertaken cataract surgery. The histories of disease in 6 months~1 year and 1 year~10 years were 30.56% respectively. Sur- gical exicision was taken in all cases. There were 2 recurrence cases. Conclusion Traumatic implantation cyst of the iris is almost see- ondary to perforating injury. Surgical excision is an effective strategy to treat this disease.
