1.Effect of Pulmonary Surfactant on Respiratory Function of Newborn Infants with Respiratory Distress Syndrome in the Advanced Stage

Objective To observe the effect of pulmonary surfactant(PS) on lung function and ventilator parameters of neonatal respiratory distress syndrome(NRDS) in the advanced stage.Methods Twenty-eight infants with NRDS were given PS in one dose by endotracheal intubation on the left side,right side,feet high with head low,and level decub respectively.The dose of PS was 100-150 mg/kg each time,each posture slow note of the drug were required 1/4,out of the straw,hand-controlled ventilation,to reduce fluid loss,with the exception of a clear airway obstruction,within 6 hours after the administration not to shoot back suction,to give mechanical ventilation after the injection.Lung function parameters were also measured:pressure of oxygen in artery[p_a(O_2)],carbon dioxide partial pressure[p_a(CO_2)],the ratio of pressure of oxygen in artery and alveolar oxygen partial pressure[a/Ap(O_2)] and oxygenation index(OI) were determined.Ventilator parameters were determined:oxygen concentration(FiO_2),oxygen peak(PIP),end-expiratory positive pressure(PEEP) and mean airway pressure(MAP) were determined.These numerical data were analyzed and compared before and after treatment with PS.Clinical manifestations,thoracic X-ray changes,survival rate and incidence rate of complications were also analyzed and compared before and after PS therapy.Results p_a(O_2),a/Ap(O_2) showed significant upgrade and OI had a decrease after PS administration in comparison with those before PS therapy.The ventilator parameters(except for PEEP) acquired were also lower after drug administration than those in before drug therapy.There were significant differences in both stages(P_a90%,respiratory sound in 24 cases enhanced,the observation of chest film after 24 h indicated that,lesions in 21 cases improved significantly,5 cases took a favorable turn.The survival rate was 85.7%.The incidence rate of complication was as follows:pneumonia was 25%,patent ductus arteriosus was 10.7%,pneumorrhagia was 7.1% and intraventricular hemorrhage was 3.6%,respectively.Conclusion Respiratory function of NRDS is significantly improved by using PS in the advanced stage,and therapeutic effect is apparent.

4.Experimental Research of Therapeutic Effect and Mechanism of Ribavirin to Viral Myocarditis

Objective To investigate the therapeutic effect and mechanism of ribavirin to mice viral myocarditis. Methods Onehundred and twenty mice were randomly divided into 3 groups. Group 1 was normal control group 1 .group 2 CVB3 control group, group 3 ribavirintherapy group. In the latter two groups, 0.2 mL CVB3(10-7.5Tcid5o/mL)was infected into abdominal cavity. Ten mice of each group were killed at the 7 th, 14 th,21 st,28 th day. Mice motality,light and electron microscape, myocardial viral tiler, myocardial pathological score,serum cTnI,malonytdialdehyde(MDA) and cardiac myocyte apoptosis rate(CMAR) were examined.Re-sults The motality,myocardial viral titer,myocardial pathological score,cTnI were lower in group 3(ribavirin therapy group) than in group 2(CVB3 control), and MDA and CMAR were no significant differences between ribavirin group and simple CVB3 group. Conclusion Ribavirin has good treatment effect to mice viral myocardilis,and its mechanism is due to decrease viral titer.
5.Normal Values of Blood Pressure and Critical Hypertension and Hypotension in Full-Term Infants within Seven Days

Objective To study the normal values of blood pressure in healthy full-term infants in the first 7 days of life,and to determine the critical hypertension and hypotension in newborn infants.Methods Omni-Trak~(TM)NVS monitor with DINAMAP was used to measure the blood pressure.Systolic,diastolic and mean blood pressure were measured every day by DLNAMAP in a cohort of 50 healthy full-term infants in the first 7 days of life.Results 1.Systolic,diastolic and mean blood pressure of healthy full-term infants were significantly influenced with the age of birth.2.Stepwise linear multiple regressions was used to examine the multiple correlations among days of life,birth weight and gestational age.3.Optimal regression equations were built respectively.SBP=32.4+0.7X_1+(3.7X_2+)(0.6)X_3(mm Hg).DBP=13.1+ 0.6X_1+2.9X_2+0.6X_3(mm Hg).MAP=17.3+0.4X_1+2.4X_2+0.8X_3(mm Hg).(3.MAP=)(DBP+)(0.45)(SBP-DBP)(mm Hg).4.The critical hypertension and hypotension in newborn infants were obtained.Conclusions (Du)ring the first 7 days of life,there is a progressive rise in blood pressure of healthy full-term infants.There are linear reliance correlations between systolic,diastolic,mean blood pressure and day of life,birth weight gestational age.The critical hypertension and hypotension in newborn infants is important for clinic diagnosis.
6.Dynamic study and screening of new markers of spermatogonial stem cells by iTRAQ protein mass spectrometry.

OBJECTIVETo study the dynamic changes in the protein marker expression in the spermatogonial stem cells (SSCs) of mice at different ages by iTRAQ protein mass spectrometry and to screen new markers using the bioinformatic proteome database.
METHODSBased on the postnatal weeks, we divided 80 healthy male C57BL/6 mice into eight age groups of equal number, harvested their testicular tissues, extracted proteins following purification of the SSCs by compound enzyme digestion and magnetic-activated cell sorting. Then we analyzed and identified proteins using two-dimensional electrophoresis, protein mass spectrometry, and protein database information.
RESULTSTotally, 248,510 mass spectra were obtained from the MS experiment and 1132 proteins were identified. By the criteria of >1.2-fold for protein abundance difference and P value <0.05, we identified 298 differentially expressed proteins and 9 currently known makers of SSCs (PCNA, GFRalpha1, CDH1, Annexin A7, UCHL1, VASA, CD49f, CD29, and PLZf). Compara- tive analysis showed different expressions of the proteins in the SSCs of the mice of different ages, and the differences in the expressions of GFRalpha1, CD49f, and CD29 were consistent with the findings in other published literature. Ten proteins (P63, CD71, CD98, K19, ACE, K18, K15, K17, SH2, and SH3) were selected as SSC markers to be further studied.
CONCLUSIONThe proteins in SSCs are differentially expressed in mice of different ages. The technology of iTRAQ protein mass spectrometry can be used to analyze and compare the proteome information of mouse SSCs, obtain differentially expressed proteins in mice of different ages, and thus offers a new ap- proach to further analysis and study of the function and roles of these differential proteins.

8.Relationship between pulse pressure and atherosclerotic renal artery.

OBJECTIVETo analyze the relationship between the pulse pressure (PP) and atherosclerotic renal artery stenosis (ARAS) in middle-age and eldery paients with hyperensio, so as to provide the basis for the diagnosis and treatment of ARAS.
METHODSWe tetro spectively analyzed the data of 257 patients with hypertension undergoing renal arteriography at General Hospital of PLA between 2009 and 2014. Their mean age was (64.09 +/- 7.81) years. According the results of arteriography, they were divided into two groups: ARAS group (n = 145), and non-ARAS group (n = 112). Their PP and multinomial clinical indexes with ARAS were statistically analyzed.
RESULTSSingle factor correlation and Logisitic analysis showed that age, PP, serum creatinine (SCr) levels, and new onset of hypertension after 50 years of age were strongly positively related to ARAS. The age and PP and SCr was a team of continuous variable.
CONCLUSIONPP was the first new risk factor of ARAS in middile-aged and elderly patients with hypertension. It is suggested that PP may be a early predictive indicator and a new therapeutic target for ARAS.

9.Effect of silencing heparanase on the biological behavior of gastric carcinoma in nude mice.

OBJECTIVETo investigate the effects of silencing heparanase (HPA) on growth, angiogenesis and metastasis of human gastric carcinoma transplanted in nude mice.
METHODSHuman gastric carcinoma SGC-7901 cells and those cells with silenced HPA (gastric carcinoma SGC-7901-HPA(-)) were separately transplanted subcutaneously in 6 nude mice. The time, size and speed of tumor growth were recorded. RT-PCR and Western-blot were used to detect the expression of HPA mRNA and protein in the subcutaneous tumors of the two groups. Immunohistochemical staining was used to detect microvessel density (MVD) in the subcutaneous tumors of the two groups. Cells of the subcutaneous transplanted tumors of the two groups were separately injected into the peritoneal cavity of nude mice, 6 mice each. The growth of metastatic tumors in nude mice was observed.
RESULTSHuman gastric carcinoma SGC-7901 cells and SGC-7901-HPA(-) cells were subcutaneously inoculated in nude mice, and tumors appeared at 4 days and 7 days after inoculation, respectively. The MVD was (20.69 ± 1.20)/HP and (11.35 ± 1.94)/HP, respectively (P < 0.05). The expressions of HPA mRNA and protein of the subcutaneously transplanted SGC-7901-HPA(-) tumor were decreased. Four voluminous metastatic tumors caused by SGC-7901 cells occurred in 3 mice in the liver, right kidney, omentum and intestine. Two smaller abdominal metastatic tumors of SGC-7901-HPA(-) cells were found in the liver and right kidney.
CONCLUSIONSilencing HPA can inhibit the tumor growth, angiogenesis and metastasis of human gastric cancer in nude mice. It suggests that HPA might become a new target for prevention and treatment of gastric cancer.

10.Detection of the serum level of interleukin-17 in the chronic hepatitis B patients

Objective To research serum level of interleukin-17 (IL-17)associated with the progression of hepatic injury in the chronic patients with hepatitis B. Methods The serum level of IL-17 was measured by ELISA and the serum levels of IL-6,IL-8 were measured by RIA in patient groups and healthy group, the patient groups including 42 mild patients, 37 moderate patients and 38 severe patients. Results IL-17,IL-6 and IL-8 levels in patient patients were significantly higher than those in healthy people(P< 0.01). There is no significant difference among mild patients and moderate patients. Compared with mild patients and moderate patients, the cytokines lever were significantly higher in severe patients(P<0.01). Conclusion IL-17 as a new cytokine probably play a multiple role as immune factor and inflammation element in the progression of the chronic hepatitis B disease. Maybe, it can provide a new approach to the therapy of the chronic hepatitis B.
