1.ANALGESIC AND ANTIINFLAMMATORY EFFECTS OF DEOXYNUPHARIDINE

Deoxynupharidine is one of the alkaloids extracted trom Nuphar pumilum ( Timm ) DC which has been used in the treatment of arthritis and lumber-leg pain in folk medicine. This article presents the results of the experimental study of the analgesic and antiinflamma-tory actions of deoxynupharidine.Deoxynupharidine, ip, 15-20mg/kg, increased significantly the pain threshold of thermal and electrical stimulation in mice, and decreased significantly the rate and frequency of the development of body twisting reaction induced by ip acetic acid. Deoxynupharidine also significantly reduced exudation of acetic acid-induced inflammation in peritoneal cavity of mice. Data also showed that deoxynupharidine 15-20mg/kg and dexamethasone 4-8mg/kg produced a similar inhibing effeet on the degree of ankle swelling induced by subcutaneous injection of egg albumin. Dose-response relationship was seen in the above experiment.These resalts provide evidences for the clinial appli cation of deoxyuupharidine in the treatments of arthrtis and other inflammatory diseases.
2.Effects of total flavonoids of Herba Epimedii on the metabolism of typeⅠcollagen and the expression of cathepsin K in the bone of ovariectomized rats

Objective To investigate the effects of total flavonoids of Herba Epimedii(HEF)on the metabolism of typeⅠcollagen and the expression of cathepsin K in the bone of ovariectomized(OVX)rats. Methods Fifty-four female SD rats were allocated into 6 groups;OVX group,sham operation group,OVX rats followed by three doses of HEF(40,80 and 160 mg?kg~(-1)?d~(-1))and nilestriol(0.1 mg?kg~(-1)?d~(-1))for 12 weeks respectively.Bone mineral density(BMD)of whole body was determined by dual-energy X-ray absoptiometry.The level of cross-linked N-telopeptide of typeⅠcollagen(NTx)in the urine were determined by ELISA.The amounts of typeⅠcollagen protein and cathepsin K protein in bone tissue were detected by immunohistochemical method and Western blotting.Results Compared with OVX group,the total BMD values in the HEF treated groups were increased(all P<0.05),and the expression levels of typeⅠcollagen in three HEF treated groups rose significantly in a dose-dependent manner after 12 week,and simultaneously,both the expression of cathepsin K in bone and the level of NTx/Cr were reduced markedly(P<0.05),being most significant(P<0.01)in the group treated with the highest dose of HEF(160 mg?kg~(-1)?d~(-1)).Conclusion HEF seems to be able to elevate BMD and improve bone quality of rats via promoting synthesis and inhibiting proteolysis and absorption of typeⅠcollagen in the bone.
3.Allelopathic effects of aqueous extracts from Panax notoginseng on three maize varieties (Zea mays).

It has been showed that there were obvious obstacle effects of Panax notoginseng replanting. Crop rotation was the main effective technique to overcome the obstacle. To find a reasonable crop rotation system for P. notoginseng, aqueous extracts from root, stem and leaf of P. notoginseng were analyzed for allelopathic effect on three maize varieties (which are often grown in regions where P. notoginseng grown). The main results were as follows: (1) Allelopathic effect of P. notoginseng stem and leaf extracts on the three other tested plants was stronger than that of root extracts; (2) Corn was more vulnerable to the effects of allelochemicals at seedling stage than at germination stage, and the corn root was more sensitive than aerial part to allelochemicals; (3) Lusan No. 3 and Yunrui No. 1 showed resistance to P. notoginseng allelopathy, with respective comprehensive sensitivity indexes (M3) of - 0.089 3 and -0.159 2, while Bainuo No. 1 is sensitive at M3 = -0.261 0. It then can be concluded that Lusan No. 3 and Yunrui No. 1 may be an alternative rotation plants for overcoming P. notoginseng continuous cropping obstacle.

4.Mechanism of protective effects of low dosage of ultrashortwave diathermy on cerebral ischemia-reperfusion injury

Objective To observe the effect of low dosage of uhrashortwave(USW) on infarction volume, B cell lymphocytoma-xl (Bcl-xl) and tumor necrosis factor alpha (TNF-?) after cerebral ischemia-reperfusion in rats and discuss its acting mechanisms. Methods Focal ischemia-reperfusion model was established in 25 rats by re- versible right middle cerebral artery occlusion with filament. The right side cerebral ischemia was lasted for 2 hours and then followed with 24 hours of reperfusion. The content of neurological deficits were evaluated by the Zea-Longa 5-degree scoring system to select rats. After surgery, the rats were divided into 3 groups: blank control group, control group and USW treatment group. The brain of all rats was taken at 24 hours after reperfusion. The cerebral infarction volume, the expression of Bcl-xl and TNF-?were measured and analyzed. Results Twenty-five rats were used in the analysis of results. Objective To investigate the effect of total flavonoids of Herba Epimodii (HEF) on the expression of core binding factor?1 (Cbfal) in the bone of ovariectomized rats.Methods Fifty-four female SD rats were allocated into 6 groups (9 in each): sham operation group,ovariectomized (OVX) group,OVX followed by three kinds of HEF doses(40,80 and 160 mg?kg~(-1)?d~(-1))and nilestriol (0.1 mg?kg~(-1)?week~(-1))for 12 weeks respectively.Bone mineral density (BMD) of whole body was determined by DEXA.The levels of osteocalcin (BGP) and estradiol (E_2) in serum were measured by radioimmunologic method.All rats were then sacrificed,and total RNA were directly isolated from the skull.Relative quantification of Cbfal mRNA expression was detected by real time quantitative PCR.Results Serum E_2 and BMD of whole body in the OVX group were lower than those in sham group significantly after 12 weeks (both P0.05).Relative quantification of Cbfal mRNA expression in the OVX group was significantly lower than that in sham group after 12 weeks (P

Objective To investigate the effects of tetramine to cardiac/skeletal muscle in the rats and elucidate the relationship of the effects and the elevated sero-enzyme.Method The 30 homogeneous SD rats were invided into three groups:the control group,the half-lethal dose group and the lethal dose group.The number of female and male rats was equal.The tetramine powder is dissolved into 0.9% NS and puured into the stomach of the objects in medication groups;0.9%NS was poured into the rats of control group.Once the rat died or one hours later,the related sero-enzyme was determined and the rat was executed,Immediately cardiac muscle and skeletal musclewas respectively drawn the materials from the rat and was pathologically examined.Results After the rats are intragastric administrated they spasm in 10-60 min;the 6 rats of the medial lethal dose group die in 20-60 min.Serum TnI/CK/AST/LDH/a-HBDH in medication groups is higher than in control group (P
8.Laparoscopic radical cholecystectomy vs open surgery for T2 gallbladder cancer

Objective To evaluate laparoscopic radical cholecystectomy for T2 gallbladder cancers.Methods A retrospective analysis was made on 44 patients with pathology confirmed T2 gallbladder cancer undergoing laparoscopic vs open radical cholecystectomy.The clinicopathological and follow-up data were compared.Results Laparoscopic cholecystectomy was performed in 32 patients,the implantation metastasis rate of this approach has no statistical differences compared with the open cholecystectomy (P =1.000).26 patients underwent laparoscopic radical resection and the remaining 18 patients underwent open radical resection.There was no statistical difference in operation time (P =0.953),blood loss (P =0.193)and postoperative complications (P =1.000),but the laparoscopic radical resection group is superior to the open group on postoperative pain grading (P =0.022),ambulation time (P =0.000),nothing per mouth time (P =0.000) and length of hospital stay (P =0.048).The mean number of lymph nodes retrieved was 5 ±4 (range 1-12) in the laparoscopic radical group and 6 ± 3 (range 1-12) in the open group (P =0.983);the 1,3,5 year survival rates was 92.3%,70.3%,61.5% and 92.3%,76.3%,69.3%,respectively(P =0.473).Conclusions Initial laparoscopic cholecystectomy does not increase the rate of seeding metastasis in the context of intact gallbladder carcinoma.Totall laparoscopic radical resection is feasible in selected T2 gallbladder cancer patients.
9.Changes of Transforming Growth Factor-?_1 and Vascular Endothelial Growth Factor in Children with Primary Nephrotic Syndrome

Objective To investigate the change of serum transforming growth factor(TGF)-?1 and vascular endothelial growth facter( VEGF) in children with different types of primary nephrotic syndrome( PNS). Methods Children involved in the experiments were divided into simple type group, 16 cases; nephrit was type group, 16 cases, collecting blood sample in prednison - pretreated stage and in prednison- treated stage;control group, 14 cases. Monoclonal EUISA detected TGF-?1 and VEGF. Results Serum level of TGF-?1 and VKGF in active stage of simple type were higher than those of remission stage. The level in nephritis type was no signifi cant difference between prednison- treated stage and prednison - pretreated stage. The level in nephritis type was significantly higher than that in simple type. Conclusion Monitoring the dynamic change of serum TGF-?1 and VEGF can assess the effect of prednison treatment,and evaluate the prognosis of nephrotic syndrome.
10.Enhanced Stability of Plasmid pcDNA3.1+ within Salmonella typhimurium by Downregulation of the Ampicillin Resistance Gene Expression

The high-copy-number plasmid pcDNA3.1+ is unstable within S almonella typhimurium. A novel plasmid pmcDNA3.1+ was constructed by removin g the promoter sequence of ampicillin resistance gene (bla gene) in plasmid pcDNA3.1+. In contrast to pcDNA3.1+, pmcDNA3.1+ was stable within Salmonel la typhimurium SL7207 in LB medium with or without ampicillin. Further experi ments showed the ?-lactamase activity of Salmonella typhimurium SL7207(pmc DNA3.1+) was apparently lowered than that of Salmonella typhimurium SL7207( pcDNA3.1+) and the high ampicillin concentration was maintained longer in LB me dium culturing Salmonella typhimurium SL7207(pmcDNA3.1+). When mice were a dministered with Salmonella typhimurium SL7207(pmcDNA3.1+) intraperitoneall y, more than 95% of Salmonella cells separated from the spleen still harbore d the plasmid pmcDNA3.1+ 7 days later; but 99% of Salmonella cells lost the plasmid pcDNA3.1+ at day 3 in mice innoculated with Salmonella typhimurium SL7207(pcDNA3.1+). By lowering the expression of bla gene, the rapid deco mposition of ampicillin in LB medium was avoided and the metabolic pressure was relieved for the host cells. This method offers a solution for the problem of t he instability of high-copy-number plasmid within Salmonella typhimurium.
