3.Neuroprotective effect screening and the mechanism of 10 kinds of coumarin derivatives.

The study reports the detection of neuroprotective effect of 10 kinds of coumarin derivatives and explores their possible mechanism. MTT method was used to screen the neuroprotective effect of 10 coumarin derivatives on neurotoxic agents (Aβ25-35 and rotenone) or OGD (oxygen-glucose deprivation). A compound with better protective effect was obtained. Then the effect of this compound on neurotoxic agents on PC12 was detected by the morphological observation. Furthermore, the effect of compound 3 on microglia with lipopolysaccharide (LPS) induced inflammation was detected. And the inflammatory factor was tested. Finally, direct free radical scavenging ability was detected. Compound 3 was found to be the best compound through three neurons toxic models. Not only compound 3 ameliorated cell viability reduced by three neurons toxic models, but also significantly inhibited the production of inflammatory factor (TNF-α and IL-1β). And its free radical scavenging ability is very good, especially the effect on superoxide anion, which is comparable with vitamin C. The significant scavenging effect of compound 3 on superoxide anion might be the mechanism of the neuroprotection. Compound 3 as a potential neural cell protective agent merits further investigation.

Forty type 2 diabetic patients with diabetic peripheral neuropathy (DPN) were assigned to two groups and treated respectively with?-lipoic acid or mecobalamin for 2 weeks.The results suggested that?-lipoic acid could accelerate the nerve conduction velocity and decrease the plasma level of endothelin and C reactive protein as well as microalbuminceria with a effect similar to mecobalamin therapy on DPN.

Objective To investigate effects of atorvastatin on the development from macrophages (HMDM) to foam cells.Methods Monocytes were isolated from human peripheral blood by Ficoll-Hypaque density gradient centrifugation and plastic adsorptive process.The isolated cells were stimulated by phorbol 12-myristate 13-acetate (PMA) (50 nmol/L) for 48 h and transformed to macrophages.Macrophages were co-incubated with 80 mg/L ox- idized low density lipoprotein (ox-LDL) and atorvastatin (0-100 ?mol/L),respectively for 0,6,12 and 24 h. Total cholesterol (TC),free cholesterol (FC) and protein (Pro) in cultured cells were quantitatively analyzed by high performance chromatography (HPLC) analysis and modified Lowry protein assay.Results When macropha- ges were incubated with 80 mg/L ox-LDL,the ratio of TC/Pro was greater than 20,and large amount of lipid drop- lets were displayed indicating the formation of foam cells.Atorvastatin decreased TC/Pro ratio in foam cells in a concentration and time dependent manner (0-100 ?mol/L)(P
7.Epidemiological study on 38 418 syphilis cases during pregnancy.

OBJECTIVETo explore the prevalence of syphilis and risk factors on pregnant women in Fujian province.
METHODSFrom July 1st, 2004 to June 30th, 2006, epidemiologic methods as questionnaires to fill in and rapid plasma reagent testing (RPR) were performed. Pregnant women with positive RPR test and then were confirmed by treponema pallidum test (TP). Intervention was provided to the pregnant women who were followed up to the postpartum periods. Results were compared during the peri-neonatal stage between syphilis-infected and non-infected women. Factors which were relative to syphilis infection were analyzed by simple and then further logistic regression analysis.
RESULTSThere were 38 418 pregnant women under study, of whom 772 were confirmed including 60 who were still pregnant. The prevalence of syphilis during pregnancy was 1.85% (712/38 418), with mostly underlying syphilis which played an important role during the neonatal stage. Main risk factors to have been found as: women's occupation, cultural background, place of living, husband's occupation, family income, having sexually transmitted disease(STD) infection. The prognosis of peri-neonatal stage was worse if the mother was having blood RPR test positive. Mortality of peri-neonatal, preterm birth rate and low-birth weight rate were found also higher among mothers when RPR was positive (P < 0.01).
CONCLUSIONThe prevalence of syphilis in Fujian province was going up yearly. 8 factors including women's occupation, cultural background, place of living, received poor health education, having multiple sexual partners, husband's occupation, family income and having STD were main risk factors in the province, the mother's status of syphilis infection would strongly relate to the peri-neonatal stage of pregnancy.

Objective To explore the maternal-fetal transmission patterns and interventional methods of syphilis during pregnancy.Methods A total of 847 cases of syphilis in pregnancy confirmed by rapid plasma reagin test(RPR)and treponema pallidum hemoagglutination test(TPHA)were treated with procaine benzylpenieillin intramuscular injection,and with erythrocin oral medication if hypersensitive to benzylpenicillin.Eight hundred forty seven cases of syphilis during pregnancy were followed up for pregnancy outcomes.And their newborn babies were tested using the RPR.The newborns with positive results were given intervention and followed up until 24 months after birth.Results(1)A total of 733 cases among the total 847 have given birth to living-babies,in which 626 cases were tested using RPR,and the positive rate was 55.1%(345/626).(2)The RPR positive rate,neonatal mortality,preterm birth rate and low birth rate in the newborn of mothers with an RPR titer higher than or at 1:8 were higher than those of mothers with an RPR titer lower than 1:8(P
9.Screening and identification of indoleacetic acid producing endophytic bacterium in Panax ginseng.

Endophytic bacteria which was producing indoleacetic acid was screened from Panax ginseng by using the Salkowski method. The active strain was also tested for its ability of nitrogen fixation by using the Ashby agar plates, the PKV plates and quantitative analysis of Mo-Sb-Ascrobiology acid colorimetry was used to measure its ability of phosphate solubilization, for its ability of potassium solubilization the silicate medium and flame spectrophotometry was used, for its ability of producing siderophores the method detecting CAS was used, for its ability of producing ACC deaminase the Alpha ketone butyric acid method was applied. And the effect on promoting growth of seed by active strain was tested. The results showed that the indoleacetic acid producing strain of JJ5-2 was obtained from 118 endophytes, which the content of indoleacetic acid was 10.2 mg x L(-1). The JJ5-2 strain also had characteristics of phosphate and potassium solubilization, nitrogen fixation, producing siderophores traits, and the promoting germination of ginseng seeds. The JJ5-2 strain was identified as Bacillus thuringiensis by analyzing morphology, physiological and biochemical properties and 16S rRNA gene sequences.

10.Preliminary study on the effects of an artificial liver support system in the treatment of medicamentous liver insufficiency.

OBJECTIVESTo study the effectiveness of an artificial liver support system.
METHODSThirty-two patients with medicamentous liver insufficiency were treated with an artificial liver support system in addition to the routine medicinal therapy. Thirty patients treated with routine medicinal therapy only served as controls.
RESULTSThe clinical symptoms (e.g. hepatic encephalopathy) and the laboratory indices (serum total bilirubin and prothrombin time) of the treatment group patients were obviously improved compared with those of the control group patients (P < 0.05). The cure rate and hospitalization days were 90.6% (26/32) and 47 days respectively in the treatment group, and 43.3% (13/30) and 72 days in the control group (P < 0.05).
CONCLUSIONUsing an artificial liver support system combined with routine medicinal therapy is more effective than using medication alone.
