2.Effect of constraint-induced movement therapy on hemiplegia in children

ObjectiveTo explore the effect of constraint-induced movement therapy on hemiplegia in children.Methods30 hemiplegia children caused by cerebral palsy or trauma were divided into 2 groups.Treatment group accepts the Constraint-Induced Movement Therapy (CIMT) combined with Bobath approach. The control group accepts the Baboth approach only. Before after 12-week treatment, Ueda Test and the Brunnstrom's test were used to classify every stage of the recovery. ResultsBoth groups improved their motor function assessed either with Ueda Test or Brunnstrom's test, which was better in treatment group than in control group.ConclusionThe CIMT with Bobath approach together can remarkably raise the rate of utilization of the hemiplegia suffering limbs as well as improve the learned non-use of the suffering parts.
3.Effects of brain-derived neurotrophic factor on the expression of caspase-2 and caspase-3 and cell apoptosis in retinal ischemia/reperfusion injury

AIM: To explore the relationship between the expression of caspase-2 and caspase-3 and the apoptosis in retinal ischemia/reperfusion (I/R) injury of rats, as well as the therapeutic effects of brain derived neurotrophic factor (BDNF)on the ischemic and reperfused retina.METHODS: This experiment was conducted at the laboratory of Affiliated Hospital of Qingdao University Medical College from February 2007 to July 2007. The models of retinal ischemia/reperfusion injury were made by transiently elevating intraocular pressure. A total of 28 rats were divided into Normal and Operative Groups. Operative group was divided into six subgroups. In each subgroup there were four rats. The left eyes of rats were used for I/R and the right eyes were used for intravitreal injection of brain-derived neurotrophic factor (BDNF) as treatment group. After reperfusion we divided our subgroups according to the reperfusion time as 1, 6, 12, 24, 48, 72 hours. The retinal ganglion cell number was counted by using optic microscope(BX-51,Olympus). Apoptosis was assessed by the terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick-end labelling (TUNEL) method, and the expression of caspase-2,caspase-3 was studied by enzyme linked immunosorbent assay (ELISA) and strept avidin-biotin complex (SABC)immunohistochemistry.RESULTS: No positive apoptotic cells were observed in the normal rats' retinae, but there were a significant number of positive apoptosis cells in 6-24 hours after transient ischemia followed by a decrease at 48 hours. The number of apoptotic cells reached a maximum at 24 hours after ischemia .The expression of caspase-2 gradually increased as early as at 6 hours, reached a peak at 24 hours, then decreased between 48 and 72 hours. Similarly, caspase-3 has the same rule with caspsae-2 in the time courses of expression in retinal tissues.BDNF administered before reperfusion inhibited the expression of apoptosis and ameliorated the retinal tissue damage. It also decreased caspase-2 and caspase-3 expression in ischemic/reperfused retina.CONCLUSION: Retinal ischemia-reperfusion can induce apoptosis of cells in the retina. BDNF rescues retinal ganglion cells (RGCs) from retinal ischemia/reperfusion injury through down-regulation of cell apoptosis and caspase-2 and caspase-3 expression. BDNF have a neuroprotective effect on retina.
4.Causes and strategies to deal with the coagulation and hemolysis of blood samples of cardiovascular disease patients before examination

Objective To study the causes of coagulation and hemolysis of blood samples collected from cardiovascular disease patients and the method to prevent its happening in order to reduce the numbers of coagulation and hemolysis samples.Methods All the blood samples of cardiovascular disease patients were randomly divided into control group( n =7396)and experimental group (n =7334 ).Control group was collected and transported according to the traditional methods,while the experimental group adopted standard control in sample collecting,transporting,laboratory transmission.Causes of the blood sample coagulation and hemolysis in two groups were observed.Results Unqualified blood samples ratio of experimental group was 0.21%significantly lower than 0.82% that of the control group ( x2 =27.599,P < 0.01 ).The ratio of coagulation and hemolysis of experimental group was 0.18%,and that of control group was 0.65%,and the difference was statistically significant (x2 =19.923,P < 0.01).Conclusions Quality control of each procedure in the blood sample before examination can effectively reduce the number of unqualified blood coagulation and hemolysis and improve the quality of overall blood samples,and further provide reliable and accurate scientific evidence for disease diagnosis.
5.Dynamic study and screening of new markers of spermatogonial stem cells by iTRAQ protein mass spectrometry.

OBJECTIVETo study the dynamic changes in the protein marker expression in the spermatogonial stem cells (SSCs) of mice at different ages by iTRAQ protein mass spectrometry and to screen new markers using the bioinformatic proteome database.
METHODSBased on the postnatal weeks, we divided 80 healthy male C57BL/6 mice into eight age groups of equal number, harvested their testicular tissues, extracted proteins following purification of the SSCs by compound enzyme digestion and magnetic-activated cell sorting. Then we analyzed and identified proteins using two-dimensional electrophoresis, protein mass spectrometry, and protein database information.
RESULTSTotally, 248,510 mass spectra were obtained from the MS experiment and 1132 proteins were identified. By the criteria of >1.2-fold for protein abundance difference and P value <0.05, we identified 298 differentially expressed proteins and 9 currently known makers of SSCs (PCNA, GFRalpha1, CDH1, Annexin A7, UCHL1, VASA, CD49f, CD29, and PLZf). Compara- tive analysis showed different expressions of the proteins in the SSCs of the mice of different ages, and the differences in the expressions of GFRalpha1, CD49f, and CD29 were consistent with the findings in other published literature. Ten proteins (P63, CD71, CD98, K19, ACE, K18, K15, K17, SH2, and SH3) were selected as SSC markers to be further studied.
CONCLUSIONThe proteins in SSCs are differentially expressed in mice of different ages. The technology of iTRAQ protein mass spectrometry can be used to analyze and compare the proteome information of mouse SSCs, obtain differentially expressed proteins in mice of different ages, and thus offers a new ap- proach to further analysis and study of the function and roles of these differential proteins.

6.Endemic fluorosis in Jilin province: analysis of surveillance data for 2006 - 2010

Objective To identify changes in the occurrence of endemic fluorosis in order to provide scientific basis for making countermeasures. Methods Five villages from 14 counties of mild, moderate and severe fluorosis affected areas were selected by stratified cluster sampling every year in the whole province during 2006 - 2010. Water and urinary fluorine were determined by ion selective electrode method(GB/T 8538-1995); dental fluorosis of children 8-12 years old was diagnosed with Dean method; skeletal fluorosis was diagnosed according to "clinical indexing standards of endemic skeletal fluorosis "(GB 16396-1996), between 2006 and 2008, and "clinical diagnosis standard of endemic skeletal fluorosis"(WS 192-2008) between 2009 and 2010. Results A total of 25 diseased villages were surveyed, 14 with water sources changed, covered a resident population of 8005 people, beneficiary population 7154, and accounting for 89.37% of the resident population; not changed villages 11. In accordance with the "State drinking water health standards", in the 14 changed villages the fluoride in drinking water was qualified (≤ 1.20 mg/L), there were 3 schools whose water fluorine content exceeded the standard; among the 11 villages that did not change water sources 7 drinking water samples fluorine content exceeded the standard. Of the 8 to 12 years old children in villages with changed water sources, 363 of them were checked and 142 dental fluorosis were found, the detection rate of dental fluorosis was 39.12% (142/363); in villages with water sources not changed, 303 children were checked, the detection rate of dental fluorosis was 43.89%(133/303). Of sixteen and elder adults in water source changed villages, 6424 people were checked and 403 skeletal fluorosis were found, skeletal fluorosis detection rate was 6.27% (403/6424); 3572 people were checked in not changed villages, the detection rate of skeletal fluorosis was 13.89%(496/3572). In water sources changed areas, geometric mean of urinary fluoride was in the normal reference value(WS/T 256-2005, 1.40 mg/L)or less. Conclusions Endemic fluorosis is decreased in water improved areas, but in unimproved areas the disease is still severe, and control of endemic fluorosis is still an arduous task.
7.Correlation analysis on combined medication with of Xiyanping injection in treatment of lung infection in real world.

To analyze the regularity in combined medication with Xiyanping injection (Xiyanping for short) in the real world by as- sociation rules. Totally 5 822 patients using Xiyanping injection was collected from the 18 Class III Grade I hospitals nationwide to study the combined medication information of the patient with lung infection and make the analysis by using association rules and Apriori. According to the results, major drugs combined with Xiyanping in treatment of lung infection included compound amino acid, inosine, coenzyme A, cytidine triphosphate, vitamin C. Common drugs combined with Xiyanping can be divided into 5 categories: nutrition support therapy (vitamin C, compound amino acid) , coenzymes (coenzyme A, cytidine triphosphate, inosine), expectorants and antiasthmatics (ambroxol, salbutamol, doxofylline), hormones (dexamethasone, budesonide), antibiotics (mainly cefminox). The main combined medicines mostly conformed to the regularity for drugs treating lung infection. In addition, there were two most common medical combination models: the model for Xiyanping combined a single medicine is Xiyanping + nutrition support therapy, while the model for Xiyanping combined two or more than two medicines is Xiyanping + nutrition support therapy + coenzyme. Pharmacologically, Xiyanping is mostly combined with western medicines with similar pharmacological effects to substitute or supplement the antibiotic effect in treating lung infection. However, further studies shall be conducted for the safety and rationality of the combined medication based on clinical practices, in order to provide reference for clinical medication.

8.Influencing factor hospitalization expenses in patients undergone transsphenoidal pituitary tumor resection

Objective To analyze the main factors that influence the hospitalization expenses in patients undergone transsphenoidal pituitary tumor resection so as to provide basis for the development of clinical pathways and the control of irrational increase in medical expenses.Methods Data of patients undergone transsphenoidal pituitary tumor resection in our hospital in the years of 2008 and 2009 was collected.Statistical analysis was performed by using multinomial linear regression analysis.Results The major factors influencing the hospitalization expenses in pafients undergone transsphenoidal pituitary tumor resection included drug fees,material fees in operation,inspection fees and length of stay.Conclusions Avoiding overtreatment,reasonably lowering the drug fees,material fees in operation and inspection fees,as well as reducing the length of stay were the key to the control of the excessive growth of hospitalization expenses in patients undergone transsphenoidal pituitary tumor resection.

10.Study of Cancer-Related Fatigue in the preoperative patients with colorectal cancer

Objective To assess the status of Cancer-Related Fatigue in the preoperative patients with colorectal cancer by the use of Identity-Consequence Fatigue Scale (ICFS),analyze impacted factors and explore corresponding nursing methods.Methods Totals of 103 preoperative patients with colorectal cancer were asked to be evaluated by ICFS,and multivariate analysis and univariate analysis were exerted on the fatigue.Results The score of ICFS was 28-119 (60.47 ± 18.16).And age (P =0.289),gender(P =0.743),insured or not(P =0.869),combined another symptoms or not(P =0.293),tumor-related TNM stage (P =0.623) and the degree of differentiation (P =0.873) took no influence on cancer-related fatigue.Conclusions The degree of the fatigue of preoperative patients with colorectal cancer have no obvious specificity and preference,nursing staff should highlight care on every patient,and strengthen their understanding and attention toward cancer-related fatigue.
