Objective:To investigate the preventive and therapeutic effects of intraperitoneal injectoin of IFN? on bronchial asthma in mice and the relevant mechanism. Methods: Thirty-six BALB/c mice were randomly equalized into 3 groups:group A (normal control group),group B (asthmatic model group) and group C (IFN? treated group). The asthmatic model was established in group B and C by immunization with ovalbumin (OVA) absorbed to aluminum hydroxide. Mice of group B and C received 0.25 ml PBS and 5 ?g IFN? intraperitoneally on days 23 to 30 once daily prior to ovalbumin challenge,respectively. To study the treatment of metastasis of experimental lung cancer with a recombinant adenovirus carrying IL 2 gene, concentrations of IL 2 and associated cytokines in murine lung lavage and blood were determined by ELISA at different times after intratracheal injection of the recombinant adenovirus. The lung metastasis nodes, survival time, survival rate were investigated in a C57BL/6 mouse model by intratracheal administration, and the NK activity and CTL activity were also determined by 51 Cr 4 hour release method. The results showed that IL 2 could be detected in lung tissue as early as 6 hours after administration of IL 2 gene, its concentration was higher in lung lavage than in peripheral blood, and was higher in experimental group than that in the control group; intratracheal use of the IL 2 carring recombinant adenovirus had significant therapeutic effect on metastasis of experimental lung cancer. Increased CTL and NK activities, longer survival time, and higher survival rate were observed in the experimental group compared with the control group. It is suggested that intratracheal usage of adenovirus vector carrying IL 2 gene might play therapeutic role on the lung cancer metastasis, indicating that gene therapy of lung tumors could be done through airway directly with recombinant adenovirus.

Objective To explore the change of somatostatin(SS)in brain areas of juvenil rats with repeated febrile seizures(FSs)and hyparthermia.Methods Fifty-one male SD rats were divided randomly into 3 groups:normal group(NC group,n=14),hyperthermic group(HC group,n=19)and febrile seizure group(FS group,n=18).FS were induced by placing rats in a bath of water.The expressions of SS in DG,CA3,CA1 and CTL were measured by immunohistochemistry.The level of SS in hippocamps of these rats were determined by radioimmunoassay.Results The results of immunohistochemistry shew in HC group,the number of the SS positive neuron were increased in DG(21.10?2.51),CA3(10.60?2.41)in FS group,which was less than that in NC group(10.50?2.12,6.90?2.02),there was no difference in CA1 and CTL.The result of Radioimmunoassay:the level of SS in hippocampus in HC group[(53.74?7.56)ng/g]was higher than that in FS group[(39.54?7.74)ng/g](P0.05).Conclusion There is different change of somatostatin content in some brain areas of rats with repeated febrile seizure,which suggest that SS can increase the affectivity of seizure and promote the seizure.

Objective To investigate the expression of cyclooxygenase-2(COX-2)in breast cancers, and understand its clinical significance.Methods The immunohistochemistry was used to determine the ex- pression of COX-2 in the 40 tissues of breast cancer and 18 adjacent noncancerous mammary tissues.Results The positive expression of COX-2 was detected in 52.5% of breast cancer tissues and in 11.1% of adjacent noncancerous mammary tissues.There was a significant difference in the expression of COX-2 between breast cancer tissues and adjacent noncancerous mammary tissues(P0.05).Conclusion COX-2 was over expressed in breast cancer and correlated with metastatic lymph nodes and HER-2/neu.The expression of COX-2 may play an important role in carcinogen- esis,progression and prognosis of breast cancer.

OBJECTIVETo explore the role of the basic fibroblast growth factor (bFGF) in the directional differentiation of bone marrow mesenchymal stem cells (BMSCs) into Leydig cells.

METHODSAfter purification and identification, we inoculated the third-generation BMSCs of SD rats onto a six-orifice board and then randomly divided them into groups A (normal saline control), B (human chorionic gonadotropin [hCG] + platelet-derived growth factor [PDGF] induction), C (hCG + PDGF + 5.0 ng/ml bFGF induction), D (hCG + PDGF + 10.0 ng/ml bFGF induction), and E (hCG + PDGF + 20.0 ng/ml bFGF induction). On the 7th, 14th and 21st day of induction, we observed the morphological changes of the cells and measured the level of testosterone (T) and expression of 3 beta hydroxy steroid dehydrogenase (3β-HSD) in the supernatant by immunofluorescence staining.

RESULTSAfter induction, the BMSCs of groups B, C, D, and E exhibited microscopic features of enlarged size, inter-connection, long-shuttle or irregular shape, adherent growth, and large round nuclei, all characteristic of Leydig cells. With the prolonging of time and enhanced concentration of bFGF, gradual increases were observed in the T level and the count of 3β-HSD-positive BMSCs in the four induction groups, with statistically significant differences between group B and groups C, D, and E (P < 0.05), as well as between group C and groups D and E (P < 0.05), but not between D and E (P > 0.05).

CONCLUSIONThe bFGF has an obvious promoting effect in the in vitro induced differentiation of rat BMSCs into Leydig cells.
