1.Ultrasonic manifestations of heptic mesenchymal hamartoma in children

Objective To describe the sonographic appearance of mesenchymal hamartoma of the liver(MHL)and to analyze the diagnostic value of ultrasound.Methods Eleven surgically and pathologically confirmed cases of MHL from January 2005 to May 2011 in the Beijing Children′s Hospital were retrospectively reviewed.Results Ultrasound examinations showed 9 cystic hamartomas,including 7 multiseptate cystic and 2 monocystic lesions.Of the 7 multiseptate cystic hamartomas,4 had a honeycomb appearance and 3 had irregularly-distributed multiple cysts with varied septations.Of the 2 monocystic hamartomas,1 had a large cystic portion while the other was mainly solid with approximately 4% cystic portion of the tumor.Two cases in this group were solid,presenting with a well-defined homogenous mass.Conclusions Ultrasonography is an effective imaging modality for the diagnosis for MHL.A mixed or a mainly-cystic liver mass found in a child less than 2 years old should be suspicious for MHL.
2.Influencing factor hospitalization expenses in patients undergone transsphenoidal pituitary tumor resection

Objective To analyze the main factors that influence the hospitalization expenses in patients undergone transsphenoidal pituitary tumor resection so as to provide basis for the development of clinical pathways and the control of irrational increase in medical expenses.Methods Data of patients undergone transsphenoidal pituitary tumor resection in our hospital in the years of 2008 and 2009 was collected.Statistical analysis was performed by using multinomial linear regression analysis.Results The major factors influencing the hospitalization expenses in pafients undergone transsphenoidal pituitary tumor resection included drug fees,material fees in operation,inspection fees and length of stay.Conclusions Avoiding overtreatment,reasonably lowering the drug fees,material fees in operation and inspection fees,as well as reducing the length of stay were the key to the control of the excessive growth of hospitalization expenses in patients undergone transsphenoidal pituitary tumor resection.
3.The pathological change of rats' benign hyperplastic prostate after radical denervation.

OBJECTIVETo study the pathological change of rats' benign hyperplastic prostate (BHP) after radical denervation.
METHODSA total of 65 male spontaneous hypertension rats (SHR) at 30 weeks age were randomly assigned into treatment group, sham surgery control group and normal control group. In surgery group, all the axonal branches of the major pelvic ganglion (MPG) supplying the bilateral prostate were truncated, followed performing of cystostomy; In sham surgery control group, only cystostomy was performed; In normal control group, no procedure was performed. The rats were sacrificed at 3, 7, 11, 15 and >or= 21 d post-operation respectively. The gross morphological changes of prostate in all animals were observed.
RESULTSIn treatment group, the prostate in 3 d post-operation showed granular solidification and shrunken volume and the changes occurred gradually over time. The glandular epithelial cells showed gradual degeneration, necrosis and detachment. The glandular epithelium became progressively thinner, the smooth muscles elongated and thinned progressively and the stromal components showed mild to moderate overgrowth. At the later stage, the glandular epithelium, glandular lumen and smooth muscles gradually disappeared and the prostate was largely replaced by connective tissues. Electron microscopic study showed that the glandular cells gradually underwent vacuolar degeneration and the structures of basement membrane became fuzzy. The smooth muscles cells degenerated overtime and the fibroblasts and collagenous fibers in the stroma overgrew slowly. At the late stage, most of the glandular cells became necrotic, the basal membrane and smooth muscle cells disappeared and collagenous fibers were highly hyperplasic. In surgery group in 3 d post-operation, the S-100 staining of nerve fiber was diffuse and disappeared after 11 d while it persisted normally in other groups. The two values in sham surgery control group showed no significant changes post-operatively.
CONCLUSIONSAfter radical denervation, the rat prostate with benign hyperplasia (gland and smooth muscles) undergoes dramatic atrophic changes and the volume decreases significantly. It suggests that this treatment may represent a novel therapy for BPH.

4.Foodborne pathogens in retail oysters in south China.

OBJECTIVETo investigate the occurrence of important foodborne pathogens in shellstock Pacific oysters in the food markets in South China.
METHODSFrom July 2007 to June 2008, retail oysters were collected in different seasons from South China and analyzed for the prevalence and levels of Listeria monocytogenes, Vibrio vulnificus and Vibrio parahaemolyticus.
RESULTSNone of L. monocytogenes could be detected in any of the 202 oyster samples tested, while E vulnificus and V. parahaemolyticus could be detected in 67 (54.9%) and 109 (89.3%) of the 122 oyster samples analyzed, respectively, with an MPN (most probable number) value greater than or equal to 3. V. vulnificus and V. parahaemolyticus with a more than 102 MPN/g were found in 36 (29.5%) and 59 (48.4%) of the 122 oyster samples, respectively. The tdh and trh genes were detected in 4 (0.3%) and 8 (0.6%) of the 1 349 V parahaemolyticus isolates, respectively. Of the 122 samples, 4 (3.3%) was positive for either tdh or trh. The levels of V. vulnificus and total V. parahaemolyticus in oysters in South China varied in different seasons.
CONCLUSIONV. vulnificus and pathogenic V. parahaemolyticus are frequently found in oysters in south China, which may pose a potential threat to public health. Data presented here will be useful for the microbiological risk assessment in oysters in China.

5.In vitro expression of hemophilia B gene mediated by lentivirus.

OBJECTIVETo construct a three plasmids lentiviral vector containing canine coagulation factor IX (cFIX) gene with ubiquinone promoter (PUB) and observe the expression of cFIX gene.
METHODSLentivirus was generated by transient three-plasmid transfection, namely, the VSV-G envelope expression cassette, the delta NRF packaging plasmid and the PTK 164 plasmid. Viral particles were used to infect the target cell, third passage mesenchymal stem cells (MSCs) and 293T cell respectively at MOI 3: 1. The cFIX activity was detected in cultured cells with one-stage clotting assay.
RESULTSThe MSCs were obtained in vitro. The lentivirus infected MSCs and 293T cells all expressed the active factor IX with the activity of (26.30 +/- 2.10)% and (19.70 +/- 1.53)%, respectively, which are significantly higher than that of control (1.00 +/- 0.05)%.
CONCLUSIONSThe lentiviral vector of three plasmids with ubiquinone promoter (PUB) was constructed and can transfect the MSCs and 293T cells.

6.Methylation of FHIT gene promoter region in DNA from plasma of patients with myelodysplastic syndromes and demethylating effect of decitabine.

This study was aimed to detect the methylation status of FHIT gene promoter region in the DNA from plasma of patients with myelodysplastic syndrome (MDS), and to investigate the demethylating effect of decitabine. Methylation-specific PCR method was used to detect the methylation status of FHIT gene promoter region in the DNA from plasma of 4 patients with MDS before and after treatment with decitabine plus semis CAG therapy (among them, 1 case of newly diagnosed MDS, 3 cases progressed into acute leukemia). The results indicated that 3 cases were found to have an increased methylation in the promoter region. After treatment with decitabine plus semis CAG, increased methylation was reversed in 2 cases. In 4 cases, 2 cases displayed clinical response. It is concluded that FHIT gene hypermethylation is associated with MDS pathogenesis. Decitabine has demethylating effect on the FHIT gene hypermethylation of plasma from MDS patients. Detecting the methylation status of FHIT gene in DNA from plasma may play a role in MDS auxiliary diagnosis or prognosis.

7.Analysis of Signal Peptide Fragment of Canine Distemper Virus Wild-type Strain Fusion Protein

A pairs of primers were designed according to the fusion protein(F)gene sequences of canine distemper virus(CDV)in GenBank.A 369 bp fragment aimed signal peptide fragment of F gene was amplified.The PCR products from viscera samples,blood,urine of fur animals including foxes,minks and raccoon dogs,which collected in the years 2005-2007,were cloned to pMD18-T Vector and sequenced.We obtained 13 positive signal peptide fragments from wild-type strains.The results indicated there was obviously genetic diversity between the wild-type strains and CDV3 and other vaccine strains.The homology with CDV3 is 80.7%-83.2%in nucleotide,and 64.8%-71.3%in amino acid.The analysis for the hydrophobic regions indicated the function of signal peptide fragment may be changed.This study can offer aca- demic data to research of CDV genetic variation and epidemiology.
8.Detection of residual leukemic cells after complete remission of patients with acute myeloid leukemia.

The aim was to study minimal residual disease (MRD) in blood and bone marrow after complete remission of patients with acute myeloid leukemia (AML) and explore the role of MRD in detecting relapse of acute myeloid leukemia. The blood and bone marrow samples from 33 AML patients who had been in complete remission were determined for residual leukemic cells (RLC) with flow cytometry. The results showed that RLC in AML group of complete remission was higher than that of normal group both in blood by (4.7518 +/- 4.1537)% vs (0.4835 +/- 0.2005)% and bone marrow by (17.9082 +/- 20.4819)% vs (0.7285 +/- 0.2209)%, while the RLC in relapsed group was higher than that in non-relapsed group both in blood by (2.233 +/- 1.5923)% vs (10.2369 +/- 9.4714)% and bone marrow by (4.779 +/- 3.0336)% vs (38.0685 +/- 19.4295)%. In conclusion, early detection of leukemic residual cells with flow cytometry contributes to treatment of relapse in time.

9.Study on the clinical effects of psychological nursing intervention on 160 cases of primary hypertension

Objective To explore the clinical effects of psychological nursing intervention on cases of primary hypertension. Methods A total of 160 patients with primary hypertension were randomly divided into the intervention group and control group with 80 patients in each group. The control group received the routine medical treatment, while the intervention group received the psychological care in addition to the routine medical treatment.The hospitalization days, the adherence behavior before and after the intervention, systolic blood pressure (SBP) and diastolic blood pressure (DBP) after the intervention were observed in patients. Results There was statistical significance in the hospitalization days between two groups [(14±6.67)d vs (20±7.32)d,P< 0.01].There was statistical significance in the compliance behaviors and decline blood pressure between the intervention group and control group(P<0.01). Conclusions Psychological care plays a very important role. in the treatment of primary hypertension
10.Effect of self-made meridian compression stocking on the bladder function recovery of patients with extensive hysterectomy

Objective To explore the self-made meridian compression stocking to extensive hysterectomy patients with bladder function recovery.Methods Sixty patients who received retained urethral catheterization for more than 1 week after extensive hysterectomy were randomly divided into control group (n =30) and experimental group (n =30).The control group received bladder training by intermittent catheter clamping before catheter removal as routine nursing care for retained urethral catheterization after gynaecologic surgery.While the experimental group,on the basis of conventional nursing,self-made meridian compression stocking was adopted.Then,the urinary retention and urinary tract infection incidence,the time of urinary catheter were observed and compared.Results Urinary retention and urinary tract infection rate respectively were 6.67% and 10.00% in the experimental group,and respectively were 30.00% and 33.33% in control group,and the differences were statistically significant (x2 =5.45,4.81 ; P < 0.05).Indwelling catheter days comparison were (9.15 ±2.03)d in the experimental group and (10.97 ± 3.38)d in the control group,with significant difference(t =2.55,P < 0.05).Conclusions Extensive hysterectomy patients wearing self-made meridian compression stocking can promote bladder functional recovery,and reduce urinary retention and urinary tract infection incidence.
