Objective To investigate the characteristic of ?-amyloid precursor protein (A?) processing in activated platelet in AD.Methods Thirty-six sporadic AD patients and 30 control subjects were included in this study.Blood was collected from the subjects to separate platelets.After treated by thrombin,the soluble amyloid precursor protein (APP) level in the snpernatants of platelets from 36 were analyzed by means of western blot with a specific antibody recognizing soluble APP.Meanwhile A? level was measured by radioimmunoassay.Results After treated with thrombin,the level of soluble APP in the supernatants of platelets in patients with AD decreased by 31.0% (P

Analyzed the prescriptions for phlegm retention syndrome that built by Ma Peizhi by the association rules and clustering algorithm, the frequency of drug usage and the relationship between drugs could be get. And from that we could conclude the experiences for phlegm retention syndrome of Ma Peizhi of menghe medical genre. The results of the analysis were that 18 core combinations were dig out, such as Citri Exocarpium Rubrum-Eriobotryae Folium-Citri Reticulatae Pericarpium. And there were 9 new prescriptions were found out such as Aurantii Fructus-Citri Exocarpium Rubium-Eriobotryae Folium-Citri Reticulatae Pericarpium. The results of the analysis were proved that Ma Peizhi of Menghe Medical Genre was good at curing phlegm retention syndrome by using the traditional Chinese medicine of mild and light, such as the medicines of mild tonification, and clearing damp and promoting diuresis.

The clinical prescriptions of hemoptysis that built by Menghe physician Ma Peizhi were collected, and were analyzed by the method of unsupervised data mining, such as association rules and clustering algorithm. From the prescriptions, we got the frequency of drugs, the association rules among drugs, 10 core drug combinations and 5 new prescriptions. Accordingly, after the analysis, Menghe physician Ma Peizhi had rich experience in the treatment of hemoptysis. His therapy for hemoptysis was clearing heat and moistening lung, dispel heat from blood to stop bleeding, and enriching yin and blood. And we can also make a conclusion that the TCM inheritance support system is of great practical value for minning the old doctors' clinical experiences.

5.Effect of knockdown microRNA-214 on drug resistance of different lymphoma cell types

Objective To explore the function of microRNA-214 on the drug resistance of different lymphoma cell types.Methods The Jurkat cells,OCI-Ly3 cells and Raji cells were selec-ted as the experimental groups.The healthy peripheral blood lymphocytes (Scr) were selected as the control group.The cells were miR-214 knockdown and stimulated with different drugs.The expression of miR-214 in different types of lymphoma ceils was detected by real-time PCR.The cell viability of lymphoma cells after knockdown miR-214 was detected by CCK-8.The drug sensitivity was detected by methyl thiazolyl tetrazolium colorimetric method(MTT) assay.The BCL11B protein levels was detected by Western blot.The luciferase reporter assay was used to detect whether BCL11B was the target gene of miR-214.Results The miR-214 expression in the experimental groups of Jurkat,Oci-ly3 and Raji cells were 27.21 ± 1.37,17.13 ±2.11,19.31± 2.46,which were significantly higher than those in control group of 1.17 ± 0.28,the difference was statistical significance (P < 0.05).After knockout miR-214,compared with the cell viability in control group (86 ± 12)%,the cell viability of Jurkat,OCI-Ly3 and Raji were (42 ± 13)%,(65 ± 11)%,(58 ± 7)%,the difference were significantly decreased (P < 0.05).The drug sensitivity of Scr cells (control group) to doxorubicin,cyclophosphamide,daunorubicin,mito-mycin C and vincristine were 0.46 ± 0.11,1.96 ± 0.13,1.15 ± 0.22,3.90 ± 0.52,0.52 ± 0.15,respectively.The drug sensitivity of Jurkat cells to five drugs were 0.32 ±0.07,0.76 ±0.16,0.73-±0.13,2.36 ±0.16,0.37 ±0.06.The drug sensitivity of OCI-Ly3 cells to the five drugs were 0.28 ±0.06,0.91 ±0.12,0.55 ±0.09,2.01 ±0.36,0.23 ± 0.07,respectively.The drug sensitivity of Raji cells to five drugs were 0.36 ± 0.04,0.82 ± 0.08,0.64 ± 0.07,3.31 ±0.21,0.35 ±0.05,respectively.Which indicated that the drug sensitivity of lymphoma cells to various drugs was significantly decreased after the inhibition of miR-214 expression.The BCL11B was the direct target of miR-214.Conclusion Inhibition of miR-214 expression could decrease the cell viability of lymphoma cells and decrease the multidrug resistance of leukemia lymphocytes,which may be related to the miR-214 knockdown on the expression of BCL11 B.
6.Clinical effect of flurbiprofen axetil injection on preemptive and postoperative analgesia in orthopedic surgery patients

Objective To determine the analgesic effects of flurbiprofen axetil injection on orthopedic surgery patients.Methods Seventy-eight orthopedic surgery patients were randomly divided into control group ( n=39 ) and test group ( n=39 ).Patients in control group were given intravenous administration of parecoxib 20 mg at 15 min before anaesthe-sia induced by propofol 1-2 mg · kg -1 and maintained by isoflurane in operation; after operation , 60 mg of parecoxib dissolved in 100 mL 0.9%NaCl was intravenously dripped.Patients in test group were given the same treatment beside the 20 mg of parecoxib was changed to 50 mg of flurbiprofen axetil.Average artery pressure ( MAP ) and heart rate (HR), visual analogue score ( VAS), Ramsay sedation score ( RSS), levels of serum C reactive protein ( CRP ) , prostaglandin E 2 ( PGE2 ) and incidence of adverse drug reactions of the two groups were observed.Results The MAP and HR, VAS, RSS of two groups showed no statisti-cal significance expect the VAS at moment post surgery; the content of serum CRP in test group at postoperative 24 h was ( 16.77 ±5.52 ) mg · L-1 , significantly lower than that of control group of ( 20.68 ±7.76 ) mg · L-1 ( P<0.05 ); the PGE2 content in test group had no significant difference with control group ( 135.1 vs 141.6 pg · mL -1 , P >0.05 ).The incidence of adverse drug reaction in test group was significantly lower than that in control group (7.7%vs 20.5%, P<0.05 ).Conclusion Flurbiprofen axetil injection can reduce the stress reaction of orthopedic surgery patients , the incidence of side effect was also lower.
7.CD133 selected stem cells from proliferating infantile hemangioma and establishment of an in vivo mice model of hemangioma

Background Infantile hemangioma (IH) is the most common benign tumor in children with prevalence in the face and neck.Various treatment options including oral propranolol have been described for IH,but the mechanism of drugs remains enigmatic.The aim of this study was to investigate the pathogenesis and establish a reliable in vivo model of IH which can provide platform for drug exploration.Methods Stem cells from the proliferating hemangiomas (HemSCs) were isolated by CD133-tagged immunomagnetic beads.Their phenotype and angiogenic property were investigated by flow cytometry,culturing on Matrigel,real-time polymerase chain reaction (PCR),immunofluorescent staining and injection into BALB/c-nu mice.Results HemSCs had robust ability of proliferating and cloning.The time of cells doubling in proliferative phase was 16 hours.Flow cytometry showed that HemSCs expressed mesenchymal markers CD29,CD44,but not endothelial/hematopoietic marker of CD34 and hematopoietic marker CD45.The expression of CD105 was much lower than that of the reported hemangioma derived or normal mesenchymal stem cell (MSC).Real-time PCR showed that the mRNA levels of vascular endothelial growth factor (VEGF),basic fibroblast growth factor (bFGF) and matrix metalloproteinase-1 (MMP-1) of HemSCs were higher than that of neonatal human dermal fibroblasts (NHDFs) and human umbilical vein endothelial cells (HUVECs).After HemSCs were cultured on Matrigel in vitro,they formed tube-like structure in a short time (16 hours) and differentiated into endothelial cells in 7 days.After 1-2 weeks of implantation into immunodeficient mice,HemSCs generated glucose transporter 1 positive blood vessels.When co-injected with HUVECs,the vascularization of HemSCs was greatly enhanced.However,the single implantation of HUVECs hardly formed blood vessels in BALB/c-nu mice (P <0.05).Conclusions HemSCs may be some kinds of primitive mesoderm derived stem cells with powerful angiogenic ability,which can recapitulate human hemangioma by co-injecting into immunodeficient mice with HUVECs.
8.Meta-analysis on curative effects of surgical procedures for intrahepatic bile duct lithiasis.

OBJECTIVETo compare curative effects of various surgical procedures of bile duct stones.
METHODSTwo thousand nine hundred and fifty-five patients with intrahepatic bile duct lithiasis who had undergone various surgical procedures were analysed with Meta-analysis. Some of these cases were reported in Chinese Medical Journals from January 1990 to March 2001 and others were from Tongji Hospital.
RESULTSThere was a significant difference between curative effects of non-hepatectomy and that of hepatectomy (chi(2) = 62.945, P < 0.01), and the outcomes of hepatectomy were much better than those of non-hepatectomy with OR(S) equalled to 0.303 (0.222 - 0.413). There was not a significant difference between curative effect of interposed jejunum and that of hepatectomy (95% CI of RR from 0.98 to 1.04). All the other operation, effects were worse than hepatectomy (upper limit of 95% CI of RR < 1).
CONCLUSIONSHepatectomy is the most ideal surgery for intrahepatic bile duct stones and operation methods should be diversified since good effect could also be obtained when other operations are performed on suitable cases.

9.Disequilibrium linkage between the polymorphism in exons 2, 3 and 4 of the MICA gene and HLA-B antigen of patient with ankylosing spondylitis.

OBJECTIVETo investigate the association between the exons 2 to 4 of the MICA gene and ankylosing spondylitis (AS).
METHODSBy PCR-SSOP, DNA samples from 56 AS patients and 112 random healthy individuals, as normal control were genotyped to analyse the polymorphism in exons 2, 3, 4 of the MICA alleles.
RESULTSMICA*008 was dominant in MICA allele,accounted for 32.14% and 30.36% in AS patients and normal controls respectively. The frequency of MICA*007 was significantly increased in AS patients, when compared with normal controls (chi-square=10.18, P<0.05, RR=2.50). No difference was found in the other MICA alleles. The haplotype analysis revealed that there were the strong linkage disequilibrium between MICA and HLA-B of AS patients, and normal controls. There was a difference in MICA*007-B27 between two groups (chi-square=18.46, P<0.05, RR=7.47). Both HLA-B27 and MICA*007 were strongly associated with AS. Stratified analysis showed that HLA-B27 was significantly relative to AS,while it was not found between MICA*007 and AS.
CONCLUSIONThe increased frequency of MICA alleles may be due to its strong linkage disequilibrium with HLA-B27.

10.Application of alpha-fetoprotein in the diagnosis of hepatocellular carcinoma.

OBJECTIVETo evaluate the usefulness of serum alpha-fetoprotein (AFP) in clinical diagnosis and screening for hepatocellular carcinoma (HCC).
METHODSTotally 290 HCC patients, 48 liver cirrhosis patients, and 49 healthy subjects were enrolled in this study. Serum AFP analysis was performed to investigate the correlation between the serum AFP level in HCC and the clinical or biochemical parameters of the disease, which included the size and number of tumor and the TNM stage. Sensitivities and specificities of AFP in HCC prediction at different cut-off levels were determined.
RESULTSThe serum AFP level was significantly higher in HCC patients than in liver cirrhosis patients (P = 0.0274) and healthy subjects (P = 0.0001). Among 290 HCC patients, 95 patients (32.8%) were AFP-negative (AFP < 20 microg/L), 195 (67.2%) were AFP-positive (AFP > or =20 microg/L). Sensitivity and specificity of AFP at 20 microg/L cut-off was 67.2% and 29.2%, respectively, and the positive and negative predictive value was 85.2% and 12.8%, respectively. Sensitivity of AFP at 400 microg/L cut-off was only 42.8%. Serum AFP levels were significantly different among HCC with different tumor size (P = 0.0009), tumor number (P = 0.0001), and TNM stage [TNM I vs. TNM III-IV (P = 0.0001); TNM II vs. TNM III-IV (P = 0.0003)].
CONCLUSIONSIncreased serum AFP level is highly suggestive in HCC diagnosis. Combined with other imaging examinations, AFP level can be used for the screening of high risk population and for the follow-up of AFP-positive patients.
