Objective:To explore the effect of nontypeable Haemophilus influenzaen(NTHi)strain ATCC49247on pri-mary rabbit tracheal epithelial(TE)cells.Methods:TE cells were isolated with low temperature protease digestion and cul-tured on collagen gel-coated membranes at an air-liquid interface in serum-free medium.Under these conditions,TE cells were proliferated and differentiated into a pseudostratified mucociliary epithelium,which were infected by NTHi.Morphologic changes of the cells were examined by scanning electron microscopy(SEM)and transmission electron microscopy(TEM)after 24h.Results:SEM showed that bacteria adhered to non-ciliated cells;death or apoptosis occurred in90%of TE cells and cil-iaries were broken.TEM showed NTHi adhered to the cell surface on which there were many microvillus.Lamellipodia and microvilli surrounded bacteria within vacuoles of airway cells.Conclusion:NTHi can attach to non-ciliated cells,the latter de-vours the bacteria by lamellipodia and microvilli.NTHi is toxic to TE cells,resulting in the death or apoptosis of TE cells.

This study is designed to obtain recombinant human acetylcholinesterase (rhAChE) and apply it in screening acetylcholinesterase inhibitors. The rhAChE was overexpressed in HEK293 cells transfected by plasmid of pCMV-AChE with the cationic liposome and rhAChE was found to be secreted into cell culture medium. AChE activity was assayed according to modified Ellman method to obtain kinetic parameters. IC so50 values for donepezil compounds of rhAChE were calculated to determine their activities of inhibition. The results showed that Km value was 151.9 micromol.L-1 donepezil inhibited rhAChE in a mixed competitive-noncompetitive way (Ki= 16.03 nmol.L-1, Ki = 18.36 nmol.L-1) and that most new compounds tested exhibited high activities of inhibition on rhAChE. The study suggests that rhAChE is available to be applied in screening AChE inhibitors in vitro.

Objective To investigate the neurodevelopmental outcomes and its risk factors of very low birth weight infant (VLBWI) and extremely low birth weight infants (ELBWI).Methods Data of 85 VLBWI and ELBWI hospitalized in Children's Hospital of Fudan University from October 2005 to November 2009 who had finished infant development test of Baley Scales of Infant Development Ⅱ (Bayley Ⅱ) for neurological development at corrected gestational age between 18 to 42 months were retrospectively reviewed.Twelve infants who accepted treatment in other hospital over 10 days were excluded; the rest 73 infants were divided into normal (neurodevelopment) group or abnormal group according to the definition of neurodevelopmental impairment reported by National Institute of Child Health and Human Development (NICHD) Neonatal Network.Potential risk factors of neurodevelopmental impairment were analyzed with Logistic stepwise regression.Results The mean gestational age of 73 infants was (30.4±2.3) weeks; among which 13 were smaller than 28 weeks,42 between 28 and 32 weeks,and 18 older than 32 weeks.The mean birth weight was (1208.0±208.5) g; among which 15 (20.6％) ＜1000 g,and 58 (79.4％) were between 1000 g and 1500 g.Four babies (5.5％) were diagnosed as movement retardation,and neurodevelopmental impairment occurred in 16 cases (21.9 ％),psychomotor developmental index ＜70 occurred in 6 cases (8.2％,one case complicating with cerebral palsy); mental developmental index ＜70 occurred in 2 cases (2.7％); both psychomotor developmental index and mental developmental index ＜70 occurred in 7 cases (9.6 ％,two cases complicating with cerebral palsy),and one case (1.4 ％) was cerebral palsy only.Blind in either eyes and hearing impairment requiring deaf-aid were not found in any of the 73 babies.Logistic stepwise regression showed that use of mechanical ventilation was related to neurodevelopmental impairment (OR =6.183,95％ CI:1.664-22.983,P =0.003).Psychomotor developmental index of infants who needed mechanical ventilation (77.5±15.1) was lower than that of infants did not need (87.3±15.1)(t=2.646,P=0.010).Conclusions VLBWI and ELBWI are in high risk of neurodevelopmental impairment,especially those who need mechanical ventilation.

Objective To study the value of measurement of sinus node artery in adults with multi-slice spiral CT(MSCT).Methods 119 cases were examined by MSCT coronary artery angiography.The diameter of the sinus node artery,the distance from the origin of the sinus node artery to the origin of the coronary artery(S-O),to the origin of left circumflex branch(LO-O),and to straightly the aortic wall erectly(S-W) and the angle between the sinus node artery and the proximal segment of the coronary artery(the angle of S-C) were measured on axial and MIP images.Results The origin,morphological characteristics of the sinus node artery were shown clearly on CT coronary artery angiography,and the diameter,length,the distance of S-W,S-O and LO-O and the angle of S-C were measured precisely.Conclusion The parameters of the sinus node artery measured by MSCT are basically corresponded with those on anatomy,MSCT may be used to show the sinus node artery.

Objective To detect the meshwork of tiny lymphatic channels at bilateral vertebral body and ventral aorta at 3.0 Tesla and to probe the relationship between its detective ratio and some diseases.Methods Fat-suppressed T2-weighted imaging in 91 patients was performed at the upper abdomen. The meshworks of tiny lymphatic channels at bilateral vertebral body and ventral aorta on contiguous axial and coronal images were reviewed. The classifications of the lymphatic meshwork were made according to its width and length on coronal images. The differences in the detective ratios of the lymphatic meshwork between control group and the risk ones including portal hypertension group, choledochus obstruction group and malignant tumour group were analyzed.Results Right- or left-sided lymphatic meshwork at vertebral body or ventral aorta were 34.07%(31/91),32.97%(30/91),32.97%(30/91) and 43.96%(40/91) respectively. The detective ratio of the lymphatic meshwork on the side of the ventral aorta was higher significantly in the portal hypertension group than that in the control or malignant tumour one (P0.05). Conclusion Meshwork of tiny lymphatic channels at bilateral vertebral body and ventral aorta allows to be detected on fat-suppressed T2-weighted imaging at 3.0 Tesla, and the portal hypertension may influence the detective ratio of the lymphatic meshwork on the side of the ventral aorta.

Cryptosporidium andersoni ATP-binding cassette (CaABC) is an important membrane protein involved in substrate transport across the membrane. In this research, the nucleotide binding domain (NBD) of CaABC gene was amplified by PCR, and the eukaryotic expression vector of pEGFP-C1-CaNBD was reconstructed. Then, the recombinant plasmid of pEGFP-C1-CaNBD was transformed into the mouse intestinal epithelial cells (IECs) to study the iron transportation function of CaABC. The results indicated that NBD region of CaABC gene can significantly elevate the transport efficiency of Ca2+, Mg2+, K+, and HCO3 - in IECs (P<0.05). The significance of this study is to find the ATPase inhibitors for NBD region of CaABC gene and to inhibit ATP binding and nutrient transport of CaABC transporter. Thus, C. andersoni will be killed by inhibition of nutrient uptake. This will open up a new way for treatment of cryptosporidiosis.

OBJECTIVETo study the fingerprint of dragon's blood resina draconis by high performance liquid chromatography.

METHODThe samples are extracted with methanol and separated on a Eclipse XDB-C18 column (4.6 mm x 150 mm, 5 microm) with the mobile phase of acetonitrile-H2O in gradient mode, and the flow rate was 1.0 mL x min(-1), the detection wavelength was 275 nm and the temperature of column was 40 degrees C. Loureirin B was used as the reference compound.

RESULTHPLC fingerprint of dragon's blood was established and the similarity of the fingerprint was compared.

CONCLUSIONThe method is simple, accurate, and can be used to control the quality of dragon's blood.

Objective To evaluate the acute and delayed killing effect of high intensity focused ultrasound (HIFU) on Echinococcus granulosus(E. granulosus)protoscolices in vitro.Methods E. granulosus protoscolices were treated with different dosage of effective power(0,25,50,100,200,250 W)and time(5,10,20,30,40,50,60 s)of HIFU in vitro to obtain the dosage-effect curves.Then the survival pmtoscolices were incubated,and the mortality of each group was counted daily.The protoscolicidal effects were investigated by trypan blue exclusion assay.Results Compared with the untreated group,the Vitality of E.granulosus protoscolices significantly decreased immediately after treated by HIFU of different dosage(F=5201.59 vs 1865.65,P＜0.05),there were the interaction both different dosage and time(F=214.50,P＜0.05).The protoscolices were broken into pieces by HIFU of 250 W×40 s,whereas the growth of the surviving protoscolices after exposed to HIFU was obvious suppressed.Both the acute killing effect and the delayed inhibitory effect showed a dosage-dependant manner.The inhibitory effect increased along with the increased dosage of HIFU(P＜0.05).The inhibitory effect in 50 W×10 s group was stronger than 25 W×20 s group(P＜0.05).The mortality was increased in parallel with the increase of HIFU dosage.Conclusions HIFU show an effective immediately killing effect,as well as a growth-inhibiting effect on the E.granulosus protoscolices in vitro.
