1.Research on immune-modulation effects and its mechanisms of Guizhi Fuling capsule and its active ingredient combination.

The effects of Guizhi Fuling capsule and its active ingredient combination within different concentration on SPL proliferate were observed by MTT method. The ratio of CD80/86, CD3CD25 and CD3CD69 was used to evaluate cell activation effects of Guizhi Fuling capsule and its active ingredient combination by FCM. Guizhi Fuling capsule with concentration of 400 mg · L(-1)can promote spleen lymphocyte proliferation, as well as the active ingredient combination, which showed the obvious dose-effect relationship. Compared with control group, the difference has statistical significance (P≤0.01). The result of FCM showed that Guizhi Fuling capsule and its active ingredient combination can promote CD80 and CD86 expression on spleen lymphocyte, and also can increase CD25 and CD69 ratio between spleen CD3+ cells. Compared with control group, the difference has statistical significance (P≤0.01). Thus, Guizhi Fuling capsule and its active ingredient combination may have immune-modulate effects, and the mechanism may have a close relationship with the lymphocyte activation.

2.Effect of LEEP knife to treat cervical lesion and its nursing

Objective To observe the effect of LEEP knife in the treatment of cervical lesions and investigate its nursing methods.Methods Totals of 270 patients with cervical lesions were treated with LEEP knife.Their operative time,blood loss,postoperative CCT abnormal number after 3 months,number of cervical stenosis and cervical repair time were observed and compared with those of 220 cases who used conventional electric knife for surgery during the same time.Results The operative time and blood loss were (6.3 ± 0.8) min and(3.2 ±0.5) ml in the LEEP knife group,less than (17.3 ± 2.1) min and (8.0 ± 1.0) ml in the conventional group,and the differences were statistically significant (t =79.311 5,69.003 9,respectively;P <0.05).The number of cervical stenosis and postoperative CCT abnormal number after 3 months were 6 and 22 in the LEEP knife group,less than 21 and 55 in the conventional group,and the differences were statistically significant (x2 =5.386 2,25.991 8,respectively;P < 0.05).The cervical repair time was (13 ± 1)d in the LEEP knife group,shorter than (22 ± 2) d in the conventional group,and the difference was statistically significant (t ==64.691 1,P <0.05).Conclusions LEEP knife is an effective way to treat cervical lesions,with effective interventions,which can reduce blood loss,shorten recovery time after cervix,and decrease the number of cervical stenosis as well as postoperative CCT abnormal number.

Objective To determine the expression of membrane-bound B lymphocyte stimulator (BLyS) protein and its mRNA in vitro of peripheral blood mononuclear cells (PBMCs) from individuals with systemic lupus erythematosus (SLE),and to investigate the role of interferon-?(IFN-?) on the expression of BLyS.Methods PBMCs were obtained from 25 SLE patients (mean age of 31+14) and 20 healthy volunteers (mean age of 28?10).They were randomized into IFN-?(5 ng/ml) group and control group.PBMCs were col- lected at 0,6,12 and 24 h for BLyS mRNA assessment using semi-quantitative reverse transcription-PCR (RT-PCR).PBMCs were also collected at 72 h for membrane-bound BLyS protein detection using flow cy- tometry (FACS) and direct immunofluorescence.Results①The expression of BLyS mRNA and membrane- bound protein in PBMCs was significantly higher in individuals with SLE compared with healthy controls (P<0.05);②IFN-?enhanced BLyS mRNA expression in PBMCs in both healthy controls and SLE patients,with the greatest effect at 6 h (stimulated vs unstimulated,0.42?0.19 vs 0.25?0.14,P<0.01;0.59?0.28 vs 0.44?0.21,P<0.01 );③IFN-?also increased the expression of membrane-bound BLyS protein in both healthy con- trols and individuals with SLE (FACs,mean fluorescence intensity,4.5+3.0 vs 3.7~2.6,P

Objective To observe the sequence of fat deposit and its relationship with insulin resistance in SD rats fed by high fat diet.Methods Normal 8-week-old male SD rats were randomly divided into normal chow (NC,n=40)and high fat diet(HF,n=40)groups.Triglyceride(TG)in serum,liver and muscle were measured;glucose infusion rate(GIR)and the mRNA level of genes related to lipid metabolism in liver and muscle were determined in different stages.GIR was detected by eugiyeemic-hyperinsulinemic clamp for evaluating the insulin sensitivity.Gene expression was determined by real-time PCR.Results(1)As compared with NC group,serum TG was not increased after high fat feeding for4 and 8 weeks,it began to increase after 12 weeks [0.52(0.15-1.00) mmol/L vs O.31(0.09-0.53)retool/L, P0.05)in skeletal muscle.After 8 weeks,the expression of ACC1 in liver in HF group was increased by 20.6%,CPT-1 was decreased by 27.1%(P
5.Investigation of satisfaction on the model of continuous scheduling and group-responsibility system

Objective To explore the satisfaction of nurses on the work and patients on the nursing service after implementing the model of continuous scheduling and group-responsibility system,so as to to provide theories for improving nursing quality and human management.Methods Self-designed satisfaction questionaire was used to investigate nurses and patients before and after implementing the model of continuous scheduling and group-responsibility system,and nurses'satisfaction on the work and patients'satisfaction on the nursing care were observed and compared.Results The comprehensive satisfaction of nurses on the work in sixth,ninth,twelfth month after implementing the model of continuous scheduling and group-responsibility system was 91.0%,91.6%,92.3%,respectively which was higher than 80.6% before implementing,and the difference was statistically significant ( x2 =4.74,5.56,6.40,respectively ; P < 0.05 ).The satisfaction of patients on nursing increased from 90.1% to 95.7% after the model was implemented three to twelve months,which was significantly higher than 78.5% before implementing( P < 0.01 ).Night shift period was shortened and the nurses' satisfaction was decreased from 76.2% to 61.8% after the model was implemented ( x2 =6.95,P < 0.01 ),while there was no difference in the situation of their mental pressure.Conclusions The scheduling revolution and group-responsibility system can provide no-gap and excellent nursing,and stimulate nurses' potential,and improve the nursing quality.But the situation of nurses' mental pressure is not improved for the lack of clinical nurses.
6.A study on clinical differential diagnosis between hemangioma and vascular malformation in infant.

OBJECTIVETo explore the main points of clinical differentiation between hemangioma and vascular malformation in infant.
METHODSBased on Mulliken and Waner's classification, from March, 1997 to February, 1999, 81 baby patients with hemangioma were included in this study. Thirty-eight cases, 43 cases received medical treatment of steroids.
RESULTSAll the patients were followed up from 5 to 7 years. Thirty-eight cases of red strawberry-like lesions limited in the skin began to involute within two years old. Of the 30 patients with strawberry-like lesions and subcutaneous mass, 20 cases involuted in varying degree; 10 cases' subcutaneous mass grew gradually and didn't involute, in 4 cases biopsy was performed, 3 cases were confirmed as hemangioma accompanied with venous malformation by pathology, 1 case was hemangioma accompanied with arteriovenous malformation. Of 13 cases with light blue or normal skin and subcutaneous mass, 7 cases involuted in varying degree; 6 cases grow gradually and didn't disappear, 2 cases were confirmed as venous malformation by biopsy.
CONCLUSIONSHemangioma in infant begins to involute within two years old. Vascular malformation or hemangioma with deep vascular malformation grows persistently and does not disappear. Skin temperature of lesion surface and dilative veins on the skin artery pulsation, are indexes compressibility, for differentiation between hemangioma and vascular malformation in clinical diagnosis.

10.Ultrastructural and immunohistochemical studies on Trichomonas vaginalis adhering to and phagocytizing genitourinary epithelial cells.

BACKGROUNDTrichomonas vaginalis (T. vaginalis) belongs to a common sexually transmitted disease pathogen causing genitourinary trichomoniasis in both sexes. We investigated the pathogenetic mechanism of genitourinary trichomoniasis.
METHODSCultured T. vaginalis bodies were injected into the vaginas of rats, or incubated with genitourinary epithelial cells of female subjects, male subjects, and sperm. The ultrastructural and microscopic changes were observed via transmission and scanning electron microscopy and through microscopic histochemistry.
RESULTSGroups of T. vaginalis adhered to PAS positive columnar cells at the surface of stratified epithelium in the middle and upper portions of the vaginas. They also traversed under these cells. The parasites were shown to be PAS, cathepsin D, and actin positive, and they could release hydrolase into the cytoplasm of adhered epithelial cells. In the amebiform T. vaginalis, microfilaments were arranged into reticular formation. Similar phenomena were found during the interaction of T. vaginalis with host cells, both in vitro and in vivo. Usually several protozoa adhered to an epithelial cell and formed polymorphic pseudopodia or surface invaginations to surround and phagocytize the microvilli or other parts of the epithelial cytoplasm. Adhesion and phagocytosis of sperm by the protozoa occurred at 15 - 30 minutes of incubation. Digestion of sperm was found at 45 - 75 minutes and was complete at 90 - 105 minutes.
CONCLUSIONST. vaginalis tends to parasitize at the fornix of the vagina, because this is the site where columnar cells are rich in mucinogen granules and their microvilli are helpful for adhesion and nibbling. T. vaginalis possesses some invading and attacking abilities. Shape change, canalization, encystation, phagocytosis, digestion, the cell coat, cytoskeleton, and lysosome all play important roles in the process of adhesion. They have two methods of phagocytosis: nibbling and ingestion. Genitourinary epithelium may be injured directly by the digestive action of hydrolases, phagocytosis, and the mechanical action of pseudopodia.
