1.Establishment of a mouse model of primary biliary cirrhosis by AMA M2 autoantigen injection.

OBJECTIVESTo establish a primary biliary cirrhosis (PBC) model by AMAM2 autoantigen injection into C57BL/6 mice.
METHODSMice of the model group were immunized intraperitonealy with 200 microl of purified recombinant AMAM2 autoantigen in complete Freund's adjuvant (CFA). Mice immunized with bovine serum albumin and CFA in the same way were used as negative controls. Sixty-six weeks later, mice were sacrificed and their sera were collected. Sera samples were assayed for AMAM2 autoantibody, alkaline phosphatase (ALP), ALT and total bilirubin (TBil). Their liver, stomach, muscle and kidney tissues were sectioned and stained using HE to observe the pathological changes.
RESULTSAntibodies to AMAM2 autoantigen were readily induced in the model group. The mice in the model group had no significant changes in the level of serum ALT and TBil but had an obvious increase of ALP (P<0.05). The stomach, muscle and kidney tissues showed no evident damage while the livers had obvious pathological changes, including bile duct degeneration or proliferation, and mononuclear cell infiltration.
CONCLUSIONThe AMAM2 autoantigen-induced PBC animal model was successfully established in C57BL/6 mice in our experiment and its characteristic biochemical and pathology are quite similar to that in the early stage of human PBC. This model may provide a useful experimental approach for further study of the pathogenesis and clinical treatment of human PBC.

2.A new sesquiterpenoid from fungus Colletotrichum sp. and its cytotoxicity.

A novel sesquiterpenoid (1) and three known compounds identified as isoaltenuene (2), altenuene (3), and alternariol 4, 10-O-dimethyl ether (4), were isolated in our investigation of the cytotoxic constituents from solid cultures of the endophytic fungus Colletotrichum sp. The structures of these compounds were elucidated through spectroscopic data analysis. All compounds exhibited cytotoxic activity against lung cancer cell line A549, breast cancer cell line MDA-MB-231 and pancreatic cancer cell line PANC-1. Compound 4 could induce the PANC-1 cells inflation or death, but couldn't induce apoptosis at the IC50 of 60.2 microg x mL(-1).

3.Clinical study of application of platform switching to dental implant treatment in esthetic zone.

OBJECTIVETo evaluate the clinical results of dental implant treatment with platform switching technique in esthetic zone and to investigate its technical characteristics.
METHODSSeventeen patients who were partially edentulous in esthetic zone were included and received a total of 26 Frialit-2 system implants in Center of Dental Implantation, Peking University School and Hospital of Stomatology from January to December 2001, and the platform switching technique was applied. The mean follow-up time was 56.3 months (24 to 74 months). Clinical examination and X-ray were conducted.
RESULTSClinical results of all implant restoration that used platform-switched abutments were satisfied. No implant loss or other severe complications occurred. The medians of marginal bone loss 1 year and 5 years after final restoration was 0.13 (-0.12 - 0.55) mm and 0.27 (-0.04 - 0.77) mm respectively. The medians of interimplant crestal bone resorption was 0.14 (0.00 - 0.30) mm and 0.21 (0.09 - 0.31) mm after 1 year and 5 years' functional loading respectively. According to Jemt classification of papilla, 23 sites reached II degree, and 10 sites reached III degree among 36 sites 5 years after prostheses. The mean value of pink esthetic score (PES) was 11.8 (10 - 14).
CONCLUSIONSPlatform switching is a simple and reliable technique for dental implant treatment, helping to control marginal bone loss and ensure an esthetic results in the esthetic zone.

4.Cytogenetic study of 1780 cases of spontaneous abortion.

OBJECTIVE:
To explore the relationship between chromosome anomaly and spontaneous abortion, and to provide useful information for genetic counseling and prenatal diagnosis in reproductive clinic.
METHODS:
A total of 1 780 patients who had a history of spontaneous abortion before 24 weeks of gestation were enrolled. The lymphocyte culture and harvest were performed according to standard methods. Karyotypes were analyzed by G-banding in all cases and C- banding in some cases in addition.
RESULTS:
Altogether 57 abnormal karyotypes were found and the overall incidence of chromosomal abnormalities was 3.20% (women 3.32%; men 2.12%). Among them 23 cases were the balanced translocation; 14 cases were the Robertsonian translocation, 3 cases were the complex chromosomal rearrangement, and the other 17 cases were the other abnormalities. In women with 1, 2, 3 or more spontaneous abortion, the incidence of chromosomal abnormalities was 1.7%, 2.3%, and 5.8%, respectively.
CONCLUSION
Translocations are the major abnormal karyotpes associated with spontaneous abortions. The chance of finding chromosomal aberration increases with the number of abortions. Chromosomal abnormalities are more common in women with 3 or more spontaneous abortions.

5.Influence of ubenimex on immunocyte functions of mice

Objective To study the effects of ubenimex on immunocyte functions of mice.Methods Antitumor activity of ubenimex was observed in implanted H22 mice.The killing activity of NK cells to target cell YAC-1, phagocytosis of peritoneal macrophage and the secretion of mac -rophage NO effect were carried out with CCK method , neutral red colori-metric method and Griess method , respectively.Lymphocyte proliferation test was done by stimulating methods in vitro and in vivo.Tumor associated macrophages(TAMs)and myeloid derived suppressor cells (MDSCs) in pe-ripheral blood and T lymphocytes , B lymphocytes and NK cell in spleen were determined by flow cytometry method.Results The 20 mg· kg-1 of ubenimex could depress the growth of H 22 and its tumor inhibitory rate was 41.71%.The killing activity of NK cells was increased by 56.5%.The 10μg· mL-1 of ubenimex significantly enhanced secretion of macrophage NO effect (P<0.01) and it also enhanced phagocytosis of peritoneal mac-rophage (P<0.05) at concentration of 100μg· mL-1.Besides the lym-phocyte proliferation was also strengthened (P<0.05).The number of T lymphocytes and CD 8+T cell in spleen was increased obviously ( P<0.01 or P<0.05).Ubenimex could adjust the number of TAMs and MDSCs in peripheral blood to normal level.Conclusion Ubenimex has antitumor growth activity through enhances non -specific immunity and specific immunity and suppresses the tumor associated cell.
6.The value of mental intervention in 64-line spiral CT for coronary angiography

Objective To evaluate the value of mental intervention in 64-slice spiral CT for coronary angiography. Methods 60 patients who suspected coronary artery stenosis were divided into two groups at random (the mental intervention group and the routine group, with 30 cases in each group). The quality of coronary imaging was assessed as excellent, good and bad according to ACC1999 by two experienced doctors. The results of the two groups were calculated by statistical analysis. Results The image quality of the mental intervention group includes: excellent 72 cases, good 15 cases, and bad 3 cases; and the image quality of the routine group includes: excellent 48 cases, good 12 cases, and bad 30 cases, respectively. The quality of imaging in the mental intervention group was obviously superior to the routine group, and there was significant difference between the two groups (P<0.05). Conclusions In 64-slice spiral CT for coronary angiography, mental intervention can raise the achievement ratio of examination and improve the image quality of coronary artery. It is the essential for the success of the 64-slice spiral CT coronary angiography.
7.Relationship between the oral language development and restoration of velopharyngeal closure function in Chinese toddlers with repaired cleft palate

Objective To evaluate how the oral language development enhances articulators to function well in cleft palate movement and velopharyngeal closure in the young Chinese children with repaired cleft palate. MethodsThe recordings of 78 cases of Chinese toddlers with repaired cleft palate were reviewed. This group of children aged from 27 months to 33 months ( average: 30 months). Transcription using Pinyin system was made. Mean utterance count (MUC) and mean special consonant correct count (MSCC) were calculated. Correlation between MUC and MSCC was statistically analyzed. ResultsThe correlation coefficient of MUC and MSCC is 0. 360 ( P < 0. 01 ). ConclusionsThere is positive correlation between the oral language development and restoration of velopharyngeal closure function. Children with repaired cleft palate should be encouraged to start oral language as early as possible and as much as possible in order to get the restoration of velopharyngeal closure function.
8.Nursing care of diabetic mellitus patients treated by the real-time dynamic insulin pump system

Objective To investigate the nursing care for the diabetic patients receiving real-time dynamic insulin pump system treating.Methods In addition to psychological care and diet instruction,the nurses actively communicated with the diabetic patients,introduced the principle and advantages of the real-time dynamic insulin pump system to promote the diabetic patients to cooperate with the medical staff.Results The blood glucose control of the diabetic patients treated with the real-time dynamic insulin pump system was well after 2.7 days.Among 7 cases,there were twice cal error alarm,4 times lost sensor,3 times weak signal,twice sensor error,1 time high blood glucose alarm,1 time low blood glucose alarm.Conclusions Development and implementation of system nursing care can achieve a satisfactory treatment effect.
9.Changes in the Non-targeted Metabolomic Profile of Three-year-old Toddlers with Elevated Exposure to Polycyclic Aromatic Hydrocarbons

Objective To investigate changes in the urinary metabolite profiles of children exposed to polycyclic aromatic hydrocarbons(PAHs)during critical brain development and explore their potential link with the intestinal microbiota. Methods Liquid chromatography-tandem mass spectrometry was used to determine ten hydroxyl metabolites of PAHs(OH-PAHs)in 36-month-old children.Subsequently,37 children were categorized into low-and high-exposure groups based on the sum of the ten OH-PAHs.Ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry was used to identify non-targeted metabolites in the urine samples.Furthermore,fecal flora abundance was assessed by 16S rRNA gene sequencing using Illumina MiSeq. Results The concentrations of 21 metabolites were significantly higher in the high exposure group than in the low exposure group(variable importance for projection>1,P<0.05).Most of these metabolites were positively correlated with the hydroxyl metabolites of naphthalene,fluorine,and phenanthrene(r=0.336-0.531).The identified differential metabolites primarily belonged to pathways associated with inflammation or proinflammatory states,including amino acid,lipid,and nucleotide metabolism.Additionally,these distinct metabolites were significantly associated with specific intestinal flora abundances(r=0.34-0.55),which were mainly involved in neurodevelopment. Conclusion Higher PAH exposure in young children affected metabolic homeostasis,particularly that of certain gut microbiota-derived metabolites.Further investigation is needed to explore the potential influence of PAHs on the gut microbiota and their possible association with neurodevelopmental outcomes.
10.Effect of baicalin on HSP70 expression of hippocampal neurons in focal brain ischemia-reperfusion injury rats.

AIMTo investigate the effect of baicalin on the hippocampal neuronal apoptosis and the expression of HSP70 in rats with focal brain ischemia-reperfusion injury.
METHODSOne hundred and twenty male Wistar rats were randomly divided into six groups:sham operated group, ischemia-reperfusion group, nimodipine group and three baicalin groups,to which baicalin was administered at doses of 50, 100 and 200 mg x kg(-1), separately. The models of focal brain ischemia-reperfusion injury induced by middle cerebral artery occlusion (MCAO) were used in this study. HE stain was used to observe the pathological changes. Flow cytometry (FCM) was used for determination of neuronal apoptosis. HSP70 protein expression of the neurons was detected with immunohistochemistry. Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of the mRNA level of HSP70.
RESULTSBaicalin can significantly relieve the pathological changes and inhibit apoptosis in hippocampus CA1 area, and at the same time increase the expression of HSP70 and HSP70 mRNA.
CONCLUSIONBaicalin can relieve brain damage induced by focal brain ischemia-reperfusion in rats, which may be related to inhibiting the process of the neuronal apoptosis. The mechanism of antiapoptosis effect of baicalin may be related to the promotion of transcription of HSP70 mRNA and increasing the expression of the protein.
