3.Neuroprotective effect screening and the mechanism of 10 kinds of coumarin derivatives.

The study reports the detection of neuroprotective effect of 10 kinds of coumarin derivatives and explores their possible mechanism. MTT method was used to screen the neuroprotective effect of 10 coumarin derivatives on neurotoxic agents (Aβ25-35 and rotenone) or OGD (oxygen-glucose deprivation). A compound with better protective effect was obtained. Then the effect of this compound on neurotoxic agents on PC12 was detected by the morphological observation. Furthermore, the effect of compound 3 on microglia with lipopolysaccharide (LPS) induced inflammation was detected. And the inflammatory factor was tested. Finally, direct free radical scavenging ability was detected. Compound 3 was found to be the best compound through three neurons toxic models. Not only compound 3 ameliorated cell viability reduced by three neurons toxic models, but also significantly inhibited the production of inflammatory factor (TNF-α and IL-1β). And its free radical scavenging ability is very good, especially the effect on superoxide anion, which is comparable with vitamin C. The significant scavenging effect of compound 3 on superoxide anion might be the mechanism of the neuroprotection. Compound 3 as a potential neural cell protective agent merits further investigation.

4.Influencing Factors of UncertaintyinIllness and Implementation of Humanistic Care Strategies in Patients with Systemic Lupus Erythematosus

Objective:To investigate the uncertainty in illness of patients with systemic lupus erythematosus (SLE) and to explore the influencing factors and humanistic care strategies.Methods:The general situation questionnaire and the uncertainty in illness scale were used to investigate the influencing factors of uncertainty in illness and positive humanistic care strategies in patients with SLE.Results:Patients with SLE were at a high level of uncertainty in illness,of which the unpredictability and uncertainty scored higher but lack of information and complexity were of lower scores.Educational status,family income,and self-perceived severity of disease were the main influencing factors of uncertainty in illness (P < 0.05).Conclusion:Patients with SLE were at a high level of uncertainty in illness,mainly from the unpredictable progress of disease and prognosis and unclear symptoms of the disease.Educational status,family income,and self-perceived severity of disease were the main influencing factors of uncertainty in illness.The clinical staff should pay attention to uncertainty in illness among patients with SLE and adopt positive humanistic care measures so as to reduce the sense of uncertainty in illness.
5.Transformation of Kentucky Bluegrasss (Poa pratensis L.) by Particle Bombardment

BADH-CMO double gene,CMO gene and DREB1A gene were transformed respectively to embryonic callus of Kentucky bluegrass by particle bombardment. Then the embryonic callas of kentucky bluegrass were put in meclium for subculture which is mixed with 100mg/L hygromycin and the meclium for plantlet regeneration which mixell with 50mg/L hy gromycin about one month. Thus,the hygromycin-selectecl plants were obtained and were transplantecl into tlowerpots. The results of the PCR and Southern blot analysis indicated that the DREB1A gene,CMO gene and BADH-CMO double-gene were integrated into the genomic DNA of Kentucky bluegrass.
6.The Preparation of the Recombinant Fusion Protein Human Retinal Pigment Epithelium-derived Factor and the Analysis of Its Angiogenesis Activity

Objective:The recombinant human retinal pigment epithelium-derived factor(PEDF)protein to be obtained and the angiogenesis of the rPEDF to be identified.Methods: PEDF gene gene was amplified by PCR and cloned into pET32a,rPEDF protein was expressed in E.coli BL21 and confirmed by SDS-PAGE and Western blot.The rPEDF was purified by Ni-NTA on denature condition.The concentration of the rPEDF was determined by Bradford method.The angiogenesis of the rPEDF was determined by chick chorioallantoic membrane(CAM) method.Results: The expression plasmid pET32a-PEDF was constructed successfully.The rPEDF was expressed with stable efficiency in E.coli BL21.The results of the CAM experiment showed that the rPEDF had notable angiogenesis effect in the concentration 0.4、0.04 ng/ml,but had no effect in 4 ng/ml.Conclusion:The PEDF gene was cloned and expressed efficiently,the angiogenesis of the rPEDF to be identified and the activity was worked in certain range.The results can facilitate studying its function and spreading its application.
7.Research progress of febuxostat derivatives as xanthine oxidase inhibitors

Febuxostat, as a xanthine oxidase inhibitor, is a classic anti-gout drug with significant therapeutic effects and good tolerability. The structures of febuxostat and its derivatives can be divided into two parts: a substituted phenyl ring and a five-membered or six-membered heterocyclic ring with a carboxyl substitution. This paper reviewed the research progress of febuxostat derivatives in recent ten years and classified the structure-activity relationships of various febuxostat derivatives. Exploring the action mechanisms and structure-activity relationships of xanthine oxidase inhibitors might be significant for the rational design and development of new anti-gout chemical entities.
8.Analasis on critical thinking ability of undergraduate nursing students

Objective To study the characteristic of ablities of critical thinking among undergraduate nursing students and provide evidence for making critical thinking training plan. Methods Critical Thinking Disposition Inventory-Chinese Version(CTDI-CV) ,established by Hongkong Polytechnic University, was used to investigate 210 undergraduate nursing students of Third Military Medical University. Results The average score of CTDI-CV was (273. 24±1. 86). Among scores of every disposition of critical thinking, the highest was in Inquisitiveness and the lowest in Truth Seeking. With the increasing of grades, students' critical thinking ability showed a declining tendency, which was statistical significance (P <0.01). Conclusions The critical thinking ability of undergraduate nursing students is generally low. It suggested that critical thinking ability should be consciously cultivated from aspects of curriculum provision, construction of curriculums,and teachiong methods, etc.
9.Effect of alleviating Venous Cannulation Pain by 1% Lidocaine Local Injection

Objective To observe the effect of 1% lidocaine local injection on alleviating 18G venous cannulation pain.Methods 100 patients were randomly divided into two groups(each had 50 cases),local anesthesia group used 1% lidocaine 0.2 ml local injections firstly,then performed 18 G vein line needle puncture.The control group performed 18G vein line needle puncture directly.The pain was assessed by visual analogue scale method(VAS).Results 25 patients felt no pain,24 felt mild pain,1 felt moderate pain and no patient felt severe pain in local anesthesia group,and the number was 0,8,31 and 11 respectively in sequence in control group,there was significantly difference(Z =-8.136,P <0.01).Conclusions 1% lidocaine local injection can alleviate the pain of 18G vein line needle cannulation.
10.Genetically modified industrial brewing yeast with high-glutathione and low-diacetyl production.

Recombinant plasmid pICG was constructed by replacing the internal fragment of a-acetohydroxyacid synthase (AHAS) gene (ILV2) with a copy of gamma-glutamylcysteine synthetase gene (GSH1) and copper chelatin gene (CUP1) from the industrial brewing yeast strain YSF31. YSF31 was transformed with plasmid pICG linearized by Kpn I and Pst I. A recombinant strain with high-glutathione and low-diacetyl production was selected. The results of fermentation in 100-L bioreactor showed that the lagering time of beer produced for recombinant strain T2 was shortened by 3 days and the shelf life of the beer was prolonged about 50%. It may be more acceptable for the commercial application, as it does not contain foreign DNA.
