2.Neuroprotective effect screening and the mechanism of 10 kinds of coumarin derivatives.

The study reports the detection of neuroprotective effect of 10 kinds of coumarin derivatives and explores their possible mechanism. MTT method was used to screen the neuroprotective effect of 10 coumarin derivatives on neurotoxic agents (Aβ25-35 and rotenone) or OGD (oxygen-glucose deprivation). A compound with better protective effect was obtained. Then the effect of this compound on neurotoxic agents on PC12 was detected by the morphological observation. Furthermore, the effect of compound 3 on microglia with lipopolysaccharide (LPS) induced inflammation was detected. And the inflammatory factor was tested. Finally, direct free radical scavenging ability was detected. Compound 3 was found to be the best compound through three neurons toxic models. Not only compound 3 ameliorated cell viability reduced by three neurons toxic models, but also significantly inhibited the production of inflammatory factor (TNF-α and IL-1β). And its free radical scavenging ability is very good, especially the effect on superoxide anion, which is comparable with vitamin C. The significant scavenging effect of compound 3 on superoxide anion might be the mechanism of the neuroprotection. Compound 3 as a potential neural cell protective agent merits further investigation.

3.Influencing Factors of UncertaintyinIllness and Implementation of Humanistic Care Strategies in Patients with Systemic Lupus Erythematosus

Objective:To investigate the uncertainty in illness of patients with systemic lupus erythematosus (SLE) and to explore the influencing factors and humanistic care strategies.Methods:The general situation questionnaire and the uncertainty in illness scale were used to investigate the influencing factors of uncertainty in illness and positive humanistic care strategies in patients with SLE.Results:Patients with SLE were at a high level of uncertainty in illness,of which the unpredictability and uncertainty scored higher but lack of information and complexity were of lower scores.Educational status,family income,and self-perceived severity of disease were the main influencing factors of uncertainty in illness (P < 0.05).Conclusion:Patients with SLE were at a high level of uncertainty in illness,mainly from the unpredictable progress of disease and prognosis and unclear symptoms of the disease.Educational status,family income,and self-perceived severity of disease were the main influencing factors of uncertainty in illness.The clinical staff should pay attention to uncertainty in illness among patients with SLE and adopt positive humanistic care measures so as to reduce the sense of uncertainty in illness.
4.Study on interventional methods and the pattern of maternal-fetal transmission of syphilis during pregnancy

Objective To explore the maternal-fetal transmission patterns and interventional methods of syphilis during pregnancy.Methods A total of 847 cases of syphilis in pregnancy confirmed by rapid plasma reagin test(RPR)and treponema pallidum hemoagglutination test(TPHA)were treated with procaine benzylpenieillin intramuscular injection,and with erythrocin oral medication if hypersensitive to benzylpenicillin.Eight hundred forty seven cases of syphilis during pregnancy were followed up for pregnancy outcomes.And their newborn babies were tested using the RPR.The newborns with positive results were given intervention and followed up until 24 months after birth.Results(1)A total of 733 cases among the total 847 have given birth to living-babies,in which 626 cases were tested using RPR,and the positive rate was 55.1%(345/626).(2)The RPR positive rate,neonatal mortality,preterm birth rate and low birth rate in the newborn of mothers with an RPR titer higher than or at 1:8 were higher than those of mothers with an RPR titer lower than 1:8(P
5.Resting-state Network of Brain in Leukoaraiosis Patients: A Magnetic Resonance Imaging Study

Objective To explore the diversity of resting-state network of brain between the patients with leukoaraiosis and the healthy people. Methods 31 patients with leukoaraiosis (patients) and 27 healthy persons (controls) were checked with resting-state functional magnetic resonance imaging (rs-fMRI), and analyzed with the independent component analysis (ICA) to explore the resting-state functional brain network. Results The resting-state brain network was found in both the patients and the controls, which was coincident with the previous studies. The active areas were the same in both groups, and the activation was weaken in the patients than in the controls, especially in quadrate gyri, posterior cingulate cortex, superior temporal gyrus, superior parietal gyrus, anterior central gyrus, post central gyrus, insula and prefrontal cortex. Conclusion There is a significant diversity of resting-state network of brain between the patients with leukoaraiosis and healthy people in the activation of active areas.
6.Effect of VEGFR1 gene silencing by shRNA on proliferation, migration and apoptosis of U937 line.

OBJECTIVETo investigate the effects of lentivirus-mediated RNA interference targeting vascular endothelial growth factor receptor 1 (VEGFR1) gene on the proliferation, migration and apoptosis of leukemic cell line U937.
METHODSShort hairpin RNAs (shRNA) targeting VEGFR-1 was synthesized and cloned into pRNAT-U6.2 lentiviral vector. The expression vectors were transfected into 293T cell line to produce packaged lentivirus. After infected with the packaged lentivirus, the expression of VEGFR-1 gene of U937 cells at mRNA and protein level was detected by real-time PCR and Western blot. VEGF production by the cells was determined by ELISA. Cell proliferation and survival under regular culture and in the presence of cytarabine (Ara-C) was determined by CCK-8 assay. Migration assays were performed by 5 µm pore transwell inserts.
RESULTSThe lentiviral shRNA vector targeting VEGFR-1 was successfully constructed and transfected into U937 cells. The shRNA vector effectively inhibited the expression of VEGFR-1 gene in U937 cell line at mRNA and protein levels. As compared to that of the control, the proliferation rate of U937-shVEGFR-1 cells reduced; The VEGF production and migrated cell number of U937-shVEGFR-1 cells decreased dramatically. After treated with Ara-C, the inhibition rate and apoptotic rate of U937-shVEGFR-1 cells increased significantly. The number of migrated cells in the KD group under regular culture and in the presence of VEGF was markedly lower than that in the NC group and CON group. Bevacizumab could decrease the number of migrated cells in the NC group and CON group, but could not in the KD group.
CONCLUSIONSLentivirus-mediated RNA interference targeting VEGFR1 gene reduces the proliferation, migration of U937 cell line and enhances its sensitivity to Ara-C.

7.CCL3 promotes hBMSC proliferation and inhibits exosome secretion

AIM:To explore the regulatory effect of chemokine CCL 3 on exosome secretion from human bone marrow mesenchymal stem cells(hBMSCs).METHODS: hBMSCs were stimulated with chemokine CCL 3 at different concentrations in vitro.The proliferation of hBMSCs was measured by CCK-8 assay and viable cell counting.Exosome se-cretion from hBMSCs was qualitatively analyzed by transmission electron microscope(TEM)and flow cytometry, and the quantitative analysis was carried out by flow cytometry and nanoparticle tracking analysis(NTA).RESULTS:Compared with control group,the viability of the hBMSCs detected by CCK-8 assay was increased when hBMSCs were treated with CCL3(P<0.05).The results of viable cell counting demonstrated that the number of hBMSCs was raised in CCL 3 group in a dose-dependent manner(P<0.05).The results of flow cytometry showed that hBMSCs expressed 3 CCL3-related spe-cific receptors,CCR1,CCR5 and CCR9.Compared with control group,the fluorescence intensity of CCR9 in CCL3 group was obviously enhanced.However,no significant difference of fluorescence intensity for CCR 5 and CCR1 was observed be-tween the 2 groups.The results of NTA demonstrated that the secretion capacity of CCL 3-induced hBMSCs was far less than that in control group(P<0.05).However, the microvesicles larger than 100 nm in CCL3 groups were increased(P<0.05).The above results indicated that the higher concentration of CCL 3 induced the lower secretion of exosomes.In addi-tion,the results of flow cytometry demonstrated that CCL 3-induced hBMSCs showed lower quantity of CD 9 +exosomes than those in control group(P<0.01).CONCLUSION:CCL3 promotes the proliferation of hBMSCs but depresses the secre-tion of exosomes in a dose-dependent manner.CCL3 affects the size distribution of exosomes and increases the number of nonfunctional microvesicles of larger than 100 nm in size.CCL3 induces the expression of CCR9 in hBMSCs.
8.Clinical features and follow-up results of boys and girls with systemic lupus erythematosus: a comparative analysis.

OBJECTIVE:
To compare the clinical features and follow-up results of systemic lupus erythematosus (SLE) between boys and girls.
METHODS:
A retrospective analysis was performed for the clinical data of 79 children (18 boys and 61 girls), aged ≤14 years, who were diagnosed with SLE from 2008 to 2018. The boys and the girls were compared in terms of initial and major clinical symptoms, injury of organs/systems, related laboratory markers, and follow-up results.
RESULTS:
As for the initial and non-initial symptoms, fever had the highest incidence rate in the boys, while facial erythema had the highest incidence rate in the girls. The boys tended to develop renal injury and hematological damage (P<0.05), with a significantly higher incidence rate of proteinuria than the girls (P<0.05), while the girls tended to develop joint pain (P<0.05). There were high abnormal rates (>80%) of anti-nuclear antibody, dsDNA, complement C3, and erythrocyte sedimentation rate in both boys and girls (P>0.05). The boys had a significantly higher disease activity than the girls at the first visit and in year 9 of follow-up (P<0.05). A one-month to ten-year follow-up showed that among the boys, 3 were lost to follow-up, 1 died, 7 were well controlled but required oral administration of large doses of hormones or immunosuppression, 2 progressed to chronic renal failure, and 1 developed lupus encephalopathy. Among the girls, 3 were lost to follow-up; 5 died; 34 were well controlled, among whom 5 were maintained on oral prednisone acetate with a dose of <10 mg, 1 was withdrawn from the drug for 1 year, and 2 were withdrawn from the drug for 2 years; 4 developed lupus encephalopathy; 1 developed depression and anxiety and had suicidal tendency in the 7th year after disease onset; 2 experienced impaired vision, blurred vision, and chloropsia; 1 developed a vascular necrosis of both femoral heads in the 3rd year of hormone administration.
CONCLUSIONS
There are differences in clinical features, several laboratory markers, and prognosis between boys and girls with SLE. Boys tend to have a high severity at disease onset, develop renal injury and hematological damage, and have poor long-term prognosis, while girls tend to have joint involvement.
9.A study on quality of life and its influencing factors of drug addicts with methadone maintenance treatment in Wenzhou

Objective To analyze the quality of life status and its influencing factors of drug addicts with methadone maintenance treatment (MMT) in Wenzhou, in order to provide a basis for developing health service measures to improve their quality of life. Methods A total of 199 drug addicts with MMT in Wenzhou were investigated by the Quality of Life for Drug Addicts (QOL-DA) and Self-made questionnaire, influencing factors on quality of life were analyzed by multiple linear regression analysis. Results Most of 199 drug addicts with MMT were male, middle aged, local residents, lower educational level, and unemployed. Most of them used traditional drugs such as heroin by intravenous injection, and the proportion of who continually used drugs more than 10 years was 78.39%. The majority of them had abandoned drug habits many times. The proportions of abandoning drug habits more than 12 months this time and self-reported involving drug abuse related diseases were 77.39%, 61.81%, respectively. Scores of physical function and hunger sensory (PH), psychological spirit and self-esteem (PS), withdrawal syndrome and toxic effects (ST), social support and operational capability (SO) were 32.96±6.75, 33.03±5.96, 47.61±8.51 and 38.42±6.86, respectively. The total score of QOL-DA was 152.01±23.55. Marital status, occupation, age of first using drug, time of drug use and drug abuse related diseases were its influencing factors. Conclusion The quality of life of drug addicts with MMT is lower. Comprehensive health service measures for influencing factors should be taken to improve their quality of life.
10.Efficacy of YZH amblyopic therapeutic apparatus and nursing in treatment of children with amblyopia

Objective To investigate efficacy and safety of YZH amblyopic therapeutic apparatus in treatment of children with amblyopia.Methods Forty patients were randomly divided into two groups.The control group received routine treatment and the intervention group was treated with YZH amblyopic therapeutic apparatus based on routine treatment.Results The effective rate was 66.67% and ineffective rate was 33.33% in the control group ( n =20,33 eyes).The effective rate was 97.37% and ineffective rate was 2.63% in the intervention group(20 cases,38 eyes).There was significant difference between the two groups( x2 =11.853 6,P < 0.05).Conclusions It is effective and safe to improve children' s vision with YZH amblyopic therapeutic apparatus.During the treatment process,proper care,active cooperation and participation of parents can improve the treatment and shorten the course of disease.
