The prostate-specific antigen (PSA) test contributes a lot to the diagnosis and treatment of prostate cancer (PCa) and, along with imaging-guided prostate biopsy, has improved the diagnosis rate of lower-risk PCa and the accuracy of its clinical staging. However, many questions and controversies remain as to the choice of optimal biopsy strategies. Scholars differ in views about how to utilize PCa-related biomarkers to optimize the detection of initial and repeat biopsies. This review focuses on the present status of and advances in transrectal ultrasound-guided biopsy for PCa.

5. Investigation of Mechanism of Premature Ovarian Failure Regulation by Kidney-tonifying Herbs and Liver-clearing Herbs in Dingjing Decoction
Chinese Herbal Medicines 2015;7(4):328-333
Objective: To investigate the mechanisms through which kidney-tonifying herbs (KTHs) and liver-clearing herbs (LCHs) in Dingjing Decoction (DJD) regulate premature ovarian failure (POF). Methods: One hundred and fifty Sprague-Dawley rats were randomly divided into five groups such as control, model, KTHs, LCHs, and DJD groups. POF-related biological molecules were examined. Factor analysis was performed to investigate the regulatory networks and key biomolecules involved in mediating POF after treatment with KTHs and LCHs. Results: The master regulatory factors in the reproductive endocrine network associated with KTHs intervention included four molecules in the pituitary-ovarian axis, cortisol (CORT) in the target gland of pituitary-adrenal axis, and some molecules in the hypothalamus. In contrast, the master regulatory factors associated with LCHs intervention included four molecules in the pituitary-ovarian axis and some molecules in the hypothalamus; No biomolecules in the pituitary-adrenal axis were involved in the LCH-mediated mechanisms. Gonadotropin-releasing hormone (GnRH), which was identified as a common biological molecule in the hypothalamus, was involved in regulating the reproductive endocrine network in association with KTHs intervention. Conclusion: KTHs directly regulates biological molecules in the pituitary-adrenal axis and indirectly regulates those in the pituitary-adrenal axis through the hypothalamus, while the LCHs only exert its effects indirectly. GnRH is the key biological molecule associated with KTHs intervention.

In order to affirm the cardioactive components in Fuzi, we identified a group of aminoalcohol- diterpenoid alkaloids in Fuzi using ultra high-performance liquid chromatography coupled with electrospray ionization mass spectrometer (UPLC-ESI-MS) method. Among a total of forty-one isolated ingredients, thirteen major aminoalcohol-diterpenoid alkaloids were identified by comparing their retention times and MS spectra with those of the reference substances. Moreover, Fuzi samples from different places of origin and with different processing methods were examined and their components displayed a pattern of high similarity, though the relative abundance varies probably due to their different processing methods. Furthermore, the cardiac effect of each identified alkaloid was individually evaluated using the isolated bullfrog heart perfusion experiment. Among the thirteen aminoalcohol diterpenoid alkaloids tested, six of them significantly enhanced the amplitude rates. Taken together, we affirm that the cardioactive components in Fuzi are aminoalcohol-diterpenoid alkaloids, shedding light on future studies of the mechanisms and development of these cardioactive compounds.

Objective:To observe the effect of electroacupuncture (EA) at Jiaji (EX-B 2) points plus hydro-acupuncture with sinomenine hydrochloride for low back pain caused by compression fractures in the elderly.Methods:Ninety-five elderly in-patients with low back pain caused by compression fractures were randomly divided into an observation group and an EA group according to the visit sequence. Both groups received the same basic treatment. In the EA group,48 cases were treated with EA at Jiaji (EX-B 2) points plus the basic therapy;47 cases in the observation group received the basic treatment plus EA and hydro-acupuncture with sinomenine hydroch|oride at Jiaji (EX-B 2) points. The levels of osteoprotegerin (OPG) and interleukin-1β (IL-1β) in peripheral blood were measured by enzyme-linked immunosorbent assay (ELISA) before and at the 21st day of treatment in both groups.Oswestry disability index (ODI) and visual analog scale (VAS) scores were used to analyze the clinical efficacy. Results:After treatment, the OPG content in the observation group was higher with statistical significance compared with that before treatment in the observation group and after the treatment in the EA group, respectively (both P<0.05);the content of IL-1β,ODI and VAS scores were lower than those before treatment in the observation group and after treatment in the EA group with statistical significances (all P<0.05).Conclusion:The combination of EA and hydro-acupuncture with sinomenine hydrochloride at Jiaji (EX-B 2) points is effective for low back pain caused by compression fractures in the elderly, and is superior to EA at Jiaji (EX-B 2) points alone.

Objective To investigate the conception implicit memory (CIM) and perception implicit memory (PIM) impairment in patients with frontal and occipital lobe stroke patients.Method Patients with frontal lobe stroke (n =23) and occipital lobe stroke (n =21) and healthy controls (n =26) were administered with a neuropsychological battery of tests including conception and perception implicit memory (CIM and PIM) tasks,as well as explicit memory tasks including immediately recall, delay recall, delay recognition. Results Compared with healthy controls, patients with frontal lobe stroke performed poor CIM test (1.96 ± 1.00 and 3.52 ±0.52,t =6.987,P <0.01),as well as its performance in explicit memory tasks including immediate recall(3.91 ± 1.53 and 5.42 ± 1.06),delay recall (6.04 ± 3.05 and 8.19 ±1.60),delay recognition (22.61 ± 4.71 and 25.38 ± 3.24 ; t =2.428,3.990,3.138 ; all P < 0.05).PIM was impaired in the patients with occipital lobe stroke (5.56 ± 8.19 and 22.12 ± 4.68,t =0.011,P <0.01),while there was no significant difference between occipital lobe stroke and healthy group in CIM task. Conclusion Frontal lobe stroke present CIM damage and PIM relative retention, while occipital lobe stroke patients perform PIM damage and CIM relative retention, confirm the dual separation in implicit memory neural mechanism.

Aim To investigate the cardiotoxicity of propofol and thiopental. Methods 4day-old contracting neonatal primary myocardial cells obtained from 2-to 3-day-oldWistar rats were divided into 5 groups, with normal contrast group, and the cellcultures in groups PL, PH, TL and TH, were treated with propofol(3 ? 10-5 and3 ? 10-4 mol? L) and thiopental (1 ? 10-5 and 1 ? 10-4 mol?L) for 8 h.The con-tractility and morphology of the cells were observed and the cytoplasmic enzyme(LDH, AST, CK and ALP) release content of myocardial cell and the concentrationof electrolytes (K +, Na +, Cl - and Ca2+ ) in the medium were measured 8 h afterintravenous anesthetics administration. Results In groupPH and TL decreasedsignificantly (P
