Objective To detect levels of serum platelet activating factor(PAF),thrombomodulin(TM) and white blood cell(WBC),platelet count(PLT) in neonates with meconium aspiration syndrome(MAS),and observe changes of mediators of inflammation and function of endotheliocute.Methods All cases were taken vein blood in 24 h and 72-96 h after birth.Surm PAF and TM were detected by EILSA technique,at the same time,blood cell counts were determined.Results PAF and WBC in neonates with MAS increased,which were relevant to the patients′ condition.TM of neonates with MAS increased significantly,especially in 72-96 h after birth and(aggrava)-ted with the patients′ condition.Conclusion Neonates with MAS have inflammatory reaction and injured endotheliocyte,which are(inte)-raction.
2.Clinical observation on electroacupuncture plus hydro-acupuncture for low back pain caused by compression fractures

Objective:To observe the effect of electroacupuncture (EA) at Jiaji (EX-B 2) points plus hydro-acupuncture with sinomenine hydrochloride for low back pain caused by compression fractures in the elderly.Methods:Ninety-five elderly in-patients with low back pain caused by compression fractures were randomly divided into an observation group and an EA group according to the visit sequence.Both groups received the same basic treatment.In the EA group,48 cases were treated with EA at Jiaji (EX-B 2) points plus the basic therapy;47 cases in the observation group received the basic treatment plus EA and hydro-acupuncture with sinomenine hydroch|oride at Jiaji (EX-B 2) points.The levels of osteoprotegerin (OPG) and interleukin-1β (IL-1β) in peripheral blood were measured by enzyme-linked immunosorbent assay (ELISA) before and at the 21st day of treatment in both groups.Oswestry disability index (ODI) and visual analog scale (VAS) scores were used to analyze the clinical efficacy.Results:After treatment,the OPG content in the observation group was higher with statistical significance compared with that before treatment in the observation group and after the treatment in the EA group,respectively (both P<0.05);the content of IL-1β,ODI and VAS scores were lower than those before treatment in the observation group and after treatment in the EA group with statistical significances (all P<0.05).Conclusion:The combination of EA and hydro-acupuncture with sinomenine hydrochloride at Jiaji (EX-B 2) points is effective for low back pain caused by compression fractures in the elderly,and is superior to EA at Jiaji (EX-B 2) points alone.

4.Quantitative study by grey system on the latent period of lung cancer induced by air pollutants.

OBJECTIVETo calculate the latent period of lung cancer induced by air pollution.
METHODSThe degree of grey incidence (DGI) between the concentrations of TSP or SO(2) and the mortality of lung cancer were assessed through a grey system model. According to the maximum values of DGI, the total latent period of lung cancer was calculated. Data was collected in H city.
RESULTSThe maximum DGI value of TSP was 0.886 while the relationship between the comparison sequence from 1985 to 1989 and the reference sequence from 1994 to 1998 was greatly correlated. The maximum DGI value of SO(2) was 0.919 while the relationship between the comparison sequence from 1986 to 1990 and the reference sequence from 1994 to 1998 was most correlated.
CONCLUSIONSThe latent periods of lung cancer induced by TSP and SO(2) were 7 and 8 years respectively in H city. The model of grey system was less affected by the confounding factors, and the calculation process was relatively simple. A feasible and useful new way was provided by the model of grey system for quantitative analysis of the latent period of lung cancer induced by air pollutants.

5.Impacts of acupuncture at twelve meridians acupoints on brain waves of patients with general anxiety disorder.

OBJECTIVETo observe the clinical efficacy of acupuncture at twelve meridians acupoints on general anxiety disorder and explore its mechanism.
METHODSEighty patients were randomized into an acupuncture group and a clonazepam group, 40 cases in each one. In the acupuncture group, acupuncture at twelve meridians acupoints was applied, meaning quick needling at the specific acupoints of each meridian, such as Lieque (LU 7) of the Lung Meridian, Hegu (LI 4) of the Large Intestine Meridian and Shenmen (HT 7) of the Heart Meridian. After arrival of qi, the reinforcing or reducing technique was applied accordingly and the needles were not retained. In the clonazepam group, clonazepam was prescribed for oral administration. The course of treatment was 6 weeks. Before treatment and in 2, 4 and 6 weeks of treatment, respectively, Hamilton Anxiety Scale (HAMA) was evaluated in two groups and the changes in the basic electrical activity of brain waves before and after treatment were observed.
RESULTSHAMA score was reduced apparently after treatment as compared with that before treatment in two groups (all P < 0.01). The improvements of the total HAMA scores in 2, 4 and 6 weeks of treatment in the acupuncture group were superior obviously to those in the clonazepam group (all P < 0.05). After treatment, the activity of brain waves was improved remarkably, manifested as reducing of wave a frequency, increasing of wave alpha rhythm and reducing of wave theta (all P < 0.05). The efficacy was similar in comparison between the two groups (all P > 0.05).
CONCLUSIONAcupuncture at the twelve meridians acupoints achieves the superior and quick effect on general anxiety disorder as compared with clonazepam and the efficacy mechanism is related to the improvements of brain waves in the patients.

6.SHENG MAI ZHUSHEYE improves the viability and movement parameters of human sperm in vitro.

OBJECTIVETo observe the effect of SHENG MAI ZHUSHEYE on the movement parameters and viability of human sperm in vitro.
METHODSWe collected sperm samples from 33 normal fertile men, divided each into two, and cultured them in vitro with SHENG MAI ZHUSHEYE + Hams-F10 and Hams-F10 alone, respectively. Then we measured the straight line velocity (VSL), curvilinear velocity (VCL), average path velocity (VAP) and the amplitude of lateral head displacement (ALH) of the sperm by computer-aided semen analysis at 0.5, 1, 2, 4, 8 and 12 h. And the sperm viability was detected.
RESULTSVCL was significantly higher at 8 h (P < 0.05) and very significantly higher at 12 h (P < 0.01) in the SHENG MAI ZHUSHEYE + Hams-F10 group than in the Hams-F10 group. VSL, VAP and ALH were significantly increased in the former group at 4, 8 and 12 h as compared with the latter (P < 0.05). The sperm viability was significantly decreased in the Hams-F10 group at 12 h (P < 0.05).
CONCLUSIONSHENG MAI ZHUSHEYE can improve sperm movement parameters and increase sperm viability in vitro.

7.Intervention of pyrrolidine dithiocarbamate on expressions of connective tissue growth factor, type I collagen, and type III collage in acute paraquat poisoned rats.

OBJECTIVETo observe the changes in the expression of connective tissue growth factor (CTGF), type I collagen (Col I), and type III collagen (Col III) among the rats with acute paraquat (PQ) poisoning and the intervention effect of pyrrolidine dithiocarbamate (PDTC) on their expression, and to investigate the mechanism of PQ-induced pulmonary fibrosis and the intervention effect of PDTC on the disease.
METHODSSprague-Dawley rats were randomly divided into control group (n = 6), PQ group (n = 36), and PQ + PDTC group (n = 36). The PQ group and PQ + PDTC group were given a single dose of saline-diluted PQ (80 mg/kg) by gavage; 2 h later, the PQ + PDTC group was intraperitoneally injected with a single dose of PDTC (100 mg/kg), and the PQ group was intraperitoneally injected with the same amount of saline. The control group was given saline (1 ml/kg) by gavage and was intraperitoneally injected with the same amount of saline 2h later. At 1, 3, 7, 14, 25, and 56 days after operation, the protein expression of CTGF was evaluated by Western blot; the mRNA expression of CTGF, Col I, and Col III was analyzed by real-time quantitative PCR; the content of hydroxyproline in lung tissue was measured, and the pathological changes of lung tissue of the poisoned rats were observed.
RESULTSThe protein expression of CTGF in the PQ group increased as the time went on, slowly from the 3rd to the 14th day and rapidly from the 28th to the 56th day, significantly higher than that in the control group at each time point (P < 0.05 or P < 0.01). The mRNA expression of CTGF in the PQ group began to rise markedly on the 1st day, increased rapidly from the 3rd to the 14th day, and remained at a relatively high level from the 28th to the 56th day, significantly higher than that in the control group at each time point (P < 0.01). The mRNA expression of Col I in the PQ group changed little on the 1st and 3rd day, increased slightly on the 7th day, and increased greatly from the 14th to the 56th day, significantly higher than that in the control group from the 7th to the 56th day (P < 0.05 or P < 0.01). The mRNA expression of Col III in the PQ group began to rise on the 1st day, reached the peak level on the 7th day, and then declined, significantly higher than that in the control group at each time point (P < 0.05 or P < 0.01). Masson staining showed that fibroblasts proliferated from the 14th to the 28th day, and collagen fibers increased gradually. Compared with the PQ group, the PQ + PDTC group showed significantly decreased protein expression of CTGF as well as mRNA expression of CTGF, Col I, and Col III (P < 0.05 or P < 0.01).
CONCLUSIONIn PQ-induced pulmonary fibrosis, the expression of CTGF keeps rising, and the collagen secretion and matrix synthesis are increased probably by upregulating the transcriptional levels of Col I and Col III; CTGF plays an important role in PQ-induced pulmonary fibrosis. PDTC can inhibit the expression of CTGF, thus reducing the lung injury in rats with PQ poisoning.

8.Influence of luxS of Streptococcus mutans on biofilm of dental plaque.

OBJECTIVETo construct luxS mutant aften luxS gene of Streptococcus mutans (S. mutans) was knocked out, and examine their ability of biofilm formation.
METHODSA recombinant plasmid containing the flanking fragment of luxS of S. mutans was transformed into S. mutans UA159, and selected by brain heart infusion (BHI) agar medium with kanamicin. The luxS mutant further confirmed via polymerase chain reaction (PCR) and the autoinducer-2 (AI-2) bioluminescence assay of Vibrio harveyi (V. harveyi), and ability of luxS mutant and S. mutans UA159 biofilm formation was examined in different phases, in BHI medium with 1% sucrose and 1% glycose by scanning electron microscopy (SEM).
RESULTSLuxS-deficient S. mutans strains were successfully constructed. Compared with S. mutans UA159, the luxS mutant maintained in BHI medium containing 1% sucrose displayed an apparent defect in biofilm formation, while they showed no significant deviation in BHI medium containing 1% glycose.
CONCLUSIONluxS gene in S.mutans can play a role in dental plaque biofilm formation, and the luxS gene is possible to regulate sucrose-dependent biofilm formation.

10.Fibrinogen beta chain gene mutation contributes to one congenital afibrinogenemia.

OBJECTIVETo identify the fibrinogen (Fg) gene mutations in a Chinese pedigree of congenital afibrinogenemia.
METHODSThe plasma Fg activity and protein of the proband and his family members were detected. Genomic DNA was isolated from the peripheral blood mononuclear cells. All the exons and exon-intron boundaries of fibrinogen gene were amplified by PCR and sequenced thereafter.
RESULTSTwo mutations, 7972 del G in FGB and T2543A in FGG, were found in the proband.
CONCLUSIONSFGG2543 is a polymorphism site, which lead to the polymorphism of gamma144 I/K. The G deletion at base 7972 of FGB contributes to the frameshift mutation after amino acid 419, resulting in the truncated beta chain without the terminal 27 amino acids. The latter may contributes to the pathogenetic mechanisms in Chinese congenital afibrinogenemia patients. The G deletion at base 7972 of FGB is identified for the first time.
