1.Value of transabdominal and transvaginal sonography,sonohysterography and hysteroscopy in evaluating uterine cavity lesions

Objective To evaluate the value of transabdominal sonography(TAS),transvaginal sonography(TVS),sonohysterography(SHG),and hysteroscopy(HS) in the detection of endometrial pathology in the patients with uterine cavity lesions. Methods Uterine abnormality and disease were routinely examined by TAS, TVS, SHG.Results TAS was performed in 88 cases,TVS in 45 cases,SHG in 39 cases,and HS in 23 cases.Comparison was made between different means,showing SHG and HS with the highest sensitivity,accuracy and positive evaluation,respectively as 94.29%, 90.24%, 94.29%,and 95.65%, 91.67%, 95.65%.Conclusions Transvaginal sonohysterography is an easy, accurate,econimical,painless and no-injury technique for detecting endomatrial pathology and endoluminal masses.
2.Expression and significance of thioredoxin system in placenta of preeclampsia

Objective To assess the expression of thioredoxin reductase(TR)and thioredoxin (Trx)mRNA,TR activity in human placenta in women with preeclampsia,and to discuss the relationship between Trx system and the pathogenesis of preeclampsia.Methods Twenty-sixwomen with preeclampsia(10 mild and 16 severe)and 28 healthy gravidas (control) were included in this study.Semi-quantitative reverse transcription polymerase chain reaction(RT-PCR)were employed to demonstrate the expression of TR and Trx in placentas.TR activity was measured spectrophotometrically.Results The expression of TR mRNA(mild:0.865±0.018;severe:0.800±0.025,P<0.05)and Trx mRNA(mild:0.873±0.008;severe:0.818±0.025,P<0.05)were lower in placenta of preeclampsia than in the control group(TR mRNA:0.893±0.023;Trx mRNA:0.918±0.023.P<0.05),and those levels in the severe preeclampsia group were lower than in the mild ones.Both the mild and severe preeclampsia groups showed lower TR activity than the control(mild:17.732±1.653;severe:15.626±1.543 vs 19.770±2.432,P<0.05),and that of the severe preeclampsia group was lower than that of the mild ones.Positive correlation was found between the TR activity and TR mRNA expression in both groups(r=0.612,P<0.001).Conclusions Abnormal decreased expression of TR mRNA and Trx mRNA and TR activity in human placenta of preeclampsia might participate and play an important role in the pathogenesis of preeclampsia.
3.Efficacy of high-dose ambroxol in elderly patients with aspiration pneumonias

Objective To investigate the efficacy of high-dose ambroxol in elderly patients with aspiration pneumonias. Methods Totally 62 patients aged 65 years and over with aspiration pneumonia were enrolled into 2 groups:conventional-dose group intravenously given ambroxol 60 mg/d (n=30) and high-dose group received 270 mg/d (n=32).The times of remission and fever disappearance,rate of adverse effects,average duration of hospitalization,rates of mortality and recurrence within 2 months were observed. Results There were no significant differences in clinical charecteristics between the two groups.The times of remission and fever disappear ance and average duration of hospitalization were lower in high-dose group than in conventional-dose group [(3.2±0.6)d,(54.2±19.5)h,(12.7±4.1) d vs.(3.8±1.1)d,(66.5±18.4)h,(13.5±3.1)d,t=2.11,2.36,2.04,all P<0.05].No differences were found in rates of mortality and recurrence within 2 months between the two groups (6.3%,3.1% vs.10.0%,13.3%,x2=0.01,0.87,both P>0.05).Adverse effects did not appear in the two dose groups. Conclusions High-dose ambroxol is efficient and safe for aspiration pneumonia in elderly patients.

8. Effects of NaCl stress on physiological characteristics of Thermopsis laceolata callus

Objective: To investgate the effects of NaCl stress on the growth and physiological characteristics of Thermopsis laceolata callus, and to reveal the physiological mechanism of T. laceolata under salty environment at the cellular level. Methods: The callus was cultured on the subculture medium with NaCl (0.2%-1.2%), and the contents of soluble sugar, soluble proteins, proline, and malondialdehyde (MDA) and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were determined and analyzed. Results: The callus growth expressed a decreasing trend with NaCl concentration increasing (≤ 0.8%) but had no significant difference to contrast. Nevertheless, the callus grew very slowly with NaCl concentration increasing (> 0.8%); the contents of soluble sugar and free proline increased firstly and then decreased, but were higher than those of the control group; the content of soluble protein and the activities of SOD, CAT, and POD increased firstly and then decreased; MDA accumulated slowly at salinity (≤ 0.8% NaCl), then increased rapidly at salinity (> 0.8% NaCl). Conclusion: T. laceolata callus has an adaptability to salty environment (≤ 0.8% NaCl), and the callus could alleviate the salty injury by increasing the contents of the osmotic regulation substances, such as proline and soluble sugar, decreasing the osmotic potential, and increasing the activities of anti-oxidative enzyme.
9.Induced differentiation of adipose-derived stromal cells into myoblasts.

This study aimed to induce the differentiation of isolated and purified adipose-derived stromal cells (ADSCs) into myoblasts, which may provide a new strategy for tissue engineering in patients with stress urinary incontinence (SUI). ADSCs, isolated and cultured ex vivo, were identified by flow cytometry and induced to differentiate into myoblasts in the presence of an induction solution consisting of DMEM supplemented with 5-azacytidine (5-aza), 5% FBS, and 5% horse serum. Cellular morphology was observed under an inverted microscope. Ultrastructural changes occurring during the differentiation were observed by transmission electron microscopy and confocal laser scanning microscopy. Cellular immunohistochemical staining was applied to determine the expression of desmin protein in cells with and without induced differentiation. Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were used to detect mRNA and protein expression, respectively, of sarcomeric and desmin smooth muscle proteins. The results showed that ADSCs were mainly of a spindle or polygon shape. Flow cytometry analysis revealed that ADSCs did not express CD34, CD45, and CD106 but high levels of CD44 and CD90, which confirmed that the cultured cells were indeed ADSCs. After induction with a 5-aza-containing solution, morphological changes in ADSCs, including irregular cell size, were observed. Cells gradually changed from long spindles to polygons and star-shaped cells with microvilli on the cell surface. Many organelles were observed and the cytoplasm was found to contain many mitochondria, rough endoplasmic reticulum (rER), and myofilament-like structures. Cell immunohistochemical staining revealed different levels of desmin expression in each phase of the induction process, with the highest expression level found on day 28 of induction. RT-PCR and Western blot results confirmed significantly higher desmin gene expression in induced cells compared with control cells, but no significant difference between the two groups of cells in sarcomeric protein expression. It was concluded that under specific induction setting, ADSCs can be induced to differentiate into myoblasts, providing a potential new option in stem cell transplantation therapy for SUI.
