2.Study on Inducing of Cord Blood Derived Endothelial Progenitor Cells into Endothelial Cells

Objective To induce cord blood derived endothelial progenitor cells(EPCs)into endothelial cells,and investigate the feasibility of these cells as the seed cells of tissue engineering cardiovascular replacement.Methods Mononuclear cells were isolated from fresh cord blood by 6%HES and density gradient centrifugation.Isolated cells were cultured in medium supplemented with vascular endothelial growth factor(VEGF).Attached cells were identied by morphology,immunofluorescence staining and flow cytometry.Results The percentage of mononuclear cells isolated from fresh cord blood was(3.4?2.1)?10~7/mL.The morphology of attached cells changed while being cultured and inducted,from small-sized round cells to spindle-like cells,to a typical cobblestone morphology,and the total number of cells was 10~7.After 7 days of culture,immunofluorescence staining showed that the vWF and VEGFR-2 were expressed.Compared with the original,cell markers CD_133 decreased(3.11%?1.05%) to(0.09%?0.02%),P
3.Construction of TK Gene-deleted PRV SH StrainContaining a Single LoxP Site

Pseudorabies virus (PRV) is a swine herpesvirus of the Alphaherpesvirinae subfamily and a pathogen of swine resulting in devastating disease and economic losses worldwide. Cre/loxP site-specific system has the character of site specific, time specific, tissue specific and high efficiency in recombination, which makes this system universal in vivo and in vitro recombination of bacteria, fungus, plants, insects and mammals. A recombinant PRV which contain a loxP site in TK locus by using Cre/LoxP recombinant system was construsted. A pair of primers were synthesized according to the pEGFP-C1 sequence published on GenBank, and were used to amplify the EGFP gene expression cassette with two loxP sites flanking each side. This target gene was cloned into pSKLR, the resulting transfer vector pSKLR-GFP-loxP was then cotransfected into 293T cells with PRV SH strain genomic DNA. The recombinant virus rPRV1 was selected and purified in TK-143 cells by choosing fluorescent expressing plaques. Cre expression vector pOG231 was cotransfected into 293T cells with rPRV1 genomic DNA. The second recombinant virus rPRV2 was obtained, which contains only one loxP site in TK locus. Sequencing results of rPRV2 TK gene indicated that 34bp loxP site was inserted into rPRV2 genome and there were 270bp deletion in TK gene. PCR amplifying different generations of rPRV2 TK gene showed that the mutant was stable when passages in RK-13 cells. TCID_ 50 assay indicated that rPRV2 grows well on RK-13 cells. The LD_ 50 test results on BALB/C mice suggested that the virulence of rPRV2 was reduced. As a conclusion, the report gene GFP expression cassette was removed successfully from rPRV1 genome and only one LoxP site was leaved in rPRV2 genome by using Cre/LoxP recombinant system.
4.Preliminary experiences with the da Vinci S surgical system in thoracic surgery

Objective The goal of this report was to evaluate the applicability of the da Vinci S surgical system in thoracic surgery.Methods The da Vinci S surgical system consists of a console, a patient cart, and a vision system.The patient cart loading with 3 robot arms and 1 central endoscope arm connects the console which commands the movements of the arm tips.The robot arm tips are introduced via small chest wall ports and attached to the arms of the robot.The surgeon, sitting at the console, manipulates highly sensitive sensors that transfer the surgeon's movements to the arm tips.The so called EndoWrist'technology offers up , down, left, right, and twist, seven degrees of movements, thus exceeding the capacity of a surgeon's hand in open surgery.17 intrathoracic lesion cases, including 12 Myasthenia Gravis, 1 diaphragm hernia, 2 esophageal cancer, 1 pulmonary cancer , 1 pneumothorax, were evaluated for clinical application of the da Vinci S surgical system.Results Out of 17 surgical procedures, 14 procedures were done using the robot from beginning to the end, including 12 thymectomies, 1 diaphragm hernia repair, 1 pulmonary bleb dissection.Only gastric mobilizations, the abdominal part procedures were done using the da Vinci S system, and the thoracic part procedures were done through small incision thoracotomy in 2 cases with esophageal cancer.One resection of left upper lobectomy had to be converted due to surgical problem.The postoperative courses were uneventful.Conclusion The da Vinci operating robot can do nearly all kinds of thoracic operations.Advanced general thoracic procedures can be performed safely and effectively with the da Vinci S robot allowing precise dissection.This benefit becomes evident most elegantly in thymectomies.The robot operation procedures can be done by the doctors with open and assisted thoracopic surgery experiences and other personnels getting trained in a short period of time, but case selection and preparing emergency thoracotomy at any time is needed to ensure the patient safety.

The paper described the rationality of traditional and modern application of prepared decoction pieces of herbal medicine on basis of application, statistics and comparison analysis of three forms of drugs of traditional Chinese herbal pieces prepared for decoction, prepared decoction pieces in small packing and granules; and illustrated different opinions correlative to the three forms of drugs; put forward the counter-measures and proposals for the problems facing the traditional Chinese herbal pieces for decoction; the paper stated clearly that the traditional Chinese herbal pieces for decoction should not be replaced, instead, the viewpoint and the reasons on its application must be holding on; and the trend of development and expectations of the Chinese herbal pieces for decoction were predicted as well.

Methods The operating principle, construction design, electronic control system and software program flowchart of a robot transfer arm for victim-transfer were introduced.Results and Conclusion The victim didn not have to change their body posture during transfer. The procedure is very simple.A push at only one key is enough,without secondary injury.
7.Identification of a Thermoacidophilic Sulfolobus sp. Isolated from a Hot Spring in Tengchong Rehai

An extremely thermoacidophilic isolate K4-1 was obtained from an acidic hot spring in Teng- chong Rehai, Yunnan province. Morphology, growth characteristics, utilization of carbon compounds, en- ergy sources and 16S rRNA gene sequence of K4-1 were studied. Cells of K4-1 are irregular cocci with monotrichous flagella. The strain grew aerobically in either a lithotrophic or a heterotrophic mode. Growth on elemental sulfur occurred through oxidation of sulfur. It grew optimally at 75?C and pH 3.5. On the basis of 16S rRNA gene sequence similarity, strain K4-1 was shown to belong to genus Sulfolobus, being related to the type strains of genus Sulfolobus (86.6%~94.3% similarity), and being most closely related to strain Sulfolobus tengchongensis RT8-4 (98.9% similarity). The GenBank accession number of strain K4-1 16S rRNA gene sequence is EU729124.
8.Hydrogenation properties of mechanically milled Mg2Nio.8Cr0.2-CoO/Al2O3 composites

Mg2Ni0.8Cr0.2-x wt.% CoO/Al2O3 (x=0.5, 1, 2 and 3) composites were prepared by mechanically milling sintered Mg2Ni0.8Cr0.2 alloy and CoO/Al2O3 compound for 45 h. The addition of CoO/Al2O3 compound resulted in the good kinetics properties of hydriding/dehydriding reaction of the composites. The composite with 1.0 wt.% CoO/Al2O3 catalyst could reach the maximum hydrogen absorption capacity (2.9 wt.%) within 5 min at 393 K under H2 pressure of 4 MPa, and can desorb rapidly at 493 K. The decomposition and synthesis of hydrogen molecule on Mg2Ni0.8Cr0.2 alloy surface was promoted by addition of CoO/Al2O3 catalyst. In addition, the formation of metallic Ni particles, strain and defects during the ball milling process also resulted in the improved hydrogenation performance of Mg2Ni-based alloys.
9.Study on the effects of watering tube with normal saline during transfusion in preventing from drugs' bad interactions

Objective To observe the significance of watering tube with normal saline during transfusion in preventing from drugs' bad interactions. Methods Totally 60 cases with acute upper respiration infection accepted transfusion were randomly divided into two groups. The study group ( n = 30) were performed watering tube with normal saline during transfusion. The control group (n = 30 ) were done with general transfusion nursing. All the subjects were observed about appearance changes of drugs together in Murphy's tube and recorded side-effects and clinical efficacy. At the same time, the satisfactions about transfusion were investigated and compared. Results The probability of appearance change in Murphy's tube in the study group (0.0%)showed significantly lower than that in the control group(30.0% ,P <0. 01 ). The incidence of side-effects in the study group( 10.0% ) showed significantly lower than that in the control group(33.3% ,P<0. 05).There showed no significant differences between the two groups in clinical efficacy and treatment days (P >0. 05). The satisfactions about transfusion,the drug dispensation and transfusion technology in the study group showed significantly higher than those in the control group ( P < 0. 05 or 0. 01 ). Conclusions The way of watering tube with normal saline during transfusion has some advantage in preventing from drugs' bad interactions. It may be used skillfully during clinical transfusion.
10.Analysis on genetic diversity of different Salvia miltiorrhiza geographical populations in China.

OBJECTIVETo research on genetic diversity of different Salvia miltiorrhiza geographical populations in China.
METHODThe genetic diversity of 27 S. miltiorrhiza geographical populations from ten provinces in China was estimated using amplified fragment length polymorphism (AFLP) markers. The data of amplified bands were analyzed by the software POPGENE and SPSS.
RESULTThe ten primers employed produced a total of 528 discernable and reproduceable amplified fragments. There were 476 polymorphic brands. The percentage of polymorphic bands with in different populations was 90.15%. Genetic diversity analysis showed that Neis gene diversity (He) was 0.261 2 and Shannon's genetic diversity index (1) was 0.403 3. The coefficient of gene similarity was 0.504 0-0.789 0 between populations. The cluster map including all samples were obtained by UPGMA. In the map, there were seven cluster groups and one individual outside the groups.
CONCLUSIONThe genetic diversity with in different geographical population of S. miltiorrhiza in China is plentiful.
