1.Comparison of retinopathy of prematurity incidences between single and multiple premature infants and analysis of related factors

Background Retinopathy of prematurity (ROP) is one of major causes of childhood blindness.Early screening and treatment of ROP is necessary.Objective This study was to analyze the incidence of ROP in single and multiple premature infants and related factors.Methods This study was approved by Ethic Committee of Suzhou Municipal Hospital,and oral informed consent was obtained from the parents of the infants before any medical examinations.Five hundred and four fetus with the gestational age of ≤34 weeks or birth weight of ≤ 2 000 g were collected in Suzhou Municipal Hospital,of whom 357 infants were single fetus birth and 147 infants were multiple fetus,including 26 twins and 21 three fetus birth.The fundus were examined by RetCam Ⅱ equipment and ROP was determined based on the fundus findings and history of birth.ROP incidences between the single fetus and multiple fetus were compared,and the inducing factors were analyzed.Results Sixty-six infants were ROP in all 504 fetus,with the incidence of 13.10%.The incidence of ROP in the single fetal group was 10.92% (39/357),which was significantly higher than 18.37% (27/147) in the multifetal group (x2=5.069,P=0.024),and risk of ROP in the multifetal group was higher than that in the single fetal group (OR =1.835).The oxygen inhalation rate was 42.02% (150/357) and the oxygen inhalation duration was 5.71 days in the single fetal group,and that in the multifetal group was 58.50% (86/147) and 9.42 days,respectively.Conclusions Incidence and risk of ROP in the multifetal infants are higher than those in the single fetal infants,and low-birth-weight,less gestational weeks and excess oxygen inhalation are influence factors.
2.Factors affecting immune reconstitution in HIV/AIDS patients after antiretroviral therapy

Objective:
To investigate the immune reconstitution of HIV/AIDS patients and its influencing factors after receiving antiviral therapy (ART) in Hangzhou City, so as to provide insights into improving the treatment effects and quality of life in HIV/AIDS patients.
Methods:
A retrospective cohort of HIV/AIDS patients who began antiviral treatment between January 1, 2016 and August 31, 2021 and had a baseline CD4+T lymphocyte (CD4) counts of less than 500 cells/μL or a baseline CD4/CD8+T lymphocyte (CD8) ratio of less than 0.8 in Hangzhou City was followed up until August 31, 2023. Demographic information, antiviral therapy in formation, CD4 counts, and CD4/CD8 were collected from the Chinese Disease Prevention and Control Information System. A good immune reconstitution was defined as having CD4≥500 cells/μL and CD4/CD8≥0.8. The immune reconstitution status of HIV/AIDS patients were analyzed, and factors affecting immune reconstitution were identified using a multivariable Cox proportional risk regression model.
Results:
A total of 3 349 HIV/AIDS patients were enrolled, with a median age at ART of 31 (interquartile range, 20) years. There were 3 075 males (91.82%), 1 600 cases with college education and above (47.78%) and 2 455 cases at WHO clinical stage Ⅰ-Ⅱ(73.31%). There were 1 368 cases with good immune reconstitution, accounting for 40.85%, and the proportion of HIV/AIDS patients with good immune reconstitution that began ART in 2016 was the highest, reaching 51.90%. Multivariable Cox proportional risk regression model identified WHO clinical stage (Ⅰ-Ⅱ, HR=2.529, 95%CI: 2.023-3.162), timely ART (HR=1.196, 95%CI: 1.027-1.394), initial treatment regimen (TDF+3TC+NVP/EFV, HR=2.185, 95%CI: 1.891-2.524; integrase inhibitors, HR=8.509, 95%CI: 6.706-10.795), baseline CD4/CD8 (≥0.1, HR: 1.600-4.515, 95%CI: 1.061-6.661), baseline hemoglobin (<90 mg/dL, HR=0.327, 95%CI: 0.121-0.880), hepatitis B infection (HR=0.619, 95%CI: 0.457-0.840) and hepatitis C infection (HR=0.308, 95%CI: 0.099-0.956) as factors affecting immune reconstitution in HIV/AIDS patients.
Conclusion
The immune reconstitution in HIV/AIDS patients after ART is associated with WHO clinical stage, timely ART, initial treatment regimen, baseline CD4/CD8, baseline hemoglobin and hepatitis B or C infection.
3.Effects of negative PTFE electrets on the apoptosis of fibroblast cells

Objective: To study the influence of negative electrets on apoptosis of fibroblast cells and to probe its mechanism. Methods: Fibroblast cell were treated with -300, -500 and -1 000V PTFE electrets for 24, 48 and 72 h, respectively, and the influence of negative electrets on cell apoptosis was studied by means of flow cytometry and transmission electron microscope. Results: Compared with control group, apoptosis cells increased from 0.5% to 10% (some even to 15%) after 24,48 and 72 h action of -300, -500 and -1 000 V electrets. After action of -500 V PTFE electrets for 48-72 h, fibroblast cells showed characteristic morphological features of apoptosis. These features included chromatin aggregation, nuclear and cytoplasmic condensation and partition of cytoplasm and nucleus into membrane bound-vesicles (apoptotic bodies). The effect of negative electrets on apoptosis was in proportion to the time and electric field intensity. Conclusion: Negative electrets can enhance apoptosis of fibroblast cells.
4.Expressions of tumor necrosis factor-α, caspase-8 and caspase-3 in lichen planus lesions

Objective To quantify the expressions of tumor necrosis factor-α (TNF-α),caspase-8 and caspase-3 in lichen planus (LP) lesions,and to investigate their significance.Methods Skin samples were collected from the lesions of 20 patients with LP and normal skin of 20 healthy human controls.Immunohistochemistry was used to determine the expressions of TNF-αt,caspase-8 and caspase-3,and terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end-labeling (TUNEL) technique to evaluate the apoptosis in keratinocytes,in these samples.Results The expression levels (expressed in integrated optical density,IOD)of TNF-α,caspase-8 and caspase-3 were (12.58 ± 2.33) × 103,(11.69 ± 3.52) × 103 and (11.45 ± 2.82) × 103 respectively in LP lesions,significantly higher than those in the normal skin ((5.12 ± 1.78) × 103,(3.87 ± 3.36)× 103,(4.76 ± 1.93) × 103,t =11.38,7.19,8.76,respectively,all P < 0.01).Elevated apoptosis index was noted in keratinocytes from LP lesions compared with those from normal skin (71.35 ± 7.93 vs.33.62 ± 8.75,t =14.29,P < 0.01).In LP lesions,the expressions of both TNF-α and caspase-8 were positively correlated with the apoptosis index of keratinocytes (r =0.72,0.75,respectively,both P < 0.01) and the expression of caspase-3 (r =0.68,0.73,respectively,both P < 0.01).Conclusion The up-regulated expressions of TNF-α,caspase-8 and caspase-3 may participate in the apoptosis in keratinocytes in LP.
5.Morphological and functional alteration of serotonin-producing intestinal enterochromaffin cells in patients with irritable bowel syndrome

Objective To investigate morphological and synthetic alteration of enterochromaffin(EC) cells of intestinal mucosa in patients with irritable bowel syndrome(IBS). Methods Fifty cases of IBS were classified into diarrhea predominant and constipation predominant in accordance with RomeⅡ criteria. Colon biopsy tissues were stained through Envision immunohistochemistry. Morphological changes of EC cells in intestinal mucosa were also studied by electron microscopy. Results EC cells were seen in the crypt of intestinal mucosa. The shapes and the number of EC cells in diarrhea predominant IBS and constipation predominant IBS increased remarkably, compared with those in controls (15.90 ?5.09, 14.73?2.73 vs. 7.27?2.50). It was shown that EC cells synthesized excessive 5 hydroxytryptamine (5 HT) by immunohistochemistry. The function of EC cells in IBS was active under electron microscopy. Conclusions Active enteral EC cells noticed in IBS indicate that excessive 5 HT may play an important role in the pathogenesis of IBS.
6.Survival analysis of HIV/AIDS patients receiving antiretroviral therapy in Hangzhou from 2004 to 2014

Objective To analyze the survival rate of HIV /AIDS patients receiving highly active antiretroviral therapy(HAART)since the implementation of the national Four Free and One Carepolicy against HIV in Hangzhou.Methods Clinical data of 2370 AIDS patients were collected from National AIDS Comprehensive Treatment Information System Treatment Library from 2004 to 2014.The data, including basic information,viral load,CD4 +T lymphocyte counts,starting time of treatment,WHO clinical stage,infection pathways and follow-up were respectively analyzed.Kaplan-Meier and Cox proportional hazards models were used to analyze the survival rate and the factors affecting survival.Results The total follow-up time was 3968.14 person years and 57 patients died in 2370 patients with a mortality rate of 1 .44 /100 person years (57 /3968.14).Kaplan-Meier method showed that the cumulative survival rates of the first,third and fifth year were 98.08%,96.20% and 95.24%,respectively.The overall mortality rate fell from 6.06 /100 person years in 2006 to 1 .44 /100 person years in 2014.The mortality rate of AIDS-related disease declined from 1 .10 /100 person years in 2009 to 0.90 /100 person years in 2014.Multivariate Cox regression analysis showed that the risk of death for patients with CD4 +T 200-349 cells/μL was 0.466 times(95%CI 0.246-0.882)as that for patients with CD4 +T cells <200 /μL.The risk of death was 3.408 times(95%CI 1 .365-8.506)in patients aged≥ 50 years,3.788 times(95%CI 1 .645-8.718)in patients aged 40 to <50 years,and 2.593 times(95%CI 1 .139-5.905)in patients aged 30 to 40 years as that in patients aged <30 years.The mortality risk for patients with baseline WHO stage Ⅲ and Ⅳ was 1 .960 times as patients with WHO stage Ⅰ and Ⅱ (95% CI 1 .117-3.439 ).Conclusions Patients with increased age,low CD4 +T counts and baseline WHO stage Ⅲ or Ⅳ are main risk factors affecting survival rate of HIV /AIDS patients,early antiviral therapy is the key for improving the survival rate of patients.
7.Effect of MoS2 content on mechanical properties and electrical wear performance of Ag-MoS2 composites

BACKGROUND: Silver-matrix composite is usually used as a kind of electrical conductive matedal, which has excellent properties in electrical and heat conducting. MoS2, which used as a common solid lubricating material, has unique anti-frictional and lubricating properties in the process of wearing. Ag-MoS2 composites using in the area of electrical sliding wear were fabricated by adding MoS2 to the silver-matrix. OBJECTIVE: Ag-MoS2 composites with different content of MoS2 were produced to gain the mixture ratio for best properties by the experiments of mechanical and wear test. DESIGN, TIME AND SETTING: The controlled experiment was performed at the Department of Matedal Science and Engineering, Hefei University of Technology from April to May 2008. MATERIALS: Silver powder (Bengbu Gold and Silver Article Factory, China) and MoS2 powder (Sinopharm Chemical Reagent Co., Ltd., China) were used in this study. METHODS: Ag-MoS2 composites for five different contents were fabricated by powder metallurgy method, which included the processes of powder mixture, drying, initial pressure, sintering, and second pressure. MoS2 contents were the MoS2 composite of 12.0%, 13.5%, 15.0%, 16.5%, 18.0% silver- MoS2. MAIN OUTCOME MEASURES: Flexural strength was measured by versatility mechanical testing machine. The microstructures of the composites after sintering were observed by an optical microscope. The electrical sliding friction and wear properties were tested by the electrical wear test machine. The worn surfaces of the samples were observed by a scanning electron microscope. RESULTS: With the increase in MoS2, the flexural strength decreased, the resistivity increased, and the wear loss decreased first, and then increased, which comes the lowest point when the MoS2 content was 15%. CONCLUSION: When the content of MoS2 is 15%, the average properties was the best.
8.Infection characteristics of newly reported HIV/AIDS cases in Hangzhou City in 2022

Objective:
To analyze the infection characteristics of newly reported HIV/AIDS cases in Hangzhou City, so as to provide the reference for effective AIDS intervention.
Methods:
Newly reported HIV/AIDS cases in Hangzhou City in 2022 were recruited. Demographic information, HIV testing status, infection routes and sexual behaviors were collected using questionnaire surveys. Blood samples were collected before antiviral treatment, and HIV-1 pol gene sequences were detected to construct molecular transmission networks. The characteristics of HIV/AIDS cases, including infection routes, time, and location were analyzed. Factors affecting infection time and location among HIV/AIDS cases were analyzed using a multivariable logistic regression model.
Results:
A total of 1 007 HIV/AIDS cases were reported in Hangzhou City in 2022, with 907 cases (90.07%) completing questionnaire surveys. Among them, 833 were males (91.84%), and 532 had out-of-province household registrations (58.65%). Ninety-one molecular transmission networks were established, and 276 cases were involved, with homosexual contact as the main infection route (199 cases, 72.10%). There were 311 recently infected cases (35.34%) and 569 previously infected cases (64.66%) among 880 cases whose infection time could be determined. There were 531 locally infected. cases (70.24%) and 225 imported cases (29.76%) among 756 cases whose infection location could be determined. Multivariable logistic regression analysis showed that the HIV/AIDS cases who were identified through voluntary counseling and testing (OR=1.826, 95%CI: 1.055-3.175) and sought sexual partners through homosexual dating apps (OR=2.461, 95%CI: 1.193-5.234) were more likely to be recently infected; the cases who lived in Hangzhou City for more than one year (>1 to 5 years, OR=2.853, 95%CI: 1.552-5.358; >5 years, OR=3.534, 95%CI: 1.382-9.804), sought sexual partners through entertainment venues (OR=3.449, 95%CI: 1.390-8.935), online/social apps (OR=2.416, 95%CI: 1.084-5.488) and homosexual dating apps (OR=3.734, 95%CI: 1.677-8.493) were more likely to be locally infected; student cases were more likely to be infected outside Hangzhou City (OR=0.115, 95%CI: 0.019-0.525).
Conclusions
The newly reported HIV/AIDS cases in Hangzhou City in 2022 were primarily infected through homosexual contact, previously and locally. Seeking sexual partners through homosexual dating apps is an important influencing factor for recent and local infections, highlighting the need for strengthening traceback investigations of related cases.
9.Screening anti-fibrosis Chinese medicinal compounds based on machine learning

Objective To establish a new-type virtual screening predictive model of Chinese medicinal compounds with anti-fibrosis effects, and to verify the predictive performance of the model.Methods The dimension reduction and characteristic optimization of molecular fingerprints were implemented by using random forest (RF) algorithm and gradient boosting decision tree (GBDT) algorithm.A hybrid model of characteristic optimization-machine learning was established, and optimized characteristics were input into logistic regression (LR) and machine learning algorithm of artificial neural network (ANN) for training.Precision, recall rate and F1 value were used for reviewing the performances of various model combinations.The virtual screening predictive model of Chinese medicinal compounds with anti-fibrosis effect was determined according to results of model performance reviewing.The predictive results of anti-fibrosis activity of Chinese medicinal compounds were compared between the virtual screening predictive model and molecular docking model for further verifying the predictive efficiency of the virtual screening predictive model.Results The precision of RF model was 0.76, recall rate was 0.75 and F1 value was 0.74 (AUC=0.818).The precision that of GBDT model was 0.76, recall rate was 0.74 and F1 value was 0.72 (AUC=0.829).The precision of ANN model was 0.75, racall rate was 0.75 and F1 value was 0.75 (AUC=0.802) , and that of model of RF+LR was 0.77, recall rate was 0.76 and F1 value was 0.75 (AUC=0.840).The precision of model of RF+LR was 0.74, recall rate was 0.84 and F1 value was 0.79 (AUC=0.850) , and that of model of GBDT+LR was 0.80, recall rate was 0.80 and F1 value was 0.79 (AUC=0.872).The precision of model of GBDT+ANN was 0.73, recall rate was 0.91 and F1 value was 0.81 (AUC=0.837).The results of molecular docking activities of Chinese medicinal compounds including curcumin, glycyrrhizic acid, hydro-xysafflor yellow A, emodine and gypenoside were accordance with the predictive results of the virtual screening predictive model.Conclusion The model based on RF+LR is better than the models established based on other methods.The virtual screening predictive model has good performance in prediction of Chinese medicinal compounds through comparing with molecular docking model.The method has feature of highthroughput screening and can make up the shortage of compound screening efficiency in molecular docking.It provides a new way for virtual screening prediction of Chinese medicinal compounds with anti-fibrosis effects.
10.Research on Traditional Chinese Medicine Professor Niu Jianzhao's Medication Rule in Prescriptions with Climacteric Syndrome Based on Data Mining

This study was aimed to explore traditional Chinese medicine (TCM) professor Niu Jianzhao's medication rule in prescription with climacteric syndrome.Prescriptions treating climacteric syndrome that built by professor Niu were collected to build a database based on TCM Inheritance Assist System.The method of association rules with apriori algorithm was used to achieve frequency of single medicine,nature,taste and channel tropism of herbs,frequency of drug combination,association rules between herbs and core herb combinations.The data mining results showed 22 types of herbs used with high frequency,28 pairs of herb combination with most frequency,20 groups of herb association rules with the confidence of more than 0.95,52 herb pairs with high association,32 groups of three herbs as the core combinations,and 7 new prescriptions.It was concluded that in professor Niu Jianzhao's treatment of climacteric syndrome,the drugs were commonly used to invigorate the kidney,liver and spleen,and in combination with promoting blood circulation and calming the nerves.The medicinal properties of used herbs were even,warm or slightly cold.The tastes of used herbs were sweet,bitter and spicy.The channel tropisms of used herbs were the liver,kidney and lung meridian.This study highlighted data mining as an important tool for expedited analysis of TCM professor Niu Jianzhao's medication rules in prescriptions with climacteric syndrome.
