Objective To observe the free radicals level in Toufeng disease,also named migraine,so to explore the pathogenesis of the Toufeng disease,as well as to provide an effective treatment for migraine with integration of traditional Chinese and western medicine.Methods The free radicals(SOD,MDA) level in the Toufeng disease onset group(100 cases) were tested and compared with the healthy group(100 cases).The free radicals(SOD,MDA) of the patient group were tested in 4 hours after headache onset while that of the healthy group were tested as soon as taken in.Serum SOD was assayed with xanthine oxidase and MDA with TBA(thiobarbituric acid).Results Compared with the healthy group,the level of SOD was significantly decreased(P
2.Ender nail fixation for pediatric femoral shaft fractures

Objective To investigate the effectiveness of Ender nail fixation in the management of pediatric femoral shaft fractures. Methods A total of 24 children with femoral shaft fracture underwent small incision Ender nail internal fixation followed by 4 weeks of monolateral hip spica-cast immobilization. Results Follow-up observations for 6~24 months revealed no non-union or delayed union. The affected limbs were found within 1 cm shorter or longer than the contralateral in 4 cases. None became lame, and the functions of lower limbs were completely recovered. Conclusions So far as indications and contra-indications of Ender nail fixation are strictly followed, this treatment for femoral shaft fractures in children of age 5~10 years gives characters of small incision, anatomic reduction, unimpaired periosteum, short hospital stay, and quick functional recovery.
3.Effect of Yixin Jiangya Capsule on Serum Insulin-like Growth Factor-1 Level in Patients of Left Ventrecular Hypertrophy of Primary Hypertension

Objective To observe the effect of Yixin Jiangya Capsule (YXJYC) on serum insulin-like growth factor-1 (IGF-1) level in patients of left ventricular hypertrophy (LVH) of primary hypertension at stageⅡ with Yinxu Yangkang and blood stasis syndrome (YXYKBSS). Methods 93 patients were randomly divided into the treated group (62 patients) and the control group (31 patients). The two groups were both treated with enalapril, and the treated group was treated with YXJYC simultaneously. Treatment course of the two groups was 6 months. Radioimmunoassay was used to examine the level of serum IGF-1 before and after treatment in the two groups, compared with that of 30 healthy persons. BACKGROUND:The mesenchymal stem calls (MSCs) have multiple-direction differentiation potential.How to induce human mesenchymal stem cells (hMSCs) into chondrogenic in vitro is an advanced researching direction.OBJECTIVE: Experimental study on chondrogenic differentiation of mesenchymal stem cells from human bone marrow in study culture medium so as to observe the characteristic of the cells in the continuous progressive process,such as culture of the primary generation and passage culture.DESIGN: A single sample and open study.SETTING : Department of Orthopaedics, Children's Hospital Affiliated to Soochow University.MATERIALS:A total of 5 children with single bone cyst were selected from Children's Hospital Affiliated to Soochow University from August 2002 to September 2003,including 3 boys and 2 girls aged from 3 to 12 years.All of them donated 5 Ml bone marrow, which was added with 1 Ml (20 U/Ml) heparinization, from which MSCs was obtained. Main reagents were detailed as follows: F-12 (Gibco, USA), fetal calf serum (Sijiqing Bioengineering Company, Hangzhou), trypsin (Sigma, USA), transforming growth factor-β1 (TGF-β1) (Sigma, USA) and vitamin C (the Third Medical Company,Nanjing; batch number: 021017).METHODS:① Standard culture medium:0.1 volume fraction of fetal calf serum,100 U/Ml penicillin,100 U/Ml streptomycin, 5.8 g/L Hepes, 2 g/L NaHCO3 and 0.3 g/L glutamine were added into F-12 culture medium (Ph 7.2-7.4). ②Study culture medium: 50 μg/L vitamin C and 1 μg/L TGF-β1 were added into standard culture medium (Ph 7.2-7.4). ③Cell culture of the primary generation: 12 Ml Hanks which contained 1 Ml of heparinization (20 μ/Ml) and 5 Ml bone marrow was centrifugated. Then the cells which had nucleolus were rinsed. Then cells were cultured in a 50 Ml plastic culture bottle at the density of 3×109 L-1 and were cultured in 6-well culture plate at the density of 1×109 L-1. One bottle and 3 well was used study culture medium,the other bottle and 3 well was used standard culture medium.The cells were cultured in incubator containing 0.05 volume fraction of CO2 and saturated tumidity at 37℃. The medium was changed every 48 hours. The cells were observed under inverted microscope. ④ Passage culture: When the primary generation cells had been cultured with standard culture medium for 10-14 days, we would find that the cells covered 80% of the bottle bottom. Then 2.5 g/L of trypsin containing 0.2 g/L of EDTA was used to digestion and passage. The third generation cells were separated at the ratio of 1:1, and were cultured in two bottles. One bottle used standard culture medium, the other used study culture medium. At the same time, the cells in 6-well plate which contains coverslip were deal with in the same way. 3-wells used standard culture medium, and the others used study culture medium. Culture time was7 days.The cells were observed under inverted microscope.⑤ Index detection:Alcian blue staining was used to show the expression of the glycosaminoglycan of the primary generation and the third generation. Type- Ⅱ collagen immunohistochemical staining was used to show the express of the type- Ⅱ collagen of the primary generation and thethird generation.MAIN OUTCOME MEASUREMENTS:① Morphological observation; ② secretion of glycosaminoglycan; ③ expression of type- Ⅱ collagen.RESULTS: ① Observation under inverted microscope: Most of the hMSCs in primary culture were spindle; a few of them were wide, flat and polygon. After passage, the cells were found in a uniform spindle shape. The cells, which were induced in TGF-β1 and vitamin C, became round or oval shape. ② The positive rate of primary generation was 82.4%, the 3rd generation which were cultured in study culture medium was 76.3% (x2 =1.14,P> 0.05), and the 6th generation was 68.5%(x2 =5.22, P < 0.05). The cells, which were culturedinstandardculturemedium, were negative. ③ The 3rd generation and 6th generation cells which were cultured in study culture medium, were found that brown-yellow granules distributed in cytoplast. It was the positive reaction. The cells, which were cultured in standard culture medium, were negative.CONCLUSION:MSCs derived from human bone marrow may be inducedinto hondrocytes under study culture medium which contained TGF-β1 and vitamin C in vitro.The express of the glycosaminoglycan and type Ⅱ collagen show the characteristic of chondrocyte.
5.Effects of fluid shear stress on the gene expression of core-binding factor alpha 1 in human osteosarcoma cells

BACKGROUND: The role of mechanical stress in the functional regulation of osteoblasts becomes an emphasis in osseous biomechanical researches recently. There are few reports on whether fluid shear stress can induce the expression of core-binding factor α1 (Cbfα1) or not and what is its rule.OBJECTIVE: To investigate the effects of fluid shear stress (FSS) on the gene expression of Cbfα1 in human osteosarcoma cells.DESIGN: A controlled observation experiment.SETTING: Department of Aerospace Biodynamics, Faculty of Aerospace Medicine, Fourth Military Medical University of Chinese PLA.MATERIALS: Subjects were human osteosarcoma cells (MG-63) METHODS: This study was carried out at the Department of Aerospace Biodynamics, Faculty of Aerospace Medicine, Fourth Military Medical University of Chinese PLA from November 2004 to April 2005. ①After cultured for 60 hours, MG-63 were treated with 0.5 Pa (0.5 Pa stress treated group ) or 1.5 Pa (1.5 Pa stress treated group ) FSS in a flow chamber for 15, 30, 60minutes, respectively. Cover glass was put in the Petri dish containing culture medium at the same time, serving as the control group of FSS treated group. ②The total RNA in cells was isolated. Reverse transcription polymerase chain reaction analysis was made to examine the gene expression of Cbfα1 mRNA. The ratio of Cbfα1 mRNA and GAPDH mRNA was calculated.MAIN OUTCOME MEASURES: The mRNA expressions of Cbfα1 at different FSS and different time.RESULTS: ① Compared with control group, Cbfα1 mRNA expression increased significantly at 30 and 60 minutes with the treatment of FSS.Within certain time range (15 to 60 minutes), Cbfα1 mRNA expression was strengthened with the elongation of time and the increase of stress level. ②The Cbfα1 mRNA expression at 30 and 60 minutes were significantly increased in 1.5 Pa stress treated group in comparison with 0.5 Pa stress treated group (P<0.01).CONCLUSION: FSS can significantly increase the gene expression of Cbfα1 in human osteosarcoma cells.
6.Effect of temozolomide on the glioma invasiveness and its possible mechanism

Purpose To explore the role of tumor necrosis factor-α( TNF-α) in the process of temozolomide ( TMZ) reduce glioma in-vasiveness and its possible mechanism. Methods C6 glioma cells of logarithmic phase were randomly divided into TMZ treatment (10, 30, 60, 120, 180, 240 min group) (n=15), dynamic monitoring content of TNF-αin the culture medium was measured by ra-dioimmunoassay, expression of p53 protein in C6 cells was detected with Western blotting method, and cell apoptosis was used with AnnexinV-FITC. A glioma invasiveness model was established in vitro and glioma invasiveness was determined by crystal violet stai-ning. Results For C6 cells, contents of TNF-αin the nutrient fluid and expressions of p53 protein in C6 cells obviously increased af-ter TMZ treatment and they achieved the peak at 120 min (P<0. 01), followed by decrease gradually. Glioma invasiveness was re-duced after TMZ acted on glioma in vitro. Conclusion TMZ can reduce glioma invasiveness by TNF-α, which this role may be is TMZ promote C6 cells release of TNF-α and increased TNF-α due to glioma cells apoptosis.
7.Effect of yixin jiangya capsule on left ventricular mass index and plasma neuropeptide Y of primary grade 2 hypertensive patients with hyperactivity of yang due to yin deficiency blood stasis

BACKGROUND: Left ventricular hypertrophy induced by primary hyper tension is a risk factor which has a dependent effect on various cardiovas cular complications. Reversal left ventricular hypertrophy has been a keyobject for treating hypertension currently. Plasma neuropeptide Y has en dothelium-derived contracting blood vessel, and has adjusting effect on va somotor function through endothelial cell to participate in onset and main tenance of hypertension. OBJECTIVE: To investigate effect of yixin jiangya capsule on left ven tricular mass index (LVMI) and content of neuropeptide Y of primary grade2 hypertensive patiepts with left ventricular hypertrophy of hyperactivity ofyang due to yin deficiency blood stasis. DESIGN: Case analysis on the basis of patients. SETTING: Department of Traditional Chinese Medicine, General Hospitalof Jinan Military Area Command of Chinese PLA; Department of InternalMedicine, Affiliated Hospital of Shandong University of Traditional Chi nese Medicine. PARTICIPANTS: Totally 93 primary grade 2 hypertensive patients withleft ventricular hypertrophy of hyperactivity of yang due to yin deficiency blood stasis were selected from General Hospital of Jinan Military AreaCommand of Chinese PLA and Affiliated Hospital of Shandong University of Traditional Chinese Medicine from March 2003 to August 2005. All cases were divided into two groups according to random digital table. There were 62 cases in observational group including 40 males and 22 females aged from 46 to 70 years with mean age of 58.94 years and coursed from 2 to 19 years with mean course of 8.93 years. There were 31 cases in control group including 19 males and 12 females aged from 44 to 69 years with mean age of 59.27 years and coursed from 2.5 to 18 years with mean course of 9.11 years. Another 30 people were regarded as healthy group.METHODS: Patients in both groups were treated with enalaprol orally with 10 mg each time of once a day. Patients in observational group were also treated with yixin jiangya capsule (component: shengrlihuang, shengshouwu, sangjisheng, chuanniuxi, gouteng, shijueming, xiakucao, tianma,mudanpi, chishaoyao, dilong, chuanshanjia, etc. Mixed according to ratio,extracted, concentrated, made of capsule, 0.5 g/pill, 4.65 g/raw drug, provided by Shandong Jinan Hongjitang Pharmacy Limited-liability Company,batch number:. 021214), 5 pills once for 3 times every day. Courses in both groups were 24 weeks respectively. LVMI and content of neuropeptide Y were measured with ultrasonic cardiograph and radio-immunity analysis method before and after treatment. Meanwhile, indexes mentioned above were also measured in healthy group so as to regard as the controls.MAIN OUTCOME MEASURFS: LVMI, content of neuropeptide Y,changes of blood pressure, adverse events and side effects.RESULTS: Totally 93 patients entered the final analysis. ① LVMI and content of neuropeptide Y were increased both observational group and control group before treatment [(147.2±16.9), (146.7±17.2), (101.8±.22.4) g/m2;(135.4±33.2), (133.8±31.1), (78.6±24.5) ng/L.; P < 0.01]. ② LVMI and content of neuropeptide Y were decreased both observational group and control group after 24-week treatment, and those in observational group were superior to those in control group [(103.9±24.6), (122.6±19.9) g/m2;(82.9±25.2), (113.7±26.2) ng/L; P < 0.05-0.01]. ③ LVMI and content of neuropeptide Y in observational group were not significantly different from those in healthy group increased both in observational group and control group after treatment (P.> 0.05), and those in control group were higher than those in healthy group (P < 0.01). ④ Descent levels of systolic pressure and diastolic pressure were not significantly different between observational group and control group [(137.4±12.8), (134.2:t:11.9) mm Hg;(80.1±8.3), (81.7±7.4) mm Hg, P > 0.05]. Blood routine and liver-kidney function were not changed obviously both groups before and after treatment, and side effect was not observed during the process.CONCLUSION: LVMI and content of neuropeptide Y of primary grade 2 hypertensive patients with left ventricular hypertrophy of hyperactivity of yang due to yin deficiency blood stasis are higher than those of healthy people. Yixin jiangya capsule can deteriorate left ventricular hypertrophy and decrease the content of neuropeptide Y on the basis of application of enalaprol.
8.Ultrastructural changes of the wall of femoral vein after simulated weightlessness in rabbits

BACKGROUND: At present, most researches on simulated weightlessness are confined to its effect on the artery. Therefore, the effect on the vein needs to be further studied.OBJECTIVE: To probe into the effect of simulated weightlessness on the ultrastructure of rabbit femoral vein and the remodeling of femoral vein.DESIGN: A completely randomized and controlled trial based on experimental animals.SETTING: Department of Aerospace and Aviation Medical Science, Fourth Military Medical University of Chinese PLA.MATERIALS: The experiment was completed in the Department of Aerospace and Aviation Medical Science, Fourth Military Medical University of Chinese PLA from December 2000 to December 2001. Altogether 24healthy male New Zealand rabbits, weighing 2.0-2.5 kg were obtained.INTERVENTIONS: The model of Head-Down Tilt( -20°) (HDT) simulated weightlessness was established in rabbits. A total of 24 healthy male New Zealand rabbits were randomly divided into control group, 10-day simulated weightlessness group and 21-day simulated weightlessness group, with 8rabbits in each group. The ultrastructure of the rabbit femoral vein was observed under transmission electron microscope.MAIN OUTCOME MEASURES: Changes and degeneration of endothelial cell, mitochondrion, internal elastic membrane, and smooth muscle.RESULTS: Cell organelles in endothelial cell of the femoral vein of HDT rabbits decreased, mitochondrion dissolved and disappeared, vacuolar degeneration in endothelial cell increased and phagolysosome in endothelial cell could be found. Internal elastic membrane became thinner and was broken. The smooth muscle layer became thinner and some smooth muscle cells became degenerated. Cell interstitial substance increased. There was significant difference in changes between 21-day and 10-day simulated weightlessness groups.CONCLUSION: The vascular remodeling of femoral vein occurs with atrophic changes during simulated weightlessness. The longer duration of simulated weightlessness is, the more obviously the structure of femoral vein changes.
9.A clinical investigation on factors influencing the prognosis of primary liver cancer patients after hepatectomy

Objective To explore factors influencing the prognosis of hepatocellular carcinoma (HCC) patients after hepatectomy. Methods From May 1994 to January 1998, 189 patients who underwent hepatectomy for HCC were enrolled for reviewing their clinical characteristics, treatment and prognosis. Twenty-two parameters contributing to long-term survival rate (SR) and disease-free SR were analysed. Results The 3,5-year cumulative SR of the whole group was 63%,45%, respectively. The 3,5-year SR and disease-free SR in the curative resection (CR) group ( n =162) were 67%,47% and 45%,26% respectively. Results showed that the way by which a tumor was found, tumor size, portal tumor thrombi, satellite nodule, TNM stage, cirrhosis type, recurrent and treatment, blood transfusion, differentiation grade,and CR were risk factors by individual variable analysis( P =0.0000~0.0034); A multivariable analysis showed that CR, tumor size, tumor finding mode and reoperation were significant factors associating with prognosis( P =0.0000~0.0024). Blood transfusion and type of cirrhosis were closely correlated with tumor-free survival ( P =0.0001). Conclusions Curative resection, tumor size, reoperation for recurrence were important factors for recurrence by multivariate analysis. The severity of concomittant liver cirrhosis and perioperative blood transfusion were closely correlated with postoperative tumor free survival.
10.Effects of Intermittent Head-up Tilt on the Endothelin Expression and Morphological Changes of Artery during Simulated Weightlessness in Rabbits.

Objective To observe the effects of intermittent 45? head-uptilt on endothelin(ET-1) expression and morphological changes of femoral and carotid arteries in rabbits induced by 21 d simulated weightlessness.Method Twenty-four healthy male rabbits were randomly divided into control group,simulated weightlessness group and countermeasure group,8 rabbits in each group.The simulated weightlessness group kept head-down tilt for 21 d.The countermeasure group was exposed to 45? head-up tilt for 2 h every day during 21 d head-down tilt.After 21 d experiment,structure of femoral artery and carotid artery were studied and ET-1 expression was tested by immunohistochemical method.Result Detachment of endothelial cells in femoral artery,necrosis of smooth muscle cells and decreased ET-1 expression were found in animals in the simulated weightlessness group,and the vessel wall became thinner.While hyperplasia of endothelial cells and thickened internal elastic lamina of the carotid artery were also observed.No significant changes were observed in femoral artery and carotid artery in animals of the countermeasure group.ET-1 expression decreased in femoral artery but increased in carotid artery in animals of the simulated weightlessness group,but no significant change was found in both femoral and carotid arteries in animals of the countermeasure group as compared with those in the control group.Conclusion Twenty-one days simulated weightlessness may induce an atrophic change and decreased ET-1 expression in femoral artery,and hyperplastic change and increased ET-1 expression in carotid artery in rabbits.Two-hour 45? head-up tilt every day may partly counteract the change in femoral artery and carotid artery induced by simulated weightlessness.
