1.Application of optimized multi-enzyme combination and sample pretreatment in proteomics.

Proteomics is a powerful subject focusing on large-scale study of protein structures and functions. A complete enzymatic digestion of protein complexes is the key step in modern high-resolution and high-throughput mass spectrometry (MS)-based identification and quantification. To achieve MS analysis, both peptide sample pretreatment and data acquisition are prerequisite in proteomic studies. In this paper, we summarized both the enzymatic proprieties of three common proteolytic enzymes, Trypsin, Lys-C and Glu-C, the optimization of multi-enzyme combination and an advanced sample pretreatment in proteomics research.

3.Uterine involution after cesarean section promoted with acupuncture: a randomized controlled trial.

OBJECTIVETo explore the effect and feasibility of acupuncture in uterine involution after cesarean section.
METHODSSeventy cases of cesarean section in primipara were randomized into an observation group (33 cases) and a control group (37 cases). In the control group, the conventional treatment was adopted after cesarean section. In the observation group, on the basic treatment as the control group, acupuncture was applied at Sanyinjiao (SP 6), Hegu (LI 4), Qihai (CV 6) and Guanyuan (CV 4). In 2 h after operation, acupuncture started, once a day, continuously for five times. The daily height of uterine fundus, daily uterine fundus decreasing degree, postpartum blood loss, lochia duration and ultrasonic B test in 42 days of postpartum were compared between the two groups.
RESULTSThe therapeutic methods of the two groups all promoted uterine contraction. The postpartum height of uterine fundus in the observation group was lower than that in the control group (all P<0.05) and the uterine fundus decreasing degree was higher than that in the control group (all P<0.05). The blood loss in 2 h, 6 to 12 h and 24 to 48 h of postpartum was less than that in the control group (all P<0.05). The lochia duration in the observation group was shorter than that in the control group (P<0.05).
CONCLUSIONAcupuncture promotes uterine contraction, reduces postpartum blood loss and lochia duration and benefits uterine involution after cesarean section.

4.Analysis on fabrication and performance of novel NO electrochemical microsensor based on Teflon/Au nanoparticles

Objective To explore the performance of the novel nitric oxide(NO) electrochemical microsensor based on Au nano-particles(nano-Au) modified glass-fiber .Methods With columnar glass-fiber as the substrate material ,the in-situ chemical seed-growth technology was adopted to fabricate one kind of electrochemical microsensor based on gold nanoparticles pole electrodes and the amperometric response method was used to investigate the various properties of the NO microsensor .Results This microsensor had high sensitivity response to NO ,its linear range was 7 .2 nmol/L -11 .7μmol/L(r=0 .998) with a detection limit of 3 .6 nmol/L .Conclusion A novel electrochemical microsensor based on nano-Au pole electrode was fabricated .The NO microsensor posseses high sensitivity ,wide linear range ,good reproducibility and excellent anti-interference ability .
5.CT features of gastric heterotopic pancreas

Objective To analyze CT findings correlated with pathologic findings in ectopic pancreas of the stomach.Methods CT scans of 15 surgically proven cases of ectopic pancreas of the stomach were reviewed,and enhanced CT scan was performed in 11 cases.CT findings were correlated with the pathologic findings.Results All cases had single lesion,and all lesions showed homogeneous density on plain scans without cystic or malignant changes.The size ranged from 1.3 to 3.1 cm,with mean diameter of (1.9? 0.2)cm.The lesions were round or oval in shape with broad base against the gastric wall.Two showed central umbilication sign.Only 2 cases were correctly diagnosed prior to operation and the rest were misdiagnosed or diagnosed indistinctly.The locations were in the gastric antrum in 11 cases,in the body in 3,and in fundus in one;The ectopic pancreas located in the greater curvature in 10,and in the lesser curvature in 5.Homogeneous or inhomogeneous strong enhancement similar to the pancreas was seen in 8 cases and they consisted mainly of pancreatic acini with the same histologic features as the pancreas.Three cases showed poor enhancement and consisted mainly of ducts and hypertrophied muscle,pancreatic acini were a minor component.Conclusion Ectopic pancreas of the stomach showed characteristic locations with the findings of snhmucosal diseases.Different enhancing patterns were correlated with their pathologic findings.
6.Changes of VEGF protein and mRNA in the vulva of rabbits after ultrasound irradiation

Objective To investigate ultrasound as a therapy for white lesions of the vulva.Methods Immunohistochemistry and hybridization in situ were used to study changes in VEGF protein and mRNA in the vulva skin of rabbits after ultrasound irradiation.Results After irradiation,VEGF protein and mRNA had increased markedly after 4 and 7 days,but their levels recovered by day 14.Conclusion Ultrasound irradiation causes local edema and hypoxia which enhance production of VEGF.VEGF promotes growth of local microvessels and increases the permeability of the endothelial cells of blood vessels.These changes improve nutrition of the mierovessels and nerve endings and ameliorate the microenvironment for tissue growth.One could speculate that ultrasound irradiation helps rehabilitation and regeneration of pathologic vulva skin.
7.Studies on genetic diversity of three Tibetan herbs.

The genetic diversity of three Tibetan herbs, i. e., Sang-Di, E-Dewa and Ye-Xingba (Tibetan names), was studied based on the field collection, specimen identification and DNA sequence analysis. Swertia hispidicalyx, Gentiana lhassica and Scrophularia dentata, as the original plants of the three Tibetan herbs, were collected and identified. The regions of ITS, matK, rbcL, rpoC1, trnL(UAA), psbA-trnH, atpB-rbcL, trnS (GCU)-trnG(UCC), rpl20-rps12, trnL(UAA)-trnF(GAA) and nadl 2nd intron were amplified and sequenced. The ITS regions of S. hispidicalyx and S. dentata were cloned and sequenced, and the sequences were classified into different genotypes. All the sequences were analyzed and compared with those of closely related species. Our studies may provide reference for the genetic diversity analysis and molecular identification of the three Tibetan herbs.

8.Using a new generation of classification system of antineoplastic drugs to guide Chinese medicine research.

A new generation of classification system of antineoplastic drugs was put forward based on comparing the first with the second generation classification system of antineoplastic drugs. Antineoplastic drugs are divided into cytotoxic drug, cells biological behavior regulator, biological response modifier and biochemical modulator. The using of biological response modifier (immune modulator for instance) and biochemical modulator are supplementary methods for Western medicine treatment in tumor, because the cytotoxic effects of Chinese herbs are lower than those of chemotherapeutic drugs. For the new breakthrough of Chinese medicine oncology research, new idea, new technology and new target should be used, the Chinese medicine mechanism should be studied from a new view. Reversing abnormal biological behavior of tumor cells by Chinese medicine could be an important breakthrough of Chinese medicine oncology research.

9.Effect of Busheng Huoxue Capsule on the quality of life of primary osteoporosis senile males.

OBJECTIVETo investigate the effect of Busheng Huoxue Capsule (BHC) on the quality of life (QOL) in senile male osteoporosis (OP) patients, and to explore its mechanisms.
METHODSTotally 200 senile primary OP patients were randomly assigned to two groups according to random digit table method, 100 in each group. Patients in the treatment group took BHC plus caltrate-D (600 mg CaCO3), while those in the control group took alendronate (70 mg per week) plus caltrate-D. The therapeutic course was 12 months for all. Chinese medical symptom score and quality of life (QUALEFFO-41) score, bone mineral density (BMD) of lumbar vertebra (L2 -L4) and left femoral neck were compared between the two groups before and after treatment. Serum free testosterone (FT) and estradiol (E2) were also measured.
RESULTSChinese medical symptom scores and QUALEFFO-41 scores, serum FT and E2 levels increased in the two groups after treatment (P < 0.05, P < 0.01). The therapeutic effect was superior in the treatment group (P < 0.05, P < 0.01). After treatment the BMD of lumbar vertebra (L2 -L4) and the left femoral neck were somewhat improved (P < 0.05), but with no statistical difference between the two groups (P > 0.05).
CONCLUSIONBHC could effectively improve the QOL of senile male OP patients, which might be correlated with elevating the BMD levels and regulating the levels of sex hormones.

10.Genotyping of K15 gene of human herpesvirus 8 in Kaposi's sarcoma

Objective To study the distribution of K15 alleles of human herpesvirus 8 (HHV-8) in Kaposi's sarcoma (KS) in Xinjiang,and to investigate the relationship between clinical profiles of KS and alleles of HHV-8 K15.Methods HHV-8 DNA was extracted with phenol-chloroform-isoamyl alcohol from 27 formalin-fixed,paraffin-embedded tissue specimens of KS.The HHV-8 K15 gene was amplified by nested- PCR,and sequenced for the identification of K15 allele.Results HHV-8 DNA could be detected in 22 (81.48%) out of 27 KS patients in Xinjiang.HHV-8 DNA was detected in all 4 patients with AIDS-related KS.Twenty viral strains were identified as P type,including all 4 from the AIDS-related KS patients;two strains were identified as M type,which were all from the classical KS patients.Conclusions In KS,most of HHV-8 K15 alleles are P type,and some are M type.The 4 patients with AIDS-related KS all carried P type of K15 allele.
